Parameters that influence the extent of site occupancy by a candidate telomere end-binding protein.

The MF3 protein specifically recognizes telomeric and non-telomeric DNA probes that can form G.G base-paired structures (Gualberto, A., Patrick, R. M., and Walsh, K. (1992) Genes & Dev. 6, 815-824). Here we further characterize the nucleic acid recognition properties of MF3 and present a mathematical analysis that evaluates the potential extent of telomere site occupancy by this factor. The substitution of dI at dG positions in telomeric DNA probes revealed that a single dG at any position within the internal repeat was sufficient for high affinity binding to MF3. The RNA analogs of high affinity DNA sites were not bound specifically by MF3, but the substitution of dU for dT in a DNA probe had little or no effect on binding. These data demonstrate that ribose ring structure is a critical feature of nucleoprotein complex formation, and this ribose specificity may enable MF3 to occupy sites of unusual DNA structure while minimizing interactions with cellular RNAs. Collectively, the nucleic acid binding properties of MF3 suggest that it may occupy a significant fraction of sites at telomere ends or other G-rich regions of altered DNA structure in vivo.

Aneurysmal bone cyst in a Holstein bull.

A 3-year-old Holstein bull was examined because of a mass involving the cranial portion of the right hemimandible and the oral cavity. The mass had been observed 2 weeks earlier. The bull had lost weight throughout the 45 days before admission, and was anorectic at the time of admission. An aneurysmal bone cyst of the mandible was diagnosed.

Giant cavernous hemangioma of the liver complicated by abscess and thrombosis.

A case of giant cavernous hemangioma of the liver complicated by abscess formation and thrombosis is presented. To our knowledge, complication of hepatic hemangioma by an abscess has not been previously reported. The clinical and radiographic features, as well as the management of giant hepatic cavernous hemangiomas is discussed.

The DNA sequence of sea urchin (S. purpuratus) H2A, H2B and H3 histone coding and spacer regions.

The DNA sequence of two cloned segments of the histone gene repeat unit of the sea urchin S. purpuratus has been determined. One sequence contains the contiguous H2B and H3 genes and their interdigitated spacer regions; the other comprises the H2A gene and flanking spacer sequences. Analysis of the coding regions reveals a methionine residue within the H2A protein. H2A, which generally lacks this amino acid, contains methionine only in a protein variant which is synthesized in early sea urchin embryogenesis. We thus conclude that the cloned DNA represents a set of genes which is active early in development. Codon selection is markedly skewed and similar for each of the three genes. The DNA sequences are co-linear with known histone protein sequences and-unlike several other eucaryotic genes-do not show any insertions in the coding regions. The spacer regions are relatively AT-rich although GC cluster are scattered throughout. Several short stretches of homology are found in regions both upstream and downstream from the protein coding segments. The conservation of these sequences and their location at analogous sites suggest that they are involved in gene transcription or in mRNA translation. No tandem or dispersed repeats were found, with the exception of the remarkable sequence having the structure located in the spacer between the H2A and H1 genes.

Increased fertility in rhesus monkeys by breeding after the preovulatory LH surge.

Fertility in a rhesus monkey breeding colony was significantly increased by caging females with males immediately after detection of the preovulatory luteinizing hormone (LH) surge. LH was measured in daily serum samples by a rapid (24-hr) radioimmunoassay which used iodinated ovine LK as tracer and an antiserum to human LH.

Design and evaluation of an instrument to measure microcirculatory blood flow and oxygen saturation simultaneously.

There are compelling clinical advantages in being able to monitor, simultaneously and continuously, blood oxygen saturation and tissue perfusion from the same volume of tissue. We describe the design of a new instrument capable of combining both functions. It uses a laser diode at 805 nm to give a blood flow index and oxygenation information at that wavelength, and a pulsed LED at 660 nm to complete the estimation of oxygen saturation. By using a compensating photodetector these readings can be made relatively insensitive to sampling site. After calibrating the instrument on forehead sites it reliably read the oxygen saturation of sites on the thigh with a bias of -0.2% and a precision of +/- 1.7% relative to an IL 282 CO-oximeter.
