Accounting for voluntary waiting period changes in US dairy herds: Adjusting daughter pregnancy rate and introducing first-service to conception.

The genetic evaluation of female fertility traits in dairy cattle in the United States has progressed over the past 2 decades, with 4 additional traits integrated into the national evaluation system since the introduction of daughter pregnancy rate (DPR) in 2004. However, concerns have arisen in the dairy sector, with reports of producers extending the voluntary waiting period (VWP), which is the time of initial breeding after calving, due to more persistent lactation yields. Since its inception, DPR calculations in the US evaluation have assumed a fixed 50-d VWP, which may not reflect modern reproductive strategies. Furthermore, producers may determine that some of their cows should have their VWP extended while others may follow the standard time. A re-evaluation of the current female fertility evaluation is necessary to ensure traits align with current management practices. Therefore, this study explores the addition of a potential new trait, First Service to Conception (FSC), along with a revised DPR formula that permits more flexibility with VWP. About 32 million records from the National Cooperator Database, covering 5 major dairy breeds (2003-2023), were used for this study. Data for cows calving before 2003 were unavailable because insemination records were not recorded prior. The findings suggest FSC enhances female fertility evaluations, providing a more comprehensive reproductive assessment independent of VWP. Another approach was an adjustment to the DPR calculation (DPRadj) to account for herd VWP on a herd-year and lactation group basis. The mean DPR value was 45.06%, while DPRadj increased the mean value to 52.58%. The mean FSC was 62.64 d. In an analysis using first lactation cows for cow traits and heifers for heifer traits, genetic correlations of FSC with other fertility traits were 0.97 with Cow Conception Rate (CCR) and DPR, 0.98 with DPRadj, 0.40 with Heifer Conception Rate (HCR), and 0.32 with Early First Calving (EFC). Predicted transmitting abilities (PTA) were calculated to assess the implications of adding FSC to, or replacing DPR with DPRadj within the CDCB multi-trait evaluation index, Net Merit $ (NM$). The highest correlation among the top 10% of bulls occurred when replacing DPR with DPRadj (0.99). Adding FSC to the original model reduced the latter correlation to 0.88, which indicates slight re-ranking of top bulls. Among animals with PTA reliability above 50%, the lowest correlation was between the original index and the index replacing DPR with FSC (0.95). This work advances genetic evaluation in dairy cattle, improving reproductive efficiency and productivity.

Dry period length to maximize production across adjacent lactations and lifetime production.

The primary objectives of this research were to determine the dry period lengths that maximize production across adjacent lactations and also dry period length that maximizes lifetime yield. Effect of days dry (DD) after lactations 1 through 3 were determined separately for both adjacent lactation sums and lifetime yield. Field data, collected through the Dairy Herd Improvement Association, on US Holstein cows first calving between January 1997 and January 2004 were utilized. Lifetime records were restricted to cows first calving no later than December 1999. Actual lactation yields, in contrast to standardized records, were used to calculate lactation sums and lifetime records. Herds were required to be on test for the entire period to avoid partial records. Another important edit was that actual calving dates had to agree with expected calving dates, based on reported days open, within 10 d. This edit ensured that the producer knew, at least at one point in time, when the cow was going to calve. Cow effects were corrected for in both the adjacent lactation and lifetime analyses. The minimum DD to maximize production across adjacent lactations depended on parity. For yield across first and second lactations, there was little loss in production with a minimum of 40 to 45 DD. Longer dry periods (55 to 65 DD) were required after second and third lactations however, presumably due to the lower persistency of second and later lactation cows. Lifetime production was maximized by 40 to 50 DD after first lactation and 30 to 40 DD after second and later lactations. Fewer DD were required to maximize lifetime yield than adjacent lactation yield because cows with fewer DD also had more lifetime days in milk. Although dry periods of 30 to 40 d can be used after second and later lactations without cost in lifetime yield, their benefit to lifetime production is minimal. Dry periods shorter than 30 d or longer than 70 d are costly to lifetime yield and should be avoided. Dry periods longer than 80 d are even more costly than dry periods less than 30 d.

Characterization of Holstein heifer fertility in the United States.

