Cloning and sequencing of the fragment of angiostatin K (1-3) gene.

OBJECTIVE: To secure anti-endotheliocyte hyperplasia functional fragment of human angiostatin K (1-3) gene. METHODS: As the template of leukocytotic cDNA, functional fragment of human angiostatin K (1-3) cDNA was amplified by PCR, cloned into the vector pGEM-3Zf, and sequenced according to Dye primer sequencing kit. RESULTS: The functional fragment of angiostatin K (1-3) cDNA (859 bp) was obtained by PCR and determined by sequencing. CONCLUSION: The angiostatin K (1-3) gene has been cloned. The gene is of great significance in tumor-antiangiogenesis therapy.

[The characteristics of luminance of color visual evoked potential in macular diseases].

OBJECTIVE: To objectively evaluate the luminance channel damage of macular diseases by color pattern reversal visual evoked potential (CPR-VEP). METHODS: CPR-VEP was performed in 38 eyes with age-related macular degeneration (ARMD) dry type, 24 eyes with ARMD wet type, 20 eyes with Stargardt's disease and 54 normal eyes. Color pattern stimuli were obtained with color monitor controlled by computer program. Color pattern reversal rate was 2 Hz and its spatial frequency was 0.53 cycle/degree. The color graphic system could present white/black (W/B), red/black (R/B), green/black (G/B), blue/black (B/B), yellow/black (Y/B). RESULTS: In normal eyes, CPR-VEP P(1) latency was negatively correlated with luminance, whereas P(1) amplitude positively correlated with luminance; there were linear regressions between the mean luminance and P(1) latency and between the mean luminance and P(1) amplitude. Also in ARMD dry type, the CPR-VEP showed that the characteristics of the luminance channel were similar to that in the normal eyes. However, these phenomena did not exist in ARMD wet type and Stargardt's disease. CONCLUSION: In ARMD dry type, there is no damage of luminance channel, but in ARMD wet type and Stargardt's disease, there is severe damage of luminance channel and the patients can not discriminate luminance difference.

[Effect of diazepam on fetus after intravenous administration during the active phase in labour].

To investigate the effects of diazepam on fetal heart rate and Apgar scoring administered intravenously during the active phase in labour, sixty-two normal term parturients during the active phase of labour with the cervical dilation to 3-5 cm were randomly divided into two groups, diazepam group (n = 32) and control group (n = 30). In diazepam group, 10 mg diazepam was administered intravenously. The concentrations of diazepam in maternal and cord serum were measured by high performance liquid chromatography after birth and the fetal heart rates were monitored by electronic monitor before or after administration of diazepam and Apgar scores were determined immediately after birth. The results showed that the mean level of diazepam in umbilical cord serum was markedly higher than that of maternal serum. Fetal heart base line mutation in diazepam group was weaker than that of the control group. Apgar scores in the neonates of diazepam group was lower than those of the control group. The results indicate that the use of diazepam in pregnant women during active phase of labour may cause depression of fetus.

[Epidemiological survey of conjoined twins in China].

A descriptive study was conducted to investigate the incidence and epidemiological characteristics of conjoined twins in China. From 1988 to 1992, data were collected from more than 500 hospital-based units involved in a monitoring program and were analyzed by the National Center for Birth Defects Monitoring. The monitored subjects were the hospitalized births from 28 weeks gestation to a period within 7 days after delivery. 106 conjoined twins were found in 3,246,408 births with an incidence rate of 0.3/10,000. The highest rate was 0.8/10,000 in He'nan Province and Anhui Province(P < 0.05), and the lowest 0.1/10,000 in Beijing and Jilin Province (P < 0.01). Conjoined twins were more common in females; the ratio of female to male was 2 to 1. No difference was observed between the urban and rural areas. The prenatal mortality was as high as 915@1000. Only 31.13% conjoined twins were diagnosed prenatally. The results indicate that the incidence of conjoined twins in China is not different from that in other countries, but the work of prenatal diagnosis in China should be improved.

