Biochemical and ultrastructural alterations in platelets, reticulocytes, and lymphocytes from rats fed vitamin E-deficient diets.

Effects of vitamin E-deficiency, dietary fat (corn oil versus lard), and incubation on ultrastructure of platelets, red and white cells, and on selected biochemical parameters of platelets and plasma were studied. Platelets from vitamin E-deficient rats had no obvious morphological defects, but reticulocytes and lymphocytes had swollen and deformed mitochondria. Fatty acid and glycogen levels of platelets were not affected by the deficiency but total lipid levels in plasma were decreased of increased depending upon the type and level of dietary fat. In comparison with supplemented controls, the proportion of stearate increased in the phospholipid fraction of plasma from vitamin E-deficient, lard-fed but not corn oil-fed rats. In platelets, total fatty acids per mg protein were 8-12% lower with lard than with corn oil as fat source. Oleate and linoleate were higher and lower, respectively, although not to the same degree, and arachidonate was not affected. With incubation of platelet-rich plasma at 37 degrees C for 6 hr, there were no obvious morphological changes in platelets from control or from deficient, lard-fed rats, but platelets from deficient, corn oil-fed rats contained mitochondria that were swollen and deformed. Incubation did not affect fatty acid, glycogen, or tocopherol levels of platelets or tocopherol levels of plasma, regardless of type or amount of dietary fat.

Improved direct determination of alpha- and gamma-tocopherols in plasma and platelets by liquid chromatography, with fluorescence detection.

Tocopherols extracted from plasma with methanol or from platelets with chloroform/methanol were injected in methanol on a reversed-phase (C18) "high-performance" liquid-chromatographic column and eluted with water/methanol (2/98, by vol) at a flow rate of 1.4 mL/min. A "high-performance" spectrophotofluorometer was used for detection. Analytical recoveries ranged from 89 to 106%. The response was linear to at least 0.3 micrograms of either tocopherol (alpha- or gamma-) applied to the column, and the limit of detection was 0.1 ng. The method was used to measure tocopherols in plasma and platelets from human subjects, and some values are presented.

[Alcohol and central and peripheral nervous system disorders].

It belongs to the definition of chronic alcoholism that also clinically established lesions of the organs can be proved. As to the nervous system the case in question are multilocular defects which concern the central and peripheral nervous system. The local accentuation, the developmental speed and the intensiveness of the alcoholic defect of the cellular metabolism lead to different courses. The suggestion of their individuality must at the same time take into consideration the character of chronic alcoholism which on principle affects the whole nervous system.

Dopamine receptor mediated inhibition by pergolide of electrically-evoked 3H-dopamine release from striatal slices of cat and rat: slight effect of ascorbate.

The dopamine receptor agonist pergolide inhibited the calcium-dependent, electrically evoked overflow of tritium from slices of the striatum of cat or rat prelabelled with 3H-dopamine. This inhibition of tritium overflow by nanomolar concentrations of pergolide was antagonized by the benzamide neuroleptic S-sulpiride (0.1 microM). In millimolar concentrations, L- ascorbate had slight or no effects on this dopamine receptor mediated inhibition, in striatal slices of either the cat or the rat. Since these same concentrations of ascorbate have been reported to completely block the specific binding of 3H-2-amino-6,7-dihydroxy-1,2,3,4-tetrahydronaphthalene (ADTN) and of 3H-apomorphine to presumed dopamine receptors, the present results suggest a dissociation between the characteristics of 3H-ADTN and 3H-apomorphine binding and the dopamine autoreceptor. Previous contradictory results concerning the existence of inhibitory dopamine receptors which modulate depolarization-evoked overflow of dopamine from the striatum of the rat are thus apparently not due to a species difference nor to the use of ascorbate, but rather to differences in experimental conditions.

Wild-type temperature-sensitive and -resistant visna viruses: isolation and biological comparison.

The plaques formed by wild-type (WT) populations of strain K485 visna (V-K485) virus, strain K796 visna (V-K796) virus, and of progressive pneumonia virus in sheep choroid plexus (SCP) cell cultures are heterogeneous in size. Plaque purification procedures showed that this heterogeneity was due to the presence of two biologically different viruses, large plaque-forming (Lpf) virus and small plaque-forming (Spf) virus. The V-K796 Lpf virus, the V-K796 Spf virus, and the V-K796 WT virus are antigenically similar to each other. Although the V-K796 Spf virus is less cytopathic than the V-K796 Lpf virus, both viruses have a 14- to 16-hour latent period in SCP cells and have similar rates of synthesis for the initial 24 to 30 hours after infection. The V-K796 Spf virus is considered a temperature-sensitive visna virus since, unlike the V-K796 Lpf virus, it does not produce plaques at 41 degrees C and it is more thermosensitive than the V-K796 Lpf variant. Population analyses of two strains of wild-type visna virus and one strain of progressive pneumonia virus demonstrated that these populations contain variants with the phenotypes of V-K796 Spf virus and V-K796 Lpf virus, that the Spf virus was not selected against in sheep, and that cultivation of the wild type visna viruses at 37 degrees C selected against the temperature-sensitive variants. The existence of the temperature-sensitive Spf viruses in these wild-type virus populations suggests that the Spf variants maintain the persistent visna virus infection in sheep and delay the development of the host's cell-mediated immune response to the viral proteins.

Comparative sensitivities of tocopherol levels of platelets, red blood cells and plasma for estimating vitamin E nutritional status in the rat.

