[Effect of osmotic stress on wheat membrane injury under different CO2 concentrations].

This paper studied the variation of active oxygen content and membrane permeability in leaves of spring wheat seedling exposed to osmotic stress under normal 350 microliters.L-1 and doubled CO2 concentration 700 microliters.L-1. The results indicate that the increasing rate of O2.- and H2O2, and the membrane permeability in wheat leaves exposed to osmotic stress in normal CO2 concentration were higher than those in doubled CO2. Accordingly, it is suggested that doubled CO2 can reduce the membrane oxidative injury caused by osmotic stress, and increase plant drought resistance.

[Changes after dynamic observation of ultrastructural cerebral infarction in renovascular hypertensive rats].

OBJECTIVE: To study the ultrastructural damages of brain tissues dynamically after middle cerebral artery obstruction (MCAO). METHODS: A middle cerebral artery obstruction (MCAO) model was reproduced in renal vascular hypertensive rats (RHR). Brain tissues in different locations were collected for eight times in row, from two hours to seven days after establishment of the animal model. RESULTS: The changes occurred in the whole brain after focal cerebral ischemia. The severity of impairment and time of emergence differed with the location sampled. The earliest in the infarcted region with necrosis was considered the major change and completely irreversible damage. The next was seen in marginal regions, mainly with microvascular collapse, formation of microthrombi, and partial necrosis of brain cells and was considered partially reversible damage. The last was in distant and contralateral hemisphere, mainly with reversible swelling of endothelial cells and astrocytes. Changes in the MCAO models reproduced by RHR models were much similar to the clinical pathological changes in cerebral infarction based on the hypertensive cerebrovascular impairment, and it was regarded as a relatively ideal model for studying focal ischemic cerebral impairment. CONCLUSION: Dynamic changes of cerebral ultrastructure in which microvascular impairment plays an important role suggest that clinical therapy cannot only be focused on infarction foci, but also on the whole brain with comprehensively effective measurements.

A preliminary investigation of Tanakan in the treatment of hypertensive arteriosclerosis and stroke in rats.

OBJECTIVE: To observe the changes in hypertensive cerebral arteriosclerosis and the incidence of stroke in rats following treatment with Tanakan, a preparation of Ginkgo Biloba (GB). METHODS: Using two-kidney, two clip renovascular hypertensive rats (RHRs) as animal models--we investigated changes in these hypertensive arteriosclerosis-associated factors: A I, A II, ALD, ET, SOD and cGMP. Blood levels were observed after oral administration of GB, at both early and late stages of hypertension. RESULTS: The degree of arteriosclerosis was more severe in both RHRs control and the later treated group than in the early treated group, with blood levels of A I, A II, ALD and ET in the former two groups significantly elevated. The degree of arteriosclerosis in the early treated group decreased and the blood levels of A I, A II, ALD and ET in this group decreased nearly to normal levels. The blood levels of SOD and cGMP were also elevated in the early treated group. CONCLUSIONS: These results reveal that it is important to treat hypertension as soon as possible, in order to reduce the occurrence of stroke.

Gonadotropin-releasing hormone agonists and laparoscopy in the treatment of adenomyosis with infertility.

OBJECTIVE: To study the role and value of gonadotropin-releasing hormone agonists (GnRH-a) and laparoscopy for the treatment of adenomyosis with infertility. METHODS: Four cases were seen with adenomyosis and infertility, 3 of these cases also presented local adenomyomata in the posterior uterine wall. GnRH-a Triptorelin (decapeptyl) or Goserelin (Zoladex) therapy was instituted for six months before laparoscopic surgery for coexisting pelvic pathologic infertility factors in one case and after laparoscopic surgery in three cases. RESULTS: All cases remained amenorrheic during GnRH-a therapy. After the GnRH-a therapy, all enlarged uterus (7-10 weeks gestation size) all decreased to normal or near normal size; menstruation returned in 80-90 days and three cases conceived within four menstrual periods after cessation of treatment. In the 4 cases one pregnancy resulted in the birth of a healthy 3150 g male newborn at 38 weeks gestation by cesarean section; one pregnancy was terminated after adenomyomectomy by emergency cesarean section at 30 weeks gestation because of threatened rupture of uterus; one case was then normal at 28 weeks pregnancy; the last case had 2 resumptive menstrual periods and was still being followed up. CONCLUSIONS: GnRH-a is markedly efficient in reducing adenomyotic uterine size, facilitates uterine or endometrial receptivity for embryos and enhances uterine ability to maintain pregnancy. For adenomyomata associated with infertility, GnRH-a instead of surgical removal of lesions, which are deep in the myometrium, may avoid uterine rupture when pregnancy occurs. For infertility, GnRH-a treatment before laparoscopic surgery greatly decreases surgical difficulties and blood loss in certain cases.

