Evaluation of acid-base disorders in dairy cows using principal component analysis and empiric equations.

Acid-base disorders were studied in two groups of dairy cows. According to conventional interpretation in the 1st group of dairy cows (n = 10) the results of the acid-base parameters indicated respiratory alkalosis and in the 2nd group (n = 10) metabolic acidosis. The data of the two groups were examined using principal component analysis. So called "reduced variables"--principal components for each animal were calculated. Each experimental animal was projected in the coordinates of 3 principal components K1, K2 and K3. The components K1, K2 and K3 comprised a 90% data variability. The importance of variables for a dividing of groups (1 and 2) was quantified. The most important were pCO2, (HCO3)' and BE. It was proved according to mathematico-empiric equations that dairy cows of 1st group suffered from acute respiratory alkalosis. In the 2nd group of dairy cows the combined acid-base disorder was confirmed using the equations for metabolic acidosis and chronic respiratory acidosis. The use of PCA method and empiric equations for acid-base imbalances shows the possibilities for gaining new knowledge how to make the diagnostic process more exact.

Paraoxon-induced decrease in the muscarinic acetylcholine receptor content in rat heart.

The regulation of the rat heart muscarinic acetylcholine receptor by injection of paraoxon (an acetylcholinesterase inhibitor) was characterized. No changes in receptor density in the four cardiac chambers were seen after two days. After three daily injections, however, significant decreases (20-25%) were observed in both atria and ventricles. These changes were associated with a 50% decrease in cardiac acetylcholinesterase activity. We were unable to demonstrate a change in the proportion or value of the apparent KDs for the two previously described affinity states of the receptor for the agonist methacholine. We found that the proportion of the high affinity form of the receptor was greater in atria than in ventricle (70 vs. 55%). Moreover, we also found that acetylcholinesterase activity was greater in the atria than ventricle. Although methacholine will produce down regulation of the muscarinic receptor in the perfused rat heart in 2.5 h (Reinhardt and Roskoski, 1983; J. Pharmacol. Exp. Ther. 226, 135), injection of large doses of methacholine failed to produce any receptor decreases within 6 h in vivo.

[Genetic markers in the blood and their relation to metabolic parameters in dairy cows].

A relationship between the genetic markers of blood (blood groups, serum polymorphic proteins) and the clinico-chemical parameters was studied in the dairy cows of the Slovak Pied breed. Antigens belonging to systems A, B, C, F, S, R, T, Z and the polymorphic traits genetically controlled from loci Tf, Cp, Am and Hb were identified in all the animals subjected to testing. The values of the parameters of acid-base balance and concentration of 13 metabolic components were repeatedly determined in the experimental period. The results of the F-test indicated that there were no significant differences in the values of any of the tested parameters between the phenotypes of the A, J, Am, Tf and Cp systems. Of the 21 parameters tested, statistically significant differences were found in 11 parameters between some alleles of the C, FV, T, Z and Hb systems.

[Genetically conditioned variability of metabolic profile parameters in dairy cows].

Individual heritability and differences in the concentration of the chemical components of the blood were studied in the dairy cows of the Slovak Spotted breed. The experiment was performed with 166 cows. The set comprised six groups of half-sisters from three stocks. The differences among the cows were statistically significant (alpha = 0.01) in the majority of the parameters studied: haematocrit, haemoglobin, pH, PO2, oxygen saturation of the blood; plasma potassium, phosphorus, magnesium, calcium, total protein, urea, glucose, alkaline phosphatase, and esterified fatty acids. The coefficients of repeatibility for the mentioned parameters ranged from 0.19 to 0.75. The heritability coefficients were calculated for the parameters in which the inter-group differences were significant: total protein (0.62), magnesium (0.57), potassium (0.51), urea (0.49), glucose (0.45), phosphorus (0.43), calcium (0.39), haematocrit (0.37), haemoglobin (0.35), pO2 (0.29). The results suggest that some of the parameters under study are under certain genetic control.
