Loss of heterozygosity on chromosomes 2p, 3p, 18q21.3 and 11p15.5 as a poor prognostic factor in stage II and III (FIGO) cervical cancer treated by radiotherapy.

Loss of heterozygosity (LOH) has been shown to be an important prognostic factor in a variety of malignant neoplasm's. Cervical cancer develops as result of multiple genetic alterations. The aim of this study was to analyze presence of LOH in cervical cancer and to identify the correlation between LOH and survival and relapse-free survival time in patients treated with radiotherapy. Studies were performed on tumor specimens and venous blood from 20 patients with cervical cancer (squamous cell carcinoma G2 and G3) in stage II and III (FIGO) treated with radiotherapy. DNA was isolated using organic extraction. Additional microcolumn purification was performed. The fluorescent multiplex polymerase chain reaction (PCR) was used to amplify 10 microsatellite loci included in commercially available human identification kits. Microsatellite marker BAT 26 was amplified in separate PCR reactions. 75% cervical cancers manifested LOH. LOH in BAT 26 analysis (chromosome 2) was present in all these specimens. 60% of the cases showed LOH at one or more of other examined loci (mostly on 3p, 18q21.3, and 11p15.5). Eight of nine cervical cancers in clinical stage III showed LOH. All cases of G3 squamous cell carcinoma of the cervix manifested LOH on 2p. Patients with LOH have worse prognosis for survival and relapse-free survival compared to patients without LOH.

Three-dimensional sonography in the diagnosis of hepatic hemangiomas.

The purpose of the study was to compare power 3-D with conventional 2-D sonography with color and power Doppler in assessment of tumor vascularity in small hepatic hemangiomas. The study group comprised 12 patients (7 males and 5 females) aged 28-60, (mean 44) with diagnosed hemangiomas. In all the patients 2-D and 3-D sonography was performed and their results were compared. Both color and power Doppler 2-D sonography failed to detect blood flow in the tumors. Blood flow signals were detected in 3-D power Doppler sonography. 3-D power Doppler is superior to 2-D color and power Doppler imaging in the detection of blood flow signal in hemangiomas.

[MRNA activity in synovial fluid after anterior cruciate ligament reconstruction].

The authors present the results of studies on the transcription level of mRNA (poly A + mRNA) in patients in whom anterior cruciate ligament reconstruction was performed. In a group of 9 patients who underwent this procedure an average increase of the concentration of mRNA was noted--from 1.5 micrograms/ml to 67 micrograms/ml in the first week following surgery. This was followed by a decrease of concentration in the third week to 37 micrograms/ml and 7.8 micrograms/ml in the fifth week. In the control group (16 patients who underwent arthroscopy with partial meniscectomy) an average increase in mRNA concentration from 1.3 micrograms/ml to 4.8 micrograms/ml in the first week after the procedure was noted. It was followed by a decrease to 1.6 micrograms/ml in the third week after surgery. The authors believe the marked increase of mRNA is a result of stimulation of synovial cells and an increase of the number of mononuclear cells in the synovial fluid. The small concentration increase of mRNA in the control group seems to be related only the increase in number of mononuclear cells in synovial fluid.

Disputable paternity expertises in the material of the Department of Forensic Medicine Medical Academy of Białystok.

Results of 1045 disputable paternity expertises analysed at the Department of Forensic Medicine, Medical Academy of Bialystok in the years 1992-1999 were reviewed. Investigation procedures involved classical serology methods, HLA testing and DNA polymorphism analyses. Numbers of expertises and exclusion rates in the consecutive years in relation to the employed research techniques are presented.

An assessment of lymphocytic population in peripheral blood of patients with renal cell carcinoma before and after embolization.

The immunophenotype of peripheral blood lymphocytes was evaluated in 50 renal cell carcinoma (RCC) patients, before and after embolization. In RCC patients we have observed significant decrease in the total lymphocyte number as well as in T lymphocyte, their subpopulations and B lymphocyte number. Ten to twelve months after the treatment by embolization, significant increase in the total number of not only lymphocytes (p < 0.001), including T lymphocytes (p < 0.001), T-helper lymphocytes (p < 0.05), cytotoxic T lymphocytes (p < 0.05) and B lymphocytes (p < 0.05), but also an increase of NK cells number (p < 0.05) were found, compared to the values obtained before the treatment and reached the level observed in healthy subjects. Cytometric evaluation of peripheral blood lymphocytic phenotype in renal cell carcinoma patients can help evaluate the immune status of RCC patients as well as assess the embolization efficiency.

The STR systems FES/FPS and F13B in a Polish population.

Allele frequencies for the STR systems FES/FPS and F13B were determined from 203 unrelated individuals from north-eastern Poland. After denaturing PAGE, 7 and 6 alleles were detected for FES/FPS and F13B, respectively. No deviations from Hardy-Weinberg equilibrium were observed.

DNA locus HLA-DQ alpha polymorphism in human population of the north-eastern Poland.

