[Cloning and fusion expression of detoxifying gene in Escherichia coli].

In insects, esterases play an important role in the degradation of organophosphate (OP) and Carbamate (CB) insecticides. The ability of esterase B1 to degrading OP and CB insecticides opens broad prospect of using it in programs to the insecticide pollution. OPs can be detoxified by hydrolysis of their phosphoester bonds, pyrethroids and some OPs like malathion, by hydrolysis of their carboxylester bonds, and diverse carbamate insecticides, herbicides and fungicides by hydrolysis of amide or other similar bonds. Most of the candidate bioremediation enzymes identified to date have been hydrolases. In recent years, we have focused our work on the transfer of a detoxifying esterase B1 gene from mosquito into E. coli and explored the possibility of using the detoxified enzymes in environmental protection. 5' initial of esterase B1 cDNA was cloned by RT-PCR and sequenced consequently. After the combination of 5' initial and 3' terminal fragment of esterase B1 cDNA, the recombinant vector pET-ESTB1 was constructed and transformed into E. coli BL21. A 60 kD protein was induced by IPTG and its expression was temperature-dependent. After 12 h induction, the target protein occupied 27% of the total protein. A pure recombinant protein was obtained by purification, and was detected with 10% SDS-PAGE. The results showed that 22.1% of malathion was degraded by crude detoxification enzyme in 15 mins, demonstrated a high degradation property. This research provides a novel approach, which takes the advantages of eucaryotes for bioremediation of pesticide pollutions.

[Characteristics of APC gene mutations in colorectal tumors].

OBJECTIVE: To investigate the mutational features of adenomatous polyposis coli (APC) gene and their possible arising mechanism in colorectal tumors. METHODS: PCR-based IVSP assay (in vitro synthesized protein test) and sequence analysis were used to determine somatic mutations of whole APC gene in 115 cases with non familial adenomatous polyposis(non-FAP) colorectal tumors. RESULTS: Seventy-six mutated cases with 101 mutations were confirmed in the study. Analysis of the spectra of APC mutations revealed that frameshift mutations were more common than point ones(56% vs 44%), ninety-one percent of them being insertions/deletions of 1-2 bp, particularly single A (61%) and 51% of point mutations locating at CpG sites. In comparison with sporadic tumors, hereditary nonpolyposis colorectal cancer (HNPCC) was found to have more frameshift mutations (69% vs 52%, P<0.05), less point mutations at CpG site (31% vs 50%, P<0.05), and a slightly lower frequency of mutations(53% vs 67%, P>0.05). There was no difference in mutational distribution of segment 3 between them (38% vs 46%, P>0.05). It was also found that 15 cases(20%) had double mutations with distance ranging from 7 to 1443 bp, suggesting the coexistance of allelic and non-allelic mutations. CONCLUSION: Based on the mutational spectra of APC gene in colorectal tumors, it can be concluded that most of gene mutations are probably related to DNA mismatch repair deficiency, and the APC mutational patterns of HNPCC and sporadic tumors are quite different. The reason of coexistance of allelic and non-allelic mutations needs to be investigated.

[The effect of p53 gene on p-glycoprotein expression and chemotherapeutic cytotoxicity of hepatocellular carcinoma].

OBJECTIVE: To test the hypothesis that wild-type p53 regulates the expression of p-glycoprotein. METHODS: Hep3B cells which lack the expression of both p53 and retinoblastoma tumor suppressor genes because of deletions, were transfected with a wild-type (wt) p53 cDNA and control vector by a liposome method. RESULTS: After G418 selection, stable wt-p53 transformants and control vector transformants (pNeo) were obtained. Northern and Western blot analysis determined the expression of p53 mRNA and protein in wt-p53 transformants, respectively. In wt-p53 transformants, induction of transcriptionally active p53 was confirmed by the increase of P21(waf1/cip1) protein. Levels of P-gp reduced in the cells expressing wild-type p53 were linked to wt-p53 activity. Cytotoxicity assays revealed that the wt-p53 transfectants were more sensitive to doxorubicin and mitomycin compared with the pNeo transformants. Flow cytometry showed that the accumulation of doxorubicin in wt-p53 transfectants was as 13 times as that of the pNeo transformants. CONCLUSIONS: Restoration of wt-p53 activity in Hep3B leads to sensitiveness to chemotherapeutic agents because of the decrease of p-glycoprotein expression.

Transient dissociation of polyribosomes and concurrent recruitment of calreticulin and calmodulin transcripts in gravistimulated maize pulvini.

