[Palatography including lingual and occlusal surface of teeth, palatogram pattern of Chinese sounds].

In order to develop a method that can determine not only the contact pattern of the tongue with the palate but also with the teeth during speech. Static palatograph recording system was used in this study. Palatograms of six normal dentulous adults were made by the recording system during pronounce the following Chinese phonetic alphabets: /S/, /X/, /D/, /J/, /N/, /K/, /R/, /L/. The contact pattern that is the contact area of tongue to palate and tongue to teeth in the palatograms were identified and analysed. The result indicated that the palatograms of the six normal adults are different which can be divided into four types: Type1/S, X/; Type2/D, N, J/; Type 3/K/; Type 4/R, L/.

[Forty six cases of application of free femoris forelateralis flap in trauma].

In serious scar or chronic ulcer caused by burn or trauma, skin graft can not be survival and the local flap can not be found in these cases. Also skin grafts can not improve function of these places. Free flap transplantation should be used in these cases. Femoris forelateralis flap has some advantages. It is hidden from view and there is no effect after it is taken. It has constant blood vessels which are easy anastomosed. So the flap is more widely used. Forty-six cases were given femoris forelateralis flaps (hand burn scar deformity 15 cases, foot scar 10, forearm or leg scar or tissue defect 8, leg or foot chronic ulcer 8, face or neck tissue defect or serious scar 8). Forty-five cases (97.8%), were survival. Only one case had necrosis. The key of survival is choice of the operative indication, excellent anastomosis technique, good fixation postoperatively, care observation, and timely handling. In vasculitis or phlebitis ulcer, the vessel condition is bad. In old people, especial with heart or vessel diseases, condition is also bad. We shoule be careful in these situations.

[Clinical significance of multidrug resistance gene(mdr1) expression in patients with acute leukemia].

OBJECTIVE: To evaluate relationship between drug resistance of leukemia cells and prognosis. METHODS: The expression of multidrug resistance gene(mdr1) mRNA was measured in 85 patients with acute leukemia and in 20 normal controls by using reverse transcription polymerase chain reaction (RT-PCR). RESULTS: The mdr1 positive rate in untreated group was 44.7%. The CR rate differed significantly between mdr1+ (23.9%) and mdr1-(88.5%)(P < 0.005). The mdr1 positive rate of refractory-relapsed group was higher than that in CR group (P < 0.001). The mdr1 mRNA expression level of refractory-relapsed group was higher than that in CR group. A gradually increasing level of mdr1 mRNA expression in CR patients indicated early relapse. The mdr1 positive rate in normal contral and long-term surviver patients was very low. The mdr1 expression was correlated with French-American-Birtish Cooperative Group (FAB) classification. CONCLUSION: The mdr1 expression correlated with chemotherapeutic effect and prognosis. It is an unfavorable prognostic factor for patients with acute leukemia.

[Development of quality of life questionnaire for Chinese cancer patients receiving chemobiotherapy].

OBJECTIVE: To develop the quality of life questionnaire for Chinese cancer patients receiving chemobiotherapy (QLQ-CCC), and to study the methodology involved in the construction of scales measuring quality of life of cancer patients. METHODS: From April, 1993 to April, 1995, QLQ-CCC had been developed mainly according to the Rolls-Royce model: 30 items were iuitially identified and tested in advance, from which the "best" items were selected to from the final format of QLQ-CCC (the fourth-generation questionnaire) including 35 questions. RESULTS: QLQ-CCC had 35 items mainly concerning the five different aspects of cancer patients. Physical aspect included 16 items, of which 9 were disease/treatment-related (specific items), and 7 non-specific items. Psychological/spiritual, social and general health aspects had 5, 5 and 9 items, respectively. CONCLUSION: Quality of life (QOL) research in cancer patients is conducted primarily through the development, implementation and assessment of QOL scales. QOL scales adopted in the developed countries, owing to difference in sociocultural background, have to be modified according to China's concrete condition. This study is an attempt to set up our own QOL scales for cancer patients.

[Effects of n-butanol extract from Cortex Ilicis rotundae on blood pressure and heart rate in rate].

It was studied that effects of n-butanol extract from Cortex Ilicis Rotundae on blood pressure and heart rate in differedt state. Results showed that the drug elicited a sgnificant depressor effect on both normal pressure and artery pressure to rise for common carotid artery blocked in rats. Of the two latter decline of percent of blood presure and active time and slowing heart rate were more significan than the former. It was same depressor effect that injected the drug into duodenum but not act to hypotensive rats. These results suggested that the drug induce significantly depressor effect and slow heart rat with the exception of hypotension.

