Calcium-binding protein-immunoreactive projection neurons in the caudal subnucleus of the spinal trigeminal nucleus of the rat.

It has been reported that calcium-binding proteins are good markers for different sets of neurons in various brain regions. We examined expression of the main calcium-binding proteins in projection neurons in the rat medullary dorsal horn (MDH) by combining immunofluorescence histochemistry for calbindin D28k (CB), calretinin (CR) and parvalbumin (PV) with the retrograde tract-tracing method. A fluorescence tracer, tetramethylrhodamine-dextran amine (TMR-DA), was injected into the parabrachial, thalamic or hypothalamic region. After such injections, a number of PV-, CR-, and/or CB-immunoreactive MDH neurons were labeled retrogradely with TMR-DA. Triple-immunofluorescence histochemistry further revealed that a number of CB-, CR-, or PV-immunoreactive TMR-DA-labeled MDH neurons showed immunoreactivity for substance P receptor (NK1), and that they expressed immunoreactivity for c-fos protein in the rats which were injected with formalin into the lips. Thus, it was indicated that some of CB-, CR-, or PV-containing projection neurons in the MDH might be involved in the transmission of nociceptive stimuli.

Primary afferent fibers of the pelvic nerve terminate in the gracile nucleus of the rat.

After injection of horseradish peroxidase (HRP) into the pelvic nerve of the rat, a small number of HRP-labeled axon terminals were found in the gracile nucleus. Double labeling experiments were also performed: Fluoro-Gold (FG) was injected into the pelvic nerve, while cholera toxin B subunit (CTb) was injected into the gracile nucleus or dorsal faciculus at the fifth and sixth cervical cord segments ipsilateral to the FG injection. About 5% of FG-labeled neurons were labeled with CTb in the L6-, S1- and S2-dorsal root ganglia ipsilateral to the tracer injection. The results suggest that some primary afferent information from pelvic visceral organs may be directly conveyed to gracile nucleus by the primary afferent neurons.

Preprodynorphin-like immunoreactivity in medullary dorsal horn neurons projecting to the thalamic regions in the rat.

Preprodynorphin (PPD)-like immunoreactive (-LI) neuronal cell bodies in the trigeminal sensory nuclear complex of the rat were found in laminae I and II of the medullary dorsal horn (MDH; caudal spinal trigeminal nucleus) and the paratrigeminal nucleus. A PPD immunofluorescence histochemistry combined with a fluorescence retrograde tract-tracing method revealed that some of the PPD-LI neurons in the MDH and paratrigeminal nucleus projected to the thalamic regions. Nociceptive nature of the PPD-LI MDH neurons projecting to the thalamic regions was also demonstrated by a triple labeling method, using the technique of the noxious stimulus-evoked expression of the immediate-early gene, c-fos. In the rats which were subcutaneously injected with formalin into the upper and lower lips, c-fos protein (Fos) was found in PPD-LI neurons which were labeled with a retrograde tracer injected into the thalamic regions.

Expression of the focal adhesion protein paxillin in lung cancer and its relation to cell motility.

Lung cancer can lead to abnormalities of the actin cytoskeleton structure which may be important in transformation. In this study, we have investigated the expression of the cytoskeletal associated protein paxillin in lung cancer. Paxillin is a 68 kDa focal adhesion protein, with four tandem LIM domains at the C-terminus, involved in growth factor receptor, integrin and oncogenic signaling such as v-src, BCR/ABL, and E6 of the papilloma virus. In non-small cell lung cancer (NSCLC) cell lines, paxillin localized to the focal adhesions. The possible role of paxillin in lung cancer cells was assessed by overexpressing green fluorescence protein (GFP)-paxillin construct in two separate NSCLC cell lines (Calu-1 and H661). Over the course of 48 h, GFP-paxillin consistently caused the cells to become round and to decrease cell motility as compared to normal controls, GFP-N-terminus paxillin, or GFP-LIM transfected cells. Because some lung cancers may be quite aggressive and metastasize quickly, which may be related to the cytoskeleton, we determined the expression of paxillin in NSCLC and small cell lung cancer (SCLC) cell lines and patient tumor tissues. Expression of paxillin in NSCLC and SCLC cell lines were determined by Northern blot and Western blot analysis. The expression of paxillin was consistently low in SCLC cell lines, whereas there was paxillin expression in NSCLC cell lines. There was a variability of expression of paxillin in NSCLC tumor tissue as compared to normal lung tissue. In contrast, by immunohistochemistry, we show that there was no detectable expression of paxillin in 5/5 SCLC patients. This data suggests that absence or low level of paxillin protein expression may cause certain lung cancers, such as SCLC, to be more motile and possibly more aggressive.

