beta-Adrenergic receptors in the developing rabbit lung.

beta-Adrenergic receptors and catecholamine-sensitive adenylate cyclase were identified and partially characterized in membrane fractions of rabbit lungs from day 25 of gestation to adulthood with the beta-adrenergic antagonists (--)-[3H]dihydroalprenolol [(--)-[3H]DHA] and (--)-[125I]iodohydroxybenzylpindolol [(--)-[125I]HYP]. beta-Adrenergic receptor number (Bmax) increased 11.5-fold during this time period, increasing progressively during the latter days of gestation and the early neonatal period, from 37 +/- 10 fmol/mg protein at 25 days gestation to 425 +/- 51 fmol/mg in the adult rabbit lung (mean +/- SD). Receptor affinity for (--)-[3H]DHA (KD = 1.8 nM) or (--)-[125I]HYP (KD - 0.104 nM) and the proportion of beta 1- and beta 2-adrenergic receptor subtypes (60% beta 1 and 40% beta 2) did not change with advancing age. Basal adenylate cyclase activity in lung homogenates decreased significantly with increasing age, whereas the activity in the presence of catecholamine or NaF remained nearly constant. Catecholamines stimulated adenylate cyclase activity at all ages studied supporting a role of the maturation of beta-adrenergic receptors in the regulation of pulmonary function.

Specific interaction of myeloma tumor cells with hapten-bearing liposomes containing methotrexate and carboxyfluorescein.

Hapten-bearing liposomes containing methotrexate and the fluorescent solute carboxyfluorescein were incubated with murine myeloma tumor cells expressing surface immunoglobulin with affinity for the hapten. Liposomes bearing the dinitrophenyl hapten became bound to MOPC 315 myeloma tumor cells, and liposomes bearing the phosphorylcholine hapten became bound in much larger amounts to TEPC 15 cells. In each case, fluorescence microscopy showed a patchy surface pattern, indicating intact liposomes at the cell surface. Few liposomes were bound to cells in the presence of excess soluble hapten or to cells lacking the relevant surface immunoglobulin. Cell-associated liposomes were quantitated by use of the fluorescence-activated cell sorter, and the pharmacological effect of the methotrexate was assessed from measurement of incorporation of radiolabeled deoxyuridine into the cells. Little inhibition of deoxyuridine incorporation was observed, because contents of the bound liposomes did not enter the cytoplasmic compartments of the cells.

Further observations on the incidence and nature of atypical creatine kinase activity.

The interference of atypical creatine kinase (CK; EC 2.7.3.2) with anion-exchange methods for the measurement of the CK-MB isoenzyme is now firmly established. False-positive results from this source are much more common than interferences caused by the BB isoenzyme. Atypical CK, at least in some patients, does not appear to be a genetic variant, nor could we relate it to a specific clinical diagnosis, to hypoxia, or to administration of a particular drug. Its behavior in an immuno-inhibition test for CK-B subunit indicates an immunological difference from the normal M subunit. Thus the change in immunological properties is associated with the altered electrophoretic properties. The variability of electrophoretic patterns suggests that atypical CK may represent multiple forms of creatine kinase rather than a unique entity.

Atypical increase in serum creatine kinase activity in hospital patients.

We use an ion-exchange column-chromatographic technique for separating creatine kinase isoenzymes in serum, and occasionally observe what appears to be sustained increase in the MB fraction. Most patients whose sera show such behavior have myocardial disease, but not necessarily a recent myocardial infarction. Electrophoretic analysis of a small sampling of such sera revealed that the apparent MB migrates atypically, appearing distinctly between isoezymes MB and MM. In another electrophoretic system, the peak might easily be mistaken for MM. This unusual isoenzyme does not appear to be "macro" creatine kinase. In laboratories that use the ion-exchange technique, the possibility of a falsely positive MB value should be considered in subjects who show persistent increases together with normal or nearly normal values for total creatine kinase activity. A suitable electrophoretic method that clearly demonstrates this unusual isoenzyme should be used in such cases, for confirmation.

Combined immunodeficiency disease: an inborn error of purine metabolism.

The recently discovered association of adenosine deaminase (ADA) deficiency and combined immune deficiency (CID) has emphasized the critical role of purine salvage in the function of lymphoid tissue. Known enzymatic properties of ADA are presented. In addition, known phenotypic data and possible genetic mechanisms for the occurrence of ADA deficiency in CID are discussed. A hypothesis based on considerations of known metabolic pathways in human erythrocytes is proposed to account for the selectivity of ADA deficiency for lymphoid tissue. Finally, some inhibitors of ADA are discussed as well as some immunosuppressive agents.
