A multivariate approach to saccharide quantitative structure-activity relationships exemplified by two series of 9-hydroxyellipticine glycosides.

Multivariate saccharide quantitative structure-activity relationships (QSARs) have been developed for two series of 9-hydroxyellipticine glycosides. In order to describe the structural variation of the glycoside moieties, thirteen chromatographic variables were used. Eleven D-glycosides and seven L-glycosides were used in separate QSARs to model 9-hydroxyellipticine glycoside antitumour activity against L 1210 leukemia. The multivariate partial least squares (PLS) method was used to establish the QSARs.

Affinity of human carcinoma cell nuclei for polyinosinic acid demonstrated by monoclonal antibodies.

The anti-nuclear cross reactivity of the monoclonal anti-actin antibodies M 372/809 was studied in some detail. The reactivity against a number of nuclear constituents was examined in the ELISA test and the capacities of these constituents to block the M 372/809 anti-nuclear and anti-actin reactions were evaluated in indirect immunofluorescence tests against tissue sections and monolayer cultures of fibroblasts and Vero cells. The repetitive polynucleotides polyinosinic and polyguanylic acid and their deoxyanalogues, actin and vimentin, were found to have the antigenic epitope. The epitope was covered or otherwise inactivated in the presence of polycytidylic acid. Using the M 372/809 antibodies as a reagent, carcinoma cell nuclei were found commonly to have an affinity for polyinosinic and polyguanylic acid. This was seldom noted with non-neoplastic cells.

Growth-activity related variation of a nuclear antigen in monolayer-culture cells quantitated by immunofluorometry and microspectrophotometry.

Antibodies of the hybridoma clone M 372/809 react with a nuclear antigen in growing cells. Using immunofluorometry and microspectrophotometry, the antigen was found to accumulate to a maximum coincident with the marked early increase in nuclear protein contents. The following DNA increase appeared when the intensity of the reaction with M 372/809 antibodies was decreasing. The antigen increase was inhibited by alpha-amanitin. It may therefore be associated with transcription early in the cell cycle.

Homicidal intoxications by lye- and parachlorcresol-containing disinfectants.

In a nursing section for 54 senile, demented inpatients for the period of 3 months, a casual worker committed homicide in 11 cases and attempted homicide in 16 cases by peroral application of lye- and parachlorcresol-containing disinfectants. Autopsies had been performed in nearly all of the fatal cases without detection of the true causes of death. The fatal intoxications could be verified through reexamination of the lung slides and through gas chromatography/mass spectrometry of the formalin-fixed tissue specimens.

Dermatitis herpetiformis: relation between circulating antibodies against reticulin and gluten, small-intestinal mucosal status and absorptive capacity.

The status of the jejunal mucosa and of the intestinal absorptive capacity were investigated and related to the occurrence of antibodies against reticulin and gluten in 55 patients with dermatitis herpetiformis (DH), 28 on a normal, 11 on a gluten-reduced and 16 on a gluten-free diet. The mucosal status was characterized on the basis of histopathological findings and the numbers of intra-epithelial lymphocytes. Absorption was evaluated by 5-h urine and 1-h serum D-xylose tests. There was a positive correlation between the degree of pathological mucosal changes, malabsorption and the occurrence of circulating antibodies against reticulin and gluten. The serum xylose test was more sensitive than the urine xylose test for screening of the relatively mild enteropathy of DH and identified 88% of the patients with an abnormal mucosal status. The serological test (antibodies to reticulin and gluten) identified 80% of such patients. Among patients on a gluten-free diet there was some discrepancy between the serum xylose and the serological test, in that 5 of the 16 patients on this diet had an abnormal serum xylose test result, but no antibodies. In DH patients on a normal diet, the presence of antibodies to reticulin and gluten provided the same information about the presence of mucosal lesions as the serum xylose test. In the whole material a combination of the serum xylose test and the serological test identified 24 of 25 patients with an abnormal mucosal status.

Gluten-free diet in patients with dermatitis herpetiformis. Effect on the occurrence of antibodies to reticulin and gluten.

In a prospective study of 51 patients with dermatitis herpetiformis (DH), the occurrence of antibodies against reticulin and gluten was investigated and the influences of gluten-free, gluten-restricted, and normal diets on the persistence of these antibodies were compared. After a period extending from 11 to 47 months, none of the patients who were eating a gluten-free diet had reticulin or gluten antibodies. The patients eating a normal diet had the same incidence of reticulin autoantibodies at the end of the study as at its beginning, and gluten antibodies developed in some additional patients during the follow-up period. Thus, we have shown that the elimination of gluten from the diet of patients with DH has an important influence on the occurrence of antibodies to both reticulin and gluten.

