OBJECTIVE: To investigate the effects of danshen(Salvia plectranthoides Griff.) and shengmaiye (Panax ginseng C. A. Mey, Ophiopogon japonicus Ker-Gawl and Schisandra chinensis Baill) on glomerulosclerosis induced by adriamycin in SD rats. METHODS: Left kidney of the animals was removed and after 7 days, adriamycin(6 mg.kg-1) was injected through tail vein, so as to establish an animal disease model. Then danshen or shengmaiye was injected to peritoneal cavity of the rats. All rats were killed by the end of the 8th week. Hemoglobin, BUN, cholesterol and protein in urea within 24 hours(ur Pro/24 h) were detected. IV collagen(IV col.) and laminin(LN) of renal cortex were determined by ELISA. The quantitation of IV col and LN in glomerular mesangial area was analyzed by computer pictures. RESULTS: Compared with model-danshen group(Group III), model-shenmaiye group(Group IV) and normal control group(Group I), BUN, cholesterol(Ch) and ur Pro/24 h were increased obviously and hemoglobin(Hb) was decreased in model control group(Group II). The values of BUN, Ch and Ur Pro were lower in Group III and Group IV than those in Group II and higher than those in Group I (P < 0.05). The quantitation of IV col and LN within renal cortex and mesangial area was less in Group III and Group IV than that in Group II and more than that in Group I (P < 0.05 or P < 0.01). CONCLUSION: Dansheng and shengmaiye may play an important role in the treatment of glomerulosclerosis in rats.
To investigate the affection of different osmotic density, pH value, and dialytic fluid with drugs on the clearance of urea and creatinine through gastrointestinal tract, we used the renal-artery-ligated rabbits as the model. After the intravenous administration of urea and creatinine through ear vein, fistulas in duodenum at about 5 cm to pylorus and in ileum at about 5 cm to the valva ileocecalis were made to carry out intestinal dialysis. The results showed that the clearance of the urea in treated groups with mannitol, regitine, and oxystarch was higher than that in the control group with normal saline solution(P < 0.05). The clearance of urea and creatinine in treated groups with atropine was higher than that in the control group(P < 0.05). There was no significant difference in the clearance of urea and creatinine among treated groups with different intestine pH value.
OBJECTIVE: The correction of ectropion of lower lid and severe infra-orbital soft tissue depression is very difficult. Former methods included simple skin graft, tubed graft, transfer of local skin flap and so on. These methods had some disadvantages, such as not enough tissue to fill the depression, too much damage done to the donor area and operation in stages required. METHODS: After investigation on the anatomy of temporal region, designed the following method. Combined transfer of the galea aponeurotica and temporal fascia was used to repair severe infra-orbital soft tissue depression and ectropion of lower lid in 6 cases. RESULTS: It was discovered that the combined transfer of the galea aponeurotica and temporal fascia was rich in blood circulation because they received blood supply from parietal branch of superficial temporal artery and could be transferred to a distance as far as 15-18 cm. The skin graft used to cover the fascia usually resulted in good survival. This technique was used in 6 cases with good success. CONCLUSION: This method had some merits such as the tissue flap had good blood supply, little damage done to the donor area, good correction of the severe depression, good appearance following correction, operation done under local anesthesia and completed in one-stage operation.
The results of antimicrobial test in vitro of water-extracts and volatile oil of the Herba Pogostemonis shown that the water-extracts posses some inhibitory activity on all the tested bacteria, including Salmonella, Escherichia coli, Shigella flexneri and Staphylococcus aureus, while inhibition to S. aureus is more stronger than to Enterobacteriaceae (P < 0.01). In addition, the activity of anti-enteropathogens of water-extracts of the herb collected from Gaoyao county is more stronger than of the herb collected from Wuchuan county (P < 0.01).
The protective action of 2 tablets of Chinese herb to 5 Chinese traditional medicines against harm of insects and mildews was tested. It was found that 2 tablets have a obvious effects of insect-repellency and mouthproof in the test with Homalomena occulta and Prunus armeniaca, the bore in the medicinal materials was decreased 94.95% and 95.55% respectively than that of check. The tablets have some effects of mildewproof in the test with Tussilago farfara.
