[Construction of recombinant E. coli with high glutathione biosynthetic activity and the biosynthetic process].

A recombinant strain E. coli II-1, which exhibited high glutathione (GSH) biosynthetic activity and high stability, was constructed by transforming plasmid pGH501 which contains gene gsh I and gsh II into a wild type strain E. coli II. 4 g/L GSH accumulated extracellularly by using toluene-treated cell. In GSH biosynthetic system, GSH production was improved by increasing the concentration of L-glutamate, while inhibited by L-cysteine if it's concentration was beyond 20 mmol/L. In GSH biosynthetic reaction, the apparent little consumption of L-glutamate and glycine was concluded experimentally to be that toluene-treated E. coli II-1 cells still contained high concentration of L-glutamate and glycine. According to the change of energy cofactor in the GSH biosynthetic process, a possible GSH biosynthetic mechanism controlled by E. coli II-1, was proposed: the energy donator of reaction catalyzed by glutathione synthetase (GSH-II) was ADP but not ATP, the reaction was rate-limited step within the whole GSH biosynthetic process, high concentration of ADP might inhibit the activity of GSH-II. Further degradation of GSH was prevented by the addition of 100 mmol/L L-serine and potassium borate mixture. In such case, 23.0 mmol/L (about 7.1 g/L) GSH accumulated at 3 h.

Pathological, ultrastructural and immunohistochemical observations of adenoma of retinal pigment epithelium.

PURPOSE: To Study the clinical, pathological, ultrastructural and immunohistchemical characters of adenoma of the retinal pigment epithelium in order to offer evidence to diagnose this tumor. METHODS: Routine paraffin slices HE stain, histochemistry PAS and VG stain, transmission electron microscopy, and immunohistochemistry for S-100 and vimentin with LSAB method were used. RESULTS: The tumor cells were oval and cuboidal in shape. Part of the tumor had a tubular arrangement. Around the sheets of tumors cells there was a large amount of uniform red stick-like substances. The above matter represented positive in PAS stain. Most of the above matter was yellow, while less of the matter showed red in VG stain. Transmission electron microscopy showed that there were tight junctions between tumor cells. Immunohistochemistry showed positive for S-100, negative for vimentin. CONCLUSIONS: The ultrastructural and immunohistochemical characters of the adenoma of retinal pigment epithelium are consistent with the retinal pigment epithelium.

[Application of corepraxia in the implantation of intraocular lens].

PURPOSE: In the cases of aphakia or cataract which complicated with iridosteresis, corectasis and pupil displacement, corepraxia was performed when implanting intraocular lens (IOL) in order to recover the normal shape and position of pupil, improve postoperative visual acuity and prevent monodiplopia and glare. METHODS: In the cases which have iridosteresis at the direction of 9-3 oclock or updrawn pupil, iris suture was performed at the incision directly(open method); while those at the direction of 3-9 o'clock, operation was performed outside the anterior chamber (close method). RESULTS: In 22 cases, the percentage which having visual acuity better than 0.5 is 65%, having nearly round shape pupil 67%, having pupil diameter less than 3 mm 73%, having normal pupil position 67%. CONCLUSION: All kinds of deformed pupil can be recovered to their normal shape, size and position by performing corepraxia while IOL was implanted. The prolene has light reaction and better effect in iris suture.

[Exogenous nitric oxide induces apoptosis in tachyzoites of Toxoplasma gondii].

OBJECTIVE: To explore whether NO is able to induce apoptosis in Toxoplasma gondii tachyzoites. METHODS: Apoptosis induced by NO in T. gondii tachyzoites was investigated by TUNEL (terminal-deoxynucleotidyl transferase mediated d-UTP nick end labeling) method, electron microscopy and agarose gel electrophoresis. RESULTS: NO donor, sodium nitroprusside (SNP), was found to induce apoptosis in Toxoplasma gondii tachyzoites in a time- and dose-dependent manner by TUNEL detection. N-acetylcysteine, a NO scavenger, could inhibit SNP-induced apoptosis in the tachyzoites while potassium ferricyanide could not induce apoptosis in the tachyzoite. Electron macroscopy showed that SNP-treated tachyzoites possessed typical morphological features of apoptosis, including chromatin condensation below the nuclear membrane, nuclear pyknosis, and formation of apoptotic body. Agarose gel electrophoresis revealed that SNP-treated tachyzoite DNA fragment exhibited characteristic "DNA ladder" after 15 to 20 h. CONCLUSION: SNP, NO donor, might induce apoptosis in T. gondii tachyzoites in terms of characteristic morphological and biochemical features.