[A clinical study of sudden death in patients with type 2 diabetes mellitus].

This study aimed to investigate the clinical characteristics of sudden death (SD) in type 2 diabetes mellitus (DM2). Clinical data from 1988 through 1996 in our hospital showed that 130 hospitalized patients with DM2 died, and 17 of them died of SD (13.08%). These 17 SD patients were pair-matched with 17 DM2 patients who were alive in the same period. The results revealed that the SD group had longer clinical DM duration, more chronic diabetic complications and higher blood pressure. In the direct causes of SD, cardiac SD accounted for 76.47%. The others were non-cardiac factors, including cerebral hemorrhage, hyperkalemia from diabetic nephropathy with renal failure and respiratory tract obstruction from lung infection. The triggering causes included eating, defecting, lung infection, strenuous attempt, hypoglycemia and surgical operation. To reduce the rate of SD in DM2, it is necessary and vital to treat the DM2 itself, and to take positive steps to prevent the onset of SD in high risk patients.

[HLA-DRB1 typing by PCR-sequence specific primers for paternity determination].

HLA-DRB1 typing by PCR-SSP technique was first used for paternity testing. The analysis of 42 paternity cases shows that this identification method is simple, fast and reliable and has high exclusion probability of paternity (66.3%). This method can also be applied to the study of transplantation, HLA associated disease and anthropogenesis.

[Radiofrequency treatment of ethmoidal nerve with allergic rhinitis under nasal endoscopy].

OBJECTIVE: To study the effect of radiofrequency treatment of ethmoidal nerve on allergic rhinitis. METHOD: Eighty-eight cases of allergic rhinitis were treated with ethmoidal nerve radiofrequency under nasal endoscopy. RESULT: It showed marked effectiveness in 73 cases (83.0%), immprovement in 11 cases (12.5%) and failed in 4 cases (4.5%) in one year follow-up. The overall effective rate was 95.5%. CONCLUSION: This result indicated that it is a choice for treating allergic rhinitis to apply ethmoidal nerve radiofrequency under nasal endoscopy.

[Experience in endoscopic sinus surgery in district hospital].

OBJECTIVE: To explore the importance and specificity of conducting endoscopic sinus surgery (ESS) in district hospital. METHOD: Seventy patients with chronic nasal sinusitis or/and nasal polyps undergone ESS from June 1997 to October 1998 were retrospectively analyzed. The curative effect was evaluated according as FESS-95 Guangzhou Standard. RESULT: In all patients, 52 cases (74.3%) were cured, 16 cases (22.9%) improved, 2 cases (2.8%) unimproved and 4 cases (5.7%) were found with complications. CONCLUSION: The results showed that intranasal endoscopic sinus surgery had a significant effect on treating nasal-sinusitis and nasal polyps. The CT types of ethmoid sinus are significant in selecting anesthesia and operation program. To get a good result, a full combined therapy in the perioperation period and a careful endoscopic follow-up postoperation are important.

[Analysis of products of thymine irradiated by heavy ion beam with capillary gas chromatography].

In this study some chromatographic methods with capillary column, such as GC, GC-MS, GC-FTIR, were used to analyze the products of thymine irradiated by 18O8+ ion beam in N2O saturated aqueous solution. Because of the complexity and small amounts of the products after irradiation by heavy ion beams, it is very difficult to separate them, while with the methods of capillary gas chromatography this problem can be overcome. From the results of GC-MS analysis we can determine the molecular weight of the product, and from the results of GC-FTIR analysis we can get structure information of the product. By this way the products such as 5,6-dihydrothymine, 5-hydroxyl-5-methylhydantoin, 5-hydroxyl-6-hydrothymine, 5-hydro-6-hydroxylthymine, 5-hydroxymethyluracil, trans-thymine glycol, cis-thymine glycol and dimers were determined. Though these products are the same as those from thymine irradiated by gamma ray in N2O saturated aqueous solution, the mechanism of heavy ion beam interact ion with thymine aqueous solution is different from that of gamma ray. The main products from thymine irradiated by 18O8+ ion beam in N2O saturated aqueous solution are formed by hydroxyl additions between 5-6 band of thymine, while those by gamma ray are dimers of thymine.

