Further characterization of a human T lymphocyte associated antigen.

A human T lymphocyte associated antigen (TLAA), defined with appropriately absorbed antisera to the lymphoblastoid cell line HSB, was originally characterized as a 150,000 mol. wt antigen using sodium deoxycholate solubilized, 125I Bolton-Hunter labelled membranes of HSB, thymus and peripheral blood lymphocytes. The TLAA, which appears to be the major membrane protein labelled with either the Bolton-Hunter, or galactose oxidase-sodium 3H borohydride methods, is weakly labelled in vitro with a 3H amino acid mixture and appears to be a minor membrane component by Coomassie blue staining of detergent solubilized HSB membranes. The antigen is trypsin-sensitive and not extractable with 3 M KCl. The glycoprotein nature of the antigen is suggested by its binding to a Lens culinaris column and its labelling by the galactose oxidase-sodium 3H borohydride and 3H glucosamine methods. Both gel filtration chromatography and sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis studies on 4% SDS gels indicate that the mol. wt. of the antigen is approximately 170,000 Daltons.

Detection of HLA-DRw (Ia-like) antigens on human T lymphocytes grown in tissue culture.

Human T lymphocytes that proliferate in the presence of conditioned medium from PHA-stimulated allogeneic peripheral blood cells were shown to express IPA antigens after the 8th culture day. Ia antigens as detected by xenogeneic antisera were present on 80 to 90% of the cultured cells which were also strongly reactive with xenogeneic antisera defining a human T cell antigen and formed E rosettes. Control cultures with PHA or no conditioned medium expressed T cell but not Ia antigens. These cultured T cells also express the same HLA-DRw determinants as the B cells of the donor they were derived from. Absorption of xenogeneic Ia, and HLA-DRw alloantisera with cultured T cells completely removed the reactivity of these sera for enriched peripheral blood B lymphocytes from normal donors.

Detection of both T-cell and Ia-like antigens on cells from patients with acute myelomonocytic leukemia and chronic myelogenous leukemia in blast crisis.

Appropriately absorbed antisera to the lymphoblastoid cell lines HSB and SB detect a human T-lymphocyte-associated antigen (TLAA) and the human Ia-like antigens, respectively. Cells from some patients with acute myelomonocytic leukemia (AMML) and chronic myelogenous leukemia in blast crisis expressed both TLAA and Ia antigens when tested in a complement-dependent microcytotoxicity assay (greater than 90% lysis with both antisera). When patients were in remission, expression of TLAA and Ia antigens returned to normal values. Quantitative absorption of anti-TLAA serum with increasing numbers of AMML cells showed that these cells could remove reactivity of the serum for both HSB and human thymocytes. Similarly, absorption of anti-Ia serum with AMML cells removed all serological reactivity when this serum was tested on chronic lymphocytic leukemia cells or normal B-cells. These serological findings were confirmed by immunoprecipitation and sodium dodecyl sulfate-polyacrylamide gel electrophoresis studies using radiolabeled antigens. Cells from an AMML patient were labeled with 125I using lactoperoxidase; both the TLAA and Ia antigens were precipitated from the resulting solubilized membrane preparation. Leukemic cells from one AMML patient and one patient with chronic myelogenous leukemia in blast crisis were studied for Ia and TLAA antigens with a double fluorescence technique. Over 80% of the cells showed dual fluorescence.

Purification and characterization of GM1 ganglioside beta-galactosidase from normal feline liver and brain.

GM1 ganglioside beta-galactosidase (GM1-beta-galactosidase) was purified from normal cat brain and liver by a combination of classical and affinity procedures. The final preparation of brain GM1-beta-galactosidase was enriched over 2000-fold with a 36% yield. However, the product was shown to contain several components by disc gel electrophoresis. GM1-beta-galactosidase was also purified from liver with greater than a 30 000-fold enrichment and 40% yield. The liver enzyme was judged homogeneous by disc gel electrophoresis at pH 4.3, 8.1, and 8.9 and by gel chromatography. Both liver and brain GM1-beta-galactosidase(s) eluted as sharp symmetrical peaks from Sephadex G-200 with molecular weights of 250 000 +/- 50 000. The apparent Km determined for 4-methylumbelliferyl beta-D-galactopyranoside (4-MU-Gal) using partially purified brain GM1-beta-galactosidase was 1.73 X 10(-4) M. Liver GM1-beta-galactosidase gave a Km with 4-MU-Gal of 3.25 X 10(-4) M and for [3H]GM1 ganglioside a Km of 4.51 X 10(-4) M was calculated. The pH optima of brain and liver GM1-beta-galactosidase using 4-MU-Gal was 3.8-4.5. By contrast, liver GM1-beta-galactosidase gave a sharp activity peak at pH 4.2 with [3H]GM1 ganglioside. Inhibition by mercuric chloride and sensitivity to hydrogen peroxide and persulfate suggest the involvement of a sulfhydryl in catalysis.

Orientation with style. Matching teaching/learning style.

Satisfaction with orientation and the link to overall job satisfaction are important because of the cost of providing new employees with orientation. Matching the teaching style of preceptors with the learning style of orientees is one way to maximize satisfaction. In this article, the author reports the results of a study conducted to determine whether there was a difference in an orientee's perceived satisfaction with orientation, based on whether the teaching style of the preceptor and the learning style of the orientee were matched or not matched. Personality type identified by the Meyers-Briggs Type Indicator (1962) was used as a measurement of teaching/learning style. The study also evaluated which of the four dimensions of personality type had the strongest relationship between matching or not matching and satisfaction. Those orientees who matched their primary preceptors on the personality preferences of introversion or extraversion reported statistically significant higher levels of satisfaction than those who did not match. Matching/not matching of the introversion/extraversion preference also showed the strongest correlation to overall satisfaction. An exploratory analysis revealed aspects of orientation having the strongest relationship to overall satisfaction. These included creativity, original thinking, experimentation, sensitivity, and flexibility.