Investigations on DNA polymorphism locus HLA-DQ alpha were carried out on a sample of 117 adult unrelated inhabitants from the north-eastern Poland. The polymerase chain reaction and the reverse dot-blot hybridisation were employed to detect 6 different HLA-DQ alpha alleles. Population data on 20 different genotypes served as a basis for statistic evaluation. The results of genotype analysis were in Hardy-Weinberg equilibrium. Other population data were compared.

Human population genetics of the VNTR locus D1S80 in the north-eastern Poland.

Population genetic studies within VNTR locus D1S80 were carried out on a population sample from the north-eastern Poland. DNA extraction was performed using the Chelex method. DNA amplification was followed by high-resolution PAGE and silver staining (AMP-FLP technique). In 116 unrelated individuals 20 alleles were observed. The D1S80 locus demonstrated a heterozygosity of 0.85. Comparison of the observable allele frequencies with population data from other Caucasian populations revealed no significant differences.

Glyoxalase I (GLO I, EC 4.4.1.5) polymorphism in hemolysates and bloodstains.

Electrophoretic patterns of glyoxalase I (GLO I) phenotypes were evaluated in hemolysates and simulated case bloodstains in relation to duration and conditions of storage and type of substrate. The bloodstains deposited on different substrates were exposed to temperatures -20 degrees C, 4 degrees C, 18 degrees C, 37 degrees C and 56 degrees C. Rapid decrease in the enzyme activity was found in the conditions of high temperature. Character of the substrate noticeably affected GLO I detectability.

Polymorphism of erythrocyte acid phosphatase system (ACP, EC 3.1.3.2) in hemolysates and bloodstains. I. Evaluation of selected methods of ACP phenotyping.

A specific ampholine mixture resulted in appreciable enhancement of discriminating features in electrophoregrams of 6 common ACP phenotypes, thus allowing unequivocal interpretation of the enzyme types in fresh blood and in bloodstains. The employed procedure resulted in marked increase in the limit of ACP phenotype detectability in bloodstains and substantial enhancement of sensitivity and efficacy of the analysis. PhastIEF appeared useful in ACP phenotyping due to its reproducibility and duration of separation. In the case of some ACP phenotypes in ageing bloodstains this technique allows even longer detectability limits compared to PAGE IEF.

Polymorphism of erythrocyte acid phosphatase system (ACP, EC 3.1.3.2) in hemolysates and bloodstains. II. comparison of ACP stability in dried bloodstains in relation to the temperature of storage and the type and character of substrate.

The aim of the study was evaluation of the usefulness of selected electrophoretic methods for ACP phenotyping in simulated casework bloodstains stored at -20 degrees C, 4 degrees C, 22 degrees C and 45 degrees C. Results confirmed faster decrease of the enzyme total activity in the conditions of elevated temperature manifesting with relatively lower fluorescence and resolution of electrophoretic bands. Preliminary evaluation of 'serological history' prior to casework bloodstain analysis is suggested. Beneficial effect of lower temperature during storage until phenotyping is emphasized.

Esterase D (ESD, EC 3.1.1.1) polymorphism in hemolysates and bloodstains. I. Usefulness of selected methods of ESD phenotyping.

The usefulness of several electrophoretic methods including Phast IEF was compared in analysis of ESD polymorphism in hemolysates and bloodstains. The most satisfactory results were obtained with the use of PAGE IEF. The technique permits unequivocal interpretation of the enzyme polymorphism in dried bloodstains as well as in fresh material. The use of the procedure described in this paper considerably increased the limits of ESD phenotype detectability. Phast IEF was shown to be reliable technique in ESD phenotyping. The advantages of this approach are reproducibility and short duration of analysis. The application of minigel IEF to ageing bloodstains may be limited by slightly decreased sensitivity and resolution when compared to standard gel IEF.

The aim of the study was evaluation of the possibility of ESD phenotyping in simulated casework bloodstains in relation to the temperature of storage and to the type and character of substrate. Bloodstained materials were exposed to extreme temperature conditions including -20 degrees C, 4 degrees C, 22 degrees C and 45 degrees C and subsequently phenotyped with the use of electrophoresis and isoelectric focusing. Presented results confirm faster decrease of the enzyme total activity in the conditions of increased temperature manifesting with relatively lower fluorescence and resolution of electrophoretic bands. Preliminary evaluation of external conditions and recognition of the type of the substrate prior to casework bloodstain analysis is suggested.

Human histocompatibility complex (HLA). The distribution of HLA-ABC antigens and structure of exclusions in investigations on disputable paternity in population of north-eastern Poland.

In the present study 80 cases of disputable paternity analysed at the Department of Forensic Medicine in Bialystok in years 1990-1993 were reviewed. Gene frequencies of HLA antigens calculated for the north-eastern population of Poland are not significantly different from those reported for all-Polish population. Similarly, the most frequent HLA haplotypes calculated per 10,000 individuals for the studied population corresponded to those reported by others for central European population. In 13 cases (16.3%) paternity of a defendant was excluded. One case (7.7%) represented exclusions based on three antigens (of loci A, B and C), four cases (30.8%)-exclusions based on two antigens (of loci A and B) and eight cases (61.5%) exclusions based on one antigen (three of locus A and five of locus B).
