A seroprevalence study of viral hepatitis E infection in human immunodeficiency virus type 1 infected subjects in Malaysia.

Hepatitis E virus (HEV) is a RNA virus transmitted enterically. A study of anti-HEV antibodies in 145 human immunodeficiency virus type 1 (HIV-1) infected subjects found that 14.4% of them were reactive to anti-HEV antibodies. Anti-HEV IgG and anti-HEV IgM was detected in 10.3% and 4.1% of the subjects respectively. Prevalence of anti-HEV (either IgG or IgM) was similar across all adult ages (p = 0.154), between the three ethnic groups (p = 0.378), and across risk groups (p = 0.120). The results showed that HEV infection in subjects recruited in this study was most likely transmitted via faecal-route.

Role of mitochondrial cytochrome c in cocaine-induced apoptosis in coronary artery endothelial cells.

Cocaine induces apoptosis in coronary artery endothelial cells. Yet the cellular and molecular mechanisms are not clear. Given that cocaine has profound toxic effects on the mitochondria, the present study examined the role of mitochondrial cytochrome c in cocaine-mediated apoptosis. Using cultured bovine coronary artery endothelial cells, we found that cocaine-induced apoptosis was dose dependently inhibited by cyclosporin A with IC(50) of 0.2 microM. The maximum of 65% inhibition was obtained with 3 microM cyclosporin A. Cocaine induced a translocation of cytochrome c from the mitochondria to the cytosol with a 1.8-fold increase in cytosolic cytochrome c levels, and a corresponding decrease in mitochondrial cytochrome c. In accordance with its inhibition of cocaine-induced apoptosis, cyclosporin A blocked cocaine-induced cytochrome c translocation. Correspondingly, cocaine-induced activation of caspase-9 preceded that of caspase-3. Caspase-8 was not activated. Cocaine also produced a dose-dependent decrease in Bcl-2 protein levels, but had no effect on Bax protein levels. The cocaine-induced decrease in the Bcl-2 protein was not affected by cyclosporin A but was partially blocked by caspase-3 inhibitor Ac-DEVD-CHO. Collectively, these data indicate that the release of cytochrome c from the mitochondria and the subsequent activation of caspase-9 and caspase-3 play a key role in cocaine-induced apoptosis in these cells. Furthermore, the down-regulation of the Bcl-2 protein may play an important role in cocaine-induced release of cytochrome c.

[The expression of transforming growth factor-beta receptor I and its mRNA in liver tissues of chronic hepatitis B and the clinical significance].

OBJECTIVE: To study the expression of transforming growth factor-beta receptor I (TGF-beta RI) and its mRNA in liver tissues of chronic hepatitis B (CHB) and the relationship between its expression and the pathological injury of hepatic tissues. METHODS: Forty-eight liver specimens from patients with CHB were observed by techniques of immunohistochemistry and in situ nucleic acid hybridization. RESULTS: TGF-beta RI was expressed in the hepatocyte, sinusoid endothelial cell and bile duct epithelial cell, with predominance in the hepatocyte and distribution in small granule or cell membrane. Increased expressing of TGF-beta RI was parallel to the degree of inflammatory activity and fibrosis (r=0.7095, P<0.05; r=0.6915, P<0.05; respectively). The number and the distribution of TGF-beta RI mRNA in the positive cells were coincident to those of its protein. The expression of TGF-beta RI was closely correlated with Tbil (r= 0.3396, P<0.05), PA (r= -0.3430, P<0.05) and ALB (r= -0.4173, P<0.01). CONCLUSION: The TGF-beta RI may involve in the pathological injury of hepatic tissues of CHB.

[Clinical study on postpartum depression and its related factors].

OBJECTIVE: The prevalence rate, related factors and clinical characteristics of postpartum depression were studied in order to promote obstetrician's understanding of the disease, and resort to preventive and therapeutic measure earlier. METHODS: Two hundred and ten puerpera were selected from September to November 1998 for mood study by self-rating depression scale, self-rating anxiety scale, Hamilton depression scale, and self-made questionnaires. RESULTS: The prevalence rate of postpartum depression was 37.14% (78/210), of which, 91.03% (71/78) was mild, and 8.97% (7/78) moderate. In addition, 10.48% (22/210) of women got anxiety after delivery. The symptoms, including decreased activity, difficulty in thinking, problems in decision-making, difficulty in doing things, feeling of uselessness and hopelessness were chief complaints of postpartum depression patients. Bad social behavior and mood controlling, inadequate psychological preparation for delivery, lack of knowledge of delivery were closely correlated to the onset of postpartum depression, which are the risk factors of postpartum depression. CONCLUSIONS: Postpartum depression is a spiritual and psychological obstacle with relatively high prevalence rate. Bad social psychological factors can bring out and aggravate postpartum depression. It is important to carry out prenatal psychological care and to propagate the knowledge of delivery, to find out and cure of postpartum depression as soon as possible.

