[Effect of stimulation of the superior and inferior spermatic nerves on testosterone secretion and testicular blood flow].

The effects of testicular nerves on regulation of androgen secretion were examined by determining testicular blood flow and testosterone in testicular vein at 10 minute interval before, during, and 10, 20, 30 min after stimulation (Pre. Est. Post. 10. Post 20. Post 30 respectively) of superior or inferior spermatic nerves (SSN or ISN) with 25 Hz high (50-70 V) or low (20-25 V) currency. Blood flow and blood samples of the testicular vein were obtained through a cannula in left side under anesthesia of pentobarbital sodium. Under the stimulation of SSN, blood flow in testicular vein decreased by 44.87% in Est with high voltage whereas with low voltage no change appeared in Est but decreased by 27.25% in the period of poststimulation; although with high voltage of stimulation of SSN a transient fall of testosterone (42.01%) was observed in Est, 2.5 times increase of testosterone occurred in the period of poststimulation, meanwhile the testosterone rose by 87.33% in Est of SSN stimulation with low voltage and was further elevated up to 4 times in post-stimulated period. Unlike the stimulation of SSN, there was no influence of stimulating ISN on testosterone with either high or low voltage whereas there was significant increase of blood flow (12.31%) in the period of Est with high voltage then it tended to decrease after stimulation. The present work supported the hypothesis that testicular nerves involve in regulation of androgen secretion in testis and provided a strong evidence for further investigation of nervous regulation of endocrine in testis.

Clinical and experimental studies on acupuncture treatment of apoplexy at the convalescent stage by consciousness-restoring method.

The consciousness-restoring method and the conventional method of acupuncture were used for treatment of 92 cases of apoplexy at the convalescent stage. They were randomly divided into 2 groups: the consciousness-restoring group (50 cases), and the conventional group (42 cases). The therapeutic results show that the cure rates in the former and latter groups are 43% and 16.67% respectively. The markedly effective and effective rates in the former group are significantly higher than that in the latter group. The experimental results demonstrate that there is remarkable improvement in the majority of criteria of blood rheology, blood lipid, micro-circulation and thrombus formation in vitro, in the former group, and the degree and extent of improvement are markedly better than the latter group.

[Unearathed ancient pulsological books and theoroy of "three parts with nine palpation sites"].

Textual research is made on the evolution of the theory of "three parts with nine palpation sites" in TCM. It is concluded that the theory, having received the influence of ancient astrology and arithmatics, was sprouted in the Eastern Han Dynasty and still is valid today. It further points out that the arrangement of channel order, and selection of channel title were all influenced by the theory of "Three Parts with Nine Palpation Sites".

[Cultivation of Polygonum multiforum].

The experiments of some cultivated medthods of Polygonum multiforum show that the direct sowing of seeds is better than the cuttage, the index of reproduction and yield are higher, and the cultivated method can decrease expense. To cultivation period of 5 years, high yield and excellent qualty is available.

[Observation on the life habit of Epicata aptera].

Epicata aptera reproduces one generation every year. Adults arise in the last ten-day period of May, mate and lay egg in the first ten-day period of June. They mate and lay egg 1-3 sequence every year, lay egg 108-269 grains every sequence. Period of their laying egg undergoes 48-78 days. Larva have six age period. They take false pupas live through the winter. Complete generations undergo 348-394 days.

[Primary analysis of the situation of hepatitis G virus infection in different groups of people].

In order to investigate the situation of hepatitis G virus (HGV) infection among different groups of people, two self-selected peptides from HGV NS5 were used to develop an indirect ELISA for anti-HGV IgG detection. 1209 sera from different peoples were detected and the positive rate of anti-HGV IgG was (3.8%). Among these groups the group of non A-E hepatitis had the highest anti-HGV positive rate (20.5%), higher than that in the groups of natural people (0.8%) and other hepatitis patients (3.3%), the difference was significant. The anti-HGV IgG positive rate in the group of HC patients 8.0% was higher than that in other hepatitis A-E patients, the positive rate (8.0%) in paid donors was higher than that in voluntary donors (0%), and that in groups with venereal diseases and HIV infection were 3.6% and 8.0%, respectively. The results suggested that there has higher rate of HGV infection in our country; HGV is the important factor causing non A-E, the paid donors is the dangerous group, they should be strictly screened, and the people with transfusion have higher rate of HGV infection. Since there is different rate of HGV infection in different human population so it is important to study further the epidemiological characteristics of HGV for its prevention.

