Autoradiographic studies on the protein synthesis and the distribution of the newly synthesized proteins in oligodendroglial cells.

The protein-synthesizing activity of oligodendroglial cells in the rat hippocampus as well as the migration of the newly synthesized proteins within these cells were examined by means of light and microscopic autoradiography when injecting 3H-leucine into the lateral ventricle. The autoradiographs of oligodendroglial cells were compared with the autoradiographs on hippocampal pyramidal cells of the same area. According to the quantitative evaluation of the autoradiographs the protein-synthesizing activity of the oligodendrocytes equals or may even exceed that of the pyramidal cells. In oligodendroglial cells most of the newly synthesized proteins migrate very rapidly from the rough-surfaced endoplasmic reticulum into the cisternae of the Golgi apparatus and disappear continuously. The proteins disappear from the Golgi apparatus simultaneously with the decrease in the radioactive concentration of the perikaryon.

Lipid composition of the mouse spleen in Rauscher leukaemia.

Lipid constituents were examined in the spleen of mice 10, 14 and 18 days after infection with Rauscher leukaemia virus. (i) The total lipid content, as related to the amount of protein, decreased with the development of the tumour. (ii) Spleen cells of control and infected mice synthesized the same lipids. (iii) In the infected spleen, free fatty acids increased by 80% and triglycerides decreased by 44% as compared to the control. The percentage distribution of phospholipid components showed a change in tumour cells: lecithin and phosphatidylethanolamine increased, lysolecithin and sphingomyelin decreased. (iv) After intraperitoneal injection of [32P] orthophosphate, in infected mice the incorporation of 32P into phosphatidylcholine and phosphatidylethanolamine increased, while 32P incorporation into lysophosphatidylcholine + sphingomyelin and phosphatidylinositol + phosphatidylserine fractions decreased.

[Methicillin levels in the human serum, pericardial effusion and myocardium].

The antibiotic level in the serum, pericardial fluid and myocardium of 11 patients prepared for heart surgery was tested by means of the agar-diffusion method after the intramuscular administration of 1 g of methicillin. The mean level after 1 to 3 hours was 14.7 mcg/ml in serum, 1.7 mcg/ml in the pericardial fluid and 8.2 mcg/g in the myocardium. In the case of drug sensitivity, the myocardial methicillin concentration is sufficient for the treatment of Gram-positive infections. If the infection is localized in the pericardium, higher doses, local applications, drug combination or complex therapy might be called for.