[Study of HSP70 gene upstream regulation element on expression efficiency of GST gene in M. smegmatis].

Four different expression vectors were constructed by cloning foreign gene which encode Schistosoma japonicum 26 kD antigen (Sj26GST) into Escherichia coli-Mycobacteria shuttle plasmid pBCG-2000 and investigated their expression efficiency in mycobacteria smegmatis. The plasmid which contains promoter of human mycobacterial tuberculosis heat shock protein 70 was firstly digested with Nco I and modified with two different ways to lead to two kinds of SD sequences, and ligated with Sj26GST encoding gene. Then, the DNA fragment contained HSP70 promoter and Sj26GST gene was obtained and cloned into E. coil-mycobacteria shuttle plasmid pBCG-2000, and finally four recombinant mycobacterial expression vectors that differenciated in SD sequence, orientation and copy number were selected. The expressed native recombinant Sj26GST(rSj26GST) was soluble and could be observed on SDS-PAGE about at the molecular weight of 26 kD obviously. Analysis with protein density scanning indicated: the expression efficiency that containing double-copy promoter-foreign gene vector was the highest and the expressed protein which was about 1.6 times than others was 28% of total protein of Mycobacteria smegmatis. The cloned direction and SD sequence had no significant effect on expression efficiency.

[Transformation of effusion and formation of granulation tissue in the pathologic process of otitis media with effusion].

OBJECTIVE: To explore the transformation of stagnant effusion (SE) and the pathologic behavior leading to granulation tissue (GT) formation in the pathologic process of otitis media with effusion (OME). METHOD: The temporoal bone slides from 306 ears with OME at the Otopathology Laboratory of the University of Minnesota USA were studied histopathologically under light microscopy. RESULT: The observations revealed a pathologic process in which the type and the condition of stagnant effusion in the middle ear cleft was variable in the dynamics of OME progression from an early pathologic stage to an advanced pathologic stage. The granulation tissue was primarily formed by two types of fibroblast proliferation in subepithelial space. The absorption and the orgnization of SE and the formation of granulation tissue maybe occur simultaneously in same areas and a same pathologic process. CONCLUSION: The granulation tissue can only form in the areas where effusion has stagnated and has been absorpted. It is very important that effusion of the otitis media should be removed completly before the formation of granulation tissue.

[Studies on the supercritical-CO2 fluid extraction and quality evaluation of ginger oils].

Essential oils from the dried rhizomes of Zingiber officinale were extracted by supercritical-CO2 fluid(SFE-CO2) and traditional water still distillation (WSD) methods, and the SFE extraction technology and quality (GC-MS, etc.) of the oils were studied. The results showed that the SFE-CO2 contains 49 constituents, such as 6-paradol, etc, the ginger peppery component of them gets to 22.90%.

[The manufacture and clinical use of the auto-sialographic instrument by remote controlling]

OBJECTIVE:The purpose of this article is to realize the auto-control of the injection of radiographic drugs in terms of the principle in remote controlling.METHODS:30 cases of sialaden diseases was studied using the auto sialographic instrument by remote controlling,compared to 67 cases studied using sialography by hand-controlling.RESULTS:The auto-sialographic instrument by remote controlling realize the telecontrol of the injection of the sialographic drugs and auto-injection,simplify the working procedure,and prevent medical staffs from X-ray radiation at work.CONCLUSION:The auto sialographic instrument by remote controlling can not only bring much convenience to medical staffs,but also improve the conformation to clinical diagnosis.

Translocation efficiency, susceptibility to proteasomal degradation, and lipid responsiveness of apolipoprotein B are determined by the presence of beta sheet domains.

