Changes in adhesion complexes define stages in the differentiation of lens fiber cells.

PURPOSE: During their differentiation, lens fiber cells elongate, detach from the lens capsule, associate at the sutures, and degrade all cytoplasmic membrane-bound organelles. Changes in the expression or organization of cell adhesion and cytoskeleton-associated proteins were correlated with these events during fiber cell differentiation in chicken embryos. METHODS: Fixed or living lenses were sliced with a tissue slicer, permeabilized or extracted with detergents, stained with antibodies or fluorescent-labeled phalloidin, and viewed with a confocal microscope. The distribution of N-cadherin in elongating and mature fiber cells was determined by Western blot analysis. Reverse transcription-polymerase chain reaction (RT-PCR) was used to determine the distribution of vinculin and paxillin transcripts. RESULTS: Staining for N-cadherin and band 4.1 protein decreased soon after fiber cells detached from the capsule. Detergent extraction of lens sections and Western blots of dissected lens regions showed that much of this decrease in staining was due to epitope masking. Vinculin immunoreactivity was barely detectable on the lateral membranes of elongating fiber cells but increased markedly once these cells reached their maximum length and formed the sutures. Staining for paxillin was also low in elongating fiber cells but increased late in fiber cell differentiation, just before the cells destroyed their membrane-bound organelles. Spectrin and ankyrin immunoreactivity did not change when fiber cells reached the sutures. Staining for F-actin increased transiently in cells that had just reached the sutures. Messenger RNAs for vinculin and paxillin were more abundant in maturing than in elongating fiber cells. CONCLUSIONS: The adhesion complexes of lens fiber cells change in organization and composition soon after these cells finish elongating and detach from the capsule. Increased staining for vinculin and paxillin defines distinct stages of fiber cell differentiation that are intermediate between the completion of cell elongation and the time when lens fiber cells degrade their membrane-bound organelles. Remodeling adhesion complexes during fiber cell maturation may assure the stability of fiber-fiber associations, once these cells are no longer transcriptionally active.

[The influence of interleukin-1 on nerve growth factor secretion in newborn rat astrocytes in vitro].

OBJECTIVE: To study the nerve growth factor (NGF) expression and the influence of IL-1 alpha or IL-1 beta on NGF secretion in newborn rat astrocytes. METHODS: Astrocytes obtained from the brain cortex of newborn rats were cultured and purified, and they were divided into three groups, experimental, control and blank groups. IL-1 alpha or IL-1 beta were added into the experimental group with 25, 50 and 100 U/ml, each group was cultured for 24, 48 or 72 hours, and then the NGF contents in cultured astrocytes suspension media were measured by a two-cite enzymelinked immunoserbent assay (ELISA). RESULTS: Astrocytes could secret NGF by themselves and each concentration of IL-1 alpha or IL-1 beta media at any testing time could enhance NGF secreting in newborn rat astrocytes in certain degrees. The effects of IL-1 beta were stronger than IL-1 alpha, the strongest effect in the unit time was observed in IL-1 beta with 50 U/ml for 24 hours. CONCLUSION: Astrocytes can express NGF, and IL-1 alpha or IL-1 beta can enhance the NGF expression in newborn rat astrocytes.

The metabolism of fatty acids in human Bietti crystalline dystrophy.