The overall object of this research was to characterize US Holstein (virgin) heifer fertility. This included investigation of factors influencing heifer fertility and estimation of heritability, as well as correlations with cow fertility and first-lactation milk yield. A secondary objective was to compare linear and logistic model estimates of fixed effects and linear and threshold model estimates of heritability. Data consisted of Holstein heifers, which were artificially inseminated, with their first breeding between March 2003 and August 2005. Herds were required to have at least 60 breedings across the 3 yr of data and an overall mean conception rate (CR) between 20 and 80%. After edits there were 537,938 breedings of 362,512 heifers in 2,668 herds from 41 states used for analysis. After edits, the overall mean CR for US Holstein heifers was 57%. Linear and logistic model estimates for all factors were nearly identical. Year of breeding accounted for the most variation in heifer CR, with heifer age and month of breeding being the next most important factors. Conception rate in heifers is maximal at an intermediate age of 15 to 16 mo. Heifers at 26 mo of age and older have roughly a 10% lower CR than heifers bred at younger ages. Although month of breeding affected heifer CR, effects are less than for cows. In contrast to cow fertility, heifer CR is nearly as good in the hotter summer months as in cooler months. Approximately 88% of US herds had a 40 to 70% heifer CR. Heritability estimates of heifer CR on first service were 0.5% from the linear model and 1.0% from the threshold model. Genetic correlation estimates of heifer CR on first service with cow CR on first service and with first-lactation milk yield were 0.39 and -0.19, respectively. Results indicated that selection on either the currently available US daughter pregnancy rate evaluations for cow fertility or on cow CR will also improve heifer fertility. Furthermore, heritability of heifer CR is lower than for cow CR and reporting of heifer breedings is currently less complete than for cow breedings. Thus, there are currently no immediate plans to implement a US genetic evaluation for heifer CR.

In situ TEM observation of the nucleation and growth of silver oxide nanoparticles.

The nucleation, growth, and coalescence of silver oxide nanoparticles have been investigated dynamically and at high spatial resolution by using the electron beam of a transmission electron microscope to stimulate and to observe the processes. Under the assumption the particles are hemispherical, the growth rate was found to be proportional to the square root of the electron irradiation time. This result suggests that the rate-limiting step is the attachment of atoms to the nanoparticles. Growth of the nanoparticles occurred by the addition of columns of atoms on {111} planes. Particle impingement resulted in interpenetration of the particles and, ultimately, the formation of a grain boundary.

TEM and HREM of diamond crystals grown on Si tips: structure and results of ion-beam-treatment.

Diamond single crystals were grown on the silicon whiskers by a hot filament chemical vapor deposition technique at the filament temperature about 2100 degrees C and the temperature of support 800 degrees C. Specimens were examined by SEM, TEM, HRTEM and SAED. When the filament temperature was about 1900 degrees C globular polycrystalline diamond particles were grown. At a support temperature more then 800 degrees C SiC nanoparticles were formed. To investigate the ion etching process of the silicon tip/diamond system, tips were treated with an Ar(+) beam with energy up to 30 kV. The results depend on fluence: at 4 x 10(18)ion/cm(2) diamonds and partially Si tips were destroyed, amorphous layer was formed (sometimes with nanometric size fragments of diamond); at 1 x 10(18)ion/cm(2) sharpened diamonds (radius of curvature about 20 nm) covered with amorphous layer (radius about 80 nm) probably with nanoclusters of diamond were observed; at 4.4 x 10(17) ion/cm(2) there was no visible tip sharpening but formation of amorphous thick layer occurred. The emission characteristics of Si tips covered with diamond were improved due to ion treatment. Since such tips in our case were covered with amorphous layer containing nanometric size fragments of diamond, we suppose this layer is responsible for electron emission improvement.

Characterization of days dry for United States Holsteins.

This research characterized days dry (DD) for modern US Holsteins. This included investigation of factors influencing DD, summarizing variation in herd means as well as within-herd variation, and estimation of parameters related to DD. Data consisted of records initiated before 2003 from cows first calving after 1996. A total of 459,075 records from 295,067 cows in 3527 herds were included. The overall mean DD was 60.5 d, and standard deviation among herd means was 5.8 d. Approximately 91% of all herds had mean DD between 50 and 70 d. There were 41 herds that had mean DD of < or =45 d and 2 herds with mean DD of < or =30 d. Some herds (125) had unusually long mean DD (exceeding 71 d). Variation across regions of the United States was minimal, although southwestern herds did show somewhat longer DD than other regions. Days open (DO) accounted for the most variation in DD with longer DO leading to longer dry periods. Heritability of DD was 7% and repeatability 12%. Error correlations indicated that longer dry periods benefit both milk yield and somatic cell score in the subsequent lactation. Genetic and permanent environmental correlations were large, which indicated that 1) bias would result if genetic evaluations for milk yield included an adjustment for DD, and 2) correction for cow effects is warranted in estimation of DD effects on performance. The relationship between DD and DO is due primarily to permanent cow effects. Improved fertility will enhance the feasibility of shortened dry periods.

