11,12-Secoprostaglandins. 2. N-acyl-N-alkyl-7-aminoheptanoic acids.

A series of N-acyl-N-alkyl-7-aminoheptanoic acids has been prepared and evaluated for their ability to mimic the natural prostaglandins in certain biological systems. These compounds can be regarded as 8-aza-11,12-secoprostaglandins and, indeed, most of them stimulate cAMP formation in the mouse ovary assay, just as is observed with the natural prostaglandins. Selected compounds from this series also have been studied and shown to have prostaglandin-like effects in vivo.

Parathyroid carcinoma in familial hyperparathyroidism.

Two families with hereditary hyperparathyroidism are described. One member of each family developed a parathyroid carcinoma. In one case this recurred locally and metastasised. This patient showed hyperplasia of one of the three other parathyroid glands. It is possible that the different parathyroid lesions found in familial hyperparathyroidism may be the result of a progression from hyperplasia to formation of benign or malignant tumours. The remainiing hyperplastic glands may be suppressed by hypercalcaemia. There was no evidence of multiple endocrine neoplasia in either family. Three members of a first family had ichthyosis and both affected members of the second had tumours of the jaw, one of which was an ossifying fibroma, suggesting a possible association of these conditions with familial hyperparathyroidism.

Ion metabolism in a halophilic blue-green alga, Aphanothece halophytica.

The intracellular ion content of the halophilic blue-green alga, Aphanthece halophytica was studied as a function of age, external sodium and external potassium concentration. Intracellular Na+ was found to be about 0.38 millimoles/g dry mass. Intracellular K+ concentrations were as high as 1 M and varied directly with external salinity. Intracellular Ca++ and Mg++ were in the range previously reported for fresh water blue-green algae despite their extremely high extracellular concentrations. Average cell size is consistent at room temperature with two exceptions. When the outside K+ is lower than 6.5 mM the cells tend to be smaller with less intracellular K+ and high Ca++. In stationary phase cultrues the cells are larger with high intracellular Mg++ and low K+.

The carriage of Candida albicans in the mouths of rats treated with tetracycline briefly or for a prolonged period.

Rats given tetracycline in their drinking water for one week were orally inoculated with Candida albicans in the following week. Colonization of the mouth by the fungus resulted, whether the rats continued to receive tetracycline or not, over a period of 22 weeks. Histological changes indicative of oral candidosis were also found both in rats maintained on tetracycline throughout the experiment and in animals given the drug only initially. It is suggested that exposure to tetracycline as tested in this experiment causes a lasting reduction in the rat's ability to expel C. albicans, or an enhancement of the organism's colonizing propensities.

The histology of prolonged candidal infection of the rat's tongue.

Long-term candidal infection of the rat's tongue has been found to produce marked epithelial hyperplasia with keratin trapped between the folds of hyperplastic epithelium. The epithelium showed atypia which did not amount to carcinoma in situ. Degenerative changes were found in the superficial layers of the lingual muscle beneath areas of candidal infection, and after a year, hyalinized fibrous tissue appeared to replace the damaged muscle.