[Effects of IFN-alpha on the expressions of perforin and granzymes in IL-2 activated lymphocytes].

OBJECTIVE: To study the effect of IFN-alpha on the expressions of perforin and granzymes in IL-2-activated-lymphocytes. METHODS: NK and LAK activities were assayed by 4 hour standard 51Cr release test, the activity of perforin was detected by hemolysis method, expression of granzyme B was measured by ABC immunohistological method, expression of granzyme A was measured by BLT method. RESULTS: IFN-alpha significantly augmented the activities of NK and LAK in IL-2 activated peripheral blood lymphocytes (PBL) after 1 day culture. Perforin activity in lymphocytes was increased after 1 day exposure to IL-2 or IFN-alpha, and was enhanced when exposed to the combination of IL-2 and IFN-alpha. After 3 day culture, the perforin activity remained high in lymphocytes activated by IL-2 alone or in combination with IFN-alpha, while declined to control level in IFN-alpha exposed group. IL-2 and IFN-alpha alone or in combination had no effect on expression of granzyme A and B. CONCLUSION: IFN-alpha enhances the cytotoxicity of lymphocytes activated by IL-2. The mechanism might be that IFN-alpha upregulates the perforin expression.

[In vitro reversal of homoharringtonine resistance by the combination of tamoxifen and verapamil].

OBJECTIVE: To investigate the reversal of homoharringtonine (HHT)-resistance by tamoxifen (TAM) or verapamil (VER) alone or in combination. METHODS: The drug-sensitivity test was performed with semisolid agar culture. RESULTS: The cytotoxicity of HHT to K562/S cells was not enhanced by TAM or VER alone or in combination,but HHT resistance in HHT resistant cells (K562/H20) was reversed by VER and TAM at nontoxic doses (4micromol/L or 8micromol/L). The IC50 of K562/H20 for HHT decreased from 446.8 +/- 0.08microg/L to 45.1 +/- 0.02microg/L in the presence of 4micromol/L of VER, to 22.4 +/- 0.03microg/L in 8micromol/L of VER, to 85.1 +/- 0.03microg/L in 4micromol/L of TAM and to 26.4 +/- 0.02microg/L in 8micromol/L of TAM. In the presence of combinations of 2micromol/L VER and 4micromol/L TAM and of 2micromol/L VER and 8micromol/L TAM, IC50 of K562/H20 decreased to 30.4 +/- 0.02microg/L and 4.3 +/- 0.04microg/L, respectively. CONCLUSION: HHT-resistance could be reversed by VER or TAM alone, and the combination of the two drugs showed a synergistic effect.

[Effect of bone marrow transplantation conditioning regimen on thyroid and adrenocortical functions in patients with blood diseases].

OBJECTIVE: In order to investigate the life quality of patients with hematological diseases after bone marrow transplantation (BMT). METHODS: Pituitary-thyroid and adrenocortical function were de tected in 29 patients with hematological diseases before and after BMT. RESULTS: All patients had normal thyroid and adrenocortical functions before BMT. As compared with pre-BMT,the median serum TSH levels were significantly increased at 3, 6, 12 (P<0.01), 24-60 (P<0.05) months post-BMT. The adrenocortical function had no change in all patients, and thyroid dysfunction were found in 6 of 24 patients detected at a median time of 6 month post-BMT. Five of the 6 thyroid dysfuntion patients developed hypothyroidism, and one hyperthyroidism. CONCLUSION: There was little effect of conditioning regimen on adrenocortical function, while thyroid dysfunction was common after BMT. Long term follow up on thyroid function in patients after BMT is necessary.

[The killing effect of IL-2 and IFN-alpha activated bone marrow on K562 leukemic cell].