[Analysis of essential oil from Artemisia annul L. by extraction of different methods].

Essential oils from Artemisia annul L. were extracted with CO2 supercritical fluid and steam distilation. The essential oils were analyzed and contrasted by GC-MS. 114 components were identified by computer index. The rate of total components by supercritical CO2 fluid extraction and steam distillation were 81.24% and 85.75% respectively. Some of them such as diphenylene-Methane, Phenyl-1-naphthylamine were discovered from the plant for the first time.

[Design and application of Chinese universal spinal instrumentation in spine surgery].

OBJECTIVE: To report the rationale and primary results of application of Chinese universal spinal instrumentation (CHN-USI) in spinal surgery. METHODS: From September 1998 to June 2000, 70 patients with scoliosis, including 30 congenital scoliosis, 36 idiopathic scoliosis and 4 other disorders, were treated with CHN-USI in our hospital. All patients got follow-up after surgery with roentgenogram. RESULTS: The average follow-up time was 8.9 months. On average, the primary scoliotic curve was reduced from 70.5 degrees (40 degrees-103 degrees) to 32.5 degrees with an average correction rate of 55.6%. The average increases in body height and the length of corrective segment were 6.5 cm and 5 mm respectively. The kyphotic curve was corrected from average 80.5 degrees to 29 degrees. No serious complication was found in our group. CONCLUSIONS: The CHN-USI which has no need of spinal fusion and will elongate together with the children's growth is a new effective instrumentation for correcting scoliosis, especially in case of growing children. It can also be used in the treatment for kyphosis, anterior derotation spondylodesis, spondylolisthesis and spinal fracture.

[Application of diapason instrumentation for treatment of unstable lumbosacral spinal stenosis].

OBJECTIVE: To observe the preliminary clinical outcome of using a new instrumentation Diapason system and to introduce the characterization and surgical technique of this new system for treatment of unstable lumbar spinal stenosis. METHODS: 16 patients with unstable spinal stenosis who were treated by decompression, posterolateral intertransverse arthrodesis and transpedicle instrumentation of Diapason system, were analyzed retrospectively. RESULTS: Lower back pain (LBP) of 16 patients were significantly alleviated after surgery (scores of LBP before operation: 47.5 +/- 0.8; scores of LBP after operation: 31.9 +/- 2.3, P < 0.001). There was no implant failure, no early or later infection and no neurological complications in 16 patients at an average of 6.2-month follow-ups. No pseudoarthrosis was observed on roentgenography. CONCLUSION: Our short-term follow-up and limited cases study showed satisfactory preliminary result of treating unstable lumbar spinal stenosis with Diapason internal fixation.

The distraction reduction fixation system and its application in spondylolisthesis.

OBJECTIVE: An internal fixation apparatus--distraction reduction fixation system (DRFS) was designed to satisfy the clinical needs for spondylolisthesis. METHODS: Since 1996, 53 patients were treated with DRFS. Among them, 35 had spondylolisthesis, 12 had lumbar canal stenosis accompanied with instability, 2 had vertebral tumors and 4 suffered from spinal fracture. The average age was 53.6 years old (ranged 24 approximately 72 yrs). The mean time for follow-up was 30.6 months (16 approximately 44 months). RESULTS: The slip rate was 0.15 +/- 0.10 before operation, and decreased to 0.09 +/- 0.07 after operation. Entire slip reposition was achieved in 19 cases (54.3%). The change in height of the intervertebral space within the fixation segments was 0.7 +/- 0.17. CONCLUSION: DRFS achieved better results for spondylolisthesis less II degree and no other adverse effects were found. Compared with other foreign and domestic techniques, it had advantages in less implants, less operation gears required and ease to utilize in operation. It was proved to be an ideal internal fixation apparatus.