Association of VDR and estrogen receptor genotypes with bone mass in postmenopausal Caucasian women: different conclusions with different analyses and the implications.

Much work has been done on the association between vitamin D receptor (VDR) genotypes and bone mineral density (BMD). Despite considerable effort, the results are inconsistent. While the VDR association remains unresolved, studies have expanded to other candidate genes (i.e., estrogen receptor (ER) genotypes), also yielding inconsistent results. A few studies have suggested that interaction effects between VDR and ER genotypes significantly affect BMD. We assessed associations of BMD with VDR BsmI genotypes, and ER XbaI and PvuII polymorphisms (denoted as ERX and ERP respectively) with spine, femoral neck, distal radius BMD, and with total body bone mineral content (tbBMC) in 108 US Mid-western postmenopausal Caucasian women. We statistically controlled for confounding factors such as height, weight, etc., in the analysis. No significant association was detected for ER genotypes with spine and radius BMD, or for VDR genotypes with femoral neck and radius BMD and tbBMC. No significant interaction between VDR and ER genotypes was detected in our sample. However, the VDR genotypes are significantly (p = 0.004) associated with approximately 5.8% spine BMD variation. Both ERX and ERP genotypes are significantly (p = 0.02) associated with approximately 3.5% femoral neck BMD variation. ERX genotypes are significantly (p = 0.03) associated with approximately 2.4% tbBMC variation. However, if the data were analyzed by simple ANOVA as in some previous studies, without adjusting statistically for confounding factors, all the significant results we found here would have gone undetected. Our findings suggest that: (1) VDR and ER genotypes may have different effects on BMD at different sites and on tbBMC; and (2) if significant factors influencing bone are not appropriately controlled, true significant associations can easily be missed. These findings may offer a partial explanation for some of the earlier inconsistent results of association studies on BMD with VDR and ER genotypes.

On the experimental design and data analysis of mutation accumulation experiments.

Characterizing deleterious genomic mutations is important. Most of the few current estimates come from the mutation-accumulation (M-A) approach, which has been extremely time- and labour-consuming. There is a resurgent interest in implementing this approach. However, its estimation properties under different experimental designs are poorly understood. By simulations we investigate these issues in detail. We found that many of the previous M-A experiments could have been more efficiently implemented with much less time and expense while still achieving the same estimation accuracy. If more than 100 lines are employed in M-A and if each line is replicated at least 10 times during each assay, an experiment of 10 M-A generations with two assays (at the beginning and at the end of M-A) may achieve at least the same estimation quality as a typical M-A experiment. The number of replicates per M-A line necessary for each assay largely depends on the magnitude of environmental variance. While 10 replicates are reasonable for assaying most fitness traits, many more are needed for viability, which has an exceptionally large environmental variance. The investigation is mainly carried out using Bateman-Mukai's method of moments for estimation. Estimation using Keightley's maximum likelihood is also investigated and discussed. These results should not only be useful for planning efficient M-A experiments, but also may help empiricists in deciding to adopt the M-A approach with manageable labour, time and resources.

The effect of overdominance on characterizing deleterious mutations in large natural populations.