Methodology for estimation of days dry effects.

The primary objective of this research was to determine if, with appropriate methodology, unbiased estimates of days dry (DD) effects on subsequent lactation milk yield can be obtained from field data, particularly when DD is correlated with cow effects. Another objective was to ascertain relevant sampling properties of designed trials for estimation of DD effects. Simulated records were used to assess methodology. Along with a model with no adjustments for cow effects, alternative models including 1) previous lactation milk yield, 2) a prior adjustment for cow effects estimated from an animal model, and 3) a combination of 1 and 2, were tested. Estimates from the unadjusted model were biased downward; however, the 3 alternative analyses provided estimates of DD effects that were essentially unbiased, with a prior adjustment for cow effects and previous milk yield in the model providing the best results in terms of elimination of bias. Therefore, DD effects can be estimated from field data without bias from cow effects.A designed trial with 2 groups and 10 or fewer cows/ group is noninformative and has an unacceptably high probability of leading to invalid conclusions. A minimum of 30 cows/group is considerably better and should be used whenever possible. Even with 30 cows/group, however, the power is low unless the difference between DD groups for yield is at least 1130 kg. Prior correction of 305-d, mature equivalent records for cow effects, using predicted producing abilities, could be done in designed trials to improve the statistical power of tests and accuracy of estimates.

Use of early lactation days open records for genetic evaluation of cow fertility.

National genetic evaluations for cow fertility were introduced by the USDA in February, 2003. These evaluations, reported as daughter pregnancy rate, are based on days open. One requirement of the evaluation system is that lactations be at least 250 d in milk (DIM) to be included for analysis. The objective of this research was to develop a predictor of days open, usable in genetic evaluation, to allow for earlier predicted transmitting abilities (PTA), especially for young bulls. The final prediction equation included an overall intercept, the effects of lactation and calving ease score, the linear and quadratic effects of age at calving, and a regression on days open based on last breeding. Data used for estimation were breeding records from 4 dairy records processing centers for the years 1995 through 1998. Genetic correlations were > or =0.91 by d 130, and phenotypic means of predicted days open were in agreement with means for final days open, indicating that the predictions were phenotypically unbiased. Comparison of mean PTA based on actual and predicted days open indicated no bias in PTA, and correlations between PTA were > or =0.92 by d 130. The earlier use of data increased reliability by about 5% for sires between the ages of 4 and 5 yr. The USDA began using predicted days open for records that are at least 130 DIM in national evaluations starting November, 2003.

A composite method for the determination of the chirality of single walled carbon nanotubes.

An approach to the unambiguous determination of the conformation of individual single walled nanotubes utilizing high-resolution transmission electron microscopy and digital image processing is described. The exit plane wave of single walled nanotubes restored from a focal series of images is used in a stepwise characterization procedure utilizing both the phase of the real space restoration and its Fourier transform. A combination of these complementary characterization steps yields an accurate measurement of the chiral vector for an individual nanotube.

Surface structure study of biological calcium phosphate apatite crystals from human tooth enamel.

A surface-structure study of human tooth enamel crystals has been carried out by high-resolution electron microscopy (HREM). The surfaces of several crystals have been examined and the surfaces of a single crystal are described here. The observations made on this crystal are similar to the observations made on the other crystals, although the difference of morphology in the crystals observed indicates that the growth control by matrix proteins takes only place on the [011 macro 0] surfaces of the crystals. The crystal described here is oriented along the [112 macro 0] direction, and the following surfaces have been analyzed: (11 macro 00), (011 macro 1), (01 macro 11) and (0001). A subsurface reconstruction is observed just below the surface of the crystal not bonded to the matrix and lying above the supporting film. Observation of the matrix surrounding the crystal shows the existence of poorly crystalline phases with a structure close to that of hydroxyapatite (OHAP). Finally, a comparison of the images of the (011 macro 0) surface with computer-simulated images calculated for several models of the OHAP surface structure shows that the surface itself is stoichiometric and contains both calcium and phosphate groups. The knowledge of the binding sites of proteins on biomineral crystals such as the ones found in human tooth enamel is of prime importance for the understanding of their growth process. In this study, the first structure determination of the surface of human tooth enamel crystals is presented.