Three-dimensional correction of scoliosis using TSRH instrumentation.

OBJECTIVE: To evaluate the results of TSRH instrumentation in the correction of coronal, sagittal and rotational deformity of scoliosis. METHODS: From January 1998 to December 1999, thirty-two consecutive patients (6 males, 26 females)with scoliosis underwent anterior or posterior spinal instrumentation and fusion using TSRH instrumentation. Of these cases, 21 were idiopathic scoliosis and 11 were congenital scoliosis. The average age at surgery was 16.4 years (range, 11 approximately 45 years). The mean Cobb angle at surgery was 71.2 degrees range, 44 degrees approximately 125 degrees) in the coronal plane, and 49. degrees range, 16 degrees aprroximately 67degrees in the sagittal plane. Rotational deformity (Nash-Moe) ranged from I to III degree. Preoperative apical translation averaged 4.8 cm (range, 3 approximately 9 cm). RESULTS: The average follow-up duration was 13.3 months (range, 10 approximately 24 months). At the final follow-up, the mean Cobb angle in the coronal plane was 26.6 (range, 10 degrees approximately 73 degrees), with a 63.8% of improvement. Sagittal alignment was well maintained with a mean Cobb angle of 28 degrees (range, 10 degrees approximatelky 45 degrees). The average correction of rotation of the apical vertebra was I degree. The average apical translation was 1.6 cm (range, 0.5 approximately 5.0 cm) representing a correction rate of 66, 7%. Complication was noted in two cases with an incidence of 3.1%, one case had superficial infection and the other one had lower hook dislocation. There was no neurologic deficit and pseudoarthrodesis in this series. CONCLUSION: TSRH instrumentation is an effective and convenient three-dimensional correction system with a lower rate of complication, which can not only correct the coronal and rotational deformity, but maintain the sagittal alignment as well.

[Preliminary investigation on the pathogeny, diagnosis and treatment of chondromalacia patella].

This paper presents the preliminary investigation on chondromalacia patella at our department in recent years. A random cluster sampling survey covering 2743 normal persons was carried out. The prevalence rate is 36.2%. It was found that, applying transmission electron microscope and immunohistochemical methods on to cartilage tissues of the abnormal region, articular cartilage necrosis was in direct proportion with the abnormal pressure, while the restoration capability of local chondrocytes was in inverse proportion with pathological changes and the pressure. The chondromalacia patella was produced by repeated abnormal stress acting on the cartilage. The stress derived from the uncongruency and the decreasing in the contact area of patellofemoral joint when the subluxation or tilt of patellae was caused by the abnormal anatomical and biomechanical relationship. The initial lesion was at the matrix of cartilage, the collagen network was disrupted, then proteoglycan was lost. The microenvironment of chondrocytes was changed with degradation of matrix. So the chondrocytes became degenerative and necrosis from superficial to deep layer, then feed back the matrix again. Finally, the total cartilage layer might disappear, and the bone under cartilage might proliferate. At late stage, the cartilage was completely destroyed and had no self-restorative ability. Therefore, early diagnosis and treatment are necessary. It is highly suggested axis radiograph of the knee with the tibiae tuberositas localization are helpful to early diagnosis. Furthermore, JKY-Muscle Rehabilitation Instrument is invented for non-operative therapy. It enhances muscle power by selective training of the vastus medialis muscle using electrical stimulator to relieve pain and correct subluxation of patella with 90% efficiency (63% of excellent-effective rate). In late stage, patellofemoral replacement is recommended. The excellent-effective rate is 86.3%.

[Determination of trace as in health-giving drink by AA-670 atomic absorption spectrophotometry attached VA-90 atomization].

