Tertiary treatment of biologically treated piggery wastewater using vibratory shear-enhanced RO membrane.

This study presents a good example for the tertiary treatment of biologically treated piggery wastewater using vibratory shear enhanced RO membrane (VSEP RO). Through a simple process combination, utilizing Bioceramic SBR(BCS) and VSEP RO, at Gimhae plant livestock wastewater is treated excellently to meet the strict effluent standards. Application of RO membrane directly to the biologically treated effluent has been successful without any pretreatment to reduce high suspended solids. The combination of VESP UF followed by RO filtration processes produced a higher recovery rate in the 3-week pilot test.

Modeling the fate and transport of organic and nitrogen species in soil aquifer treatment process.

Soil aquifer treatment (SAT) is a promising technique for wastewater reclamation and reuse. This treatment strategy takes advantage of physicochemical and biological processes in the subsurface. The model employed in this study is based on MODFLOW-SURFACT (HydroGeoLogic, Inc.), a three-dimensional model for variably saturated flow and reactive mass transport. The model accounts for reactions including the nitrification of ammonium, the denitrification of nitrate, and the oxidation of organic carbon. Concentration of dissolved oxygen and biomasses involved in aerobic and anaerobic biological reactions forms the basis for estimates of nonlinear reaction rates formulated using a multiple-Monod expression. Illustrative simulations were conducted in a two-dimensional cross-sectional domain, with unsaturated and saturated zones. They examine the effects that site and operational conditions have on the performance of a SAT system. The parameters and conditions of concern included length of the wet/dry cycle, ground surface condition, and infiltration rate. From the simulations, we found that organic carbon was effectively removed in all cases. The availability of oxygen was a key factor in predicting the production and removal of nitrate. Overall, the model successfully described the fate and transport of the key constituents during the wet/dry operational periods in both unsaturated and saturated subsurface.

Evaluation of wastewater effluents for soil aquifer treatment in South Korea.

Soil batch and column experiments were performed to characterize the wastewater effluents from seven different wastewater treatment plants in the Jonnam province, South Korea, with the purpose of evaluating the effluents for possible application of a soil aquifer treatment (SAT) in Korea. Batch experiments were conducted to measure the biodegradable dissolved organic carbon (BDOC) while 1 m soil columns, for simulating SAT, were employed to further analyze dissolved organic carbon (DOC) removal. The soils were collected from a river bottom in Jonnam. The BDOC fractions and the residual DOC concentrations for the effluents ranged from 19.3 to 59.9% and from 1.0 to 7.5 mg/L, respectively, depending on the reaction time. Applying the tentative criteria based on the data obtained for the BDOC and residual DOC, three effluents, from Gwangju, Hwasoon, and Jangsung, were found to be the most suitable for SAT applications. It was also concluded that the site characteristics should be also considered with regard to the retention time when evaluating the feasibility of SAT application in a certain region.

Use of microwave pretreatment for enhanced anaerobiosis of secondary sludge.

This work elucidates the effects of pretreatment of secondary sludge by microwave irradiation on anaerobic digestion. The soluble chemical oxygen demand (COD) concentration increased up to 22% as microwave irradiation time increased, which indicated the sludge particles disintegrated. Three identical automated bioreactors with working volume of 5 l were used as anaerobic digesters at mesophilic temperature (35 degrees C). The reactors were separately fed with sludge with microwave pretreated- and control- sludge at different hydraulic retention times (HRT). The volatile solid (VS) reduction in the control operation was approximately 23.2 +/- 1.3%, while it was 25.7 +/- 0.8% for the reactors with the pretreated sludge. The average biogas production rate with the pretreated sludge at 8, 10, 12, and 15 days HRTs was 240 +/- 11, 183 +/- 9, 147 +/- 8, and 117 +/- 7 ml/l/d respectively, while those with the control sludge were 134 +/- 12 and 94 +/- 7 ml/l/d at 10 and 15 days HRTs. Maximum rates of COD removal and methane production with the pretreated sludge were 64% and 79% higher than those of the control system, respectively.

Origins of filter effluent particles: experimental study of particle deposition and detachment.

This paper investigates the relative roles of particle deposition and detachment in controlling the origin of filter effluent particles. A conceptual mathematical model was developed and laboratory-scale experiments were conducted. Laboratory experiments were performed using three sizes of fluorescent microspheres (FMs), to determine the fraction of filter effluent particles that are filter influent particles that were never removed, as well as the fraction of filter effluent particles that were detached after deposition. Experimental results indicated that particle detachment is significant beginning from the early phase of filtration. FM removal increased with filter run time, depth and particle size. For each size FM at one filter depth, FM removal increased with filter runtime to a maximum due to ripening and then decreased with filter runtime after ripening due to limited pore space remaining in the filter. The fraction of effluent particles that were detached particles increased with particle size and filter bed depth.