[Retrospective analysis of 266 cases of pregnancy complicated by heart disease].

OBJECTIVE: To investigate the influence of cardiac function of pregnant women on outcome of their pregnancies. METHODS: Clinical data of 266 cases of pregnant women with heart diseases from January, 1993 to March, 1999 were analyzed retrospectively. RESULTS: (1) The highest incidence of cardiac disease with grade I to II of heart function was due to myocarditis, sequelae of myocarditis and abnormal cardiac rhythm (179 cases, 67.29%). The incidence of congenital heart disease is on the second (41 cases, 15.41%) which is higher than that of rheumatic heart disease (28 cases, 10.91%). (2) Grade III and IV cardiac function was mainly related to congenital and rheumatic heart diseases during pregnancy. The rate of grade I/II heart function in pregnant women after heart operations was 95.45%. (3) The incidence of premature delivery, low weight of newborns and perinatal mortality of mother were higher in women with grade III and IV heart function than in those with grade I and II heart function. CONCLUSIONS: The incidence of myocarditis and sequelae of myocarditis is increasing. Pregnancies complicated with congenital and rheumatic heart diseases are still major causes of perinatal morbidity and mortality of mother and baby. Heart operation can obviously improve the heart function. More effective managements should be adopted.

[The study of T helper cell subsets and relationship to relative cytokines and ventilatory function in asthmatic patients].

OBJECTIVE: To investigate the changes of T helper cell(TH) subsets and relationship to the relative cytokines, serum total IgE, eosinophil count and ventilatory function in asthmatic patients. METHODS: 20 asthmatic patients and 10 healthy subjects were selected. The counts of the TH subsets, the level of IL-4 and IFN-gamma in the supernatant of peripheral blood mononuclear cell(PBMC), serum total IgE concentration and eosinophil count in peripheral blood were determined in acute attack and stable patients with asthma and healthy subjects. RESULTS: The TH2/TH1 ratio in the acute attack and the stable patients with asthma were increased significantly, compared with that in normal group (P < 0.01). The relation analysis showed that the value of TH1 had significantly positive relation with that of IFN-gamma, the value of TH2 had significantly positive relation with that of IL-4. The total serum IgE had significantly positive relation with the level of IL-4. FEV1 had significantly positive relation with that of TH1, negative relation with that of TH2, and TH2/TH1 significantly. The total serum IgE and the number of eosinophils had significantly positive relation with that of TH2. CONCLUSION: The balance of TH in asthmatic patients deviates significantly. The deviation of balance of TH has relation with severity of asthma.

[Analysis and comparison of dental arch symmetry between different Angle's malocclusion categories and normal occlusion].

OBJECTIVE: To analyze and compare dental arch symmetry between different Angle's malocclusion categories and normal occlusion. METHODS: 300 subjects were divided into four different malocclusion groups and normal occlusion group. Data points representing the dental arches on the casts were precisely marked by YM-2115 three Dimension Measuring Machine. Arch forms were fitted with conic sections, and then dental arch symmetry was analyzed by rotation angle. RESULTS: There were no absolute symmetric dental arch, and the direction of asymmetry was unsteady. In normal occlusion, dental arch was generally symmetric, but there was asymmetry in a very small degree. As to malocclusion, the frequency and degree of dental arch asymmetry were higher than that of normal occlusion, and the lower arch showed more asymmetry than the upper arch except ClassIII. The frequency of asymmetry in ClassII2 and ClassIII groups was higher than that in other malocclusion groups. CONCLUSION: The frequency and degree of dental arch asymmetry were different in different malocclusion groups.