PURPOSE: To investigate the role of abnormal lipid metabolism in Bietti crystalline dystrophy. METHODS: Cultured human lymphocytes and fibroblasts from patients with Bietti crystalline dystrophy (BCD) were incubated in the presence of [(14)C]18:3n-3 or [(14)C]18:2n-6. Incorporation into the cellular lipid pools and further metabolism by desaturation or elongation were monitored by thin-layer chromatography and HPLC. Results were compared with those in normal control subjects and patients with Wolman disease (WD). RESULTS: Pulse-chase experiments with labeled fatty acids in all groups showed that, after 1 hour, radioactivity was largely confined to the triacylglyceride (TG) and choline phosphoglyceride (CPG) pools. However, after several hours, radioactivity was transferred from the TG and CPG pools, some going to the serine and ethanolamine phosphoglyceride (SPG and EPG) pools. Fibroblasts from all groups showed direct transfer of fatty acids (FAs) into CPG and EPG. Incorporation of labeled FAs into the EPG pool paralleled extensive desaturation and elongation of 18:2n-6 to 22:5n-6 and 18:3n-3 to 22:6n-3. Fibroblasts from patients with WD (a lysosomal acid lipase deficiency characterized by excessive lipid accumulation), showed higher incorporation of 18:2n-6 into TGs than did normal or BCD fibroblasts. Conversely, fibroblasts from patients with BCD showed lower conversion of 18:3n-3, but not of 18:2n-6, into polyunsaturated FAs (PUFAs) than those of normal subjects or patients with WD. This was true for total FAs, CPGs, and EPGs. Similar results were found in both fibroblasts and lymphocytes; however, unlike fibroblasts, lymphocytes from normal subjects showed similar levels of incorporation of FAs into EPGs and CPGs. In contrast, incorporation of 18:3n-3 into EPGs was decreased in lymphocytes from patients with BCD. CONCLUSIONS: BCD is characterized by a lower than normal conversion of FA precursors into n-3 PUFA, whereas there is a higher than normal level of n-6 and n-3 FAs incorporation into TGs in cells from patients with WD. These findings raise the possibility that abnormal lipid metabolism associated with BCD is the result of deficient lipid binding, elongation, or desaturation in contrast to the lysosomal acid lipase deficiency found in Wolman disease.

Current practice and future directions in pathology and laboratory evaluation of the sentinel node.

We developed the techniques of lymphatic mapping and sentinel node (SN) biopsy to improve the management of patients with high-risk (thick and deep) primary melanoma. The SN is the first lymph node on the direct lymphatic drainage path from the primary tumor. This node is uniquely immune-modulated by the primary tumor and is the node most likely to contain the earliest stages of metastases. Accurate assessment of the SN requires careful evaluation of multiple sections removed from the areas of the node most likely to contain tumor. These sections are stained with hematoxylin and eosin and by immunohistochemistry with antibodies directed to tumor-associated markers (S-100, HMB-45, and Melan-A/MART-1) in the case of melanoma and to cytokeratins for breast cancer. Studies are in progress to determine whether molecular biology techniques will detect additional nodes that contain truly occult tumor deposits.

[Complications of third ventriculostomy for hydrocephalus].

OBJECTIVE: To explore the reason of complications of third ventriculostomy and the method for reducing complications. METHODS: Fifteen third ventriculostomies and the related complications were analyzed retrospectively. RESULTS: Two patients had ventricular infection, one subdural fluid collection, one obstraction of fenestration site by neoplasm, one delayed herniation due to hematoma-occupied third ventricle. The complications were due to the inferior quality of the equipment and poor experience. CONCLUSIONS: Advanced equipment and understanding of possible complications can reduce complications.

[Caloric value and energy allocation of Chloris virgata in northeast grassland].

The rules of seasonal changes in caloric values of individual plant, stem, and leaves of Chloris virgata were similar, which had two peak values from early July to early August, and then decreased gradually. Those of inflorescence assumed U shape, and had two peak values in early August and middle September, respectively. The seasonal changes in caloric values of dead standing were irregular, and the maximum value was appeared in early August. The seasonal changes in existent energy value of the aboveground parts in Chloris virgata population presented double peak curve. The two peak values were appeared in early August and early September respectively, and the maximum value was 7381.27 kJ.m-2 in early September. The energy allocation in different seasons was leaf > stem in early July, stem > leaf > dead standing in middle July, stem > leaf > inflorescence > dead standing in August, stem > inflorescence > leaf > dead standing in early September, and stem > inflorescence > dead standing > leaf in middle September. The vertical structure of energy in the aboveground parts was that the energy value gradually increased from the earth's surface to 20 cm high, and then decreased. The maximum value, which accounted for 25.75% of energy in the aboveground parts, was appeared in the layer of 10-20 cm high. In the underground parts, the energy value progressively decreased with the increase of depth, and the maximum value, which accounted for 74.21% of energy in the underground parts, was appeared in the layer of 0-10 cm depth.

[Adsorption of weak deep blue GR on hydroxy-aluminum montmorillonite].

Hydroxy-alumium montmorillonite may serve as potential adsorbents for organic pollutants from wastewater. It was synthesized by titrating Na-montmorillonite with hydroxy-aluminum solution (OH-/Al3+ molar ratio was 2.4). From powder XRD data, it was shown that the interlayer(d001) of montmorillonite was increased greatly from 12.58A to 18.63A. Seven modified montmorillonite samples, including hydroxy-aluminum montmorillonite, were used to remove the dye of weak deep blue GR from water. The results showed that hydroxy-alumium montmorillonite had very high adsorption ability, only 0.0200 g could treat 25 ml weak deep GR with 95% removal. The different adsorption mechanisms of weak deep blue GR on modified montmorillonite: surface adsorption, ion-exchanging and partion, were suggested.