Failure to detect immunologic stigmata in schizophrenia.

The sera of 17 patients with acute or chronic schizophrenia were tested for antibodies to a human brain extract or to Molt cells; a human T-cell line, with negative results. The level of in vivo lymphocyte stimulation was investigated by determination of the proportion of peripheral blood lymphocytes in the G1, S or G2 + M phases of the cell cycle and by their uptake of 3H-thymidine. There was no sign of increased lymphocyte mitotic activity by these two tests. Therefore, immunologic stigmata could not be detected by these methods in these schizophrenic patients.

Gluten-free diet in chronic active hepatitis associated with intestinal villous atrophy.

Three patients out of 16 with chronic active hepatitis exhibited villous atrophy in biopsy specimens from the upper jejunum. These patients were put on a gluten-free diet for one year, and the intestinal changes normalized in two of the patients, but did not heal in the third patient. The levels of alanine aminotransferase and IgG, did not decrease under the gluten-free diet. The liver disease of the patients with intestinal changes ran a serious course: one patient died in hepatic coma, and one patient developed portal hypertension with recurring hematemesis. These complications did not appear in the patients with healthy intestine. It is suggested that the three patients suffered from both chronic active hepatitis and coeliac disease, which appeared concomitantly on the basis of a genetic disposition for both diseases.

On the biochemical nature of the 'Sm' nucleoplasmic antigen.

The nucleoplasmic autoantigens RNP and Sm are of particular interest because of their associations with certain symptoms of mixed connective tissue disease and systemic lupus erythematosus. The RNP is generally thought to be a ribonucleoprotein and there is evidence that its RNA may be single-stranded. Experiments presented in this report are in support of the concept that the Sm-antigen may also be an RNA protein. Purified Sm-anti-Sm precipitates were shown to have high RNA contents and treatment of Sm-antigen with RNAase in a hypotonic medium strongly reduced in antigenicity. The latter effect indicates that the Sm-antigen may in contrast to the RNP contain double-stranded RNA, a possibility also suggested by the finding that the Sm-antigen was soluble in 2 M LiCl. The Sm-antigen was found further to differ from RNP in being selectively absorbed in BD-Sephadex, while RNP remained active in the supernatant. Cytochemical studies involving stimulation and inhibition experiments with lectins and RNA polymerase inhibitors showed that the Sm-antigen was, in distinction to RNP, sensitive to rifampicin but not to alpha-amanitine. This suggests that the RNAs of the nucleoplasmic antigens may be synthesized by different RNA polymerases.

Some characteristics of immunofluorescence tests for antibodies against gluten, using wheat grain sections or gliadin coated sepharose beads.

Antibodies against gluten and gliadin were determined by the indirect immunofluorescence technique, using frozen wheat grain sections or gliadin-coated Sepharose beads. The methodological characteristics and diagnostic usefulness of the two techniques were evaluated. The reproducibility of both was improved by introducing a fluorometric reading-off procedure. Antibody quantitations were preferably performed as end-point titrations. The two techniques had different dose-response relationships. The grain section technique was more discriminative for small variations in antibody concentration than the bead technique. The latter was, however, more reproducible. Besides antibodies against gliadin, a number of patients with gluten enteropathy had antibodies against the main septa of wheat grains. Fluorescence intensity was preferably expressed in multiples of background intensity. A reaction was visually perceived when the fluorometrically-measured fluorescence intensity reached 2.5 times the background intensity. Using this value as the limit for positive reactions, antibodies were demonstrated in 81% of the cases with verified gluten intolerance, compared with 28% in cases with other intestinal allergies and 8% in normals. The diagnostic specificity of both the grain section and Sepharose bead technique for gluten enteropathy increased with increasing antibody concentration and was apparently 100% when the fluorescence intensity produced by a 1/10 serum dilution reached a value 7 to 8 times that of the background. Antibodies against reticulin were demonstrated in 1/4 of the cases having anti-gluten antibodies but in none of those with non-gluten-induced gastro-intestinal symptoms. Antibodies of the IgG class against cow's milk were demonstrated more often and in higher titre in cases with anti-gluten antibodies than in those without them.