[Status of methylation of promoter of mis-match repair gene hMLH1 in lung cancer].

OBJECTIVE: To study the relationship between status of methylation of hMLH1 promoter and expression of its protein in lung cancer. METHODS: Methylation of the hMLH1 gene promoter and hMLH1 protein was examined in 50 primary lung cancer tissues by Hpa II digestion-PCR and immunohistochemistry. RESULTS: The incidence of the hMLH1 promoter methylation was 32.0% (16/50). Methylation frequeney did not correlate with sex, smoking and clinicopathological characteristics. Negative expression of hMLH1 protein occurred in 14/46 tumors, of which 11 tumors had hMLH1 promoter methylation (78.6%, 11/14), while only 5 had methylation in 32 specimens positive for hMLH1 protein expression. CONCLUSION: In about one-third of lung cancer tissues examined there exists methylation of hMLH1 promoter, which correlates well with the absence of expression of hMLH1 protein.

Effects of xin mai tong capsule on vasoregulatory peptides in the patients of coronary heart disease.

In order to inquire into the therapeutic effects of Xin Mai Tong Capsule ([symbol: see text]) on coronary heart disease with myocardial ischemia, 40 patients were randomly divided into two groups (Xin Mai Tong group and the control group). The plasma endothelin (ET) levels in the two groups of patients were markedly higher than that of the healthy people (P < 0.001), and the calcitonin gene related peptide (CGRP) was similar to that of the healthy people (P > 0.05). After treatment, ET and symptomatic scores in the two groups decreased markedly (P < 0.01), and their S-T segments were elevated obviously (P < 0.01). But the decrease of ET and symptomatic scores and elevation of S-T segment in Xin Mai Tong group were superior to those of the control group (P < 0.05-0.01). The CGRP level in the control group did not vary obviously post-treatment, but it increased markedly (P < 0.01) with the addition of Xin Mai Tong Capsule in Xin Mai Tong group.

Long-term potentiation induced by nicotine in CA1 region of hippocampal slice is Ca(2+)-dependent.

AIM: To observe the effects of Ca2+ on hippocampal long-term potentiation (LTP) induced by nicotine in CA1 region of rat hippocampal slice. METHODS: Extracellularly recorded population spikes (PS) of the pyramidal cell layer in the hippocampal CA1 region in vitro. RESULTS: Nicotine 1 mumol.L-1 induced LTP in the hippocampal CA1 region. It did not induce LTP in CA1 region when CA2+ was removed from artificial cerebrospinal fluid (ACSF). Nifedipine 1 and 10 mumol.L-1 partly inhibited LTP induced by nicotine, and thapsigargin 1 and 10 mumol.L-1 completely inhibited LTP induced by nicotine. CONCLUSION: LTP induced by nicotine in hippocampal CA1 region is Ca(2+)-dependent. Both Ca2+ influx and Ca2+ release participate in the induction of LTP.

Safety and immunogenicity of lyophilized, live attenuated hepatitis A vaccine in non-human primate model.

OBJECTIVE: To evaluate the safety and immunogenicity of lyophilized, live attenuated hepatitis A vaccine (H2 strain) in rhesus monkeys. METHODS: Nine adult rhesus monkeys were used as experimental animals. The rhesus monkeys without anti-HAV were divided randomly into the aqueous vaccination group (4 rhesus monkeys), the lyophilized vaccination group (3 rhesus monkeys), and the control group (2 rhesus monkeys). Monkeys were inoculated by intramuscular injection, with control monkeys being inoculated with Minimum Essential Medium Eagle (MEM). Following vaccination, the monkeys were observed for the development of diarrhoea and other adverse side-effects, such as changes in appetite, frequency of defaecation and stool consistency for seven days. At the weeks 2, 3, 4, 6, 8, 10 and 12 positnoculation, the peripheral blood was collected from all animals and assayed for anti-HAV and alanine aminotransferase (ALT) and aspartate aminotransferase (AST), at weeks 0, 4 and 8 postinocuation, needle-biopsy specimens were taken at weeks 0, 4, 8 and 12, all monkeys were sacrificed and tissue samples were taken from liver, lung, heart, kidney and brain for pathological examination at week 12. RESULTS: Animals were immunized with a dose of 7.0 logTCID50/ml which is stable after freeze-drying. During the 12-week observation, no animals showed abnormal elevations of liver enzymes (ALT and AST) and no change in appetite or activity. Two monkeys (one in the aqueous group and the other in lyophilized group) showed possible lesions at week 8. The lyophilized vaccine, in addition to eliciting an anti-HAV IgG response similar to aqueous vaccine (P > 0.05), also showed IgM anti-HAV response at week 2 which was not observed with aqueous vaccine. CONCLUSIONS: These results demonstrate that lyophilized, live hepatitis A vaccine is safe and highly immunogenic in primates, supporting its further evaluation in human clinical studies.

