Three new bioactive bis-adjacent THF-ring acetogenins from the bark of Annona squamosa.

Continuing work on the bioactivity-directed fractionation of the bark of Annona squamosa has resulted in the discovery of three new Annonaceous acetogenins, (2,4-cis and trans)-squamolinone (1), (2,4-cis and trans)-9-oxoasimicinone (2), and bullacin B (3). Compounds 1-3 are all adjacent bis-THF ring acetogenins with 2 representing the first bis-ring acetogenin to contain a carbonyl along its aliphatic chain. Compound 3 was selectively cytotoxic in a panel of six human tumor cell lines with a potency of nearly a million times that of adriamycin against the MCF-7 (human breast adenocarcinoma) cell line.

New bioactive triterpenoids from Melia volkensii.

Bioactivity-directed fractionation of the root bark of Melia volkensii resulted in the isolation of two new natural products, meliavolkinin (1) and melianin C (3), together with two known compounds, 1,3-diacetylvilasinin (2) and melianin B (4). Jones oxidation of 4 gave compounds 3, 23,24-diketomelianin B (5), and 16,23,24-triketomelianin B (6). The structures of the new compounds were elucidated by spectral and chemical data. Compounds 1-6 all showed marginal cytotoxicities against certain human tumor cell lines, while 5 showed selective cytotoxicities for the human prostate (PC-3) and pancreatic (PACA-2) cell lines with potencies comparable to those of adriamycin.

Annonaceous acetogenins as natural pesticides: potent toxicity against insecticide-susceptible and -resistant German cockroaches (Dictyoptera: Blattellidae).

Six compounds, representing the mono-tetrahydrofuran (THF) (gigantetrocin A, annomontacin), adjacent bis-THF (asimicin, parviflorin), and nonadjacent bis-THF (sylvaticin, bullatalicin) classes of annonaceous acetogenins, were compared with technical grades of synthetic amidinohydrazone (hydramethylnon), carbamate (propoxur, bendiocarb), organophosphate (chlorpyrifos), and pyrethroid (cypermethrin) insecticides to determine their dietary toxicities to insecticide-resistant and insecticide-susceptible strains of the German cockroach, Blattella germanica (L.). Differential susceptibility occurred among B. germanica nymphs of both strains to this variety of the acetogenins and the 5 conventional synthetic insecticides. The speed of kill (LT50) values against insecticide-susceptible and insecticide-resistant 2nd and 5th instars permitted ranking of all 11 compounds. The adjacent bis-THF acetogenins showed the highest potency among the 3 acetogenin classes. The acetogenins caused high percentages of mortality and delays in development of the 5th instars of both strains. Insecticide-susceptible nymphal development was mainly affected by gigantetrocin A and annomontacin, whereas insecticide-resistant nymphal development was mainly affected by gigantetrocin A and bullatalicin. Most tested acetogenins performed better than the conventional insecticides against both stages of both strains. No growth regulation effects were caused by any of the compounds tested. Low resistance ratios were obtained for most compounds (except chlorpyrifos). Low resistance ratios values for 2nd instars ranged from 0.9 to 2.2 with the natural acetogenins and from 1.0 to 3.8 with the synthetic compounds; the 5th instars ranged from 0.2 to 3.9 with the natural acetogenins and from 0.6 to 8.0 with the synthetic compounds. Insecticidal properties and characteristics of acetogenins and the possible use of acetogenins in baits for cockroach control are discussed.

Structure-activity relationships of diverse Annonaceous acetogenins against multidrug resistant human mammary adenocarcinoma (MCF-7/Adr) cells.

Fourteen structurally diverse Annonaceous acetogenins, representing the three main classes of bis-adjacent, bis-nonadjacent, and single-THF ring(s), were tested for their ability to inhibit the growth of adriamycin resistant human mammary adenocarcinoma (MCF-7/Adr) cells. This cell line is resistant to treatment with adriamycin, vincristine, and vinblastine and is, thus, multidrug resistant (MDR). Among a series of bis-adjacent THF ring acetogenins, those with the stereochemistry of threo-trans-threo-trans-erythro (from C-15 to C-24) were the most potent with as much as 250 times the potency of adriamycin. A spacing of 13 carbons between the flanking hydroxyl of the THF ring system and the gamma-unsaturated lactone seems to be optimum with a spacing of 11 and 9 carbons being significantly less active. Several single-THF ring compounds were also quite potent with gigantetrocin A (11) being the most potent compound tested. The acetogenins may, thus, have chemotherapeutic potential, especially with regard to MDR tumors.