In order to investigate the mechanism of Centipede on the function of animal gastrointestinal digestion, we studied the effects of cetipede aqueous extract on animal digestion, secretion and intestional movement. The results showed that the Centipede extract increased the volume of gastric juice and acid, activity of pepsin, volume of pancreatic juice and protein in rat. The Centipede extract also accelerated the mice intestinal movement. The above results revevaled that Centipede could regulate the function of the gastrointestinal digestion, secretion and movement.
The constituents of essential oils from the leaves of Psidium guajava Linn were analyzed by GC-MS qualitatively and quantitatively. Sixty compounds of the essential oils were identified at rate 90.56%. The major components were caryophyllene (18.81%), copaene (11.80%), [1aR-(1a alpha, 4a alpha, 7 alpha, 7a beta, 7b alpha)]-decahydro-1,1,7-trimethyl-4-methylene-1H-cycloprop[e] azulene(10.27%), eucalyptol(7.36%).
The chemical constituents of the volatile oil of the stems and leaves of Pogostemon cablin collected from Gaoyao county, Guangdong province, have been studied by means of GC-MS. Sixty four compounds have been identified. The main constituents are pogostone (30.99% in stems, 21.31% in leaves, the same below), patchouli alcohol (10.26%, 37.53%), transcaryophyllene (4.92%, 6.75%), alpha-guaiene (2.27%, 6.18%) and seychellene (1.56%, 1.99%), etc.

The effects of the three extracts (decoction, oil-free decoction and volatile oil) of Herba Pogostmonis from Gaoyao and Wuchuan on digestive system were compared. The results demonstrated that the decotion and oil-free decoction from Gaoyao and Wuchuan could depressed gastric evacuation in mice. All the three extracts from Gaoyao and Wuchuan could inhibit the ACh-as well as BaCl2-induced spasmodic and automatic contraction of isolated rabbit intestine. The decoction and oil-free decoction from either Gaoyao or Wuchuan inhibited the normal intestinal propulsion. They also increased gastric secretion of acid and activity of pepsin and amylase. These suggest that both of Herba pogostmonis relief gastrointestinal spasm and improve the digestion. The total effect of Herba pogostmonis from Gaoyao was more potent that from Wuchuan.
Based on the perturbation expansion and the observed energy levels for 2p(3)3s and 2p(3)3d of O I -like ions, a recursion formula for the energy levels has been derived. It is found that this method is considerably simple and accurate.
A sideband on the low energy side of the local-vibrational-mode main absorption band of carbon acceptor in GaAs was observed. The difference between the peak frequency of the main absorption band and that of the sideband is 3.5 cm(-1). The integrated absorption of the sideband varies with temperature and has an effect on the temperature dependence of the main absorption band. It is likely that the sideband originates from the anharmonicity of the local vibrational mode itself.
Using an ethnomedical-based drug discovery program, two previously unknown compounds (SP-18904 and SP-18905) from Pycnanthus angolensis were isolated that lower glucose concentrations in mouse models of type 2 diabetes. SP-18904 and SP-18905 are terpenoid-type quinones that significantly lowered plasma glucose concentration (p <.05) when given orally to either ob/ob or db/db mice, both of which are hyperglycemic and hyperinsulinemic. The antihyperglycemic actions of SP-18904 and SP-18905 were associated with significant decreases in plasma insulin concentrations (p <.05), suggesting that both compounds lowered glucose by enhancing insulin-mediated glucose uptake. This was supported by the insulin suppression test in ob/ob mice. Studies in hyperglycemic, insulin-deficient mice and in vitro experiments on 3T3-L1 adipocytes further supported this conclusion. As such, these two terpenoid-type quinones represent a new class of compounds of potential use in the treatment of type 2 diabetes.
