The tumor suppressor p53 and primary response gene Egr-1 are nuclear transcription factors with regulatory roles in signal transduction pathways mediating cellular proliferation and growth arrest as well as the complex genetic programs controlling differentiation and programmed cell death. We identified a physical association between these regulatory proteins in vitro and in vivo. Recombinant p53 and Egr-1 fusion proteins complexed with in vitro translates of Egr-1 or p53, respectively, or with these respective proteins in cell lysates. This protein-protein interaction was detected in vivo by immunoprecipitation and Western blot analysis of serum-activated cellular lysates with high levels of induced Egr-1 and of human lung cancer cell lines with constitutive overexpression of Egr-1 and mutant p53. A p53 mutant at codon 154 did not bind Egr-1, while p53 proteins with point mutations at residues 156, 246, 247, and 273 associated with this zinc finger transcription factor. p53 bound full-length Egr-1 and an Egr-1 mutant with a deletion of the 5' transactivation region but did not associate with Egr-1 protein lacking an internal segment that included the first two zinc finger domains, suggesting that binding may require the presence of intact zinc finger motifs. A variant-sized Egr-1 protein expressed by lung fibroblast cell line MRC-9 was also bound by p53. The interaction of these regulatory proteins may alter multiple features of their biological activity especially with regard to the specificity of transcriptional control.
PURPOSE: To learn whether nicotinic cholinergic receptors modulate postnatal eye growth and influence the course of form-deprivation myopia. METHODS: One-week-old White Leghorn chicks wore a unilateral goggle to induce form-deprivation myopia. Other chicks were never goggled. Nicotinic antagonist drugs were administered by intravitreal injection, usually daily or every other day to the goggled eye or to one eye of never-goggled chicks. After 1 week, the eyes were studied by refractometry, A-scan ultrasonography, and caliper measurements. RESULTS: The relatively non-subtype-specific channel-blocking nicotinic antagonists chlorisondamine and mecamylamine each inhibited the development of form-deprivation myopia but with complex multiphasic dose responses. Chlorisondamine was the most effective. Mecamylamine, at the lowest tested doses, tended to stimulate the growth response and myopic refractive shift of goggle wearing. Methyllycaconitine competitively inhibits nicotinic receptors containing the alpha7 and alpha8 subunits, which are highly expressed in chick retina. It showed a less dramatic but still significant inhibitory effect on myopia. The effects of dihydro-beta-erythroidine, a competitive antagonist relatively selective for nicotinic receptors with alpha3 or alpha4 subunits and particularly for alpha3beta2-containing receptors, were the weakest and inhibited primarily axial elongation. Chlorisondamine but not mecamylamine also affected nongoggled eyes, inhibiting growth and shifting refraction toward hyperopia, but chlorisondamine also induced degenerative changes to the retinal pigment epithelium (RPE). CONCLUSIONS: Nicotinic receptors are involved in eye growth control. Nicotinic antagonists affect the development of form-deprivation myopia and perhaps the growth of nongoggled eyes. The differences in drug activity and multiphasic dose-response curves may reflect the complexity of nicotinic receptor subtypes associated with the eye and/or pharmacokinetic differences between the individual drugs. Although another tissue(s) cannot be completely excluded by these data, the site of action of these agents may be neural retina or RPE.
OBJECTIVE: To study the effect of iron metabolism on patients with hepatitis B. METHODS: Hemoglobin (Hb), serum ferritin (SF), transferritin (TRF), serum iron (SI), and total iron binding capacity (TIBC) were detected in 103 patients with hepatitis B and 20 healthy adults. RESULTS: The severer the hepatocyte damage was, the higher the SF, SI and the lower the Hb, TRF, and TIBC were. Furthermore, it seems more obvious among fulminant hepatitis and liver cirrhosis. CONCLUSIONS: The overload of iron may enhance the hepatocyte damage induced by HBV. Therefore, to detect serum markers of iron metabolism is helpful to evaluate curative effect and prognosis of hepatitis B.