[A prospective study on the association between underweight and mortality from all cause].

OBJECTIVE: To explore the association between underweight and mortality from all cause. METHOD: We examined the relationship of underweight and mortality among subjects who participated in the survey of cardiovascular risk factors (in 1984-1986 and in 1992). Relative Risks (RRs) were estimated from the COX proportional hazard model by using a BMI between 18.5-24.9 kg/m2 as reference category. RESULTS: During an average 12.40 years follow-up, we documented 307 deaths of which 158 were from cancer, 93 from cardiovascular diseases, and 56 from other causes. We observed a L-shaped relation between BMI and overall mortality. (P for trend < 0.01); In multivariated analyses, we found that the RR of mortality for a BMI of < 18.5 kg/m2 was 1.58 (95% CI 1.16-2.14) as compared with the risk among subjects with BMI of 18.5-24.9 kg/m2. The RR of overall mortality among subjects with BMI of < 18.5 kg/m2 was still significant after the exclusion of subjects who died from cancers during the first five years follow-up. CONCLUSION: Total mortality was increased among underweight people, and this association can not been completely explained by smoking and early deaths from cancer.

Characterization of hemagglutinin gene of influenza A virus subtype H9N2.

OBJECTIVE: To determine the origin of human influenza A (H9N2) virus and the relationship among H9N2 strains isolated from different hosts, on the basis of molecular biology. METHODS: Viruses were passed in embryonated hen eggs, and virion RNA was extracted from allantoic fluid and reverse transcribed to synthesize cDNA. cDNA was amplified by PCR and the PCR product was purified with a purification kit. Afterwards RNA sequence analysis was performed by dideoxynucleotide chain termination and a cloning method. Finally, phylogenetic analysis of the sequencing data was performed with MegAlign (version 1.03) and Editseg (version 3.69) softwares. RESULTS: The amino acid sequences at the cleavage site between HA1 and HA2 domains of H9N2 viruses isolated in China are R-S-S-R. One pigeon strain contains seven potential glycosylation sites on the HA protein molecule, while all others have eight. There are 2 to 15 differences of amino acid sequences distributed at 24 different positions on the HA protein molecules among six H9N2 viruses. The H9N2 viruses with multiple lineages of HA genes were co-circulating in China recently. CONCLUSION: The highest possibility is that human influenza A (H9N2) virus was derived from Chicken H9N2 virus, and not derived from pigeon H9N2 virus. However, it is still unknown whether the H9N2 virus could transmit from person to person. The H9N2 viruses with multiple lineages of HA genes are co-circulating in China.

The relationship between plasma apolipoprotein A-IV levels and coronary heart disease.

OBJECTIVE: To evaluate the relationship between plasma apoA-IV levels and coronary atherosclerosis and to explore its relation to other risk factors. METHODS: Using ELISA techniques, plasma apoA-IV levels were quantified in 181 patients who underwent coronary angiography (CAG). Patients were divided according to their coronary status into a coronary heart disease (CHD) group (stenotic lesion on CAG, n = 118) and a control group (normal CAG, n = 63). The severity of atherosclerosis was assessed by stenosis scoring of the different lesions. Other parameters, including apoA-I, apoB, Lp(a), HDL-C, LDL-C, TG, and TC, were measured as well. Univariate, logistic regression analyses were used to define the relationship between coronary atherosclerosis and plasma apoA-IV levels. RESULTS: When compared with the control group, plasma apoA-IV levels were found to be lower in the CHD group. There was a weak negative correlation between plasma apoA-IV levels and the severity of coronary atherosclerosis. ApoA-IV was found to be a relatively independent risk factor for CHD. We also found a positive correlation between apoA-IV and triglyceride levels. CONCLUSIONS: ApoA-IV may be important in the prediction of CHD and coronary atherosclerosis severity. It may also play an important role in the metabolism of triglycerides.

Histochemical study of transmyocardial laser channels in dogs.

