A new method for culture of larvae used in diagnosis of ruminant gastrointestinal strongylosis: comparison with fecal cultures.

A study was carried out in order to compare two techniques for culture of strongyle larvae: the usual fecal culture and a new one where eggs were extracted from feces and larvae were grown in a more defined medium (agar plate fortified with Earle's medium and yeast extract). The efficiency of "on-agar" cultures was better than that of the fecal cultures with a difference of 26% for Trichostrongylus colubriformis and 32% for Ostertagia circumcincta. The variability observed between the number of larvae collected from each culture was low (8% on average) in comparison with that of fecal cultures (15% on average) thus demonstrating a better reproducibility. On the other hand growth of the larvae was similar to that obtained in fecal medium. This new technique avoids the difficulty of controlling the conditions found in fecal cultures (particularly moisture) and could become a more accurate method for diagnosis than the conventional methods. A morphometric study of infective larvae derived from the two methods of culture was carried out. Despite the more defined conditions of development in the "on-agar" cultures, it was not possible to obtain sufficiently homogeneous populations with regard to their measurements so as to distinguish larvae merely with the help of these criteria.

[Roos' axillary approach to the brachial plexus].

An anatomic study of five cadavers, that were dissected on a level with the two shoulders, was made to explore the possibilities of the Roos ' axillary surgical approach. This surgical approach permits almost complete resection of the first rib and dissection of plexus brachialis's inferior roots. (T1, C8 and C7).

Preliminary report of an exposure-based, case-control monitoring system for discovering occupational carcinogens.

There is no evidence concerning human carcinogenic potential of most substances to which workers are exposed. A case-control type method has been developed to discover heretofore unsuspected occupational carcinogens. Incident cases of cancer of several sites are ascertained and the subjects are interviewed to obtain detailed job histories. The job histories are subsequently examined by chemists and engineers who infer for each job a list of potential chemical and physical exposures. These lists of exposures then become the basis of case-control type statistical analyses, wherein each site included and each exposure noted in any history are evaluated for possible association. Controls can be selected in the general population, among non-cancer hospital patients or, as we do, among other cancer patients. Preliminary findings from a pilot study in Montreal show that 1) an efficient case ascertainment system and an acceptable interview procedure can be set up, 2) job histories provided by respondents are quite valid, 3) chemists' translations of job histories into histories of chemical exposures seem to be meaningful and useful, and 4) at least one carcinogenic agent, cigarette smoking, is detectable whether a population control series or a cancer control series is used.

Ultrastructural localization of Ag-NOR stained proteins in the nucleolus during the cell cycle and in other nucleolar structures.

EM investigation of Ag-AS-NOR staining after short glutaraldehyde prefixation followed by Carnoy fixation maintained good ultrastructural preservation and reactive selectivity. This enables exact localization of silver deposits both in the fibrillar centers of typical or segregated nucleoli during interphase, and in chromosome NORs during mitosis. These results argue in favour of the possibility that fibrillar centers are the interphasic counterpart of chromosome NORs. Special structures such as nucleolar blobs and remnants usually considered to be of nucleolar origin, were also stained. - These findings seem to indicate a relationship between the distribution of the silver-stained proteins, the arrangement of the nucleolar structures and the degree of nucleolar activity resulting from the experimental conditions. These results are of interest at the time when the concept of the nucleolar matrix is gradually emerging.

A study of ovine gastro-intestinal strongylosis in a sheep flock on permanent pasture. II. Population dynamics of parasites on the pasture in 1977 and an epidemiological interpretation.

An overall study of gastro-intestinal parasitism was initiated in a flock of sheep in the Western central region of France (Limousin) comprising studies on the animals and on the pasture. Results of the study on animal parasitism and the possibilities of using pepsinogen blood level measurements were published previously (Hubert, Kerboeuf and Gruner, 1979). This report is concerned with the distribution of parasites on the pasture and a discussion on the epidemiology of these strongyles. The pasture was contamined throughout most the year, first by the ewes and then by the lambs from the second month onwards of their presence on the land. Maximum contamination occurred in Summer, but a precocious wave of Ostertagia and Haemonchus larvae on the pasture was responsible for the high incidence of O. circumcincta infections. Unfavourable summer weather depressed the development of eggs and two secondary waves of larvae at the end of August and the end of September brought about peaks of H. contortus and Trichostrongylus in lambs at the end of the summer. Far less Nematodirus were found than normal. In comparison with the weight gain of lambs treated regularly, untreated animals suffered a drop in their rate of growth from the beginning of July to the beginning of September corresponding to the ingestion of a great number of infective larvae. These preliminary results for the year 1977 show the necessity of continuing these observations for several years to enable the effect of preceding year's contamination to be properly assessed.

[Ribosomal crystallization and hibernation in a viviparous lizard].

Part of the ribosomes crystallize in the ovarian follicles during hibernation. The association between ribosomes crystals and endoplasmic reticulum membranes depends on the cell types of the follicle. The cytoplasm of oocytes with a nucleolus shows ribosome crystals. In larger oocytes, without nucleolus, there is no ribosome crystallization. The ribosomes which crystallize would be newly synthesized.

Study of gastro-intestinal stronglyosis in a sheep flock on permanent pasture. 1. Sheep parasitism in 1977.

An epidemiological study aimed at defining gastro-intestinal parasitism in sheep qualitatively and quantitatively was carried out in the Limousin area in France. Five lambs were sacrificed regularly every 4 weeks from May till January of the following year. Complete parasite counts were performed on these animals as well as measurements of the plasma level of pepsinogen. In order to define the influence of parasitism on the growth of the animals, the weight gains of lambs treated regularly and untreated lambs were compared. The results of post-mortem examination show that parasitism is mainly linked to the incidence of strongyles. Other observations in the flock have shown that Moniezosis may also play a non-negligeable part. The parasitic level, already high after one month at pasture, reached its maximum in July-August 1977 and was marked by a very high level of parasitism. Among these species found, two played an essential part: Ostertagia circumcincta and Haemonchus contortus. A study of the growth of the lambs showed that within 6 months, the weight gain of the treated animals was twice that of the untreated animals. The effects of parasitism were worsened by the low fodder value of the pasture and the occurrence of overgrazing. The blood pepsinogen measurements confirmed the usefulness of this technique in establishing the mean level of infestation of a flock by parasites of the abomasum. It seems that in some cases it could help to define the succession of peaks of infestation by different species.

[Ultrastructural identification of the nucleolus organizer by the silver staining technic].

The nucleolus organizer regions can be selectively stained in metaphase chromosome preparations by the Goodpasture and Bloom's technique which was adapted to electron microscopy analysis of cells during interphase. Using this technique, a selective accumulation of silver grains was observed over nucleolus light areas. This selective accumulation allows the identification of the interphase fibrillar centers as the nucleolus organizer regions. Ultrastructural relationships between fibrillar centers and dense fibrillar component are discussed.