[Clinical observation on the mental symptoms of functional dyspepsia and its treatment].

OBJECTIVE: To explore the relationship between the mental symptoms and the functional dyspepsia (FD), and the effect of Xiaoyu decoction (XYD) plus psychotherapy on FD. METHODS: SCL-90 scale and FD symptom scale were used to estimate the condition of 56 healthy subjects and 56 patients of FD before and after 4 weeks treatment with XYD plus psychotherapy. RESULTS: There was significant difference in SCL-90 scales between the healthy subjects and the FD patients before treatment (P < 0.01). After treatment, the mental symptoms and the symptom of FD in the patients were markedly improved, as compared with those before treatment, the difference was significant (P < 0.01). CONCLUSION: Mental symptoms, such as depression and anxiety exists widely in patients of FD, are closely related to FD. XYD plus psychotherapy could treat it effectively.

[Separation of thoracopagus conjoined twins].

OBJECTIVE: To sam up the experience in diagnosis and separation of a thoracopagus conjoined twins. METHODS: Thoracopagus twin boys were admitted for evaluating possible separation and repair at age of 21 days. X-rays, CT, MRI, and ultrasound scan of torso showed separate and normal gastrointestinal and biliary tracts, little shared vasculature, but shared pericardium, diaphragm and liver parenchyma. The chest junctions were at the sternums and the most inferior two pairs of the ribs. One twin (twin A) had a protruding heart with VSD and ASD. RESULTS: The infants were separated successfully at age of 45 days. The chest defect of twin A was closed with aid of a polyester pericardial patch graft and a silicon supporter. Managing anesthesia met some difficulty in understanding of the anatomical and physiological consequences of the complex anomaly. They are had brain edema and multiple system organ failures after separation, and were cured under intensive care. They are alive and well 6 months after surgery. CONCLUSIONS: Careful preoperative evaluation proper anesthesia and surgical technique, and postoperative care are essential to the successful separation of the conjoined twins.

[Investigation on E. coli O157 in Fujian, China].

OBJECTIVES: To understand the distribution of Escherichia coli O157 (E. coli O157) in human beings, livestock and fowls in Fujian Province, and biological features of the isolated strains. METHODS: Fecal specimens from diarrheal patients, livestock and fowls and meat specimens in Putian and Fuzhou were collected for detection of E. coli O157 during 1997 - 1998. RESULTS: A total of 2,725 specimens were detected, and 76 strains of E. coli O157 were isolated from pigs, pigeons, cattle, chickens, ducks, et al., as well as from the diarrheal patients. These strains could be classified into three types according to their serological tests, including 33 strains of O157: H7, 21 strains of O157: NM, and 22 strains of O157: H?. Detecting rate of E. coli O157 from pigs was the highest. The isolated strains could cause diarrhea in human and obvious pathological changes in the intestines of mice, but could not produce cytotoxin Vero, or heat-stable or heat-liable enterotoxin LT or ST. CONCLUSIONS: There were three types of E. coli O157 in Fujian. But, pathogenic factors of O157: H7 and O157: NM were different from those reported abroad. It suggested that there possibly existed some new pathogenic factors.

[Supercritical extraction of Alpinia officinarum and GC-MS analysis of its components].

In order to find the effective components and new pharmaceutical effect of Alpinia officinarum, and further exploit it, we extracted its essential oils with the technique of Supercritical-CO2 fluid, then confirmed there were 27 components in Pot I And 111 components in Pot II by using the chemical component analysis and the computer information retrieval in the GC-MS system.

[GC-MS analysis of essential oil components of Alpinia oxphylla].

Essential oil Components of Alpinia oxphylla were analyzed by way of applying gas chromatography, mass spetrography and computer interpretation; essential oil was abtained by supercritical CO2 fluid, thus we got 193 peaks in oil from Pot I and 210 peaks in oil from Pot II as well as identify 139 and 155 compounds respectively.

