Delta-9-tetrahydrocannabinol: its effect on hypothalamo-pituitary system in male rats.

Subcutaneous administration of delta-9-tetrahydrocannabinol (delta 9-THC) in adult male rats caused a decrease in serum luteinizing hormone (LH) levels with unchanged serum prolactin, pituitary LH and pituitary prolactin content. Response of pituitary to in vitro gonadotropin releasing hormone (GnRH) remained unaltered while the response to in vivo GnRH treatment was markedly increased with the drug indicating the pituitary to be functionally normal. Differences in the in vitro and in vivo response could be due to the endogenous steroid levels. The hypothalamic LH-RH content concommitantly increased. Delta-9-tetrahydrocannabinol may inhibit the release of luteinizing hormone releasing hormone (LHRH).

Production and purification of Penicillium citreoviride toxin and its effect on TPP-dependent liver transketolase.

The production isolation and purification of a yellow mycotoxin from Penicillium citreoviride NRRL 2579 in different culture media was described. When injected subcutaneously to albino rats it alters the kinetic pattern of transketolase (EC 2.2.1.1) in liver in vivo in a competitive manner. In vitro, the inhibition is noncompetitive in nature. However, addition of thiamine diphosphate (TPP) to the in vitro system relieved the inhibitory effect. These findings suggested a relationship between citreoviridin-induced beriberi and the probable antithiamine effect of the toxin.

Antiandrogenic effect of delta-9-tetrahydrocannabinol in adult castrated rats.

In ventral prostate tissues of adult castrated rats, delta-9-tetrahydrocannabinol (THC) at 10 mg/kg for 7 days counteracted the testosterone-induced changes of (i) DNA, RNA and protein content, (ii) the tartrate-insensitive form of acid phosphatase, and (iii) the isoenzyme variant of acid phosphatase associated with the secretory type enzyme. These results indicate that THC acts anti-androgenically and directly at the level of male accessory sex organs.