Alternatives to the mutation-accumulation approach have been developed to characterize deleterious genomic mutations. However, they all depend on the assumption that the standing genetic variation in natural populations is solely due to mutation-selection (M-S) balance and therefore that overdominance does not contribute to heterosis. Despite tremendous efforts, the extent to which this assumption is valid is unknown. With different degrees of violation of the M-S balance assumption in large equilibrium populations, we investigated the statistical properties and the robustness of these alternative methods in the presence of overdominance. We found that for dominant mutations, estimates for U (genomic mutation rate) will be biased upward and those for h (mean dominance coefficient) and s (mean selection coefficient), biased downward when additional overdominant mutations are present. However, the degree of bias is generally moderate and depends largely on the magnitude of the contribution of overdominant mutations to heterosis or genetic variation. This renders the estimates of U and s not always biased under variable mutation effects that, when working alone, cause U and s to be underestimated. The contributions to heterosis and genetic variation from overdominant mutations are monotonic but not linearly proportional to each other. Our results not only provide a basis for the correct inference of deleterious mutation parameters from natural populations, but also alleviate the biggest concern in applying the new approaches, thus paving the way for reliably estimating properties of deleterious mutations.

Recovery of mutations of different sizes from a population sample of DNA sequences under variable mutation rates across sites.

Mutations may be classified according to their positions of occurrence in the genealogy of the sampled DNA sequences from a population. A mutation is said to be of size i if it has i descendants in the sample. Such classifications for mutations may yield detailed insights into the evolutionary history and properties of the population. Statistical methods based on such classification have been developed and shown to be efficient and powerful. However, the utility of these statistical methods critically depends on reliable and robust recovery of mutations of different sizes. We investigated the distributional changes of mutations of different sizes due to genealogy reconstruction using the unweighted pair-group method with arithmetic mean (UPGMA) and the performance of maximum-parsimony method in inferring mutations of different sizes on a given topology. Genealogy reconstruction by UPGMA was found to change the distribution of mutations of different sizes on constructed topologies. Multiple hits at some nucleotide sites made it difficult to infer mutations of different sizes with the maximum-parsimony method, even when the true topology was designated. These results suggest that while the newly developed statistical methods employing information on mutations of different sites are powerful, they also impose significant new challenges for developing methods to accurately recover mutations of different sizes from population DNA sequence data.

Skeletal muscle oxidative capacity, fiber type, and metabolites after lung transplantation.

Lung transplant (LTx) recipients have a low peak work rate, peak oxygen consumption (V O2peak), and early lactate threshold on incremental exercise. We hypothesized that LTx recipients have reduced oxidative function and altered fiber type proportion in peripheral skeletal muscle. Seven stable LTx recipients and seven age- and sex-matched control subjects were studied. Incremental exercise testing with arterialized venous sampling and a resting quadriceps femoris punch muscle biopsy were performed. Muscle specimens were analyzed for fiber type proportion, metabolites, oxidative and glycolytic enzyme activities, and mitochondrial ATP production rate (MAPR) using standard techniques. The results showed that mean V O2peak in LTx recipients was 52% of control subjects. Compared with the control subjects, LTx skeletal muscle exhibited: (1) a lower MAPR; (2) lower activity of the mitochondrial enzymes glutamate dehydrogenase (GDH), citrate synthase (CS), 2-oxogluterate dehydrogenase (OGDH), and 3-hydroxyacyl-CoA-dehydrogenase (HAD). There was no difference in the activities of anaerobic enzymes, except for higher phosphofructokinase activity; (3) a lower proportion of type I fibers; (4) a higher lactate and inosine monophosphate (IMP) content and a lower ATP content at rest indicating a high reliance on anaerobic metabolism. The reduced type I fiber proportion and severely reduced mitochondrial oxidative capacity may play an important role in exercise limitation after LTx.

[Scanning electron microscope observation on endosperm starch grain characters in multiplasmic maize].