The dynamics of polyribosome abundance were studied in gravistimulated maize (Zea mays) stem pulvini. During the initial 15 min of gravistimulation, the amount of large polyribosomes transiently decreased. The transient decrease in polyribosome levels was accompanied by a transient decrease in polyribosome-associated mRNA. After 30 min of gravistimulation, the levels of polyribosomes and the amount of polyribosome-associated mRNA gradually increased over 24 h up to 3- to 4-fold of the initial value. Within 15 min of gravistimulation, total levels of transcripts coding for calreticulin and calmodulin were elevated 5-fold in maize pulvinus total RNA. Transcripts coding for calreticulin and calmodulin were recruited into polyribosomes within 15 min of gravistimulation. Over 4 h of gravistimulation, a gradual increase in the association of calreticulin and calmodulin transcripts with polyribosomes was seen predominantly in the lower one-half of the maize pulvinus; the association of transcripts for vacuolar invertase with polyribosomes did not change over this period. Our results suggest that within 15 min of gravistimulation, the translation of the majority of transcripts associated with polyribosomes decreased, resembling a general stress response. Recruitment of calreticulin and calmodulin transcripts into polyribosomes occurred predominantly in the lower pulvinus one-half during the first 4 h when the presentation time for gravistimulation in the maize pulvinus is not yet complete.

[Long-term morphological changes in mucosa of intestinal urinary reservoir].

OBJECTIVE: To investigate the relationship between long-term morphological changes in the mucosa of the intestinal urinary reservoir (IUR) and postoperative complications. METHODS: A total of 25 patients undergoing the replacement of bladder with IUR were followed up. Of these 25 patients, 15 had colonic pouch and 10 ileal pouch. Tissues of IUR were biopsied examined with microscope and transmission electron microscope. RESULTS: In patients with colonic pouch, the glandular depth was (0.17 +/- 0.04) mm and microvillous height (0.35 +/- 0.05) micron, which were significantly lower than those (0.42 +/- 0.04) micron and (0.69 +/- 0.08) mm in the control group. In those with ileal pouch, the microvillous height was (0.70 +/- 0.14) mm, (0.54 +/- 0.10) mm and (1.99 +/- 0.14) mm in the short-term group, long-term group and control group, respectively. As compared with the control group, mucous atrophy in these patients was more severe. Tight junctions of the epithelium in both kinds of pouches were well preserved even in the long-term cases. No remarkable malignant changes were found in the IUR mucosa in all the patients. Meanwhile, no special morphological changes were seen in the patients with metabolic acidosis or pouch calculus. CONCLUSIONS: IUR mucosa has some morphological "adaptive" changes after the operation. Intestinal segments are ideal bladder substitutions.

[Analysis of relationship between risk factors of malignant transformation of oral leukoplakia and the LSCP system].

OBJECTIVE: To explore the clinical significance of the LSCP (lesion size, site of lesion, clinical aspects, pathological aspects) classification and staging system through a comprehensive analysis of relationships between the risk factors of malignant transformation of 209 cases with oral leukoplakia and the LSCP classification and staging system. METHODS: Single factor Chi-square test was first performed to examine the associations between LSCP stages (I, II, III, IV) of oral leukoplakia and each of risk factors, including sex, site, size, numbers of lesion, alcohol and tobacco consumption, clinical classification and histopathological classification respectively, to select the most significant factors which influence the LSCP classification and staging. Then, the association of these selected factors with LSCP stages of oral leukoplakia (stage IV vs. I, II and III) was evaluated using multiple logistic regression analysis. RESULTS: Sex, site of lesions, clinical aspect and histopathological features of lesions were chosen as risk factors incorporated into the multiple logistic regression models. The results demonstrated that the risk of oral leukoplakia of the female patients classified as LSCP stage IV was 2.49 times as high as that of the male patients. The risk of lesions occurring in tongue and/or that floor of mouth was higher than that in other sites. Among the different clinical subtypes of lesions, the verrucous leukoplakia was the highest and 10.00 times as high as that of the homogeneous one. Among the different histopathological types, when hyperplasia and mild dysplasia were set as the basic level, the risk of severe dysplasia classified as LSCP stage IV was the highest and 499.55 times as high as the basic level, while that of moderate dysplasia was 276.48 times as high as the basic level. Pathological features with moderate and severe dysplasia were the most important contributory factors to the LSCP staging. CONCLUSIONS: The LSCP classification and staging system provides a comprehensive description of the features of oral leukoplakia, which is helpful in evaluating the overall risk of malignant transformation of oral lesion, and is valuable in clinical follow-up and developing the best treatment plan.

Prevention and treatment of hepatitis B relapse after liver transplantation.