The Annonaceous acetogenin bullatacin is cytotoxic against multidrug-resistant human mammary adenocarcinoma cells.

Cytotoxic effects of the Annonaceous acetogenin, bullatacin, were studied in multidrug-resistant (MDR) human mammary adenocarcinoma (MCF-7/Adr) cells vs. the parental non-resistant wild type (MCF-7/wt) cells. Bullatacin was effectively cytotoxic to the MCF-7/Adr cells while it was more cytostatic to the MCF-7/wt cells. ATP depletion is the mode of action of the Annonaceous acetogenins, and these agents offer a special advantage in the chemotherapeutic treatment of MDR tumors that have ATP-dependent mechanisms.

Pathology of familial Alzheimer's disease with Lewy bodies.

The neuropathological findings of three cases from two pedigrees with early onset familial Alzheimer's disease (FAD) are reported. Affected members of the first family, including cases 1 and 2 reported here, are known to have 717 valine to isoleucine mutation of the amyloid precursor protein (APP) gene, while the genetic background of the disease has not been clarified yet in the second family. In all three cases, in addition to the classical histological findings associated with Alzheimer's disease (AD), both nigral and cortical Lewy bodies (LBs) occurred. The association of LBs with AD type pathology, which may be observed in both sporadic and familial AD, raises important nosological issues. These include a possible overlap between AD and other neurodegenerative conditions presenting primarily with LBs. In this respect the clinically and neuropathologically distinct disease entity described under the terms of "senile dementia of the Lewy body type" and "Lewy body variant of AD" may be especially important. The occurrence of LBs in association with severe AD-type histological changes in sporadic and especially in some familial AD cases indicate that these inclusions may be another expression of the altered cytoskeleton in AD.

Two new bioactive Annonaceous acetogenins, rollitacin (1) and rollinacin (2), along with one known acetogenin, javoricin, were isolated from the ethanolic extract of the leaves of Rollinia mucosa. Compounds 1 and 2 exhibited selective inhibitory effects among six human solid tumour cell lines. The structural elucidations of 1 and 2 were achieved by various spectroscopic analyses and chemical derivatizations.

Additional bioactive annonaceous acetogenins from Asimina triloba (Annonaceae).

Trilobalicin (1), a new nonadjacent bis-THF ring annonaceous acetogenin, 2,4-cis- (2) and 2,4-trans-trilobacinone (3), the ketolactones of trilobacin, an adjacent bis-THF ring acetogenin, were isolated from the stem bark of Asimina triloba (L.) Dunal (Annonaceae). Their structures were established based on chemical and spectral evidence. The relative stereochemistry of 1 was determined as trans/threo/threo/trans/erythro from C-10 to C-22 by comparisons of NMR data with those of model compounds. Compound 1 is the first example of a nonadjacent bis-THF acetogenin being isolated from the title species and represents a new type of these compounds. Bioactivities of these new structures against brine shrimp larvae and six human solid tumor cell lines were determined, and cytotoxic selectivities were shown for the lung (A-549) and breast (MCF-7) cell lines with up to a million times the potency of adriamycin.

4-deoxyannomontacin and (2,4-cis and trans)-annomontacinone, new bioactive mono-tetrahydrofuran annonaceous acetogenins from Goniothalamus giganteus.

4-Deoxyannomontacin (1) and a mixture of (2,4-cis and trans)-annomontacinone (2), new bioactive mono-tetrahydrofuran (THF) gamma-lactone and keto-lactone acetogenins, respectively, as well as five known mono-THF acetogenins [xylomaticin, longifolicin, longicoricin, (2,4-cis and trans)-gigantetrocinone, and (2,4-cis and trans)-gigantetroneninone], were isolated from the bark of Goniothalamus giganteus (Annonaceae) by activity-directed fractionation using the brine shrimp lethality test (BST). The structures were elucidated based on spectroscopic and chemical methods. The absolute stereochemistries of 1 and 2 were determined by the advanced Mosher ester method and by circular dichroism (CD). Determination of the absolute stereochemistry at C-10 as R for 1 is the first example of the direct determination of the absolute stereochemistry of a carbinol position isolated from other functional groups in the annonaceous acetogenins. 1 and 2 showed selective and potent cytotoxicities to certain human tumor cell lines and were comparable to the activity of rotenone against yellow fever mosquito larvae.

Isolation of new bioactive annonaceous acetogenins from Rollinia mucosa guided by liquid chromatography/mass spectrometry.