Angiosarcoma is the most common malignant neoplasm of the heart. However, to the authors' knowledge, no cytogenetic study of cardiac angiosarcoma has been reported. In the current study, an angiosarcoma from the right atrium of a 29-year-old man was investigated. Examination of tissue sections indicated that the tumor was a high grade epithelioid angiosarcoma of the heart. Cytogenetic analysis of tumor cells revealed a hyperdiploid clonal population with chromosomal numerical changes and one structural rearrangement, which was defined as: 55, XY, +der(1;17) (q10:q10), +2, +7, +8, +8, +19, +20, +21, +22. Multicolor fluorescent in situ hybridization on paraffin-embedded tissue sections illustrated polysomy of chromosome 8 in tumor cells. In addition, immunohistochemical analysis showed high expression of mutated p53 gene products in tumor cell nuclei. These findings demonstrate the involvement of chromosomal anomalies and gene mutation in cardiac angiosarcoma and suggest they play a role in neoplasia of the heart.
A cloned cDNA encoding a PLA2 from Agkistrodon halys Pallas was found to have conservative residues Glu53 and Trp70 but with Lys56 and Lys67 substituted by Thr56 and Asp67, respectively, when compared with sequences of other class II PLA2 with anticoagulant activity. It was inserted into a temperature-sensitive bacterial expression vector and effectively expressed in Escherichia coli RR1. The protein was produced as insoluble inclusion bodies and recovered by centrifugation after enzyme digestion. By washing to partial purification, the expression product was refolded and was purified by FPLC superose 12 to appear as a single band in SDS-PAGE. The recombinant protein proved to have obvious enzymatic, anticoagulant and hemolytic activities, which were removed after modification by p-BPB. These findings suggest that the pharmacological activities of this recombinant PLA2 may be related to its catalytic activity and warrant further research on the structure-function relationships of the pharmacological site of the PLA2 from Agkistrodon halys Pallas.
The blood lead (PbB) levels of pregnant women and their babies were followed up since the third month of pregnancy until their babies were 2 years old. The general condition of delivery, family history of the pregnant women, feeding and development of infants were investigated. Prenatal and postnatal PbB levels and their effects on infant growth were analysed. The result showed a significant negative correlation of PbB level of pregnant women near delivery, and in umbilical cord with the growth of infants. Multiple regression analysis showed a negative correlation between PbB level and height, weight of infant at 1 and 2 years old.

OBJECTIVES: The diagnostic accuracy and usefulness of an ultraportable hand-carried echocardiography system were investigated for assessing ventricular systolic function and severity of mitral valvular regurgitation. METHODS: The study population consisted of 77 consecutive patients (47 men, 30 women, mean age 63 +/- 15 years). Left ventricular end-diastolic dimension, left ventricular end-systolic dimension and left ventricular ejection fraction were measured using the hand-carried echo system and the data were compared with measurements by the conventional echocardiography system using simple linear regression analysis. Left ventricular wall motion was compared between the systems using a 16-segment model recommended by the American Society of Echocardiography. Severity of mitral regurgitation was assessed by the distance of the regurgitant signal in the left atrium. RESULTS: Left ventricular end-diastolic dimension, left ventricular end-systolic dimension and left ventricular ejection fraction showed good correlations between hand-carried and conventional echo systems (r = 0.94, 0.91 and 0.81, respectively; each p < 0.0001). The accuracy for assessing left ventricular wall motion was 94% (449 of 480 segments). The echo systems also showed the same degree of diagnostic accuracy for severity of mitral regurgitation. CONCLUSIONS: The hand-carried echo system provides accurate assessment of left ventricular function and mitral regurgitation simular to conventional echo machines.