OBJECTIVES: To investigate the effect of orthotopic liver transplantation (OLT) on hepatitis B(HB)-related diseases and the efficiency of prevention and treatment with lamivudine on recurrence of hepatitis B posttransplant in China. METHODS: Orthotopic liver transplantation (OLT) under veno-venous bypass was performed in 10, of whom 9 males had hepatitis B and 1 female had hepatocellular cancer (HCC) without HB pretransplant. Eight of the 9 males had fulminant hepatitis B (FHB) and they all had preoperative serious jaundice, ascites and coagulopathy. Six had encephalopathy; 1 was associated with acute renal failure, and 1 with gastrointestinal hemorrhage. Seven of the 10 patients had lamivudine to prevent reinfection with hepatitis B. RESULTS: Of the 8 patients who have survived for 2-12 months, 7 have survived for 6-12 months. Two died, one of recurrent FHB and the other from multi-organ failure (MOF). Seven preoprative HB patients of the 8 survivors have excellent liver function through 1 has positive HBsAg 6 months after OLT. One of the 8 survivors, the female with HCC pretransplant, suffered hepatitis B 6 months after OLT and her hepatic function has been gradually improving with lamivudine therapy. CONCLUSIONS: OLT is an effective therapy for certain cases of FHB and HCC and lamivudine may prevent recurrence of hepatitis B after OLT.

Treatment of hepatic failure with artificial liver support system.

OBJECTIVE: To assess the effectiveness of artificial liver support system (ALSS) treatment in patients with hepatic failure. METHODS: 235 cases of hepatic failure were treated with ALSS in our hospital. All data were analyzed by SPSS. The effectiveness of ALSS treatment was compared according to different stages (i.e., early, middle and end stages). RESULTS: 108 patients survived after therapy of ALSS. After each ALSS treatment, the liver function of these patients was greatly improved, the serum endotoxin and HBV-DNA concentrations were significantly decreased, and the serum concentration of aromatic amino acids (AAA) such as methionine decreased while the ratio of branched chain amino acids and aromatic amino acids (BCAA/AAA ratio) increased; patients treated with ALSS in the early or middle stages of disease had much higher survival rates than patients in the end stage of disease. CONCLUSION: ALSS is a reliable therapy for advanced liver diseases and treatment at early or middle stages is appropriate.

[Comparison of tumor suppressor gene deletion between squamous cell carcinoma and adenocarcinoma lung cancer in Chinese].

OBJECTIVE: To prove the hypothesis that there may be different tumor suppressor genes (TSGs) involved in genesis of squamous cell carcinoma (SCC) and adenocarcinoma (ADC) in non-small cell lung cancer (NSCLC) in the Chinese. METHODS: Twenty-two microsatellite markers flanking to or within 13 identified or candidate TSGs were used to detect loss of heterozygosity (LOH) in these regions in the specimens of 28 SCC patients and 13 ADC patients. RESULTS: Our data demonstrated frequent allelic loss of FHIT, p53, IFNA, VHL and p16 both in SCC and ADC. Three genes, PRLTS, PTEN and p57, showed statistically significant differences in the LOH frequency between SCC and ADC. CONCLUSION: The comparison of LOH frequencies between SCC and ADC indicates possible correlation between specific tumor suppressor genes and each type of lung cancer. The results also suggest that more attention should be paid to the PRLTS gene, which may play an important role in the development of ADC.

[In vitro study of the killing activities on H-2(d) murine normal and tumor cells by Ly49A gene transfected lymphocytes].

OBJECTIVE: To observe the killing activities of Ly49A gene transfected lymphocytes of C57BL/6 mice to normal and tumor cells of BALB/c mice. METHODS: pLXSN-Ly49A retrovirus vector was constructed and packaged with PA317 cell line. The lymphocytes of C57BL/6 mice were transfected by culture with virus producing PA317 cells. The Ly49A expression rate on the transfected lymphocytes was detected by flow cytometry and the killing activities of the transfected lymphocytes to normal and tumor cells of BALB/c mice were assayed by MTT method. RESULTS: The Ly49A expression rates of C57BL/6 mice lymphocytes transfected with pLXSN-Ly49A for 24 hours, of those transfected with pLXSN and nontransfected control were (46.67 +/- 0.35)%, (18.73 +/- 0.85)%, and (19.60 +/- 0.27)%, respectively. The killing activity of the transfected lymphocytes to 4T(1) tumor cells remained almost the same as that of the control (P > 0.05), but to normal fibroblasts decreased sharply (inhibiting rate 22% - 25%). CONCLUSION: The Ly49A transfected C57BL/6 mice lymphocytes could kill BALB/c mice tumor cells as effectively as the control did, but the activity decreased sharply to normal BALB/c mice cells, which would be instructive for resolving graft versus host disease after allogeneic bone marrow transplantation.

[Effect of PCNA antisense oligooxynucleotides and VEGF antisense oligoxynucleotides on growth of hepatocellular carcinoma transplanted in nude mice].

