Successful desensitization in case of solar urticaria with sensitivity to UVA and positive passive transfer test.

A severe case of solar urticaria induced by UVA with positive results of passive transfer test (3 recipients) and reverse passive transfer test (2 recipients) is reported. Desensitization treatment by irradiation of particular parts of the body with UVA and UVB (to accelerate tanning) was fully effective. A marked increase of the tolerance to UV remained during the period of over five years of follow-up.

Allergy to fish in fish meal factory workers.

Fifty-one fish meal factory workers in four Polish harbour cities were investigated. They had been employed in the fish meal factory for 1--33 years and 10.7 years on average. All but seven of these were smokers. The investigation consisted of respiratory symptoms questionnaire, chest X-ray, physical examination, respiratory function assessment, intradermal skin tests to ten fish genera, total serum IgE and serum precipitins to fish antigen assessment. Chronic bronchitis was diagnosed in 29 investigated patients. In 12 (23%) of the patients, according to their case history or to skin tests, allergy to fish was suspected, but was confirmed by elevated serum IgE levels in only two of these. In no cases were serum precipitins against fish antigen found. In six patients, either obstructive or restrictive changes were diagnosed according to conventional spirometry, but by MEF50% VC in 17 subjects (33%) slight airways obstruction was diagnosed and can be regarded as the result of the habit of smoking. According to the performed investigation, only two workers presented a full clinical and immunological picture of allergy to fish. From the practical point of view, this type of professional activity is less dangerous than might be theoretically expected.

Both strands of polyoma DNA are replicated discontinuously with ribonucleotide primers in vivo.

Nascent polyoma DNA molecules were isolated after pulse-labeling of infected murine 3T6 cells with [3H]thymidine. The extent of digestion of these DNA molecules by spleen exonuclease was increased by exposure to alkali or RNase, suggesting that ribonucleotides were present at or near the 5' terminal of the newly synthesized pieces of DNA. Intermediates shorter than 300 nucleotides were hybridized to the separated strands of restriction enzyme fragments of the polyoma genome: 2.5 to 3-fold more radioactivity was found in the strand whose synthesis is necessarily discontinuous (the lagging strand) than in the strand whose synthesis is potentially continuous (the leading strand) than in the strand whose synthesis is potentially continuous (the leading strand). Separation of the strands of [5'-32P]DNA molecules showed that the excess [3H]thymidine in lagging-strand molecules was not simply the result of an increased number of molecules. Therefore, assuming equivalent efficiencies of labeling, lagging-strand pieces must be slightly longer than those with leading-strand polarity. The presence of ribonucleotides on the 5' termini of molecules with both leading- and lagging-strand polarity was demonstrated by (i) release of 32P-ribonucleoside diphosphates upon alkaline hydrolysis of [5'-32P]DNA separated according to replication polarity and (ii) the change in the degree of self-annealing of nascent molecules upon preferential degradation of DNA molecules possessing initiator RNA moieties by spleen exonuclease. We conclude that replication of polyoma DNA in vivo occurs discontinuously on both sides of the growing fork, using RNA as the major priming mechanism.

[Experimental studies on cardiopulmonary relation under hypoxia, hypercapnia and airway obstruction in dog (author's transl)].

On 18 dogs the effects of hypoxia (12, 10, 7.5% O2) and hypoxia and hypercapnia (10% O2 + 5, 7.5, 11% CO2) on breathing, pulmonary vascular resistance, cardiac output and on the arterial and venous oxygen pressure were investigated. The relationship between arterial oxygen pressure to Pap as to vascular resistance is not in the mean linear. There are good, bad and nearly nonresponders in respect to the vascular resistance on the dogs. Therefore, calculation and evaluation of mean values are less helpful. A similar variation on men could help to understand also better the large scatter which is always shown on correlations between PO2a : Pap or PO2a : vascular resistance. Also the different pattern of the V/Q relationship may be influenced by the amount of the vascular response to local alveolar hypoxia. An increase of PCO/a potentiates the local vasoconstrictor effect of hypoxia. Mainly this was seen in the range between hypocapnia and moderate rates of hypercapnia. Hypoxia shows an increase of breathing frequency, hypercapnia shows and increase in tidal volume. Also under hypercapnia there are nonresponders and better responders in response to the vascular resistance in the lung circulation. Mechanical airway obstruction is followed by an increase of tidal volume; obstruction caused by acetylcholine is followed by a decrease of tidal volume and an increase of breathing frequency. Changes in the arterial blood gases are caused by changes in the ventilation perfusion relationship only under acetylcholine obstruction. Influences on the lung circulation by the airway obstruction under these experimental conditions as direct effects are small or not existent.

