Antibodies against purified pigeon IgA in pigeon breeders' disease.

It is demonstrated that previously described PDF1-A antigens from pigeon dropping extract contain pigeon IgA. The sera of patients with pigeon breeders' disease contain precipitating antibodies against pigeon IgA, in contrast to the sera of pigeon breeders suffering from unrelated forms of pulmonary dysfunction. The degree of complement consumption by PDF1A antigens, pigeon serum, and pigeon IgA turned out to be correlated. No correlation was found between precipitating anti-pigeon IgA antibodies and complement consumption by pigeon IgA in patients' sera.

Quantitation of lysozyme in the urine of the dog for the diagnosis of proximal tubular damage in renal disease.

The serum and urine LZM levels were determined with the lysoplate assay in 26 normal experimental dogs and in 21 dogs with various renal diseases. In the dogs with decreased glomerular filtration the serum LZM levels were higher than in normal dogs (less than or equal to 1.3 mg/l). No LZM could be demonstrated in the urine of normal dogs. The presence of LZM in urine was associated with proximal tubular damage, which indicates that LZM-uria can be used as an index of renal tubular damage.

Lysozyme and its presence in bovine milk and serum.

The literature on the presence of lysozyme in various biological fluids in physiological and pathological conditions is reviewed. Preliminary results of lysozyme estimations in bovine milk show that lysozyme levels are definitely higher in colostrum and mastitis milk than in normal milk. The biological role of the enzyme in the milk still has to be defined. On the other hand, lysozyme may interfere with microbiological screening techniques for penicillin in cow's milk; the lytic effect of purified human lysozyme on B. stearothermophilus var. calidolactis was demonstrated microscopically.

New immunofluorescent antibody test for diagnosis of candidiasis using macrophages-engulfed antigens as substrate.

A new method for detection of antibodies to polysaccharidic antigens of Candida albicans is described. The method is based on the selective uptake of carbohydrate-rich antigens by mouse peritoneal macrophages, the antibodies being revealed by indirect immunofluorescence after incubation of fixed, antigen-containing macrophages with sera (MU/IFA test). This test shows greater sensitivity than counterimmunoelectrophoresis individuals. The combination of CIE using cytoplasmic antigens and MU/IFA using surface antigens and MU/IFA using surface antigens is useful in a considerable majority of cases for diagnosis of clinical infection.

Protein A reactivity of various mammalian immunoglobulins.

Serum samples and immunoglobulin fractions of eight mammalian species were applied to a Sepharose--protein A column. As with the human immunoglobulin subclasses IgG1, IgG2 and IgG4, all examined IgG classes and subclasses were bound to a greater or lesser extent to protein A. However, the binding of IgG1 of ruminants was very poor. Polyclonal IgM and IgA of the pig, the dog and the cat may be separated in protein A reactive and protein A non-reactive fractions. In addition, monoclonal canine IgM and IgA partially reacted with protein A. In combination with methods such as ammonium sulphate precipitation, ion exchange chromatography and gel-filtration, affinity chromatography with protein A is recommended for the rapid purification of certain Ig (sub)classes of a number of mammalian species.

Pigeon IgA: a major antigen in pigeon breeder's disease.

Pigeon IgA has been isolated from crop-milk and an antiserum was produced. Extracts of the dust in pigeon coops contained detectable amounts of pigeon IgA, but not IgG. The sera of patients with pigeon breeder's disease contained precipitating antibody against pigeon IgA.

Skin grafting and graft rejection in goats.

As part of a study to define the major histocompatibility complex of the goat and to show certain aspects of its biological function, a skin-grafting technique is described which might also be applicable for similar studies in other ruminant species.

Quantification of turkey immunoglobulins IgA, IgM and IgG in serum and secretions.