Comet assay and cytokinesis-blocked micronucleus test for monitoring the genotoxic effects of X-ray radiation in humans.

OBJECTIVE: To assess the genotoxic effects of X-ray radiation on human populations. METHODS: The single cell gel electrophoresis (SCGE) and cytokinesis-blocked micronucleus (CBMN) test were applied as biological dosimeters to detect DNA damage and abnormalities in human peripheral lymphocytes of subpopulation exposed to X-ray radiation. The subjects were divided into four groups: 12 radiation-patients; 13 intervention-radiation-therapy doctors; 32 radiation-diagnostians; 28 controls. RESULTS: The average comet lengths of the four groups were 128.17 +/- 4.49 microns, 88.09 +/- 5.39 microns, 72.68 +/- 2.57 microns and 32.87 +/- 0.57 microns, respectively. The difference in average comet length between any two groups was highly significant (P < 0.01). The average micronucleated cell (MNC) rates (@1000) of the four groups were 12.33 +/- 0.85, 9.75 +/- 1.02, 8.48 +/- 0.66 and 3.18 +/- 0.36, respectively. The difference of MNC rates of Group 1 vs 3, 1 vs 4, 2 vs 4 and 3 vs 4 was highly significant (P < 0.01), and the difference of Group 1 vs 2 was significant (P < 0.05), but there was no difference of MNC rate in Group 2 vs 3 (P > 0.05). CONCLUSIONS: This study showed that both the comet assay and the CBMN test could be used to monitor populations exposed to X-ray radiation, but the comet assay seems to be more sensitive than the CBMN test.

[Difluoromethylornithine synergizes with antisense bcl-2 RNA in the induction of apoptosis of HL60 cells].

OBJECTIVE: To study the combined effect of difluoromethylornithine (DFMO) and bcl-2 antisense RNA in the induction of apoptosis of HL60 cells. METHODS: HL60 cells were transfected with recombinant retroviral vector expressing bcl-2 antisense RNA. The growth characteristics of transfectants were studied by morphological observation, growth curve, FCM analysis, colony formation in soft agar, DNA electrophoresis, molecular hybridization and immunocytochemical staining. RESULTS: The recombinant retroviral vectors expressing bcl-2 antisense RNA and the cell line producing recombinant virus were constructed. Although the expression of bcl-2 at mRNA and protein levels in the HL60 cells transfected with antisense bcl-2 were down-regulated, there was no change in cell growth, cell cycles and expression of ornithine decarboxylase (ODC) compared with the parental cells albeit some decrease in colony formation. In addition, transfection with antisense bcl-2 did not induce marked apoptosis whereas treatment of the transfectant with low concentration (0.2 mmol/L) of DFMO resulted in enhanced expression inhibition of bcl-2 protein, inhibition of cell growth and induction of apoptosis. CONCLUSION: DFMO synergizes with antiense bcl-2 RNA in the induction of apoptosis of HL60 cells.

[Effect of xinmaitong capsule on total ischemia burden in coronary heart disease patients with myocardial ischemia and analysis of its therapeutical mechanism].

OBJECTIVE: To assess the effect of Xinmaitong (XMT) capsule in treating ischemical cardiac disease. METHODS: Sixty coronary heart disease (CHD) patients with myocardial ischemia were divided randomly into two groups. XMT group (30 cases) was treated with XMT plus western medicine, and control group (30 cases) with western medicine alone. The changes of the scores for clinical symptoms, the total ischemia burden (TIB), the plasma endothelin (ET), the nitric oxide (NO), the superoxide dismutase (SOD), and the malonyldialdeyde (MDA) levels were observed before and after treatment. RESULTS: After treatment with XMT the scores of clinical symptoms, TIB, ET and MDA levels were significantly decreased (P < 0.01), the levels of NO and SOD were significantly increased (P < 0.01) in the XMT group, comparing with the control group these changes were statistically different (P < 0.01). CONCLUSIONS: XMT capsule can act against myocardial ischemia effectively, one of the mechanisms of which is protecting the function of vascular endothelium and resisting lipid peroxidation injury. The effects of adding XMT capsule on conventional treatment of western medicine were better than those of using western medicine alone.

[Effect of tsaoko-anemarrhenae decoction on intracerebral c-fos, c-jun mRNA expression in interrupting pentylentetrazol kindled epileptic rat model].