A new method has been developed for the determination of trace metal As in health-giving drink by AA-670 atomic absorption spectrophotometry attached VA-90 atomization. The method is convenient to determine element in water. The results show that the method has good precision and accuracy. The detection limit is 2 ng.L-1 for As. The relative standard deviation is 5.4%-6.1%, and the rates of recovery is 87.5%-115%. This method has been applied to the determination of health-giving drink with satisfactory results.

[On the characteristics of velocity modulation laser spectroscopy].

The technical properties of velocity modulation laser spectroscopy were investigated using the spectrum of A2 pi u-X2 sigma g+ (2-0) band of Meinel system of N2+. The dependence of spectral intensity on discharge current, the pressure of the gas mixture and the pump velocity of the vacuum pump were studied. Making uses of the results of these studies, the sensitivity and resolution of the spectrometer are improved by above 3 times, and economized the experimental expenses can be reduced to as 3/4 time as normal, which confirms that the results of this paper is of considerable uses.

[Assignments of FIR-LMR spectra of CF X 2Pi (nu = 1) and MIR-LMR spectra of NO X 2Pi (nu = 1<--0)].

According to the theories of Zeeman effect and the principle of laser magnetic resonance (LMR), a method and model for the analysis of the laser magnetic resonance spectra were developed and successfully employed to assign the FIR-LMR spectra of CF X2 Pi (nu = 1) and the MIR-LMR spectra of NO X 2Pi (nu = 1<--0). This work provided an efficient and accurate approach for primary analysis of the LMR spectra of novel radicals.

[Detection of genital tract cytomegalovirus and Chlamydia trachomatis infection in patients with tubal pregnancy].

OBJECTIVE: To investigate the influence of genital tract cytomegalovirus and chlamydia infection on the incidence of tubal pregnancy. METHODS: Ninety-five women with tubal pregnancy (study group) and 42 women with ovarian cysts (control group) were selected for this study. Serum cytomegalovirus (CMV) IgM was identified by enzyme-link-immunosorbent-assay (ELISA). Cervical secretions, endometrium, and salpinx tissue were tested for CMV gH gene and chlamydia heat shock protein (HSP) gene by nest polymerase chain reaction (PCR) and PCR respectively. RESULTS: CMV IgM was positive in 14 (15%) women with tubal pregnancy, and 1 (2%) in the control group. CMV gH gene was detected in 18 women (19%) and chlamydia HSP gene in 25 (26%) of the tubal pregnancy group, and 2 (5%) and 2 (5%) in the control group respectively. There were significant differences between the study and the control group (P < 0.05) with respect to detection of CMV IgM, CMV gH gene and chlamydia HSP gene. CONCLUSIONS: CMVand chlamydia trachomatis genital tract infection were more common in women with tubal pregnancy than that in the control group. Genital CMV and chlamydia trachomatis infection may associate with an increased incidence of tubal pregnancy.

Temperature-sensitive mutants of p53 homologs.

Two homologs of the p53 tumor suppressor, p63 and p73 have recently been discovered. These proteins have activities similar to p53 in cell culture but have distinct developmental functions in vivo. We found that temperature-sensitive mutants of certain p63 and p73 isoforms can be created by single amino acid substitutions of an alanine residue corresponding to alanine 135 of murine p53. The mutants (p63gamma-Pro167, p73alpha-Leu156 and p73beta-Ile156) can be controlled by temperature shift between 32 degrees C and 39 degrees C. They can be stably expressed in p53-null H1299 cells at 39 degrees C, become transcriptionally activated at 32 degrees C, and induce expression of p53-responsive genes MDM2 and p21WAF1. Activation of p73beta-Ile156 in H1299 cells inhibits cell division but induces significant increase in cell size (hypertrophy), whereas activation of p73alpha-Leu156 and p63gamma-Pro167 induces apoptosis. These mutants may be useful tools for gaining further insight to the functions of p53 homologs.

Oligonucleotide-capped gold nanoparticles for improved atomic force microscopic imaging and enhanced selectivity in polynucleotide detection.