Ribonucleotides in DNA newly synthesised in 3T6 cells in vivo.

Within the field of DNA replication, considerable interest has focused in recent years on the mechanism of initiation of synthesis of DNA molecules. In vitro replication systems from Escherichia coli have been instrumental in uncovering a priming function fo9r ribonucleotides on the earliest intermediates of DNA polymerisation in vitro and in identifying the proteins involved. In vitro replication systems from mammalian cells that permit the use of the phosphate-transfer method for detection of RNA-DNA junctions as well as direct labelling of the RNA moiety of the molecules have suggested a similar role for ribonucleotides in DNA synthesis in eukaryotes. However, the existence of this mechanism in mammalian cells in vivo has not been established. Here we report the first evidence that a significant proportion of the earliest intermediates in mammalian DNA polymerisation in vivo do, in fact, possess ribonucleotides, presumably because their synthesis was initiated with one or more ribonucleotides.

[Clinical-functional emphysema diagnostics and immunological correlations (author's transl)].

By application of the correlation flow resistance/intrathoracic gas volume the lung emphysema is differentiated in quality within the scope of chronic obstructive lung diseases. For these groups immunoglobulins, the alpha1-trypsin activity in serum and the activity of further protease inhibitors were determined. There are no substantial variables in the different groups with regard to immunoglobulins. In the emphysema group alpha1-antitrypsin was a little reduced. This group included 2 patients with an alpha1-antitrypsin deficit. The two-dimensional separation of the inter-alpha-trypsin big inhibitor shows 3 areas which decrease significantly in case of severe emphysemata. The irritation of the inter-alpha-trypsin big inhibitor, i.e. in its turnover, parallel to the increasing degree of emphysema formation, is discussed.

Most short DNA molecules isolated from 3T3 cells are not nascent.

The population of short DNA molecules (less than 10(3) nucleotides) in 3T3 cells has been studied using in vivo and in vitro pulse labeling techniques and in vitro end-labeling. There is a large number of molecules of less than 100 nucleotides present in equal numbers in both Go and S phase cells. In S phase cells, most of these molecules are not replicating intermediates because they do not become density-labeled after a moderate period of substitution of BrdUMP, although they are detected by end-labeling in vitro. This population includes the nascent Okazaki pieces that can be labeled in a short pulse with [3H]dThd or [3H]dTTP, however, these represent less than 10% of the total population. Alkaline hydrolysis of the molecules that had been end-labeled with 32P using [gamma32P]ATP and polynucleotide kinase did not reveal significant release of [32P] 2'(3'), 5' ribonucleoside diphosphates.

Fatigue of airway obstruction during long-term exposure to allergen aerosol.

The response to prolonged antigen exposure and the potentiation of airway resistance increase to ACH challenge, after this exposure, were studied on sixteen boxer dogs. One group of animals presented fatigue to A.E. after 3 hours of exposure. This group developed an increased response to ACH aerosol after fatigue to antigen was present. In a second group of dogs, absence of fatigue during prolonged exposure to allergen was observed. A growing tendency of Edyn (as an index of airway resistance) was observed after 5 hours of exposure. The therapeutical influence of bilateral vagus blockade was tested in these last animals. Blockade of nervus vagus released airway obstruction during prolonged allergen exposure and no bronchoconstriction was observed after ACH challenge during blockade.
