Affinity changes of rat striatal dopamine receptors in vivo after acute bupropion treatment.

The effect of bupropion (Wellbutrin) was studied on in vivo [3H]spiperone binding in rat striatum and cerebellum. The compound increased binding dose dependently in striatum whereas no effect was observed in cerebellum. Saturation analyses of in vivo binding in the striatum revealed an increased affinity of the receptors rather than changes in the number of binding sites. These results are the first demonstration of an increased sensitivity of central dopamine receptors after acute bupropion treatment.

[Dye concentration measurements for the determination of leakage during and following normothermic cytostatic perfusion in the treatment of tumors of the extremities].

The regional normothermic cytostatic perfusion with 3000 mg Methotrexate and 1.5 mg Vincristine under rescue conditions (leucovorine) during 120 minutes was carried out in patients with malignant melanoma stage II by Sylven or other tumors of the lower extremities in the surgical university hospital Halle since 1980. Dye concentration measurements with indigo carmine for monitoring the isolation between systemic circulation and perfusion circulation were carried out during and after cytostatic perfusion. The measurements took place with a Spekol 10 of the VEB Carl Zeiss, Jena, by wave-length of 605 nm. The leakage streams of 30 perfusions amounted from 0 to 46% during the phase - regional perfusion. The dye quantity which was secreted by the kidneys during 16 hours post perfusion despite plentiful lavage of the extremity (MTX quantity) amounted from 2 to 7%.

Dopamine modulates cholecystokinin release in neostriatum.

Peptides of the cholecystokinin family, but mainly the sulphated octapeptide (CCK8), have been found in brain extracts of several species. High amounts are present in axons and nerve endings in the rat neostriatum (caudate-putamen) and a role for cholecystokinin as a neurotransmitter in this functionally important area is possible. We have incubated slices of rat caudate-putamen (CP) to study the release of cholecystokinin-immunoreactivity (CCK-IR) in vitro. The release of CCK-IR was induced by veratridine. It was dependent on the presence of Ca2+ in the incubation medium and was blocked by tetrodotoxin. We now present evidence that dopaminergic agonists added to the slices modulate the veratridine-induced release via different groups of receptors. Receptors which mediate an enhancement of the release of CCK-IR seem to be located on afferent axons and nerve endings and are possibly of the D-2 subtype. Receptors which mediate an attenuation of the release are probably situated on cells intrinsic to the CP. These receptors seem to be coupled to adenylate cyclase and might thus be of the D-1 subtype. There is also evidence that endogenous dopamine when released enhances the secretion of CCK-IR.

In vivo [3H]spiperone binding to the rat hippocampal formation: involvement of dopamine receptors.

The existence of DA receptors in the rat hippocampus was demonstrated with an in vivo [3H]spiperone radioreceptor assay. Kinetic studies revealed that maximum binding of [3H]spiperone in hippocampus was much smaller than in striatum and frontal cortex but much higher than in cerebellum. In inhibition studies of [3H]spiperone binding, all neuroleptics tested were active in hippocampus as well as in striatum. In contrast, 5HT antagonists were definitely less potent in these two brain regions than in frontal cortex. Finally, even when 5HT receptors were blocked, dipropyl-ATN and haloperidol remained fully effective in hippocampus, striatum, but also in frontal cortex although to a lesser degree. From these results it was concluded that [3H]spiperone binds mainly to DA receptors in hippocampus as well as in striatum, whereas both 5HT and DA receptors are present in frontal cortex.

[Malignant melanomas of fingers and toes (author's transl)].

From 1968 to 1978 more than 500 patients suffering from histologically verified malignant melanoma were seen. Only in 12 cases (appr. 2%) the tumour was situated on the tip of the fingers and toes. In such cases nowadays amputation and exarticulation of the whole extremity is no longer practiced. It is sufficient only to remove the corresponding finger or toe. Additional chemotherapy should supplement surgery.

[Melanocytoblastoma in urology].

On the basis of special literature presence and frequency of autochthonic melanomas in the uro-region are described. It is referred to 1 own case. The sequels of metastasizing are given in the chapter "secondary localisation" with the comprehensive reason for the decision, again to remove again an already functioning renal graft, when it becomes known, that a metastasising melanocytoma was present in the donor of the organ.

[Endometriosis of the small intestine].

The isolated endometriosis is a rare sickness whose existence is little thought of although it shows relative specific symptoms. With the aid of literature -- survey and according to personal experience on two patients suffering from isolated endometriosis of ileumregion, were clinical treatments carried out. Surgical and hormonal treatments as well as combination of both, have special indications for this kind of sickness.

Isolation and characterization of temperature-sensitive mutants of Escherichia coli with altered ribosomal proteins.

The ribosomal proteins of temperature-sensitive mutants of Escherichia coli isolated independently after mutagenesis with nitrosoguanidine were analyzed by two-dimensional gel electrophoresis. Out of 400 mutants analyzed, 60 mutants (15%) showed alterations in a total of 22 different ribosomal proteins. The proteins altered in these mutants are S2, S4, S6, S7, S8, S10, S15, S16, S18, L1, L3, L6, L10, L11, L14, L15, L17, L18, L19, L22, L23 and L24. A large number of them (25 mutants) have mutations in protein S4 of the small subunit, while four mutants showed alterations in protein L6 of the large subunit. The importance of these mutants for structural and functional analyses of ribosomes is discussed.