OBJECTIVE: To understand the main causes of deaths affecting population health and the trend of mortality of cardiovascular disease (CVD) in Beijing area during 1984 to 1998. METHODS: Monitoring data for cardiovascular diseases in some areas of Beijing during 1984 to 1998 were collected from MONICA project and used to analyze the distribution of disease, mortality caused by cardiovascular diseases and its age-adjusted mortality. Yearly average change in cardiovascular disease-specific mortality was calculated with regression analysis. RESULTS: Cardiovascular disease-specific mortality ranked the first place of the causes of deaths in Beijing population during 1984 to 1998, accounting for 46.0% of the total deaths. Cancer ranked the second, accounting for 26.5% of the total. Overall mortality, cardiovascular-specific mortality and cerebro-specific mortality all lowered significantly during the past 15 years, with yearly average reductions of 2.9%, 2.7% and 3.5%, respectively. Mortality of coronary heart disease significantly rose. CONCLUSIONS: Cardiovascular disease was the top cause of death in Beijing population during 1984 to 1998. Mortality of coronary heart disease significantly rose in the past 15 years. It is a pressing matter of the moment to prevent cardiovascular diseases in the population.
The objective of this study was to determine the effects of mechanical vibration loading on DNA and proteoglycan syntheses in cultured rabbit articular chondrocytes. Chondrocyte culture plates were placed in a vibratory apparatus and subjected to a mechanical vibratory load at various frequencies and periods in culture. Mechanical vibration was applied at a sinusoidal waveform of 1.4 g acceleration with frequencies of 200, 300, 400, 800, and 1600 Hz. 3H-Thymidine and H2(35)SO4 incorporation were used to detect radiolabeled DNA and proteoglycan syntheses, respectively. A frequency of 300 Hz showed a time-dependent augmentation of DNA synthesis and gave a maximal increase at day 3 with periodic vibration (8 h per day) and at 72 h or longer with continuous vibration. It also promoted proteoglycan synthesis in long-term culture (from 3 to 15 days) by periodic vibration. However. frequencies above 400 Hz suppressed biosynthesis. These results suggest that mechanical vibration at certain frequencies may modulate biosynthetic response of articular chondrocytes.
In this study, surface modification by electroplating Rh on 316L stainless steel was carried out in order to improve the corrosion resistance and blood compatibility of implants. XPS was used to characterize the Rh film. The corrosion behavior of 316L stainless steel with electroplated Rh and non-treated was studied in Tyrode's biological solution. Additionally, the haemocompatibility of 316L stainless steel with electroplated Rh was evaluated by preliminary hemolysis test. The results showed that the corrosion resistance of 316L stainless steel with electroplated Rh was improved, and hemolysis decreased, which indicated that electroplating Rh can improve the blood compatibility of 316L stainless steel.
Polylactide and its copolymers are polymer materials that can be degraded by organisms and have good biocompatibility. Since the polymer materials have no toxicity and can be absorbed, greater and greater attention has been paid to the related researches and development, and they are increasingly applied in various areas, especially medicine and pharmacy. There are three methods for preparing polylactide and its copolymers. The tests for evaluating the degraded property of polylactide and its copolymers have been existing for more than thirty years, but all of them have their defects. The property and technological level of polylactide and its copolymers remain to be improved for use in some areas. Once the manufacture of poly lactide is industrialized, it can be of wide application in the fields of medicine and degraded plastics.
Based on the electrophysiologic mechanism of cardiac electrical action and the theory of voltage effect, the limit amplitude technique was employed to obtain the best clinical record effects. The results showed that limiter could limit the oversize signals overlapping each other and make the tiny signals magnification without distortion, such as the His bundle deflection in His Bundel Electrogram. At the same time, all signals recorded at multichannel physiologic recorder were clear and did not interfere with each other. Furthermore, the limiter not only improved the recording quality but also protected the cardiac electronic equipment because overdue stimulation signal was limited at a given electronic level during the recording of transesophageal pacing.
