T cell memory for the cytotoxic response to hapten-modified target cells.

This study describes the development of memory and cytotoxic murine T cells against syngeneic haptne N equals[N-(3-nitro-4-hydroxy-5-iodophenyl-acetyl)-Beta-alanylglycylglycyl] associated antigen. Memory activity in this system had the following characteristics. a) In vitro challenged cells primed in vivo resulted in an augmented cytotoxic response compared to cells primed in vitro. b) The augmented cytotoxic response in vitro was antigen-specific for both target cells in the lytic reaction and stimulator cells in the secondary response. c) Memory activity was long lasting (at least 2 months). d) Memory cells were not cytotoxic. e) Memory activity as well as the cytotoxic cells generated in a secondary response in vitro were T cell dependent, These findings are consistent with the results of others who have investigated T cell dependent memory in other cell-mediated reactions.

Thymus-independent antigens: the preparation of covalent, hapten-ficoll conjugates.

A general approach is presented for covalently attaching carboxyl and primary amino groups to Ficoll and other high molecular weight polysaccharides in a readily controlled manner. The polysaccharides are omicron-carboxymethylated with chloroacetate in NaOH solutions. Amino groups are next introduced by monoamide formation with ethylenediamine using a water-soluble carbodiimide. Reactions are carried out in aqueous solutions; excess reactants and by-products are removed by dialysis. Three different ways for coupling the 2,4-dinitrophenyl hapten to the amino derivative of Ficoll are described. Preliminary evidence indicates that 2,4-dinitrophenyl-Ficolls, prepared by procedures developed in this study, are potent thymus-independent antigens which can specifically stimulate B cells both in vivo and in vitro. Hapten-Ficoll conjugates should be exceptionally useful for studies of B lymphocyte activation because of the ready availability of the starting material and the ease with which a wide variety of hapten derivatives may be prepared.

Enhanced immunogenicity of chemically-coated syngeneic tumor cells.

The immunogenicity of the EL-4 strain of mouse lymphoid leukemia cells in syngeneic C57BL/6 recipients was enhanced by in vitro coating of the cells with either Concanavalin A or dinitrophenylaminocaproate. The tumor-specific immune response was quantitated using a recently developed assay that involves in vitro cell-mediated cytotoxicity.

Structural proteins of adenovirus-associated viruses.

The structural proteins of adenovirus-associated virus (AAV) types 1, 2, and 3 were analyzed by acrylamide gel electrophoresis. In each case, one major protein (C) and two minor proteins (A and B) were identified. Component C had an estimated molecular weight of 62,000 daltons, and the molecular weights of components A and B were found to be 87,000 and 73,000 daltons, respectively. Coelectrophoresis of adenovirus and AAV proteins revealed an overlap only between the adenovirus fiber-penton component and the AAV C polypeptide. Among AAV serotypes, homologous components were electrophoretically identical, except that the C component of AAV-2 was of slightly lower molecular weight than the C components of AAV-1 and AAV-3. The relative incorporation of (14)C-arginine and (14)C-mixed amino acids into the three polypeptides of AAV-2 was similar, indicating an absence of an arginine-rich component. In addition, AAV-2 was found to have a substantially lower arginine content than helper adenoviruses.

Genetic control of the immune response of guinea pigs to limiting doses of bovine serum albumin: relationship to the poly-L-lysine gene.

The response of strain 2 guinea pigs to limiting doses of bovine serum albumin is under dominant genetic control linked with the poly-L-lysine gene. Inbred strains 2 and 13 guinea pigs make similar antibody responses to 10 mug bovine serum albumin, whereas strain 2 but not strain 13 animals produce significant amounts of antibodies in response to 0.1 mug. The relationship between the presence of the poly-L-lysine gene and the ability to respond to limiting doses of bovine serum albumin has also been investigated in random-bred Hartley strain guinea pigs.