Apolipoprotein (apo) B100 is an atypical secretory protein in that its translocation across the endoplasmic reticulum membrane is inefficient, resulting in the partial translocation and exposure of apoB100 on the cytoplasmic surface of the endoplasmic reticulum. Cytosolic exposure leads to the association of nascent apoB with heat shock protein 70 and to its predisposition to ubiquitination and proteasomal degradation. The basis for the inefficient translocation of apoB100 remains unclear and controversial. To test the hypothesis that beta sheet domains present in apoB100 contribute to its inefficient translocation, we created human apoB chimeric constructs apoB13,16 and apoB13,13,16, which contain amino-terminal alpha globular domains but no beta sheet domains, and apoB13,16,beta, which has an amphipathic beta sheet domain of apoB100 inserted into apoB13,16. These constructs, along with carboxyl-terminal truncations of apoB100, apoB34 and apoB42, were used to transfect HepG2 and Chinese hamster ovary cells. In contrast to the lack of effect of proteinase K on apoB13,16 and apoB13,13,16, the levels of apoB34, apoB42, and apoB13,16,beta were decreased by 70-85% after proteinase K-induced proteolysis in both HepG2 and Chinese hamster ovary cells. Either oleic acid or proteasomal inhibitors (N-acetyl-leucinyl-leucinyl-norleucinal and lactacystin) significantly increased the cell levels of apoB13,16,beta, apoB34, apoB42, and full-length apoB100 but had no effect on the cell levels of apoB13,16 and apoB13,13,16. When HepG2 cells were incubated with a microsomal triglyceride transfer protein inhibitor, the cellular levels of apoB13,16,beta, apoB34, and apoB42 were decreased by 70-80%, whereas the levels of apoB13,16 and apoB13,13,16 were unaffected. The effects of microsomal triglyceride transfer protein inhibition were reversed by lactacystin. Our results clearly demonstrate that the translocation efficiency, susceptibility to proteasomal degradation, and lipid responsiveness of apoB were determined by the presence of a lipid binding beta sheet domain. It is possible that beta sheet domains may at least transiently facilitate the interaction of apoB with the lipid bilayer surrounding the translocation channel.

Autocrine transforming growth factor alpha provides a growth advantage to malignant cells by facilitating re-entry into the cell cycle from suboptimal growth states.

CBS human colon carcinoma cells are poorly tumorigenic in athymic nude mice, whereas FET colon carcinoma cells are non-tumorigenic. Both cell lines have well differentiated properties in tissue culture. Transforming growth factor alpha (TGF-alpha) was ectopically expressed by stable transfection of a TGF-alpha cDNA under repressible tetracycline control. The TGF-alpha-transfected cells showed enhanced clonal initiation and shortened lag phase growth in tissue culture without an alteration in doubling time in exponential phase relative to untransfected cells. Furthermore, the TGF-alpha transfectants showed increased independence from exogenous growth factors in clonal growth assays and induction of DNA synthesis after release from quiescence. Growth factor independence was associated with sustained epidermal growth factor receptor activation in quiescent TGF-alpha-transfected cells and the requirement of exogenous insulin for stimulation of quiescent cells to re-enter the cell cycle. Higher cloning, reduced lag time in tissue, and the acquisition of growth factor independence for DNA synthesis without a change in doubling time of TGF-alpha-transfected cells indicate that autocrine TGF-alpha functions by facilitating re-entry into the cell cycle from sub-optimal growth states rather than promoting or controlling the proliferation of actively cycling cells. The modulation of growth regulation by autocrine TGF-alpha was associated with increased malignant properties as TGF-alpha transfectants showed increased tumorigenicity in athymic nude mice. The administration of tetracycline reversed the effects of TGF-alpha expression in these cells both in vivo and in vitro, indicating that the alterations of the biological properties were due to the expression of TGF-alpha. Since these cells are continuously grown in a completely chemically defined medium without serum supplementation, it was possible to assign the mechanism underlying the generation of growth factor independence to the replacement of a requirement for exogenous insulin in parental cells by autocrine TGF-alpha.

Tumor necrosis factor-beta is associated with thymic apoptosis during acute rejection.