Microscopic origin of surface-plasmon radiation in plasmonic band-gap nanostructures.

We report spatial domain measurements of the damping of surface-plasmon excitations in metal films with periodic nanohole arrays. The measurements reveal a short coherent propagation length of a few microm inside nanohole arrays, consistent with delays of about 10 fs in ultrafast transmission experiments. This implies that the transmission spectra of the entire plasmonic band-gap structure are homogeneously broadened by radiative damping of surface-plasmon excitations. We show that a Rayleigh-like scattering of surface plasmons by the periodic hole array is the microscopic origin of this damping, allowing the reradiation rate to be controlled.

Inhibitory effect of cyclo-oxygenase-2 inhibitor on the production of matrix metalloproteinases in rheumatoid fibroblast-like synoviocytes.

Cyclo-oxygenase (COX)-2 has been associated with inflammation in rheumatoid arthritis (RA), but its role in joint destruction remains unclear. In this study, we investigated the effect on cultured rheumatoid fibroblast-like synoviocytes (FLS) of the selective COX-2 inhibitor celecoxib on the expression of matrix metalloproteinases (MMPs), which play an important role in tissue degradation and angiogenesis in rheumatoid synovium. Treatment with nontoxic doses of celecoxib resulted in dose-dependent inhibition of MMP-1, -2, and -3 secretion from FLS when measured by enzyme-linked immunosorbent assay. Celecoxib suppressed proinflammatory cytokines (tumor necrosis factor-alpha and interleukin-1beta) induced augmentation of the gelatinolytic activity on zymography. These results suggest that COX-2 inhibitors might influence matrix degradation or angiogenesis in RA by downregulating the expression of various MMPs in rheumatoid FLS.

Limits on diffuse fluxes of high energy extraterrestrial neutrinos with the AMANDA-B10 detector.

Data from the AMANDA-B10 detector taken during the austral winter of 1997 have been searched for a diffuse flux of high energy extraterrestrial muon neutrinos. This search yielded no excess events above those expected from background atmospheric neutrinos, leading to upper limits on the extraterrestrial neutrino flux measured at the earth. For an assumed E-2 spectrum, a 90% classical confidence level upper limit has been placed at a level E2Phi(E)=8.4 x 10(-7) cm(-2) s(-1) sr(-1) GeV (for a predominant neutrino energy range 6-1000 TeV), which is the most restrictive bound placed by any neutrino detector. Some specific predicted model spectra are excluded. Interpreting these limits in terms of the flux from a cosmological distributions of sources requires the incorporation of neutrino oscillations, typically weakening the limits by a factor of 2.

Attosecond active synchronization of passively mode-locked lasers by balanced cross correlation.

A balanced cross correlator, the optical equivalent of a balanced microwave phase detector, is demonstrated. Its use in synchronizing an octave-spanning Ti:sapphire laser and a 30-fs Cr:forsterite laser yields 300-attosecond timing jitter measured from 10 mHz to 2.3 MHz. The spectral overlap between the two lasers is strong enough to permit direct detection of the difference in carrier-envelope offset frequency between the two lasers.

Preliminary study of interfacial shear strength between PMMA precoated UHMWPE acetabular cup and PMMA bone cement.

Followed by successful demonstration of high interfacial tensile strength in a new design of cemented all-polyethylene acetabular cup, interfacial shear strength was investigated in this study, with the use of canine-size prototypes of polymethylmethacrylate (PMMA) precoated UHMWPE acetabular cups. In addition to the PMMA precoated prototypes, three different types of controls were also prepared and tested: grooved UHMWPE cups, PMMA (bone cement) cups, and noncoated, plain UHMWPE cups. The interfacial shear strength of the precoated prototypes was 10.1 +/- 0.69 MPa (n = 6), whereas it was 24.3 +/- 0.78 MPa (n = 2) for the PMMA cup, 6.95 +/- 0.21 MPa (n = 2) for the grooved UHMWPE cup, and 0.34 +/- 0.47 MPa (n = 2) for the UHMWPE cup. These results indicate benefits of the PMMA precoating to stabilize the polyethylene acetabular cup securely when applied with bone cement in simulated clinical applications. Analysis of the failed PMMA precoated UHMWPE prototype cups suggested that the chemically induced bonds between precoated PMMA layer and bone cement played a key role in developing high shear strength. After the interfacial shear test of the PMMA precoated prototypes, major disruptions at the interface between treated UHMWPE and precoated PMMA layer were observed by scanning electron microscopy (SEM), which was a unique failure pattern, not found with other prototypes.

