Transmission and scanning electron microscopy findings in 37 patients with monocytic leukemia.

The ultrastructural features of the monocytic blast cells of 37 patients with M4 and M5 types of leukemia were examined with transmission (TEM) and scanning (SEM) electron microscopes. The cells showed a large variety in nuclear size and shape, dependent on the stage of cell differentiation. Nuclear pockets, appendices and bridges were observed. The cytoplasm contained perinuclearly located fibrils, occasionally elongated mitochondria and Auer bodies. The surface ultrastructure of the cells was similar to that of healthy monocytes, but often numerous blebs were observed.

Elemental X-ray microanalysis of mouse peritoneal and bone marrow derived cultured mast cells.

Elemental X-ray microanalysis of intact mouse peritoneal cells revealed significantly high K alpha sulfur and chloride peaks, while phosphorus, sodium and calcium displayed relatively low K alpha peaks. Bone marrow derived cultured mast cells exhibited a low K alpha peak for sulfur and a high K alpha peak for chloride. Peritoneal macrophages which served as controls differed from peritoneal mast cells by their high phosphorus and chloride content and a relatively low sulfur peak. The finding of two different spectra for the mast cells may provide additional parameters for discrimination of mast cell subsets.

Familial fragility on chromosome 16 (fra 16q22) enhanced by both interferon and Distamycin A.

A family with a "fragile site" at 16q22, inducible by both interferon and Distamycin A, is reported. Immunological problems were found in the family. In a sibship of ten, eight children had died in infancy. Our study led to the conclusions that interferon and Distamycin A induce fragility at the same site, which has the same characteristics as the spontaneous fragile site; that a viral hypothesis for this fragility may be supported; and that immunoincompetence of one kind or another must be considered in families presenting a fragile site at 16q22.

The correlation of increased serum prolactin levels with decreased sexual desire and activity in elderly men.

Serum prolactin levels and sexual function were evaluated in 28 men from 60 to 64 years of age and in 44 men from 65 to 70 years of age. All subjects were married, physically healthy, and had no psychopathology or marital problem. About a third of the men aged 60 to 70 years suffered from impotence. No obvious correlation between elevated levels of serum prolactin and impotence was obtained. Subjects aged 65 to 70 who had decreased libido exhibited a significant elevation of serum prolactin levels, while subjects of the same age group who had reserved (normal) libido appeared to have low serum prolactin levels. Nine of ten men aged 60 to 70 years with serum prolactin levels above 40 ng/ml reported decreased libido. Potent men of both age groups (60-70 years) with high prolactin levels showed a tendency to have a decrease in frequency of sexual intercourse. Thus, it seems that mild hyperprolactinemia in aging men may be associated with decreased sexual desire and frequency of sexual activity.

Birth defects monitoring.

Population monitoring of birth defects provides a means for detecting relative changes in their frequency. Many varied systems have been developed throughout the world since the thalidomide tragedy of the early 1960s. Although it is difficult to pinpoint specific teratogenic agents based on rises in rates of a particular defect or a constellation of defects, monitoring systems can provide clues for hypothesis testing in epidemiological investigations. International coordination of efforts in this area resulted in the founding of the International Clearinghouse for Birth Defects Monitoring Systems (ICBDMS) in 1974. In this paper we will describe the functions and basic requirements of monitoring systems in general, and look at the development and activities of the ICBDMS. A review of known and suspected environmental teratogenic agents (eg, chemical, habitual, biological, physical, and nutritional) is also presented.

Diagnostic efficacy of echography in cholestasis.

The usefulness of hepatobiliary echography was assessed prospectively in 32 patients with cholestatic jaundice. The predictive reliability of the method for detecting major bile duct obstruction was 93% and for excluding this possibility was 88%. Echography altered diagnostic thinking significantly in 22 patients (69%). Correct management decisions were made promptly in all but 1 patient who underwent unnecessary laparotomy for acute hepatitis. Laparotomy was correctly avoided in 2 patients and invasive investigations were not required in a further 12. It is concluded that hepatobiliary echography is a useful and cost-effective method in the diagnosis and management of the jaundiced patient.

Distribution of tritium-labelled 8-methoxypsoralen in the rat, studied by whole body autoradiography.

The distribution of 3H-8-methoxypsoralen has been studied in rats by whole body autoradiography. The highest organ concentration were seen 1 hour after dosing. The most pronounced accumulation was found centriblobular in the liver, corticomedullary in the kidneys, and in the cortex of the adrenals. The concentrations in these organs were approximately six times higher than in the blood. Other organs showed concentrations similar to blood levels. Illumination with UVA (10 J/cm2) increased the concentrations in the subcutis.

Application of the Process Scoring System to waking, dream and therapy reports.

Extended the Process Scoring System, originally developed for measuring dream dynamics and content, to waking and therapy experiences and applied it in a study of 5 new and 5 experienced Ss in an intensive outpatient psychotherapy. Two areas were examined: First, differences on process and content variables related to length of time in therapy and, second, the parallel relationship in these variables between waking, dream, and therapy experiences. The Process Scoring System proved useful in measuring these variables in all three types of experiences. The results indicated significant differences between the new and experienced patients and measurable interaction between the waking, dream, and therapy areas.

Inattentiveness in schoolchildren with epilepsy.

Different aspects of "inattentiveness" or related behavior were assessed in 71 children with epilepsy and in 35 nonepileptic children attending ordinary school. Both groups were of similar age and eduational circumstances. Epileptic boys were significantly more inattentive and overactive than nonepileptic boys according to their teachers and parents, and they performed significantly less well on tests of sustained attention and perceptual accuracy. No such differences were seen between epileptic and nonepileptic girls. This contrast between the epileptic groups was not attributable to drug effects or EEG abnormalities. Auditory stimulation reduced the perceptual accuracy of nonepileptic children but appeared to have a beneficial effect on epileptic groups. Teachers' ratings of inattentiveness were not consistently correlated with other measures. These findings conform with other evidence which suggests that epileptic boys are behaviorally more vulnerable than epileptic girls but not simply as a reflection of sex related differences in children in general. The results also suggest that teachers' statements about "inattentiveness" are difficult to interpret and that problem behavior at school needs to be specified precisely.

Accumulation of 8-methoxypsoralen in the rat retina.

The distribution of 3H-8-methoxypsoralen (3H-8-MOP) has been examined by autoradiography in albino and pigmented rat eyes. Very high concentrations were seen in the pigmented layer of the retina, ciliary body and iris in pigmented rats. Cornea concentrations in both rat strains were about twice that of blood, and lower concentrations were seen in the lens.

Psychophysiological correlates of the spontaneous K-complex.

A pilot study covering three nights and two studies were carried out to investigate the psychophysiological correlates of the spontaneous K-complex in relation to intense psychotherapy. The pilot study produced evidence that the K-complex was not consistent and stable within or across nights and that its variability might be sensitive to psycho-emotional influences. In the first major study 2 subjects were recorded non-consecutively over a 3-wk. period during experience of intensive therapy while in the second 6 subjects were studied before and after two therapeutic sessions. Results indicated that nights after therapy differed significantly from baseline nights and a significant interaction took place between subjects and therapy on one or more variables of the K-complex and several eye movement indices. Some relationship to length of time in therapy was also noted. These findings were interpreted to indicate a possible relationship between complete expression of feeling and the occurrence of phasic events in the sleeping EEG.