OBJECTIVE: To evaluate the killing effect of IL-2 and IFN-alpha activated bone marrow cells on K562 cells. METHODS: Semi-solid colony assay was used. RESULTS: Bone marrow from leukemia patients in remission was activated in vitro with IL-2 for 3 days. The activated bone marrow (ABM) displayed killing effects of 0.31 approximately 2.30 logs on K562 cells, this killing effects was further increased to 0. 30 approximately > 3.15 logs when IFN-alpha added with IL-2 to the marrow for activation. IL-2 alone or in combination with IFN-alpha showed no inhibition of CFU-GM and K562 cells. Compared with IL-2 or IFN-alpha alone, the combination of the two cytokines could more effectively maintain the killing effect of ABM on leukemic cells. CONCLUSION: IFN-alpha can augment the purging effect of IL-2 ABM and combination of the two cytokines can effectively maintain the cytotoxicity of ABM.

[Studies on expansion ex vivo of murine bone marrow cells and its hematopoietic reconstitution capacity].

OBJECTIVE: To investigate the effects of stem cell factor (SCF) in combination with interleukin-1 (IL-1) or/and interleukin-3 (IL-3) on ex vivo expansion of 5FU treated bone marrow cells and hematopoietic recovery in lethally irradiated mice transplanted with the expanded cells. METHODS: 5FU treated bone marrow cells (d3-5FU-BMC) were cultured in a cytokines-containing medium, and the net increments of CFU-GM and high proliferative potential colony forming cell (HPP-CFC) were evaluated. RESULTS: CFU-GM increased by 33.7 +/- 18.1- or 18.1 +/- 6.3- fold, and HPP-CFC by 17.8 +/- 10.5- or 12.7 +/- 9.1- fold, respectively, in cultures containing SCF with IL- or IL-3, as compared with that in control; while SCF alone had little effect. Compared with fresh d3-5FU-BMC, transplantation of the expanded bone marrow cells accelerated the recovery of recipients' peripheral blood cell counts by 1 approximately 3 days and increased the survival rate of the transplanted animals (d3-5FU-BMC group 50% vs expansion group 8U approximately 100%). CONCLUSION: SCF in combination with IL-1 or IL-3 synergetically ex vivo expands hematopoietic cells. Transplantation of the expanded bone marrow cells accelerates the recipient's hematopoietic reconstitution.

Interleukin 1 beta converting enzyme-like proteases are essential for p53-mediated transcriptionally dependent apoptosis.

p53-mediated apoptosis in baby rat kidney (BRK) cell lines transformed by E1A and p53(val135) requires a transcriptionally functional p53. Coexpression of the E1B 19K protein in BRK cell lines transformed by E1A and p53(val135) rescues cells from p53-mediated apoptosis, and this is paralleled by the absence of both lamin and poly(ADP-ribose) polymerase cleavage. Therefore, the role of interleukin 1 beta converting enzyme (ICE)-like porteases in p53-mediated, transcriptionally dependent apoptosis was investigated. The ICE-like protease CPP32 was proteolytically activated during p53-mediated apoptosis in BRK cells, and this required a transcriptionally competent p53. Substitution of the p53 transactivation domain with the transactivation domain of herpes simplex virus VP16 (VP16/p53) resulted in accelerated kinetics of both apoptosis and Bax induction. Moreover, apoptosis induced by p53, VP16/p53, and Bax was abrogated by Z-VAD.FMK, an inhibitor of ICE-like proteases. These results indicate that all apoptotic pathways downstream of p53-mediated transcription converge upon the activation of ICE-like proteases.

Chronic obstructive pulmonary disease: role of bronchiolar mast cells and macrophages.