The use of tocopherol levels of either platelets or red blood cells was tested as an index of tissue vitamin E status. Male Wistar rats were fed 0, 5, 10, 20, or 50 ppm of vitamin E (d-alpha-tocopheryl acetate) ad libitum for 10 wk. Over the dosage range from 0 to 20 ppm, response in tocopherol content of most tissues including platelets and red blood cells were linear. Over the whole range from 0 to 50 ppm, responses were curvilinear both directly and logarithmically for all tissues. In a comparison of reproducibility of responses of platelets, plasma and red blood cells, the variation of alpha-tocopherol content within groups fed the same diet was consistently lowest for platelets. Sensitivity (linear slope/SD) for reflecting vitamin E intake was higher for platelets than for either red blood cells or plasma. In vitro spontaneous hemolysis of red blood cells was less than 10% with 10 ppm of vitamin E, greater than 85% with 0 ppm, and was variable (2 to 78%) with 5 ppm. Lipid levels in plasma increased significantly as vitamin E was increased from 5 to 50 ppm of the diets. Over this range, levels of cholesterol and phospholipid increased 20% and levels of triglyceride increased almost 200%.

Tryptophan malabsorption in dementia. Improvement in certain cases after tryptophan therapy as indicated by mental behaviour and blood analysis.

Twenty-four senile dementia patients in the age range 65-85 years were subjected to a new sensitive tryptophan loading test: Dopa-Tryptophan loading ("Dop-Try loading") in order to determine whether Try malabsorption occurs in senile dementia and if so how frequent this is. For comparison the same loading test was performed on 23 subjects in the same age grouping but with normal metal function. The differences in Try absorption were statistically analyzed and this revealed that absorption was less effective in the senile dementia group. Four patients with Try malabsorption were treated with 3 G 1-Try + 50 Mg dl-5-hydroxytryptophan (5-HTP) per day orally for 20-30 days and four patients with normal absorption were treated in the same way. The Dop-Try loading test was repeated after this treatment. Two patients showed improved absorption and coincident mental improvement, while one showed decreased absorption and mental deterioration. One died during the course of the treatment. None of the four patients with normal absorption showed mental improvement after the treatment. This pilot study suggests that increase in Try absorption following the Try treatment is a necessary condition for improvement in the mental state. The study is being continued with larger numbers of patients.

[Adrenoleukodystrophy (author's transl)].

A report on clinical course and morphological findings of a case of adrenoleukodystrophy in a 10-years-old boy is given. The brain showed the typical changes of a sudanophilic, inflammatory leukodystrophy in all lobes. Pathognomic intracytoplasmatic inclusions in the perivascular macrophages were found by electron microscopic examination. The adrenal glands showed a severe atrophy, but it was impossible to detect typical cytolpasmatic inclusions within the adrenocortical cells. By thin-layer chromatography an accumulation of cholesterol esters with very long chain fatty acids was found in the demyelinated areas. Findings diagnostic facilities and theories of pathogenesis are discussed.

Lymphokine (interleukin-2) production by mitogen-stimulated human lymphocytes in small reactors.

Human spleen cells or peripheral blood lymphocytes (PBL) were stimulated with the T-cell mitogens concanavalin A or phytohemagglutinin to produce the lymphokine interleukin-2 (IL-2). Culture conditions were optimized using 250-1000 ml reactor vessels. Optimal IL-2 release was found at a low speed of stirring allowing circulation of the medium without suspending the agglutinated cells, and at an oxygen concentration of about 30-45% of saturation. Cell viability was suboptimal at these conditions.

Glycosides for testing glycosidases: vinyl, 1-ethoxyethyl, and 1-ethoxybut-3-enyl D-glucopyranosides.

The reaction of ethyl vinyl ether and 2,3,4,6-tetra-O-acetyl-beta-D-glucopyranose (1) in the presence of Hg(OAc)2 and toluene-p-sulphonic acid as catalysts yielded the acetylated vinyl, 1-ethoxyethyl, and 1-ethoxybut-3-enyl glycosides in varying proportions. Crystalline 1-ethoxybut-3-enyl 2,3,4,6-tetra-O-acetyl-beta-D-glucopyranoside (2), vinyl 2,3,4,6-tetra-O-acetyl-alpha-D-glucopyranoside (3), and 1-ethoxyethyl 2,3,4,6-tetra-O-acetyl-beta-D-glucopyranoside (4) were isolated by chromatography. Compound 4 was also prepared by the reaction of 1 with cold acetaldehyde diethyl acetal containing a trace of acetic acid, and its alpha anomer (5) by the reaction of 1 with boiling acetaldehyde diethyl acetal containing a trace of acetic acid. Each deacetylated D-glucoside was cleaved by the corresponding D-glucosidase, to yield D-glucose and either acetaldehyde (from deacetylated 3--5) or but-3-enal (from deacetylated 2).

Flow-through fluorocytophotometry of different brain tumours.

The flow-through cytophotometric method was used to investigate the single-cell DNA content in 60 tumours of the CNS and allied structures (9 meningiomas, 5 ependymomas, 15 astrocytomas, 11 anaplastic astrocytomas, 8 glioblastomas, 7 medulloblastomas, 2 oligodendrogliomas, 1 anaplastic oligodendroglioma, and 2 neurinomas). The cytophotometric parameters were correlated with morphological and clinical data of the tumours. The results are summarised in Tables 1--7. With regard to the DNA content of their cell nuclei, the gliomas present a behaviour similar to tumours in other organs, but in the present study the cytophotometric signs of malignancy in gliomas are not so evident as in carcinomas or sarcomas. The information obtained by flow cytophotometric methods may be helpful in diagnosing the degree of malignancy in brain tumours.