OBJECTIVE: To investigate the molecular biological mechanism of anti-epileptic effect of Tsaoko-Anemarrhenae decoction (TAD) on epileptic model rats. METHODS: The pentylenetetrazol (PTZ) induced epileptic Sprague-Dawley rats were selected as the animal model. The effects of TAD on the c-fos, c-jun gene expression in the rats' brain were observed by in situ hybridisation. RESULTS: The c-fos, c-jun gene expression was obviously increased, TAD could effectively block the gene expression of c-fos, c-jun, showing better antiepileptic effect. CONCLUSION: The mechanism of TAD anti-epileptic effect might be correlated to the decrease of c-fos, c-jun gene expression.

[Tracheal transplantation].

OBJECTIVE: To treat long tracheal cancer by tracheal transplantation. METHODS: In March 17, 1999, a patient received a total laryngeal resection and a resection of 8 cm upper and middle trachea, which was replaced by a 6 cm long trachea of another donor. The donor trachea was dipped first in preservation liquid A for sterilization for 24 hours, then in preservation liquid B for over 48 hours for destructing MHC. Pedunculated greater omentum and pedunculated greater pectoral musculus supplied the blood for the donor trachea. One side of the donor was anastomosed with the lower trachea, and the other with the neck skin. RESULTS: The patient survived 300 days after operation. The auto tracheal membrane covered the donor surface; auto- and xeno trachea was well anastomose. The new trachea grew very well. CONCLUSION: Trachea transplantation is possible the best method to treat long tracheal disease if the transplantable trachea is less 5 cm.

[Adsorption behavior of chromate onto activated carbon particles].

In this paper, the adsorption behavior of chromate onto activated carbon particles was studied as a function of pH, flow rate and the concentration of tested anion. The results showed that chromate anion was not only reduced but also adsorbed by activated carbon particles, the percentages of reduction and adsorption were strongly dependent on the pH value and the flow rate. In the medium of pH < 10, chromate anion could be adsorbed directly onto the activated carbon particles and the adsorbed chromate could be eluted from activated carbon particles by 1 mol/L NaOH without valence change. The reduced product of chromate anion was mainly Cr (VI), Cr(VI) could be adsorbed in the pH > 4 medium by the activated carbon particles and eluted with 1%H2SO4.

[A study of the immune in formation of calcium bilirubinate gallstones in different rabbit models--the changes of the immunoglobulins in serum and bile and the immunoglobulins forming cells in the gallbladder mucoderm].

The changes of Ig-forming cells in the gallbladder mucoderm and the immunoglobulins in serum and bile were studied in the rabbit models. One hundred rabbits were randomly divided into the control group (Con, n = 10), simple biliary obstruction group (BO, n = 45) and biliary obstruction and infection group (BOI, n = 45). The results showed that the concentrations of serum immunoglobulin A (IgA) in BO and BOI groups increased remarkably in all phases (P < 0.001), The concentrations of serum IgG in both groups increased with the formation of gallstones. The IgA and IgG contents of bile samples in BO and BOI groups with negative bacterial culture were much higher than those of control group (P < 0.05), but the Ig content of bile with positive culture was slightly lower than that of the control group. Only a few Ig-forming cells were found in the gallbladder mucoderm of normal rabbits. The counts of Ig-forming cells in the mucoderm in BO group remained unchanged, but increased much higher in BOI group (P < 0.001), especially in IgG formation. This experiment demonstrates the immunoglobulins of serum and bile change significantly during the formation of gallstones. The Ig content of bile has a relationship with bacterial infection. IgA plays an important role in gallstone formation. The gallbladder of rabbit may be an important place of Ig-formation. The quantity of Ig forming cells in biliary tract may have a close relationship with the gallstone formation.

[Analysis of imidacloprid and damanlin by high performance liquid chromatography].

A new method for determination of imidacloprid and damanlin by using HPLC was established. It was carried out on a mu Bondapak C18 column (3.9 mm i.d. x 300 mm) with ACN-MeOH-H2O (60:10:30, V/V) as eluent and detected at 240 nm using photodiode array detector. The results of imidacloprid and damanlin were 3.71% and 15.65% respectively. The standard deviations for imidacloprid and damanlin were 0.88% and 0.76% respectively.

[Study on extraction process for psoralen in compound prescription by orthogonal design].

The study on water volume, extraction time and times for the extraction of psoralen in compound prescription Yiniao Tong capsule has been carried out by orthogonal design. The process condition has been determinated, extracting prescription herbs with water for 3 times(2 hour and 16 time amount of water all told). The determination method used in this experiment was TLC-scanning.