Reversed-phase high-performance liquid chromatography (RP-HPLC) fractionation, monitored by liquid chromatography/electrospray mass spectrometry (LC/ESI-MS), led to the isolation of two new bioactive annonaceous acetogenins, rollidecin C (1) and rollidecin D (2), from the bioactive aqueous methanol fraction of the leaves of Rollinia mucosa (Annonaceae). The structures were confirmed by analyses of the 1H and 13C NMR data. In addition, a known adjacent bis-tetrahydrofuran (THF) acetogenin, desacetyluvaricin (3), was isolated from this plant for the first time utilizing the LC/ESI-MS monitoring approach. Compound 1 exhibited selective cytotoxicity toward the colon tumor cell line (HT-29), while 2 showed only borderline cytotoxicity in a panel of six human tumor cell lines.

Screening for Annonaceous acetogenins in bioactive plant extracts by liquid chromatography/mass spectrometry.

The Annonaceous acetogenins are amenable to analysis by liquid chromatography/mass spectrometry (LC/MS) using the electrospray positive-ion mode. Under conditions of atmospheric pressure in-source collision-induced dissociation (APICID), the acetogenins reproducibly provided characteristic ion patterns and fragment ions. Accordingly, the presence of these biologically interesting acetogenins and other derivatives in plant extracts and chromatographic fractions can be readily detected by analyzing selected ion chromatograms. LC/MS screening of a bioactive crude methanol-soluble fraction of Rollinia mucosa detected the presence of some 40 known acetogenins in this plant extract, in addition to four new acetogenins of diverse structure. This rapid and relatively uncomplicated selected ionization procedure should also prove suitable for the screening of many other structural families of natural products.

Biologically active acylglycerides from the berries of saw-palmetto (Serenoa repens).

Brine shrimp lethality-directed fractionation of the 95% EtOH extract of the powdered, dried berries of Serenoa repens (Bart.) Small (saw-palmetto) (Palmae) led to the isolation of two monoacylglycerides, 1-monolaurin (1) and 1-monomyristin (2). Compounds 1 and 2 showed moderate biological activities in the brine shrimp lethality test and against renal (A-498) and pancreatic (PACA-2) human tumor cells; borderline cytotoxicity was exhibited against human prostatic (PC-3) cells. The fruits and extracts of saw-palmetto are taken orally as an herbal medicine to prevent prostatic hyperplasias.

Novel mono-tetrahydrofuran ring acetogenins, from the bark of Annona squamosa, showing cytotoxic selectivities for the human pancreatic carcinoma cell line, PACA-2.

The bark of Annona squamosa yielded three new mono-tetrahydrofuran (THF) ring acetogenins, each bearing two flanking hydroxyls and a carbonyl group at the C-9 position. These compounds were isolated using the brine shrimp lethality assay as a guide for the bioactivity-directed fractionation. (2,4-cis and trans)-Mosinone A (1) is a mixture of ketolactone compounds bearing a threo/trans/threo ring relationship and s double bond two methylene units away from the flanking hydroxyl. The other two new acetogenins differ in their stereochemistries around the THF ring; mosin B (2) has a threo/trans/erythro configuration across the ring, and mosin C (3) possesses a threo/cis/threo relative stereochemistry. Also found was annoreticuin-9-one (4), a known acetogenin that bears a threo/trans/threo ring configuration and a C-9 carbonyl and is new to this species. The structures were elucidated based on spectroscopic and chemical methods. Compounds 1-4 all showed selective cytotoxic activity against the human pancreatic tumor cell line, PACA-2, with potency 10-100 times that of Adriamycin.

(2,4-cis and trans)-gigantecinone and 4-deoxygigantecin, bioactive nonadjacent bis-tetrahydrofuran annonaceous acetogenins, from Goniothalamus giganteus.

Two new acetogenins, (2,4-cis and trans)-gigantecinone (1), isolated as a mixture, and 4-deoxygigantecin (2), a known acetogenin whose absolute stereochemistry has not been determined previously, were isolated using activity-directed fractionation, from the bark of Goniothalamus giganteus. A key step in solving their absolute stereochemistries was the preparation of 1,4-diol formaldehyde acetal derivatives (1b and 2a). Using the advanced Mosher ester method and circular dichroism, the absolute stereochemistries of 1 and 2 were revealed and were found to be the same as that of gigantecin (3), which supports a common biogenetic origin. Both 1 and 1b showed potent and selective cytotoxicities against the PC-3 human prostate adenocarcinoma cell line. Against yellow fever mosquito larvae, 1 and 2 were more potent than rotenone in pesticidal activity. Longimicin C and a mixture of (2,4-cis and trans)-isoannonacin were also isolated for the first time from this species.