To establish a cause-effect relationship between the human mismatch repair pathway deficiency and the observed phenotypes, a hMSH2 deficient HeLa cell line (HeLa-MSH2-) was established by transfecting the HeLa cells with an antisense RNA expression plasmid. The expression plasmid was constructed by inserting an 851 bp fragment of hMSH2 cDNA into the polyclonal site of the vector pREP9 in a reversed orientation. The production of the mismatch binding protein, hMSH2, was inhibited in HeLa-MSH2- cells, as demonstrated by Western blotting and band shift assay of its whole cell extract. The growth rate of this cell line was not different from the parental HeLa cells soon after transfection. However, the rate was faster after 10 subcultures. The spontaneous mutation frequency at the hypoxanthine phosphoribosyltransferase (HPRT) locus increased markedly, but no N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) tolerance appeared in this cell line. Our results clearly demonstrated several molecular events happened after the inhibition of a major mismatch recognition protein, hMSH2, in the mismatch repair pathway, mimicking carcinogenesis processes.
Cryptolepine (1) is a rare example of a natural product whose synthesis was reported prior to its isolation from nature. In the previous paper we reported the discovery of cryptolepine's antihyperglycemic properties. As part of a medicinal chemistry program designed to optimize natural product lead structures originating from our ethnobotanical and ethnomedical field research, a series of substituted and heterosubstituted cryptolepine analogues was synthesized. Antihyperglycemic activity was measured in vitro and in an NIDDM mouse model to generate the first structure-bioactivity study about the cryptolepine nucleus.
The interaction of ethanol and neurotrophin-mediated cell survival was examined in primary cultures of cortical neurons. Cells were obtained from rat fetuses on gestational day 16 and maintained in a medium supplemented with either 10% or 1.0% fetal calf serum (FCS). Exogenous nerve growth factor (NGF; 20 ng/ml), brain-derived neurotrophic factor (BDNF; 20 ng/ml) or neurotrophin 3 (NT-3; 20 ng/ml) was added to the cultures alone, or in combination with ethanol (400 mg/dl). The number of viable neurons was determined after a 48 h treatment with a growth factor and/or ethanol. The effects of ethanol on the expression of high affinity neurotrophin receptors (trkA, trkB, and trkC) and the low-affinity receptor (p75), were analyzed using Western immunoblots. In untreated cultures, 22.7% and 26.3% of the cells raised in a medium containing 10% and 1.0% FCS, respectively, were lost. Only NGF prevented the death of the cultured cortical neurons. Ethanol was toxic; it caused a 23.5% and 16.7% loss of cells (for cells grown in a medium containing 10% and 1.0% FCS, respectively) beyond that occurring 'naturally' in an untreated culture. Ethanol completely blocked the NGF-mediated cell survival. In general, BDNF and NT-3 did not offset the toxic effect of ethanol. Immunoblotting studies showed that the expression of p75 was significantly (p < 0.05) lower (40%) in ethanol-treated cultures, but ethanol did not affect trk expression. Thus, ethanol has specific effects upon NGF-mediated cell survival and the effects on the low affinity receptor imply that p75 specifically plays an important role in NGF signaling.
Among glycosaminoglycans, polysulfated heparin chains provide the greatest challenge to characterization due to high polarity, structural diversity, and sulfate lability. The present report demonstrates how electrospray mass spectrometry (ESMS) can be used to derive compositional information from pure and mixed fractions of heparin tetra- to decasaccharide fragments prepared by controlled digestion of heparin with heparinase I. It also describes an improved procedure for fractionation of heparin oligosaccharides up to octadecasaccharides. Ammonium salts prove to be superior to sodium salts, particularly for analysis of mixed components. In the mass spectrum of a hexasaccharide fraction, the identification of six major mass peaks that represent the known hexasaccharide structures confirms that ESMS analysis of heparin oligosaccharide fragments gives a close representation of their constituent composition. In addition to the previously identified components, several unreported oligosaccharides were detected in the spectra of octa- and decasaccharide fractions. The ESMS identification of the three major species in a decasaccharide fraction was confirmed after HPLC subfractionation and reanalysis. ESMS detection sensitivity of low picomole amounts of oligosaccharides can be readily achieved.