Vascular and metabolic response to isolated small muscle mass exercise: effect of age.

To determine the effect of age on quadriceps muscle blood flow (QMBF), leg vascular resistance (LVR), and maximum oxygen uptake (QVO2 max), a thermal dilution technique was used in conjunction with arterial and venous femoral blood sampling in six sedentary young (19.8 +/- 1.3 yr) and six sedentary old (66.5 +/- 2.1 yr) males during incremental knee extensor exercise (KE). Young and old attained a similar maximal KE work rate (WRmax) (young: 25.2 +/- 2.1 and old: 24.1 +/- 4 W) and QVO2 max (young: 0.52 +/- 0.03 and old: 0.42 +/- 0.05 l/min). QMBF during KE was lower in old subjects by approximately 500 ml/min across all work rates, with old subjects demonstrating a significantly lower QMBF/W (old: 174 +/- 20 and young: 239 +/- 46 ml. min-1. W-1). Although the vasodilatory response to incremental KE was approximately 142% greater in the old (young: 0.0019 and old: 0.0046 mmHg. min. ml-1. W-1), consistently elevated leg vascular resistance (LVR) in the old, approximately 80% higher LVR in the old at 50% WR and approximately 40% higher LVR in the old at WRmax (young: 44.1 +/- 3.6 and old: 31.0 +/- 1.7 mmHg. min. ml-1), dictated that during incremental KE the LVR of the old subjects was never less than that of the young subjects. Pulse pressures, indicative of arterial vessel compliance, were approximately 36% higher in the old subjects across all work rates. In conclusion, well-matched sedentary young and old subjects with similar quadriceps muscle mass achieved a similar WRmax and QVO2 max during incremental KE. The old subjects, despite a reduced QMBF, had a greater vasodilatory response to incremental KE. Given that small muscle mass exercise, such as KE, utilizes only a fraction of maximal cardiac output, peripheral mechanisms such as consistently elevated leg vascular resistance and greater pulse pressures appear to be responsible for reduced blood flow persisting throughout graded KE in the old subjects.

Expression and secretion of the heterodimeric protein interleukin-12 in plant cell suspension culture.

It has been suggested that plant cell culture is the most suitable system for producing small-to-medium quantities of specialized, expensive, and high-purity proteins. Here, we report that a heterodimeric protein, human interleukin-12 (hIL-12), was expressed and secreted into culture medium in a biologically active form. A transgenic plant expressing hIL-12 was constructed by sexual crossing of plants that expressed each subunit of the protein. From a piece of transgenic plant, callus was induced and cell suspension culture was established. The biological activity and amount of hIL-12 secreted into culture medium were analyzed using bioassays and ELISA. Analysis of cellular localization demonstrated that the protein was secreted into the culture medium together with its intrinsic signal peptide.

Idiopathic thrombocytopenic purpura: better therapeutic responses of patients with B- or T-cell clonality than patients without clonality.

Results of recent studies of the pathogenesis of idiopathic thrombocytopenic purpura (ITP) have suggested activated helper T-cells drive B-lymphocytes to produce antibodies. Twenty-eight children and 85 adults with ITP entered this study. We performed polymerase chain reaction (PCR) using framework III variable region (V(H) FRIII)- and joining region (J(H))-specific primers to analyze immunoglobulin heavy-chain gene rearrangement (IgH GR) for B-cell clonality. We used multiplex PCR to analyze T-cell receptor (TCR) gamma-chain gene rearrangement (TCRgamma GR) for T-cell clonality. We diagnosed 10 cases as acute ITP and 97 cases as chronic ITP. The IgH GR result was positive in 77.8% of the acute-form cases and in 58.8% of the chronic-form cases. The TCRgamma GR result was positive in 11.1% of the acute cases and in 10.6% of the chronic cases. There was no difference in frequency of clonality between the acute and chronic forms. After treatment the platelet count normalized in 81.8% (36/44) of the chronic ITP cases with B-cell clonality and in 88.9% (8/9) of the chronic ITP cases with T-cell clonality, compared with a normalized platelet count in 46.2% (12/26) of the chronic ITP cases without clonality. The patients with T- or B-cell clonality appeared to have better therapeutic responses than patients without clonality. In conclusion, T- and B-cell clonality may play a positive role in determining therapeutic response.