Nerve growth factor and basic fibroblast growth factor protect rat cerebellar granule cells in culture against ethanol-induced cell death.

Neuronal cell loss is one of the most debilitating effects of fetal ethanol exposure. Cultures of cerebellar granule cells are a useful model to investigate ethanol neurotoxicity, because ethanol depletes cell numbers in these cultures, which also occurs in vivo. The primary goal of the present study was to identify and characterize agents that can ameliorate the ethanol-induced cell death that occurs in this culture system. Growth factors, such as nerve growth factor (NGF), basic fibroblast growth factor (bFGF), epidermal growth factor (EGF), and insulin-like growth factor-I (IGF-I) can prevent neuronal degeneration after toxic insult in various experimental paradigms. These growth factors were investigated in the current study to determine whether or not they can mitigate ethanol-induced death of cerebellar granule cells in culture. Results indicate that NGF and bFGF significantly reduced the ethanol-induced cell loss. Both the NGF- and bFGF-mediated neuroprotection required protein and RNA synthesis, because actinomycin D (RNA synthesis inhibitor) and cycloheximide (protein synthesis inhibitor) blocked their neuroprotective effects. In addition to its neuroprotective effect, bFGF also had a neurotrophic effect and could enhance cell survival in the absence of ethanol exposure. NGF did not have a neurotrophic effect. Neither EGF nor IGF-I was neuroprotective, although the latter did have a substantial neurotrophic effect. In conclusion, bFGF and NGF have long been recognized for their role in enhancing neuronal cell survival and differentiation. This study suggests that these growth factors can also provide neuroprotection against ethanol-induced cell death.

Differential sensitivity of human neuroblastoma cell lines to ethanol: correlations with their proliferative responses to mitogenic growth factors and expression of growth factor receptors.

Early ethanol exposure depletes neurons in the developing nervous system, however the effects on neuronal precursors are not homogeneous. Some cells are more susceptible to ethanol toxicity than others. Growth factors are important mitogens for neuronal precursors. We tested the hypothesis that the differential sensitivity of neuronal precursors to ethanol is determined by their responses to growth factors using an in vitro model (SH-SY5Y, SK-N-SH, and IMR32 neuroblastoma cells) of neuronal precursors. The three cell lines were raised in a medium containing 10% or 0% fetal calf serum. Cells were exposed to ethanol and/or a growth factor. These factors included basic fibroblast growth factor, epidermal growth factor, insulin-like growth factor-I, nerve growth factor, and platelet-derived growth factors AA and BB. The numbers of cells per culture were counted both before and after 3 days of ethanol and/or growth factor treatment. In addition, the effect of ethanol exposure on the expression of receptors for these growth factors was examined. Neuroblastoma cells displayed differential sensitivity to ethanol. The growth of SH-SY5Y and SK-N-SH cells was inhibited by ethanol in a concentration-dependent manner. Ethanol did not affect cell viability. Thus, this inhibition resulted from a reduction of cell proliferation. In contrast, IMR32 cells were not affected by ethanol (even at concentrations as high as 800 mg/dl). The response to growth factors was also heterogeneous. In serum-supplemented medium, SH-SY5Y and SK-N-SH cells were stimulated by all of the tested growth factors. For cells raised in a serum-free medium, only the nerve growth factor was ineffective. IMR32 cells, however, were unaffected by most of these growth factors, regardless of the medium conditions. Ethanol blocked the action of all growth factors tested. In general, all cells expressed the specific receptors for the six growth factors. Only the expression of the basic fibroblast growth factor, insulin-like growth factor-I, and nerve growth factor receptors were reduced by ethanol exposure. In summary, neuroblastoma cells exhibit differential susceptibility to ethanol, and this correlates with their response to mitogenic growth factors. Some growth factors are a target of ethanol toxicity. These heterogeneous effects seem to parallel ethanol-induced changes of proliferating neuronal precursors in vivo.

Detection of c-myc gene expression in nasopharyngeal carcinoma by nonradioactive in situ hybridization and immunohistochemistry.