OBJECTIVE: To study the inhibitory effect of PCNA antisense oligonucleotides or/and VEGF antisense oligonucleotides on growth of human hepatocellular carcinoma in nude mice. METHODS: Human hepatocellular carcinoma transplanted subcutaneously in nude mice model was established, and subsequently was divided into four groups: PCNA-ASODN group, VEGF-ASODN group, combined group and control group. Different treatments were given respectively. The weigh and volume of subcutaneous tumors was measured, and the morphological changes of tumor cells was observed under microscope. RESULTS: The growth inhibitory rate in the PCNA-ASODN group was 72.8%, 44.9% in the VEGF-ASODN group, and 87.2% in the combined group. The combined group which was treated with PCNA-ASODN and VEGF-ASODN showed more effective inhibitory than the PCNA-ASODN group or the VEGF-ASODN group respectively. CONCLUSIONS: The growth of human hepatocellular carcinoma in nude mice can be inhibited by PCNA antisense oligonucleotides and VEGF antisense oligonucleotides respectively. Better inhibitory effect can be obtained if PCNA antisense oligonucleotides and VEGF antisense oligonucleotides are combined in use.

[Study of peak expiratory flow in normal young, middle-aged and old persons in Chengdu area].

OBJECTIVE: To build the normal ranges and regression equations of peak expiratory flow (PEF) in normal young, middle-aged and old persons. METHODS: PEF, FVC, V75, V50, V25 were measured in 393 normal young, middle-aged and old persons with the youngest aged 20 years and the eldest 84 years. Regression equations of PEF were built for younger group (20 yr-49 yr) and elder group (50 yr-84 yr) respectively. RESULTS: PEF, FVC, V75, V50, V25 were decreased with increase of age. The regression equations showed that both in younger and elder groups, PEF was negatively correlated with age in male (P < 0.0001-0.002, r = 0.32-0.39) and positively correlated with FVC in female (P < 0.0001-0.0005, r = 0.32-0.59). PEF was highly correlated to V75 with r = 0.87-0.94 in male and r = 0.93-0.96 in female. The coefficient of variation (CV) of PEF was in normal range (16%-24%) for all age groups except male and female above 70 years old with CV of 30% and 27% respectively. This confirmed the data was reliable. CONCLUSION: The normal ranges and regression equations of PEF constructed as a result of this study for normal humans of 20-84 years old can be put to use in clinical work and public health.

[Study of peak expiratory flow in 728 normal adolescents in Chengdu area].

OBJECTIVE: This study was carried out to build the normal values and regression equations of peak expiratory flow (PEF) in normal adolescents. METHODS: PEF and routine ventilatory functions, FVC, V75, V50, V25 were measured in 728 normal adolescents aged 8-19. RESULTS: The PEF and FVC, V75, V50, V25 rose with the increase of age. The regression equations showed that PEF was highly positively correlated with age and FVC both in male and female adolescents, and also positively correlated with body weight in males; the coefficients of multiple correlation were 0.85 and 0.78 in male and female ones respectively. Correlations of PEF with V75, V50, V25 were also high, especially with V75 of which the correlation coefficients came up to 0.98 both in male and female adolescents. The coefficients of variation were 15%-20% both in male and female adolescents, and were in normal ranges. CONCLUSION: The normal ranges and regression equations of PEF provided for normal adolescents can be used to evaluate the pulmonary functions of patients who are in the same age-group suffering from bronchial asthma and other respiratory diseases.

[Application of posterior canal wall reconstruction and mastoid obliteration].

OBJECTIVE: To explore the obliteration of the old mastoid cavity and reconstruction of the posterior canal wall after radical mastoidectomy. METHOD: Posterior canal wall reconstruction and mastoid obliteration of 11 ears of 10 patients with old mastoid cavity for 1 to 6 years after radical mastoidectomy were done using the pedicle postauricular composite skin flap (PPCSF), and their tympanoplasty were performed at the same time. Follow-up was performed at 3 to 30 months. RESULT: All PPCSFs survived. The old mastoid cavity of 11 ears vanished and reconstructed posterior canal wall remained in normal position without retraction. The drying ear time took 2 to 3 weeks postoperatively, and the symptoms of the radical cavity disease after radical mastoidectomy were cleared up nearly, and the average hearing threshold decreased 13.6 dB HL. The otorrhea and perforation of tympanic membrane resulted from infection happened to 1 of 11 ears three month after the operation. CONCLUSION: The PPCSF is an effective method in the obliteration of old mastoid cavity and reconstruction of the posterior canal wall. It can reduce drying ear time postoperatively, treat the radical cavity disease after radical mastoidectomy and is beneficial to the tympanoplasty at the posterior external at the same time.