Optimal enhancement of in situ hybridization for the detection of porcine circovirus 2 in formalin-fixed, paraffin-wax-embedded tissues using a combined pretreatment of thermocycler and proteinase K.

Optimal enhancement of the hybridization signal was developed for the detection of porcine circovirus (PCV) 2 in formalin-fixed, paraffin-wax-embedded tissues from pigs with postweaning multisystemic wasting syndrome. The hybridization signal obtained after thermocycler pretreatment was very uniform across the section, whereas the signal obtained after either proteinase K or microwave pretreatment not only was weaker but was of variable intensity across sections. Thermocycler pretreatment combined with brief proteinase K digestion can enhance signal detection for target viral nucleic acid in formalin-fixed, paraffin-wax-embedded tissues. A strong hybridization signal was detected in the cytoplasm of macrophages and multinucleated giant cells in lymph node and spleen without background staining and morphological damage. The technical improvement results, therefore, in an identical background at the same time as an increased signal and, thus, may help detect lower levels of PCV2 DNA in formalin-fixed, paraffin-wax-embedded tissues.

Ripe fruit of Solanum nigrum L. inhibits cell growth and induces apoptosis in MCF-7 cells.

Solanum nigrum L. (SNL) has been traditionally used as a herbal plant, whose fruit is believed to have anti-tumor properties, although the mechanism for the activity remains to be elucidated. In this study, we prepared an ethanol extract from ripe fruits of SNL and investigated the mechanism involved in its growth-inhibitory effect on MCF-7 human breast cancer cells. Results from proliferation assay using tritium uptake showed that the proliferative capacity of MCF-7 cells was strongly suppressed in the presence of SNL ethanol extract. This was further confirmed through MTT assay and trypan blue exclusion experiments, which showed a very close correlation between the SNL extract concentration and the surviving cell numbers. The SNL extract-mediated suppression of cell growth was verified to be apoptotic, based on the appearance of DNA laddering, increase in DNA fragmentation, and low fluorescence intensity in nuclei after propidium iodide staining of the cells. Furthermore, the SNL extract was revealed to be a potential scavenger of hydroxyl radicals and DPPH radicals rather than superoxide anions. Collectively, our findings suggest that SNL fruit extract could be used as an anti-oxidant and cancer chemo-preventive material.

Topoisomerase II-alpha (topoII) and HER2 amplification in breast cancers and response to preoperative doxorubicin chemotherapy.

A significant proportion of breast cancers with HER2 amplification have simultaneous amplification of topoisomerase II-alpha (topoII). Amplification of HER2 and topoII was assayed using a novel chromogenic in situ hybridisation (CISH) method. HER2 and topoII amplification status and the response to preoperative doxorubicin chemotherapy were analysed in 67 locally advanced breast cancer patients. Response to chemotherapy was increased in the cases with coamplification of HER2 and topoII (18/19), whereas the response rate was significantly decreased in the cases without HER2 and topoII amplification (17/36). The 12 cases with HER2 amplification alone showed an intermediate response rate (9/12). The findings of the current study indicate that topoII amplification may play a role in determining chemosensitivity of breast cancers to doxorubicin chemotherapy.

Association of porcine circovirus 2 with porcine respiratory disease complex.

A retrospective study was performed on natural cases of porcine respiratory disease complex (PRDC) to determine the association and prevalence of PRDC with porcine circovirus 2 (PCV2) and other co-existing pathogens in Korea. Histologically, alveolar septa were markedly thickened by infiltrates of mononuclear cells. Moderate to marked multifocal peribronchial and peribronchiolar fibrosis were present and often extended into the airway lamina propria. Among the 105 pigs with PRDC, 85 were positive for PCV2, 66 were positive for porcine reproductive and respiratory syndrome virus (PRRSV), 60 were positive for porcine parvovirus (PPV), and 14 were positive for swine influenza virus (SIV). There were 80 co-infections and 25 single infections. A co-infection of PCV2 with another additional bacterial pathogen is frequently diagnosed in PRDC. The combination of PCV2 and Pasteurella multocida (38 cases) was most prevalent followed by PCV2 and Mycoplasma hyopneumoniae (33 cases). The consistent presence of PCV2, but lower prevalence of other viral and bacterial pathogens in all pigs examined with PRDC, has led us to speculate that PCV2 plays an important role in PRDC.