OBJECTIVE: To investigate the c-myc gene expression in nasopharyngeal carcinoma (NPC) at mRNA and protein levels, and to evaluate the relationship between the degree of expression and the clinicopathological data. METHODS: The expression of c-myc specific mRNA and protein was detected using in situ hybridization with digoxigenin-labeled probe and immunohistochemistry in 12 cases of normal nasopharyngeal epithelium (NE) and 48 cases of NPC. The intensity of staining for each sample was assessed as negative, weak, moderate and intense. Statistical analyses were performed using the Chi square test and the rank sum test. RESULTS: For normal NE 41.7% (5/12) had weak (4/12) or moderate (1/12) staining for c-myc protein and 41.7% (5/12) had weak staining for c-myc mRNA. In contrast, 89.6% (43/48) of NPC showed some degree of c-myc protein positive and 87.5% (42/48) showed some degree of c-myc mRNA positive. In most samples, the expression of c-myc protein was consistent with that of c-myc mRNA. C-myc protein and mRNA expression levels in NPC were significantly higher than those in normal NE, and correlated with early recurrence, but they were not significantly related to patient's age, sex, EBVCA-IgA titre, disease stage, lymph node status and metastasis. CONCLUSIONS: C-myc gene over-expression may be involved in the pathogenesis and early recurrence of NPC. Further studies are needed to substantiate the preliminary findings.

[The peripheral blood levels of TNF alpha and PLA2 activities in dialysis and non-dialysis patients with uremia].

The levels of tumor necrosis factor a (TNF alpha) and phospholipase A2 (PLA2) in peripheral blood were determined by means of the ELISA and improved micromethod in 31 patients with urmia and 12 normal healthy persons. The results showed: 1. In non-dialysis group and during a single dialysis session, a significant increase in the levels of TNF alpha was observed. 2. In non-dialysis and peritoneal dialysis patients, serum levels of PLA2 were significantly increased compared to normal subjects. The results suggest that acute complications induced by hemodialysis are related to increasing levels of TNF alpha, souble PLA2 in cleared by hemodialysis.

[A preliminary study on p53 gene expression and infection of human papilloma virus in laryngeal squamous cell carcinoma].

Using immunohistochemical techniques with p53 monoclonal antibody DO-7 and polymerase chain reaction with type specific primers, we detected the expression of p53 of laryngeal and hypopharyngeal squamous cell carcinoma (L-HSCC) in 42 patients, tissues around tumor in 25 patients, human papilloma virus (HPV) 16/18 DNA in paraffinembedded carcinoma tissues from 13 patients with laryngeal squamous cell carcinoma (LSCC). The results showed that overexpression of p53 was detected in 54.8% (23/42) of L-GSCC and 20% (5/25) of hyperlasia epithelia, respectively. There was no correlation of p53 overexpression with clinical stages and histological grading of tumors (P > 0.05). HPV16 DNA encoding E6 protein was detected in 23.1% (3/13) LSCC tissues by PCR. The results suggest that overexpression of p53 and HPV infection are not only associated with pathogenesis of this kind of cancer but also cooperated during carcinogenesis.

Experiments on the transmission of Babesia major and Babesia bigemina by Haemaphysalis punctata.

Experiments on the transmission by Haemaphysalis punctata of three large Babesia strains were carried out. Three Babesia-free batches of laboratory reared H. punctata ticks were infected with two strains of Babesia major, B. major (Xingjiang strain), isolated with adult ticks of H punctata and B. major (Henan strain), isolated with H. longicornis) and a strain of Babesia bigemina by feeding them on the calves infected by inoculation of blood stabilates. H. punctata was shown to be capable of transmitting the B. major strains transovarially. The larvae, nymphs and adults developed from female ticks engorged on the calf infected with B. major (Xingjiang strain) transmitted the pathogen to splenectomised calves with prepatent periods of 15, 11 and 12 days, respectively. The calves infested with larvae and nymphs died of babesiosis with parasitemias of 400 and 710 per 1000 erythrocytes. The calf infested with adult ticks survived babesiosis, but the number of erythrocytes and the amount of haemoglobin were reduced greatly. H. punctata transmitted B. major (Henan strain) in the same way. The prepatent periods of the calves infested with larvae, nymph and adult ticks were 9, 10 and 12 days, respectively. Calves infested with larvae survived, but those infested with nymphal and adult ticks died of babesiosis with parasitemias of 410 and 100 per 1000 erythrocytes, respectively. H. punctata ticks did not transmit the B. bigemina strain to splenectomised calves. There were no clinical symptoms and no parasites were discovered in the blood films during a 2 month observation period after the calves were infested with larval, nymphal and adult ticks derived from female ticks engorged on calves inoculated with B. bigemina.