BACKGROUND: Intrathymic events undergoing allograft rejection remain undefined. The present study investigated the role of tumor necrosis factor-beta on acute thymic involution in rat hepatic allograft recipients during rejection. METHODS: Apoptosis and cellular phenotypic changes in the thymus were studied after hepatic transplantation. RESULTS: Thymocytes in both the medulla and cortex were sparse during acute rejection. Phenotypically, CD4+CD8+ T cells decreased significantly, whereas there were relative increases in CD4-CD8-, CD4+CD8-, and CD4-CD8+ T cells in untreated allograft recipients. Additionally, thymic apoptosis was found by in situ DNA end labeling and electron microscopy. Apoptotic cells were predominantly distributed in the cortex. Biologic lymphotoxin (tumor necrosis factor-beta)/tumor necrosis factor-alpha cytotoxic activity in the serum was significantly increased in untreated hepatic allograft recipients. Tumor necrosis factor-beta mRNA was detected in untreated allograft livers, and intraperitoneal administration of recombinant human tumor necrosis factor-beta induced extensive apoptosis of thymocytes in vivo. In contrast, no significant thymic involution was observed in donor-specific blood transfusion-treated allograft and isograft recipients. Intraperitoneal administration of rabbit anti-human tumor necrosis factor-beta polyclonal antibody or recombinant human interleukin-10 inhibited thymic apoptosis in untreated hepatic allograft recipients. CONCLUSIONS: Allograft rejection, but not donor-specific transfusion-induced immunologic unresponsiveness, is associated with thymic involution, a process that may be mediated by tumor necrosis factor-beta.

A unique amyloidogenic apolipoprotein serum amyloid A (apoSAA) isoform expressed by the amyloid resistant CE/J mouse strain exhibits higher affinity for macrophages than apoSAA1 and apoSAA2 expressed by amyloid susceptible CBA/J mice.

CBA/J and other inbred strains of mice that express the amyloidogenic apolipoprotein serum amyloid A (apoSAA) apoSAA2, together with apoSAA1, are susceptible to amyloid A (AA) amyloidosis, whereas CE/J mice that express a single unique isoform, apoSAACEJ, are resistant. Studies indicate that CBA/JxCE/J hybrid mice that express apoSAA2 in the presence of apoSAACEJ are protected from amyloidogenesis. To define a mechanism by which expression of apoSAACEJ may protect from AA formation in the presence of apoSAA2, binding of recombinant apoSAA (r-apoSAA) isoforms, validated by N-terminal sequencing, to a murine macrophage cell line was investigated. Maximal specific binding occurred after incubation of radiolabeled apoSAA with IC-21 macrophages (1x105 cells/ml) for 30 min at 4 degreesC. The binding of 125I-r-apoSAA1, 125I-r-apoSAA2 and 125I-r-apoSAACEJ was specific and saturable, with an affinity (Kd) of about 2.8, 3.2 and 1.3 nM, respectively, and approximately 2-4x106 sites per cell. Competitive binding experiments indicate apoSAACEJ binds with higher affinity to macrophages than does either apoSAA1 or apoSAA2. We suggest that greater cellular affinity of apoSAACEJ compared to apoSAA2 may contribute to protection from AA amyloid in certain CBA/JxCE/J hybrid mice by interfering with interaction of apoSAA2 by macrophages and hence either membrane associated or intracellular degradation.

Analytical shape computation of macromolecules: I. Molecular area and volume through alpha shape.

The size and shape of macromolecules such as proteins and nucleic acids play an important role in their functions. Prior efforts to quantify these properties have been based on various discretization or tessellation procedures involving analytical or numerical computations. In this article, we present an analytically exact method for computing the metric properties of macromolecules based on the alpha shape theory. This method uses the duality between alpha complex and the weighted Voronoi decomposition of a molecule. We describe the intuitive ideas and concepts behind the alpha shape theory and the algorithm for computing areas and volumes of macromolecules. We apply our method to compute areas and volumes of a number of protein systems. We also discuss several difficulties commonly encountered in molecular shape computations and outline methods to overcome these problems.