Solid-gas reactions of complex oxides inside an environmental high-resolution transmission electron microscope.

In a gas reaction cell (GRC), installed in a high-resolution transmission electron microscope (HRTEM) (JEOL 4000EX), samples can be manipulated in an ambient atmosphere (p<50mbar). This experimental setup permits not only the observation of solid-gas reactions in situ at close to the atomic level but also the induction of structural modifications under the influence of a plasma, generated by the ionization of gas particles by an intense electron beam. Solid state reactions of non-stoichiometric niobium oxides and niobium tungsten oxides with different gases (O2, H2 and He) have been carried out inside this controlled environment transmission electron microscope (CETEM), and this has led to reaction products with novel structures which are not accessible by conventional solid state synthesis methods. Monoclinic and orthorhombic Nb(12)O(29) crystallize in block structures comprising [3x4] blocks. The oxidation of the monoclinic phase occurs via a three step mechanism: firstly, a lamellar defect of composition Nb(11)O(27) is formed. Empty rectangular channels in this defect provide the diffusion paths in the subsequent oxidation. In the second step, microdomains of the Nb(22)O(54) phase are generated as an intermediate state of the oxidation process. The structure of the final product Nb(10)O(25), which consists of [3x3] blocks and tetrahedral coordinated sites, is isostructural to PNb(9)O(25). Microdomains of this apparently metastable phase appear as a product of the Nb(22)O(54) oxidation. The oxidation reaction of Nb(12)O(29) was found to be a reversible process: the reduction of the oxidation product with H(2) results in the formation of the starting Nb(12)O(29) structure. On the other hand, the block structure of Nb(12)O(29) has been destroyed by a direct treatment of the sample with H(2) while NbO in a cubic rock salt structure is produced. This in situ technique has also been applied to niobium tungsten oxides which constitute the solid solution series Nb(8-n)W9(+n)O47 with 0< or =n< or =4. All of these phases crystallize in the threefold tetragonal tungsten bronze (TTB) superstructure of Nb(8)W(9)O(47) (n=0). In the main reaction, these phases decompose in a gas plasma (O2, H2 or He, p=20mbar) into WO(3-x), which evaporates and solidifies again near the irradiated crystallite, and (Nb,W)(24)O(64), which crystallizes in a 2a superstructure of the TTB type observed here for the first time in the system Nb-W-O. Nb(8)W(9)O(47), Nb(7)W(10)O(47) and Nb(6)W(11)O(47) always react in this way, independent of the applied gas. On the other hand, the treatment of Nb(5)W(12)O(47) (n=3) and Nb(4)W(13)O(47) (n=4) in an oxygen atmosphere often caused a different reaction: these phases have been oxidized and a heavily disordered bronze-type structure has been formed. The oxygen excess in these products is largely accommodated in segregated domains of WO(3).

Captopril lowers coronary venous free radical concentration after direct current cardiac shocks.

STUDY OBJECTIVES: Direct current (DC) shocks to the heart cause morphologic and functional myocardial damage. Previous studies have suggested that acute DC shock injury is free radical mediated and that the administration of antioxidant enzymes superoxide dismutase and catalase can reduce the level of DC shock-induced free radicals. Angiotensin-converting enzyme (ACE) inhibitors are clinically used drugs that may scavenge free radicals or reduce free radical generation. The objective of our study was to determine whether the ACE inhibitor captopril lowers free radicals after DC shocks. DESIGN: In six open-chest dogs, we administered 100-J DC shocks to the epicardium, before and after administration of captopril, 3 mg/kg. We used electron paramagnetic resonance measurements of arterial and coronary venous ascorbate free radical (AFR) as a real-time marker of free radical generation (total oxidative flux). MEASUREMENTS AND RESULTS: Captopril resulted in a significant lowering of coronary venous AFR concentration: the peak rise in AFR after 100-J shocks was 17.3+/-3.4% (mean +/- SEM before captopril vs. 3.2+/-4.0% after captopril; p<0.05). CONCLUSIONS: Captopril lowers coronary venous AFR concentration after high-energy epicardial shocks.

Spontaneous gingival bleeding in an otherwise asymptomatic patient.

