Hemostasis in experimental pulmonary injury.

Fibrin Seal (consisting of fibrinogen, cold insoluble globulin, factor XIII, antiplasmin, platelet growth factor, thrombin, and calcium chloride), cryoprecipitate, at Avitene were applied to areas of pulmonary wedge resections in dogs and cynomologus monkeys in an attempt to stop bleeding and air leakage. In the control groups, hemorrhage persisted. Comparing the three hemostatic agents used. Avitene was the least effective followed by cryoprecipitate. Fibrin Seal was most effective. In all cases, it eliminated both air leakage and bleeding.

Studies on osteoporosis VII. Effect of 17 beta-hydroxy-4-estren-3-one 17-decanoate on experimental osteoporosis.

Total body neutron activation analysis was employed to measure total calcium in C3H/St (Ha) female mice. Ninety-nine percent of total body calcium is in bone and loss of calcium was used as an index of osteoporosis. Heparin (500 U/kg B.I.D.) treatment for three months resulted in significant osteoporosis. 17 beta-Hydroxy-4-estren-3-one 17-decanoate 1.5 mg/kg or 4 mg/kg twice monthly prevented this heparin accelerated osteoporosis. The results suggest that 17 beta-hydroxy-4-estren-3-one 17-decanoate may be capable of preventing bone loss and partially increasing bone mass in patients exposed to osteoporosis inducing regimens.

Studies on platelet aggregation inhibitors in vivo. X. Relationship to thrombolysis.

Human labeled fibrin clots were inserted into femoral veins of stumptailed monkeys (Macaca arctoides). Thrombolysis was slightly increased by treatment with the platelet aggregation inhibitor pentoxifylline. This agent significantly potentiated the thrombolytic effect of urokinase activated human plasminogen. Pentoxifylline was also found to release plasminogen activator activity into the circulation.

Study of platelet aggregation in vivo. IX. Effect of nafazatrom on in vivo platelet aggregation and spontaneous tumor metastasis.

Nafazatrom (Bay g 6575) was explored for its ability to inhibit platelet aggregation. In vitro, it had no effect on ADP, serotonin, epinephrine, or collagen induced platelet aggregation in platelet rich plasma of monkeys. On the other hand, in vivo it was a powerful inhibitor of ADP induced platelet aggregation as measured by the in vivo platelet aggregation recording instrument described previously (Ambrus et al., 1976). This effect was potentiated by dipyridamole. On the other hand, following parenteral administration of Bay g 6575, no ex vivo inhibition was noticed of ADP, serotonin, epinephrine, and collagen induced platelet aggregation. The hypothesis was presented that Bay g 6575 acts by increasing prostacyclin synthesis and/or release or interferes with its decomposition. This may explain in vivo activity; rapid decomposition may explain inability to demonstrate ex vivo activity. This also explains potentiation by the phosphodiesterase inhibitor dipyridamole. Bay g 6575 also was highly effective as a platelet aggregation inhibitor in monkeys after oral administration. In mice, Bay g 6575 increased circulation time of intravenously injected polyploid Ehrlich ascites tumor cells. In Furth-Wistar rats implanted with Furth-Columbia Wilms' tumor, in A/J mice implanted with C1300 neuroblastoma and in Wistar rats implanted with SMT-2A (Kim) breast cancer, Bay g 6575 significantly reduced spontaneous pulmonary metastasis. On the other hand, no effect was seen in the metastatic rate of NIH renal adenocarcinoma in BALB/cCr mice.

Studies on osteoporoses. VIII. Effect of estrogen on steroid induced osteoporosis.

Neutron activation analysis was employed to determine total body calcium in 5-8 month and 15-18 month old C3H/St(Ha) and C57/BL6 (JACOBS) female mice. Ninety-nine percent of calcium is in bone and loss of calcium was used as an index of osteoporosis. Prednisolone (50 mg/kg q.d.) treatment for three months resulted in significant osteoporosis in the C3H/St(Ha) mice of both age groups but did not cause significant bone loss in the C57/BL6 (JACOBS) mice of either age group. Premarin (20 mg/kg q.d.) failed to prevent steroid accelerated osteoporosis.

Diminished red blood cell deformability in uncomplicated human malaria. A preliminary report.

