Use of PPD and phosphatide antigens in an ELISA to detect the serological response in experimental bovine tuberculosis.

Five calves from tuberculosis free herds were each inoculated intranasally with 10(6) viable organisms of a field isolate of Mycobacterium bovis. Four of the calves developed acute tuberculosis. ELISAs employing protein and phosphatide extracts of M bovis as antigens were used to monitor the humoral response of the infected calves. Fourteen days after infection there was a dramatic increase in the level of antibodies demonstrated by the phosphatide antigen. This increase coincided with the first appearance of signs of the disease. The results suggest that the phosphatide antigen may be of potential value in detecting a humoral response, if present, in cattle infected with M bovis. The tests employing the protein antigen demonstrated a humoral response in only one of the infected calves and emphasises the importance of antigen selection to detect antibodies in tuberculous animals.

Excretion of Mycobacterium bovis by experimentally infected cattle.

Three groups, each of five calves, four to seven months old, were inoculated intranasally with different numbers of Mycobacterium bovis. Infection was established readily in the calves which received an inoculum containing either 10(6) or 10(4) colony forming units (cfu). After every infection there was a lag period during which the organisms could not be isolated from specimens of nasal mucus. All the animals excreted M bovis and the time of commencement, quantity and duration of excretion appeared to be related to the inoculation dose. Excretion continued for many weeks, and for two calves excretion became intermittent over many months. All the calves which were given inocula of 92 cfu failed to develop the disease and no immunological responses were detected; however, M bovis was isolated from nasal secretions from one of these animals 100 days after inoculation.

Pitfalls in preoperative staging in prostate cancer.

Clinical staging in 60 patients with adenocarcinoma of the prostate was compared with pathologic staging to identify factors which may contribute to staging errors. Understaging was directly related to tumor stage and was documented in 0 per cent of A2, 26.5 per cent of B1, and 66.7 per cent of B2 patients. Capsular invasion was found in 11.8 per cent of B1 and 52.4 per cent of B2 patients, seminal vesicle extension in 17.7 per cent of B1 and 52.4 per cent of B2 patients, and lymph node metastases in 2.9 per cent of B1 and 28.6 per cent of B2 patients. The majority of patients who had unnoticed gross extension of the tumor beyond the prostate underwent transurethral resection of the prostate or failed irradiation therapy prior to radical prostatectomy. The results suggest that intraprostatic or periprostatic changes caused by previous treatment to the prostate may interfere with the preoperative staging.

Excretion of Leptospira interrogans serovar hardjo following calving or abortion.

Leptospira interrogans serovar hardjo was demonstrated in the vaginal discharges of 12 experimentally infected heifers for up to eight days after abortion or calving. Organisms were recovered from the oviducts of heifers examined at slaughter eight to 22 days after calving or 32 to 91 days after infection. They were also recovered from the uteri of four heifers eight to 22 days after calving.

Bovine leptospirosis: microbiological and serological findings in normal fetuses removed from the uteri after slaughter.

Leptospires were demonstrated by a combination of the direct fluorescent antibody technique and culture in six (4.6 per cent) of 130 fetuses from apparently normal uteri collected at an abattoir. Fetal ages ranged from the fifth to the seventh months. In addition, leptospires were isolated from the oviducts of two uteri whose fetuses were not infected. All the strains isolated were similar antigenically to serovar hardjo. Leptospiral antibodies were not detected in any of the fetuses.

Bovine leptospirosis: serological findings in aborting cows.

The antibody titres of 149 cows aborting leptospire infected fetuses and 156 cows aborting fetuses not apparently infected with leptospires were determined by the microscopic agglutination (MA) test. There were differences in the distribution patterns of MA titres between the two groups of cows but these differences would only be of very limited value in the diagnosis of leptospiral abortion. Although 81.2 per cent of aborting cows which had a titre of 1:1000 or greater had leptospire infected fetuses, they represented 46.3 per cent of cows aborting leptospire infected fetuses. Among cows which aborted infected fetuses, 22.8 per cent had no detectable antibodies. The complement fixation and plate agglutination tests were of less value than the MA test.

Bovine leptospirosis: microbiological and serological findings in aborted fetuses.

Leptospiral infection was diagnosed in 41.6 per cent of 245 randomly selected aborted bovine fetuses and 68.9 per cent of 103 aborted fetuses from farms with abortion problems. Diagnosis was based on combined leptospiral isolates from fetal kidney and eye, and fetal serology and immunofluorescence. Fifty-six of the 58 strains of leptospira isolated belonged to the Hebdomadis serogroup and were similar to serovar hardjo while one of the remaining two strains belonged to the Icterohaemorrhagiae serogroup and the other to the Canicola serogroup. These findings indicate that serovar hardjo infection is a major factor in the aetiology of bovine abortion and that it is possible to isolate this organism from fetal kidney by the careful application of dilution culture technique.

Chemodectoma involving the cavernous sinus and semilunar ganglion.

This paper reports a case of paraganglioma involving the cavernous sinus and semilunar ganglion. The origin of the tumor cells, from paraganglia in this region, is also discussed. Recognition of paraganglia in this area may aid the clinician and pathologist in the differential diagnosis of tumors in this region.

Evaluation of outpatient client progress: time-series telephone interview data.

A cost-efficient information system that allows for the continual monitoring of the verbal reports of mental health outpatient behavior was designed and implemented. Verbal descriptions of patients' problem behaviors were collected during baseline, therapy, and post-therapy periods using a telephone interview procedure. Descriptive data were presented for 25 patients, demonstrating that: (a) adequate baseline measures can be taken to evaluate therapy interventions, (b) there are wide differences among patients' reports as to the change in frequency of problems that occurs, and (c) patients' reported satisfaction with therapeutic services may be inconsistent with their verbal reports of behavioral change.

Evaluation of a live Salmonella choleraesuis vaccine by intranasal challenge.

Weaned pigs were immunised with a live attenuated Salmonella choleraesuis vaccine. The protective immunity induced was compared with that of unvaccinated pigs by intranasal challenge with a field strain of S choleraesuis. Of 18 unvaccinated pigs, three died as a result of challenge from S choleraesuis and 10 of the survivors developed chronic lung lesions. None of the vaccinated pigs died as a result of challenge and two only showed macroscopic evidence of salmonellosis on autopsy three weeks after challenge. It was concluded that subcutaneous vaccination of pigs prevented death and clinical illness but did not prevent invasion of tissues following intranasal challenge with S choleraesuis.

Antibodies to Leptospira in the sera of aborted bovine fetuses.

Antibodies to Leptospira serotypes were detected in sera from 15 (6.9 per cent) of 218 aborted fetuses, but were not detected in the sera from 196 non-aborted fetuses. Fourteen (6.4 per cent) sera from the aborted fetuses had antibodies to strains belonging to the Hebdomadis serogroup and 1 (0.5 per cent) had antibodies to a strain belonging to the Icterohaemorrhagiae serogroup. These antibodies were predominantly immunoglobulin M whereas in the dams, antibodies belonged to both immunoglobulin M and G classes.
