A technique of protein-DNA flow cytometry was applied to characterize cell cycling, and to assess the cytotoxicity of cladribine (2-chloro-2'deoxyadenosine) toward seven dissociated cultures of human primary brain tumors (anaplastic astrocytoma and glioblastoma multiforme) grown in vitro from surgical biopsies. Control cytograms were suggestive of that a clonogeneic fraction of the cell population consists mainly of cells with low protein content, which do not require increase in protein content before entering the S phase of the cell cycle. Following 24 or 48 hours exposure to cladribine, 1 nM approximately 1 microM, no cytotoxic effect was evident in 4 cultures, whereas in two cases dose-dependent progressive block of the phase of the cell cycle was noted. In one case a massive cytotoxic effect resulted in disintegration of culture exposed to 100 nM of the drug. However, the treatment with cladribine was ineffective in a patient bearing the tumor which was the source for the last culture, suggesting that cytotoxicity in vitro may not be predictive of clinical response.

Allografts of brain stem from 20-day-old fetuses to nucleus caudatus of adult rabbits were performed. To prevent graft rejection immunosuppression with 2-CdA and cyclosporine A was transiently induced. Graft survival were assessed by histological and electrophysiological techniques. Both morphological (synaptogenesis, myelinization) and functional (generation of rhythmic neuronal activity) signs of graft maturation were found after nine weeks. The data suggest that transient immunosuppression used is sufficient to induce tolerance to neural graft, and no interference with maturation of implanted fetal tissue occurs.
Cytogenetic examinations were carried out in 24 untreated patients with non-Hodgkin, non-Burkitt lymphoma. 10-20 G-banded metaphases, obtained from short-term cultures of unstimulated lymph++ node, bone marrow and blood cells were analyzed in each case. In 18 patients only, the obtained metaphases were suitable for cytogenetic analysis. In 11 patients (group A) karyotype was normal or only single, +non-clonal aberrations were observed. In 7 patients (group B) clonal aberrations were found, among them, in 3-structural changes of chromosome 1, but with different breakpoints: 1p31, 1p31, 1p36. The group of patients with chromosomal aberrations showed statistically significantly shorter survival time than the group without aberrations (p = 0.04). In the former group more patients had low grade malignancy lymphoma. Our observations confirm those data from the literature which indicate that the presence of chromosomal aberrations is a factor of poor prognosis, independent of other clinical and histopathological prognostic factors in non-Hodgkin Lymphoma.

Cytogenetic analyses were carried out on peripheral blood and bone marrow cells of 31 chronic myeloid leukemia (CML) patients who presented with blastic, accelerated, or chronic phases. The percentage of cytoplasmic nonspecific cross-reacting antigen (cNCA, a marker of myelocytic differentiation)-containing cells was determined in the same blood or bone marrow samples. The patients were divided in two groups according to cytogenetic results: those with aberrations in addition to the Philadelphia chromosome (Ph1) and those with Ph1 only. Among the additional aberrations such changes, not typical of CML, were found: del(2)(p21), t(6;11)(q25;q23), and t(12;?)(p13;?). The survival time and the percentage of cNCA-positive cells of patients in blastic and accelerated phases were compared between the above-mentioned two groups of patients using the Student t test and the Kaplan-Meier estimator. The percentage of cNCA-positive cells was significantly lower and the survival time significantly shorter in the group of patients with additional aberrations. The probability of survival according to the Kaplan-Meier estimator was also lower for this group. These data suggest that the immunologically determined lower degree of maturity, that characterized cells bearing additional aberrations, coincides with and/or results in more rapid progression of the disease.
The aim of this work is to analyse chromosome aberrations and cell cycle kinetics in workers occupationally exposed to organic solvents, which contain benzene among other agents. In the exposed group structural chromosome aberrations were found in 2%, while in the control group in 0.9% of analysed metaphases. Detailed band localization of breakpoints in chromosomes of patients in both groups is presented. No significant differences in cell cycle kinetics in individual members of exposed and control groups were found.
Cytogenetic analysis was carried out on peripheral blood lymphocytes of 33 workers professionally exposed to benzene for 10-26 years. In the examined group structural chromosome aberrations were found in 147 metaphases (4.7%). Statistical analysis has shown that the distribution of breakpoints was not random. Chromosomes 2, 4 and 9 are almost twice as susceptible to breaks, while chromosomes 1 and 2 are almost twice as susceptible to gaps, as would be expected based on a random distribution of damage among chromosomes.

The analysis of cardiac electric field interference on rabbits with additional heart implanted to abdominal cavity resulted in explanation of ecg variations during the first hours of newborn's life. The investigation of multipole and multidipole fields in spherical chamber proved that the disturbation of axial symmetry of added up elementary dipoles evokes significant multipole components in the heart.