[The histological changes after preserved human amniotic membrane transplantation for conjunctival reconstruction of rabbits eyes].

PURPOSE: To investigate the histological changes after preserved human amnotic membrane transplantation of rabbit eyes. METHODS: The conjunctiva of 12 New Zealand albino rabbits' right eyes were excised partly. Then the preserved human amniotic membrane were transplanted on the sclera. Eyes were enucleated at 1 week, 3 weeks, 5 weeks and 12 weeks (n = 3 for each) after surgery, and they were examined histologically with staining of HE, PAS, and transmission electronic microscopy. RESULTS: At 1 week after transplantation, the human amniotic membrane firmly attached on the sclera, with the dead epithelial cells remaining on its surface. At 3 weeks, conjunctival epithelization was completed on the human amniotic membrane. Abundant PAS-positive goblet cells were recognized. The amnion began resolve. At 12 weeks, the amnion dissolved. On transmission electronic microscopy, the cell polarity is prominent. The epithelial cells contained a large number of mitochondria and endoplasmic reticulum. Microvilli were pronounced on the apical cell membrane. There are many intracellular desmosomes and hemidesmosomes. CONCLUSION: Preserved human amniotic membrane is a valuable conjunctival replacement material. After transplantation, the histology of new tissue is as same as normal conjunctive.

[Application of cellulose acetate membrane with the technique of impression cytology combined with immunohistochemical staining for detection of dry eye].

PURPOSE: To evaluate the conjunctival cells by impression cytology with cellulose acetate membrane. METHODS: The expression of the TGF-beta 1 in conjunctival epithelium of 24 patients with dry eye were studied by impression cytology combined with immunohistochemical staining and observed by microscope. RESULT: The cellulose acetate membrane is very transparent under the microscope. Cell's membrane by TGF-beta 1 staining is clear, cytoplasm brown and nuclei blue. CONCLUSION: The method of impression cytology with cellulose acetate membrane is simple and has no injury to the ocular surface. Combined with immunohistochemistry staining, it is suitable to observe the conjunctival cells by this technique which has an important value for the examination of the ocular surface disease.

[The culture and cryopreservation of human conjunctival epithelium in vitro].

PURPOSES: To detect the best method of culture and preservation of human conjunctival epithelia. METHODS: Human conjunctival epithelia were cultured by tissue inoculation, mechanical separation or enzyme digestion with 0.25% trypsin. The cultured cells were identified through their morphology, growing features and immunohistochemical staining. The third and fourth passage confluent cells were frozen in liquid nitrogen, and resuscitated 30 days later. RESULTS: Cells from tissue digested with 0.25% Trypsin grew much better than those from tissue inoculation. No cell outgrew in mechanical separating group. The cultured cells spread along the flask in polygonal shape and positive in pan-Keratin staining. Ninty percent of the cryopreserved cells were successfully resuscitated. CONCLUSION: Tissue digestion with trypsin is the best approach to culture human conjunctival epithelium, and the cultured cells can be cryopreserved in liquid nitrogen.

[Areas of brain involved in immunoregulation].

OBJECTIVE: To study the location of brain areas involved in immunoregulation. METHODS: Immunohistochemistry and immunofluorescence were used to detect the different distribution of cytokines immunopositive cells in the brain of rats immunized via both intraperitoneal and subcutaneous injections. RESULTS: The cytokines immunopositive cells were distributed in the supraoptic (SO) and paraventricular (PV) nuclei of the hypothalamus, the anterior hypothalamic (AH) nucleus, arcuate and median eminence, the lateral hypothalamic nucleus (LH) and the amygdaloid nuclear complex while only the appearance of the cytokines immunopositive cells in LH and amygdaloid nuclear complex in hypothalamus was related with immunization status of the animals. Double-labelling results showed that the cytokines immunopositive cells were neurons. CONCLUSIONS: We have observed that neurons of the LH and amygdaloid nuclear complex in hypothalamus as a main source of the neuroimmunoregulation played a key role in neuroimmunoregulation and they participated in the neuroimmunoregulation at an early stage of the immune response.