In this article, endosperm starch grains of eleven lines in multiplasmic maize were studied by scanning electron microscope. The results show that different cytoplasm has different effects on the characters of starch grain. The starch grains of three sweet corn cytoplasmic lines (su1, sh2, btl) are mainly spherical and packed tightly, which means they have some degree of similarity. While the grains of four cytolasmic male sterility lines (T, S, C, 21A) are mostly irregular and packed loosely except (T) Mo17, in which grains are tightly packed. The average diameter of these eleven varieties is ranged from 9.78 microns to 14.69 microns. Through the study of endosperm starch grain shape and size, we tried to explore the relationship between the property of starches and the quality of seeds. And this will significantly prompt the development of maize genetics and breeding.

Effect of tetrandrine on morphine dependence in isolated guinea pig ileum.

AIM: To evaluate the effects of tetrandrine (Tet) and nimodipine (Nim) on the morphine (Mor) withdrawal response in the isolated guinea pig ileum. METHODS: The withdrawal contracture was elicited by addition of naloxone (Nal) (1 mumol.L-1) to the isolated naive ileum incubated with Mor (3 mumol.L-1) at 37.5 degrees C for 4 h or to the ileum obtained from Mor-dependent guinea pig. RESULTS: When Nim (0.01, 0.05, and 0.1 mumol.L-1) or Tet (1, 10, and 50 mumol.L-1) was added 1 min before Nal in the naive ilea bathed in Krebs solution containing Mor, or when the ilea from Mor-dependent guinea pigs were incubated with Nim (0.01, 0.05, and 0.1 mumol.L-1) or Tet (1, 10, and 50 mumol.L-1) for 15 min, or when Nim (5 and 10 mg.kg-1, i.p.) or Tet (15 and 30 mg.kg-1, i.p.) was administered in vivo to Mor-dependent guinea pigs, the Nal-precipitated withdrawal contracture was significantly decreased in a dose-dependent manner. CONCLUSION: Tet and Nim, Ca2+ channel blockers, could inhibit the Nal-precipitated Mor withdrawal response in the isolated guinea pig ileum.

Alterations of the PPP2R1B gene in human lung and colon cancer.

The PPP2R1B gene, which encodes the beta isoform of the A subunit of the serine/threonine protein phosphatase 2A (PP2A), was identified as a putative human tumor suppressor gene. Sequencing of the PPP2R1B gene, located on human chromosome 11q22-24, revealed somatic alterations in 15% (5 out of 33) of primary lung tumors, 6% (4 out of 70) of lung tumor-derived cell lines, and 15% (2 out of 13) of primary colon tumors. One deletion mutation generated a truncated PP2A-Abeta protein that was unable to bind to the catalytic subunit of the PP2A holoenzyme. The PP2R1B gene product may suppress tumor development through its role in cell cycle regulation and cellular growth control.

Projections from the caudal spinal trigeminal nucleus to commissural interneurons in the supratrigeminal region: an electron microscope study in the rat.

Electron microscopic double-labeling study in the rat indicated that projection fibers from the caudal spinal trigeminal nucleus (Vc) were distributed ipsilaterally within the supratrigeminal region (STR) capping the trigeminal motor nucleus (Tm) and made synaptic contact with neurons projecting to the contralateral Tm. Nociceptive inputs to the Vc may reflexly control, via interneurons in the STR, the activities of Tm neurons innervating the masticatory, tensor tympani, and/or tensor veli palatine muscles.

Immunocytochemical localization of mu-opioid receptor in primary afferent neurons containing substance P or calcitonin gene-related peptide. A light and electron microscope study in the rat.

Co-expression of mu-opioid receptor-like immunoreactivity (MOR-LI) with substance P (SP)- or calcitonin gene-related (CGRP)-LI was observed in rat trigeminal and dorsal root ganglion neurons. In particular, MOR-LI was found in axon terminals with SP- or CGRP-LI in laminae I and II of the medullary and spinal dorsal horns. MOR may be implicated in modulation of release of SP and CGRP from primary sensory afferents.