Chronic obstructive pulmonary disease (COPD) is considered to be caused in part by smoking-induced inflammation, but it is unknown which inflammatory cells within the small airways are associated with the obstruction. We investigated the inflammatory infiltrate in the small airways of 16 current or ex-smokers with COPD (FEV1 < or = 75% predicted) and 15 without COPD (FEV1 > or = 85% predicted) in pneumectomy specimens that were removed for lung cancer. Mast cells, macrophages, neutrophils, eosinophils, T cells, and B cells were identified using immunohistochemistry on formalin-fixed, paraffin-embedded specimens. These cells were quantified in the epithelium and the remainder of the airway wall. The number of mast cells and macrophages in the epithelium, but not in the remainder of the airway wall, was significantly increased in patients with COPD. Neutrophil and T cell numbers did not differ between the groups. Only few B cells and eosinophils were present in both groups. Smoking history, perioperative steroid usage, tumor localization, or reversibility in the FEV1 to salbutamol could not account for the observed differences. We conclude that the number of epithelial mast cells and macrophages is increased in the bronchioli in smokers with airflow limitation, suggesting a role in development of COPD.

[Clinical significance of detection of PML-RAR alpha fusion gene of APL during differentiation therapy with ATRA].

OBJECTIVE: To characterize of the expression patterns of PML-RAR alpha fusion gene during differentiation therapy with ATRA. METHODS: A "neated" RT/PCR was established and subsequently used to study on 74 Acute Promyelocytic Leukemia (APL). RESULTS: Two major isoforms (type S and L) were detected in leukemia cells from 44 novel patients with APL. 19 of 44 (43.2%) patients with type S isoforms were accompanied with poor prognosis. 15 cases were detected sequentially by the RT/PCR method for dynamic observation of MSD during postremission therapy with ATRA and chemotherapy alternatly. The positive rate was 46.7% in 6 months after CR, then the positive rate curve tended to down wards gradually from 33.5% (12 months after CR) to 26.7% (24 months after CR), 11.1% (36 months after CR), 8.3% on 48 months after CR. Other 30 cases of APL which are long-term survivals and received the same postremission program were detected by the RT/PCR method. The expression of the fusion gene reversed positivty in two cases on 56 and 60 months respectively, then both relapsed. One case was detected regularly once a year for 3 times. In another case, the fusion gene was detectable whoever was no any abnormal evidence in peripheral blood and bone marrow 26 cases were negative and had a long-term survival CCR of 47-114 months (medium 66). CONCLUSION: The detection of RT/PCR PML-RAR alpha fusion gene plays an important role in clinical diagnosis, evaluation of prognosis and prediction of relapse in APL.

Brain substrates activated by electroacupuncture of different frequencies (I): Comparative study on the expression of oncogene c-fos and genes coding for three opioid peptides.

Low and high frequency electroacupuncture (EA)-produced analgesia have been shown to be mediated by different brain substrates and different opioid peptides. In this study, Fos-like immunoreactivity (FLI) and in situ hybridization of the three opioid mRNAs were used to examine the effect of low (2 Hz) and high (100 Hz) frequency EA on neuronal activities, and the expression of opioid genes. 2 Hz and 100 Hz EA induced a markedly different spatial patterns of Fos expression in the rat brain, suggesting there are distinct neuronal pathways underlying EA of different frequencies. Likewise, 2 Hz and 100 Hz EA exert differential effects on opioid gene expression: while 2 Hz EA induced a more extensive and intensive preproenkephalin (PPE) mRNA expression than 100 Hz EA, it had no effect on preprodynorphin (PPD) mRNA expression which was significantly increased by 100 Hz EA stimulation. In contrast, EA of both frequencies did not affect POMC mRNA expression.

Brain substrates activated by electroacupuncture (EA) of different frequencies (II): Role of Fos/Jun proteins in EA-induced transcription of preproenkephalin and preprodynorphin genes.

Antisense oligodeoxynucleotides (ODNs) of c-fos and/or c-jun were used in this study to investigate the role of Fos and Jun proteins in electroacupuncture (EA)-induced transcription of the opioid genes, preproenkephalin (PPE), preprodynorphin (PPD) and proopiomelanocortin (POMC). As the results showed, EA-induced Fos and fun expression was blocked efficiently and specifically by e-fos and c-jun antisense ODNs, respectively. This treatment significantly prevented EA-induced PPD, but not PPE, mRNA expression. These results suggest that Fos and Jun proteins are involved in PPD rather than PPE gene transcription activated by EA stimulation.