This case is presented to challenge the reader to formulate a differential diagnosis for a patient who visits the dentist with spontaneous, continuous gingival bleeding. When this situation occurs, it is serious and requires immediate attention and a specific treatment plan to arrive at the underlying diagnosis and control the bleeding. The signs and symptoms of a patient with gingival bleeding are presented for diagnosis; the history and management are detailed, and may be useful in diagnosing and treating similar patients.

Incidence of deep venous thrombosis and pulmonary embolism in otolaryngology-head and neck surgery.

OBJECTIVE: Postoperative deep venous thrombosis and pulmonary embolus are major causes of morbidity and mortality in patients undergoing surgical procedures. In contrast to other surgical fields, the incidence of these life-threatening conditions has not been studied in our specialty. The purposes of this study were to elucidate the incidence of deep venous thrombosis and pulmonary embolus in patients after otolaryngologic operations and to identify specific risk factors that may contribute to the development of these conditions. METHODS: A retrospective analysis was done of 12,805 total operations on adults done by the Department of Otolaryngology at our institution from January 1987 to December 1994 to determine the number of patients in whom postoperative deep venous thrombosis and pulmonary embolus developed. Patients in whom a postoperative thromboembolic event developed after an otolaryngologic surgical procedure were identified by the medical records department with use of an abstracting database. This search cross-referenced disease-specific codes for otolaryngologic procedures with the codes for deep venous thrombosis and pulmonary embolus to identify the 34 patients in this report. Results (rounded to the nearest decimal point) were then categorized according to the different subspecialties within otolaryngology, and appropriate statistical analysis tests were performed on the resulting data. RESULTS: Thirty-four patients with postoperative deep vein thrombosis were identified during the study period, for an overall incidence of 0.3%. Of these 34 patients, 24 also had a pulmonary embolus for an overall incidence of 0.2%. The incidence of deep venous thrombosis (and pulmonary embolus) in the subspecialties was as follows: head and neck surgery, 0.6% (0.4%); otology/neurotology, 0.3% (0.2%); head and neck trauma and plastic surgery, 0.1% (0.1%); and general otolaryngology, 0.1% (0.04%). Only the patient's age and the presence or absence of pneumatic compression devices were identified as independent risk factors for the development of a thromboembolic event. CONCLUSIONS: Postoperative pulmonary embolus is a rare occurrence in the field of otolaryngology-head and neck surgery. When it does occur, it causes significant morbidity and increases the cost of care for that patient. We discuss our approach to categorizing patients into low-, intermediate-, and high-risk groups, as well as prophylaxis against pulmonary embolus.

HREM study of irradiation damage in human dental enamel crystals.

Several phenomena have been observed during the examination of human dental enamel crystals (mainly constituted by hydroxyapatite (OHAP] by high-resolution electron microscopy (HREM) at 300 and 400 keV: orientation-dependent damage in the form of mass loss from voids or uniform destruction of crystal structure, beam-induced diffusion creating outgrowths at the crystal surfaces, recrystallization of the bulk crystal and crystallization of the inorganic components of the matrix surrounding the crystals. These beam-induced crystals have the CaO structure. The phenomena observed are most likely due to various electron-crystal interaction mechanisms (ballistic knock-on damage, electronic excitations, temperature rise, etc.). In this paper, the contribution of the ballistic process to the phenomena observed is discussed. The quantitative description of the knock-on collisions rests on the McKinley-Feshbach cross-section formula. The minimum ion displacement energies which appear in this expression have been estimated on the basis of the electrostatic ion binding energies, and the covalent bond energies if required. It is shown that hydroxyl, calcium and oxygen ions can effectively be displaced by the incident 300 and 400 keV electrons. Thus, the formation of CaO crystals by the combination of calcium and oxygen ions diffusing from their initial sites inside the OHAP lattice can tentatively be explained.

Compositional variations in apatites with respect to preferential ionic extraction.

Detection of ionic losses from the apatitic structure (Ca10(PO4)6(OH)2) by high-resolution electron microscopy was investigated theoretically. Linear image analysis showed the need for an objective aperture of at least 3.7 nm-1 to visualize four different coordinates (CaII, OH-, P and mid-point between CaII-P bond). High-resolution image analysis and plotting of OH- column intensity against specimen thickness showed an inverse proportionality between composition and OH- image intensities for very thin specimens (less than 2 nm). Image intensity variation would be detectable experimentally, but the preparation of such thin specimens by ultramicrotomy is impossible.