OBJECTIVE: Laser-induced myocardial injury was investigated in a canine transmyocardial laser revascularization model. METHODS: A total of 23 channels were created in the left ventricles of 5 animals with a CO2 laser. Four animals were sacrificed immediately and one at week 2. Lactate dehydrogenase and succinate dehydrogenase in channel regions were examined with a Nitro-BT method staining. RESULTS: Immediately after the procedure, laser channels and surrounding injured myocardium could be stratified from the center to the border into 5 concentric zones, i.e., vaporized zone, carbonized zone, enzyme-disappeared zone, enzyme-decreased zone and enzyme-concentrated zone. Two weeks later, the phenomenon disappeared. CONCLUSIONS: Immediately after transmyocardial revascularization, there is a ring of enzyme-concentrated zone at the outermost region of injured myocardium. Two weeks later, enzyme activities of reversibly injured myocytes appear to have recovered.

Gene transfer and expression in rat anastomotic artery in vivo using adenoviral vector.

OBJECTIVE: To observe the efficiency and time course of gene expression and the safety of adenoviral vector mediated gene transfer in vivo. METHODS: After soaking soluble stents in a high concentration of glucose solution containing Adv5-CMV (cytomegalovirus) (control group) or Adv5-CMV/LacZ (treatment group) for 30 minutes, the stents were inserted into the lumina of cut rat carotid arteries and end-to-end anastomoses of the cut carotid were performed with standard microvascular surgical techniques. On days 2, 7, 14, 28, 60 and 90 after gene transfer, anastomotic arteries of the two groups were observed. On days 7 and 14, the ascending aortas, hearts, brains, livers, lungs, spleens and kidneys of the treatment group were observed. All samples were analyzed for the presence of beta-galactosidase activity and histochemical staining. RESULTS: beta-galactosidase activity was not detected in the carotid arteries of the control group and organs not directly exposed to adenoviral vector of the treatment group. The amount of beta-galactosidase activity (x 10(-3) U/g tissue) in the treatment group on the 2nd, 7th, 14th, 28th, 60th and 90th day after gene transfer was 3.87, 11.38, 9.8, 6.43, 3.18 and 2.43, respectively. Microscopic examination of sections from vessels of the control group and from the aortas, hearts, brains, livers, lungs, spleens or kidneys of the treatment group revealed no X-gal staining. Microscopic examination of carotid arteries of the treatment group revealed blue-staining in all anastomotic arteries and in all layers of the arterial wall observed on days 7 and 14 after gene transfer. CONCLUSION: Adenoviral vector can effectively infect blood vessels in vivo. After adenoviral vector mediated direct gene transfer into anastomotic rat carotid arteries, recombinant gene expression began on day 2, peaked between days 7 and 14, prominently declined after day 28, and persisted at low levels more than three months. A recombinant gene could be delivered to a specific site by direct gene transfer in vivo by adenoviral vector infection.

[Study on effect of delivery way on thyrotropin levels in pregnant women and their newborns].

OBJECTIVE: To investigate the effect on thyrotropin (TSH) levels of pregnant women and their newborns in different delivery ways. METHODS: The serum TSH levels of 213 pregnant women and the umbilical cord blood TSH levels of their newborns (140 cases from normal vaginal delivery, 38 cases from cesarean section, 35 cases from low forceps delivery) were tested by immunoradiometric assay. Urinary iodine was detected from pregnant women before delivery. RESULTS: In pregnant women, the mean serum level of TSH in the forceps group [(4.13 +/- 0.69) mU/L] was significantly higher than those of the normal birth group [(2.58 +/- 0.87) mU/L, P < 0.01] and the cesarean section group [(2.81 +/- 0.45) mU/L, P < 0.01]. There was no difference in TSH level between the normal birth group and the cesarean section group. In newborns, the mean TSH level in the forceps group [(8.85 +/- 2.48) mU/L] was significantly higher than those of the normal birth group [(5.36 +/- 2.23) mU/L, P < 0.01] and the cesarean section group [(3.84 +/- 2.16) mU/L, P < 0.01]. The mean TSH level in the normal birth group was significantly higher than that of the cesarean section group (P < 0.05). The serum TSH levels of pregnant women were positively correlated with the umbilical cord blood TSH levels of their newborns in three groups. CONCLUSIONS: The contraction of uterus is the stimulus for the surge in TSH levels of pregnant women and their newborns after delivery. It's reliable that the TSH levels of newborns could be evaluated by monitoring the TSH levels of pregnant woman.

[Study on intergenera fusion of protoplasts from Rhizobium leguminosorum and Sinorhizobium xinjiangnesis].