The immunoglobulins IgA, IgM and IgG of the turkey were quantitated in individual serum samples as well as in pooled sera. The variability of Ig levels in normal, healthy birds was quite high: IgA: mean value 0.633 mg/ml (4.0 -x - 2.5 -x); IgM: mean value 4.37 mg/ml (0.5 -x - 1.4 -x) and IgG: mean value 8.92 mg/ml (0.6 -x - 1.7 -x). Immunoglobulin levels in egg-yolk, egg-white, bile and some intraocular tissues were quantitated as well. An interesting finding was, that the forementioned variability was by far not so high with respect to IgG levels in 20 egg-yolk samples: mean value 5.1 mg/ml (0.86 x- - 1.17 -x). Though IgG and IgM could be detected in pooled turkey bile, IgA predominated in this secretion. In aqueous humor, iris tissue and vitreous body only IgG could be detected.

[Immunoglogulins in swine. (A) A review supplemented by a report on findings in personal studies (author's transl)].

the literature is reviewed and the importance of studying immunoglobulins in farm animals, particularly swine, is stressed. Possible methods of clinical immunological investigation in pigs are suggested. The literature as well as personal studies show that the immunoglobulin titre in the serum of piglets continues to be very low over a prolonged period from the seventh day post partum. This incomplete state of immunity should be taken into account on weaning piglets.

[Immunoglobulins in swine. (B) Immunohistological studies on the localization of cells containing immunoglobulin in the tissues of the small intestine and spleen (author's transl)].

Antisera to porcine immunoglobulins, conjugated with flurescein isothiocyanate, were used in studies on the localization of cells containing immunoglobulin in the tissues of the intestine and spleen in pigs. Cells containing IgM and IgA were found to be present in the lamina propria of the intestinal mucosa, the villous membrane showing fluorescence due to liberated immunoglobulins. Cells containing IgG and IgM were detected in the marginal areas of the splenic lymph nodes but could not be definitely identified as mature plasma cells.

Urinary lysozyme phenotypes in monocytic leukemia.

A method of two-dimensional electrophoresis has been devised to allow the study of the electrophoretic mobility of urinary lysozyme. Three different phenotypes have been defined in a study of thirteen purified lysozymes obtained from different patients with monocytic leukemia.

[Protothecosis in cattle. Report of a case (author's transl)].

A brief review of the classification of the genus Prototheca is followed by a report on a case of protothecosis in cattle. A pure culture of Prototheca was isolated from the milk of the affected quarter. Despite the fact that the alga showed some sensitivity of nystatin in vitro, the mastitis failed to disappear on treatment with this agent. In the discussion, the incidence of Prototheca infection in man and animals and the appearance of infections with this facultatively pathogenic alga are reviewed.

Three immunoglobulin classes in the pigeon (Columbia livia).

Three classes of immunoglobulins have been identified in the pigeon. IgG and IgM were purified from pigeon serum whereas IgA was isolated from pigeon hepatic bile. Pigeon IgG and IgM had the same properties and immunohistological distribution as their chicken homologues. Pigeon IgA was identified on the following grounds: (1) it contains the same light chains as pigeon IgM and IgG; (2) it is relatively abundant in exocrine secretions such as bile, egg white, cropmilk and intestinal fluid, whereas it is present only in small amounts in serum; (3) it occurs in the cytoplasm of the majority of the immunocytes from the intestinal mucosa; (4) its electrophoretic mobility and molecular size are similar to those of chicken IgA. Surprisingly, no immunoglobulin-containing cells could be detected in sections of the wall of the cropmilk gland, despite the high IgA content of the cropmilk.

Multiple myeloma, IgA cryoglobulinemia and serum hyperviscosity in a dog.

Clinical signs of hyperviscosity syndrome in a 6-year-old dog included listlessness, polydipsia, anorexia, vomiting, and recurrent bleeding from the gums. Fundoscopy showed the typical retinal changes associated with this syndrome. A polymeric IgA cryoglobulin characterized in the dog's serum was later isolated and structurally studied. It was found to contain J chain but no secretory component. The heavy and light chains were covalently bound.
