Purification and characterization of a GTP-binding protein with a molecular weight of 20,000 in bovine brain membranes. Identification as the rho gene product.

We have determined that there are at least six GTP-binding proteins (G proteins) with Mr values between 20,000 and 25,000 in the crude membrane fraction of bovine brain and have purified one of them with a Mr of about 24,000 (24K G) to near homogeneity (Kikuchi, A., Yamashita, T., Kawata, M., Yamamoto, K., Ikeda, K., Tanimoto, T., and Takai, Y. (1988) J. Biol. Chem. 263, 2897-2904). In this study, we have purified another G protein with a Mr of about 20,000 (20K G) to near homogeneity and have characterized it. 20K G bound maximally about 1.0 mol of [35S]guanosine 5'-(3-O-thio)triphosphate (GTP gamma S)/mol of protein, with a Kd value of about 50 nM. [35S]GTP gamma S binding to 20K G was inhibited by GTP and GDP, but not by other nucleotides such as ATP, UTP, and CTP; it was also inhibited by pretreatment with N-ethylmaleimide. 20K G hydrolyzed GTP to liberate Pi, with a turnover number of about 0.01 min-1, and was not copurified with the beta gamma subunits of the regulatory G proteins of adenylate cyclase. 20K G was not recognized by the antibody against the ADP-ribosylation factor for the stimulatory regulatory G protein of adenylate cyclase. Peptide map analysis showed that 20K G was not a proteolytic product of 24K G. The partial amino acid sequence of 20K G was almost identical with that deduced from the rho gene. The amino acid composition of 20K G was similar to that of the rho gene product. These results suggest that 20K G is the rho gene product and that this G protein is present in bovine brain membranes.

Increased 1,2-diacylglycerol content in myopathic hamster hearts at a prenecrotic stage.

1,2-Diacylglycerol (DAG) has been suggested to be a secondary messenger. In this study, we determined the amount of 1,2-DAG in heart tissue from Syrian hamsters with hereditary cardiomyopathy at 30 days (prenecrotic stage) and 90 days of age by thin-layer chromatography with flame ionization detection (TLC-FID). Myocardial triglyceride contents were higher at 30 days of age and lower at 90 days of age compared to the levels in age-matched normal hamsters. Decreases in major species of phospholipids in hearts were observed only at 90 days of age. However, elevated 1,2-DAG content in myopathic hearts was found at 30 days of age, whereas there was no difference between the two groups at 90 days of age. It is suggested that the increase in 1,2-DAG at the prenecrotic stage is involved in the pathogenesis of the cardiomyopathy.

The carboxyl third of tau is tightly bound to paired helical filaments.

To obtain definitive evidence that tau is a component of paired helical filaments (PHF) in Alzheimer's disease, we fractionated and sequenced PHF-derived peptides according to a previously described procedure. In the PHF digest, we found four independent tau peptides that were located in the carboxyl third of tau. Subsequent extensive analysis of the PHF digest did not provide any other tau peptides. The conventional PHF antiserum and a new antiserum directed toward formic acid-denatured PHF reacted with the distinct CNBr fragments of tau localized on the carboxy-terminal portion of tau by protein sequencing. From these observations, we conclude that the carboxyl third of tau is tightly bound to PHF.

[Carcinoma of Papilla Vater with celiac axis stenosis--application of percutaneous transluminal angioplasty].

In a case of carcinoma of Papilla of Vater with angiographically proved celiac axis stenosis, PTA (Percutaneous transluminal angioplasty) was performed to restore sufficient blood flow, followed by standard pancreaticoduodenectomy. This would be the first case to have PTA applied to the cancer-bearing celiac axis stenosis. The patient was 58 year old female, presented with acute abdominal pain and vomiting. DIC revealing protruding defect in the choledochus and hypotonic duodenography showed elevated tumor at Papilla of Vater. Aortography revealed 75% circumferential stenosis at the celiac artery and superior mesenteric angiography visualized all the branches of the celiac axis via dilated pancreaticoduodenal arcades. PTA with Grüntzig Dilaca (4.3Fr), 5 atm for 60 seconds serially every two min for 4 times was performed with the immediate relief of the stenosis down to 25%. Intraoperatively 70-80ml/sec flow confirmed, pancreaticoduodenectomy was done without sacrificing curability. PTA would be superior strategy to restore sufficient blood flow in cases with GI anastomoses, compared with other bypass or reimplantation surgery.

Purification of atrial natriuretic peptide receptor from bovine lung. Evidence for a disulfide-linked subunit structure.

The receptor for atrial natriuretic peptide (ANP) was purified to apparent homogeneity from bovine lung by a combination of detergent extraction, ammonium sulfate precipitation, and affinity chromatography on ANP-Affi-Gel 10. The Mr of the purified receptor is about 140,000 as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. After reduction, the protein migrated as a single band with an Mr near 70,000. NH2-terminal sequence analysis of the purified material revealed only one sequence, indicating that the ANP receptor is composed of two probably identical subunits held together by disulfide bond(s), although it remains possible that one of the subunits is blocked at the NH2 terminus. Antibody was produced to the nonreduced Mr = 140,000 species and shown to interact with detergent-solubilized forms of the lung and kidney ANP receptor.

Ubiquitin is a component of paired helical filaments in Alzheimer's disease.

Paired helical filaments (PHF), which constitute a distinct type of pathological neuronal fiber, are the principal constituent of neurofibrillary tangles that occur in the brain of patients with Alzheimer's disease. Their insolubility in sodium dodecyl sulfate and urea has prevented the analysis of their subunit composition by gel electrophoresis. A monoclonal antibody (DF2) was isolated that specifically labeled PHF at both the light and electron microscopic levels. It labeled a small polypeptide (5 kilodaltons) that was shown to be ubiquitin in immunoblots of the soluble fraction of brain homogenates. To obtain direct evidence that ubiquitin is a component of PHF, PHF were treated with concentrated formic acid and digested with lysylendopeptidase; ubiquitin-derived peptides were then identified by reversed-phase high-performance liquid chromatography. Two fragments in the PHF digest were identified as derived from ubiquitin by protein sequencing. This procedure should make possible definitive identification of other PHF components.