A novel assay for selective determination of polynucleotides using atomic force microscopy in conjunction with the formation of the probe/target/DNA-gold nanoparticle sandwich structure at a gold surface is described. A 17-mer probe was attached to the surface for subsequent hybridization with a polynucleotide target. Due to the flat orientation of the probe-target hybrid with respect to the surface and the spatial obstruction of the unhybridized probes near the hybrids, the AFM images are not clear. The hybridization efficiency was estimated to be about 1.1% since certain surface features could not be resolved. The utilization of 30-mer-capped gold nanoparticles not only provides another dimension of selectivity, but also reorients the previously formed probe-target hybrid in such a way that the strands of the target become tethered with respect to the surface. This reorientation improves the resolution in imaging the hybridized target molecules and provides an accurate determination of the hybridization efficiency (16%).

Stable SNARE complex prior to evoked synaptic vesicle fusion revealed by fluorescence resonance energy transfer.

Although it is clear that soluble N-ethylmaleimide-sensitive factor (NSF) attachment protein receptor (SNARE) complex plays an essential role in synaptic vesicle fusion, the dynamics of SNARE assembly during vesicle fusion remain to be determined. In this report, we employ fluorescence resonance energy transfer technique to study the formation of SNARE complexes. Donor/acceptor pair variants of green fluorescent protein (GFP), cyan fluorescent protein (CFP), and yellow fluorescent protein (YFP) are fused with the N termini of SNAP-25 and synaptobrevin, respectively. In vitro assembly of SNARE core complex in the presence of syntaxin shows strong fluorescence resonance energy transfer (FRET) between the CFP-SNAP-25 and YFP-synaptobrevin. Under the same conditions, CFP fused to the C terminus of SNAP-25, and YFP- synaptobrevin have no FRET. Adenovirus-mediated gene transfer is used to express the fusion proteins in PC12 cells and cultured rat cerebellar granule cells. Strong FRET is associated with neurite membranes and vesicular structures in PC12 cells co-expressing CFP-SNAP-25 and YFP-synaptobrevin. In cultured rat cerebellar granule cells, FRET between CFP-SNAP-25 and YFP-synaptobrevin is mostly associated with sites presumed to be synaptic junctions. Neurosecretion in PC12 cells initiated by KCl depolarization leads to an increase in the extent of FRET. These results demonstrate that significant amounts of stable SNARE complex exist prior to evoked synaptic vesicle fusion and that the assembly of SNARE complex occurs during vesicle docking/priming stage. Moreover, it demonstrates that FRET can be used as an effective tool for investigating dynamic SNARE interactions during synaptic vesicle fusion.

A modified p53 overcomes mdm2-mediated oncogenic transformation: a potential cancer therapeutic agent.

The antiproliferative activities of wild-type (wt) p53 are inhibited by mdm2 (murine double minute2) oncogene product. We tested growth suppression activity of p53 14/19, an engineered p53 variant, which does not bind mdm2 and is completely resistant to the inhibition by mdm2. p53 14/19, unlike wt p53, suppressed the growth of cancer cells that contain amplified mdm2 oncogene efficiently by direct DNA transfection or adenovirus-mediated gene transfer. In addition, p53 14/19 also inhibited the growth of several different cancer cell lines expressing low levels of mdm2 oncogene product as efficiently as wt p53. We further examined the antioncogenic potencies of p53 14/19 in the rat embryo fibroblast cotransformation assay. Addition of wt p53 failed to cause any significant decrease in ras plus mdm2 foci counts. In contrast, cotransfection of p53 14/19 with ras and mdm2 significantly reduced foci number. In similar experiments, cotransfection of wt p53 or 14/19 p53 resulted in significant inhibition of oncogenic transformation in rat embryo fibroblast mediated by an activated ras plus c-myc, adenovirus E1A, or human papillomavirus E7 oncogenes. Therefore, these results suggest that p53 14/19 modified tumor suppressor gene may be a promising therapeutic agent for human cancers that express abnormally high levels of mdm2 oncogene product.