Six patients with active P. falciparum malaria (two resistant to chloroquine therapy), one with treated and apparently cured P. falciparum malaria, one with active P. vivax malaria and two controls from the same geographic area of Amazonian Brazil were studied. All malaria patients had impaired red cell deformability. This was not correlated with drug resistance or number of parasitized cells. The hypothesis is presented that the presence of parasites in red blood cells and related metabolic effects produce decreased red cell deformability and may lead to microvascular perfusion deficiencies including cerebral malaria.

The use of FEIBA (factor eight inhibitor bypassing activity) in cataract extraction in hemophilia A patient with inhibitor.

A 34-year-old patient, with severe factor VIII deficiency (anti-hemophilic factor deficiency) and inhibitor against factor VIII, underwent a phacoemulsification with posterior capsulatomy and thermal iridectomy under general anesthesia. FEIBA (Factor Eight Inhibitor Bypassing Activity) was used to achieve hemostasis. The surgical procedure was successful and no complication was ensued. A brief management of hemophilia A patient requiring surgery is discussed.

Studies on osteoporoses. XII. Effect of imidazoquinazolinones on experimental osteoporosis. A preliminary report.

In vivo neutron activation analysis was employed to determine total body calcium content in C3H/St(Ha) female mice. Ninety-nine percent of calcium is in bone and loss of calcium was used as an index of osteoporosis. Heparin (500 U/kg b.i.d.) treatment for three months resulted in significant osteoporosis. BL3459 and BL4160 (6 mg/kg i.m. q.d.) prevented this heparin induced osteoporosis. BL4160 appeared to be more effective than BL3459 in this regard but the difference was not statistically significant.

Studies on osteoporosis VI. Effect of human and salmon calcitonin on experimental osteoporosis.

Neutron activation analysis was employed to determine total body calcium in C3H/St (Ha) female mice. Ninty-nine percent of total body calcium is in bone and loss of calcium was used as an index of osteoporosis. Heparin (500 U/kg B.I.D.) treatment for three months resulted in significant osteoporosis. Both human and salmon calcitonin (4 MRC U/Kg B.I.D.) prevented heparin induced osteoporosis. The results suggest that calcitonin may prevent pathological fractures in patients on anticoagulants and other osteoporosis inducing regimens.

Platelet cancer cell interaction in metastasis formation. Platelet aggregation inhibitors: a possible approach to metastasis prevention.

Abnormal platelet aggregation on circulating and lodged cancer cells may play an important role in the early stages of metastasis formation. The immediate drop in the number of circulating platelets following intravenous injection of Walker-256 carcinosarcoma cells in rats represents the experimental counterpart of the morphologic finding of tumor cells associated with tumor clusters in the pulmonary arterioles and capillaries.

Depletion of phenylalanine in the blood of phenylketonuric patients using a PAL-enzyme reactor. An in vitro study.

Blood samples obtained from eight adult phenylketonuric individuals had a mean phenylalanine level of 25 mg/dl. When these samples were circulated through multitubular enzyme-reactors prepared with immobilized phenylalanine ammonia lyase an average of 77% of phenylalanine was metabolized within 30 minutes. We conclude that phenylalanine in human phenylketonuric blood is just as susceptible to metabolism by PAL-enzyme reactors as phenylalanine that is added to normal blood, or that is circulating in dogs and monkeys made hyperphenylalaninemic by experimental means.

Cyclic nucleotide phosphodiesterases in normal and malignant human tissues.

Biopsy samples were obtained from tumor tissue and surrounding normal tissue from the same organs of the same patients in 25 patients. Adequate studies could be performed on all parameters in 18 tissue pairs: 12 malignant melanomas, 4 sarcomas, and 2 others. Significantly higher levels of cAMP low affinity phosphodiesterases were found in the tumors as compared to the controls. Similar differences were seen in the high affinity cAMP phosphodiesterases, but these were of borderline significance. High affinity cGMP phosphodiesterase levels did not differ significantly between normal and neoplastic tissue samples. Low affinity cGMP phosphodiesterases were not detectable in either tissues. These studies together with earlier experimental findings suggest possible auxiliary therapeutic trials with phosphodiesterase inhibitors.