The objective of the present study was to investigate the regulation of P-glycoprotein by cyclosporine, a known inhibitor of CYP3A, at different dosage levels and lengths of treatment. Rats were given various doses of cyclosporine through oral administration or subcutaneous injection. Each treatment group was studied for 28 days or 28 days followed by 14 days of olive oil vehicle dosing. In each group, rats administered vehicle alone served as the controls. At the end of the study, liver microsomes were prepared and hepatic P-glycoprotein levels were quantified by Western blot analysis. Significant induction of hepatic P-glycoprotein was found in rats given cyclosporine. Rats administered 30 mg/Kg/d orally and 15 mg/Kg/d subcutaneously showed an increase in hepatic P-glycoprotein by 93% (p = 0.0011) and 136% (p < 0.001), respectively. Low doses of cyclosporine also induced P-glycoprotein but not to a significant extent, indicating a dose-dependent effect. The pattern of induction of P-glycoprotein was not, however, dependent on the route of administration. Fourteen days after the discontinuation of cyclosporine treatment, P-glycoprotein levels returned to near the control values. As a drug efflux transporter, the induction of P-glycoprotein by cyclosporine may decrease the hepatic metabolism of P-glycoprotein substrates. Therefore this induction of hepatic P-glycoprotein and suppression of hepatic CYP3A may have a coordinate effect on the metabolism of cyclosporine. These data may help explain the large variability associated with cyclosporine absorption, metabolism, and circulating blood levels.
OBJECTIVE: To establish a rapid and simple method with high efficiency in detecting point mutation of genomic DNA. METHODS: Four DNA fragments that were different from each other in only one based were amplified by using primers with artificial point mutation based on the sequence of exon 7 of p53 gene, and then were separated by capillary electrophoresis(CE). The neutral coated capillary and 4% linear polyacrylamide gel buffer were used, and the wave length of ultraviolet detector was 254 nm. RESULTS: A homozygous 196 bp DNA fragment can be separated into one dsDNA peak and two ssDNA peaks within 25 minutes, and the heterozygous 196 bp DNA fragments that were made with mixed wild type and mutated DNA can be separated into one dsDNA peak and three ssDNA peaks. The three ssDNA fragments that differ in one nucleotide can be easily separated with good resolution. CONCLUSION: CE technique is rapid, sensitive, accurate and well reproducible. It is an efficient and reliable method for rapidly screening point mutation.
Giant cell reparative granuloma (GCRG) is an uncommon non-neoplastic lesion that typically occurs in the mandible and maxilla: however, its involvement with the temporal bone is rare. It is usually misdiagnosed as a giant cell tumor. Although regarded as a benign process, GCRG may be locally aggressive. In this paper, we describe two cases of GCRG of the temporal bone and review the pertinent literature published in English. The clinical course, histological evaluation, diagnosis, differential diagnosis, treatment and prognosis of GCRG of the temporal bone were investigated.
A link between insulin metabolism and colorectal cancer has been hypothesized, supported by a series of potential physiological mechanisms, and confirmed by a number of reports in experimental animals. However, the evidence in humans is limited and mostly indirect. The aim of the present report is to analyze whether individuals with a cluster of metabolic abnormalities associated with abnormalities in insulin metabolism experience higher mortality for colorectal cancer than those without this cluster of metabolic abnormalities. A total of 21,311 men and 15,991 women 20-69 years of age were followed-up for an average of 7 years as part of the Risk Factors and Life Expectancy Project, a pooling of a number of epidemiological studies conducted in Italy. Our analyses indicate that participants with high levels of blood glucose and a cluster of metabolic abnormalities linked to insulin resistance experienced a significant increased risk of colorectal cancer mortality compared with participants without the cluster. For the presence of the cluster of metabolic abnormalities, the calculated hazard ratios and 95% CIs were 2.96 (1.05-8.31) for men, 2.71 (0.59-12.50) for women, and 2.99 (1.27-7.01) when both sexes were combined. These associations were independent from the potential confounding effect of age, drinking of alcoholic beverages, and smoking. Our findings are supportive of the hypotheses that glucose metabolism hyperinsulinemia, insulin resistance, and metabolic abnormalities associated with it may play a significant role in the etiology of colorectal cancer.
