Construction of shuttle vectors useful for transforming Clostridium acetobutylicum.

Plasmids pIM13, pT127 and pBC16 delta 1, introduced by transformation into Clostridium acetobutylicum N1-4081, were shown to replicate in, and to confer antibiotic resistance upon this new host. Recombinant plasmids were constructed by inserting erythromycin-resistant plasmid pIM13 into the unique ClaI site of pBR322 or by ligating a tetracycline-resistant determinant of plasmid pT127 to HindIII-linearized pIM13. The hybrid plasmids replicated and expressed erythromycin resistance in C. acetobutylicum strain N1-4081 and in Escherichia coli or Bacillus subtilis, indicating that they might be useful as shuttle vectors for transferring genes between these strains. The efficiency and stability of different replicons in C. acetobutylicum were compared.

[Internal urethrotomy with endoscopic resection of the callus. 32 cases with a follow-up exceeding 5 years].

Thirty-two patients were operated for urethral stricture by internal urethrotomy with endoscopic resection of the callus. Complications consisted of one urethral perforation, one urethral haemorrhage and one case of acute epididymitis. With a follow-up of 5 to 15 years, the authors obtained 85% of good results for strictures less than 1.5 cm long. The success rate was 66% for strictures between 1.5 and 2 cm. The recurrence rate was high for strictures longer than 2 cm (75%). The indications for this technique therefore depend more on the length of the stricture than on its cause or its site. Lastly, endoscopic resection of the callus improves the results or urethrotomy alone by 15%.

[Cystic prostatic utricle in adults. Apropos of 3 cases].

Three cases of cystic prostatic utricle, without any other urogenital abnormality, have been observed in patients of about thirty years old. Two of them were revealed by urinary and ejaculatory obstructive symptoms. The third, discovered by chance, contained calculi. In the three cases, treatment consisted of transurethral endoscopic resection of the anterior lining of the cyst which permitted ample drainage and ensured lasting healing.

[Cure of complex post-irradiation uro-genital fistulae].

The authors report two cases of complex radiation induced urogenital fistulae (vesicovaginal and combined rectovaginal), which were closed successfully at the second attempt with complete restitution of bladder function, without any urinary diversion using colpocleisis in one patient. They mention the necessity of bringing a new source of blood supply into the fistula repair area. Then, they review the different extensive procedures used to ensure this result: small bowel graft, omentopexy to reinforce the suture lines, fibroadipose tissue from the labia majora (Martius' operation). Lastly, basedow their experience and a review of the literature, they propose a therapeutic strategy based on a transperitoneovesical approach with the combined used of isolated small bowel and omental pedicle graft.

Pulmonary complications of hormone treatment in prostate carcinoma.

Fever, dyspnea, dry cough and interstitial fibrosis occurred in a man with prostate carcinoma treated with nilutamide and buserelin for two and one-half months. The diagnosis of drug-induced fibrosis was established on the basis of chronologic and semiologic events. This is the first such case reported.

Lectin-like components in the macronuclear replication bands of Euplotes eurystomus.

Upon incubation with fluoresceinylated neoglycoproteins, isolated macronuclei from the ciliated protozoan Euplotes eurystomus display different labelling patterns depending on the nature of the sugar bound to the neoglycoproteins. Specific sugar-binding components (i.e., lectin-like molecules) are associated with presumed nucleoli and with the macronuclear replication bands. This is the first demonstration that DNA synthesis and sugar-binding components are co-localized in an eukaryotic cell.

Evidence for the existence of a nucleolar skeleton attached to the pore complex-lamina in human fibroblasts.

This work deals with the types of nuclear skeletal structures obtained from human fibroblast nuclei isolated by different procedures. It is confirmed that, in somatic vertebrate cells, the pore complex-lamina is always observed, whereas the presence of internal nucleolar and extranucleolar residual structures depends upon the method of nuclear isolation used. Furthermore, the results reported here argue for the existence of a nucleolar skeleton different from the nucleolar matrix often observed in different cell types by other investigators. The conditions of nuclear isolation which allow us to visualize this nucleolar skeleton without any other internal residual structures are described. The attachment of the nucleolar skeleton to the lamina suggested by the present data is considered in relation to the in situ position of nucleoli near the nuclear envelope.

Detection of sugar-binding sites in the fibrillar and the granular components of the nucleolus: an experimental study in cultured mammalian cells.

The intranucleolar distribution of sugar-binding sites (i.e., lectin-like molecules) was analyzed in segregated nucleoli of actinomycin D-treated HeLa cells. The detection of sugar-binding sites was performed by incubation either of permeabilized nuclei in the presence of fluorescein-labeled neoglycoproteins or of ultrathin sections cut through in situ-fixed nuclei in the presence of gold-labeled neoglycoproteins. In the former case, the fluorescent nucleolar components were identified by comparison with the nucleolar components of similarly treated cells observed in electron microscopy. For the first time, this study reveals the presence of sugar-binding sites in both the fibrillar and the granular components of the nucleolus. In view of the data already reported on the biochemical composition of the nucleolus, some of our results led us to conclude that the nucleolar sugar-binding sites are lectin-like proteins. These proteins could be associated with preribosomes since the nucleolus is the site of both synthesis and stockage of ribosomal precursors. Some results from this study, however, show that the possibility of a relationship between some lectins and a structural component cannot be excluded.

Changes in the response of Haemonchus contortus eggs to the ovicidal activity of thiabendazole during the course of infection.

Hatching rates and susceptibility to thiabendazole were studied on Haemonchus contortus eggs collected after either a single, a challenge or a trickle infection of sheep with a non-resistant strain. Considerable changes were observed both in hatching rates and LC50 values for thiabendazole during the course of infection. Peaks in LC50 suggested that a susceptible strain can produce eggs at least tolerant and perhaps slightly resistant at some days pi even without any previous contact with anthelmintic. The peak obtained after a single infection was more transient than in the other cases thus suggesting that the immune response of the host may facilitate the selection of tolerant or resistant sub-populations of worms or of eggs. The inheritance of this feature was not clearly demonstrated. Nevertheless, the LC50 of a selected sub-population collected from an immune lamb and given to another lamb was higher and more constant than that of the original strain.