Penicillin and chloromycetin were regarded as the sign of resistance to antibodies of R. leguminosorum USDA2370 and S. xinjiangnesis CCBAU110 respectively. Using the protoplast fusion technique, USDA2370 and CCBAU110 were successfully fused. Fusion hybrid can inoculate in the leguminous of parental strains respectively. There were apparent differences between parents and fusion hybrid in cell morphology, colony and pattern of whole-cell protein. The values of DNA homology between fusion hybrid and USDA2370 and CCBAU110 were 56.6% and 10.2% respectively.

[The application of laser therapy plus endobronchial stenting in treatment of complete airway obstruction].

OBJECTIVE: To assess the efficacy of laser therapy plus endobronchial stenting in the treatment of complete airway obstruction. METHODS: 17 cases with complete airway obstruction were treated with Nd: YAG laser and endobronchial stenting from April 1998 to November 2000. The patients' dyspnea, airway gauge, pulmonary function and complications were observed. RESULTS: The airways were all restructured following a single or multiple laser treatment; the airway gauge enlarged and the average diameter increased (5.82 +/- 1.01) mm. Immediate relief of dyspnea was achieved in sixteen patients and dyspnea index lessened one grade in 4 patients, 2 grades in 11 and 3 grades in one. Obvious improvement of forced expiratory volume in one second (FEV(1)) was noted in all patients;the improvement averaged (0.49 +/- 0.18) L. Endobronchial stents were placed for 14 patients after laser treatment. After stent placement, the airway was further opened and the average airway gauge increased (3.50 +/- 0.94) mm further in diameter. Patients' dyspnea and FEV(1) were further improved; dyspnea index lowered one grade in eight patients and 2 grades in two patients. FEV(1) improved further (0.41 +/- 0.20) L in average. Complications after laser therapy included hypoxemia in 15 patients, hemorrhage in 2 and postoperative dyspnea in 3. Follow-up duration ranged from 2-20 months. Complications after stenting included recurrent pneumonia in 4 patients, airway obstruction due to granuloma or tumour growth in 5 and stent migration in 1. CONCLUSION: Laser therapy and endobronchial stent placement were effective in the treatment of patients with totally occluded airway. Nd: YAG laser can be first chosen to restructure the occluded airway, then endobronchial stent must be used to further enlarge the airway and maintain the therapeutic effect.

[Biosorption of heavy metals by bacteria isolated from activated sludge].

In this paper, biosorption efficiency of heavy metals by bacteria isolated from activated sludge was tested. It was shown that Pseudomonas pseudoalcaligenes and Micrococcus luteus GC subgroup B had high biosorption capacities for Cu(II) and Pb(II) ions. The adsorption of the two ions on Pseudomonas pseudoalcaligenes and Micrococcus luteus GC subgroup B exhibited Langmuir adsorption behavior with regression coefficient of 0.99. The pH value was the most important effect factor on biosorption of metal ions. The biosorption kinetic curves of Cu(II) and Pb(II) ions by cells showed two processes: rapid sorption on cell surface and slowly longer-term transformation into cells. Cu(II) ion can be desorbed effectively from cells of Micrococcus luteus GC subgroup B.

[A study for the relationship between serum calcitonin gene-related peptide and coronary arteriopathy].

OBJECTIVE: To investigate the relationship between serum calcitonin gene-related peptide (CGRP) and coronary angiopathy. METHODS: In a cross-sectional study, serum CGRP levels of 119 patients with coronary heart disease (CHD), 78 with non CHD and 39 healthy subjects were measured with radioassay method. The information on conventional risk factors were collected by interviews. RESULTS: The development of coronary angiopathy was correlated with smoking, diabetes and aging. The mean serum CGRP level was significantly lower in CHD than in NCHD patients [(35.5 +/- 48.8) mumol/L vs (63.8 +/- 47.3) mumol/L, P < 0.01). There was no difference between the mean serum CGRP level in NCHD patients and healthy controls. By multivariate logistic regression, the odds ratios (OR) of aging, smoking and diabetes were all > or = 1 (P < 0.01), that means they are independent risk factors for the development of coronary angiopathy. The OR of CGRP was < or = 1, indicating that it was an independent protective factor. CONCLUSIONS: It is shown that aging, smoking and diabetes are all independent risk factors for the development of angiopathy, while CGRP is an independent protective factor for the development of coronary angiopathy.
