Synthesis and biological activity of a linear fragment of the atrial natriuretic factor (ANF).

A linear fragment of the atrial natriuretic factor, ANF(106-125), unable to form an intramolecular cystine bridge, was synthesized by the solid-phase method. The fragment showed smooth muscle relaxant activity in the rabbit aorta and chick rectum assays, an inhibitory effect on aldosterone secretion from bovine adrenal zona glomerulosa cells, and had affinity for specific ANF receptors located in zona glomerulosa cell membranes. The potency of ANF(106-125) in these four assay systems was about two to three orders of magnitude lower than that of ANF(103-125) which contains the intact cyclic structure. The obtained results indicate that the disulfide linkage stabilizes the bioactive conformation of ANF peptides but is not an absolute requirement for biological activity.

[Modifications of disulfiram and its metabolites in blood in alcoholic hepatopathy (author's transl)].

In cirrhotic, steatosic and healthy subjects, the authors studied the metabolism of disulfiram (TETD) administrated orally (500 mg) 3 consecutive days. Carbon disulfide (CS2) and the whole TETD, diethyl dithiocarbamate (DDC) disulfides, were determined by a gas chromatographic method. The evolution of metabolites is similar in steatosic and healthy subjects. In cirrhotics the CS2, DDC and disulfides level increase within the 3 days. This phenomenon may be related with hepatic toxicity of TETD and with the persistence of disulfiram-alcohol reaction. The authors suggest to use low doses of disulfiram in cirrhotic.

Acute bronchoconstriction caused by Beclovent and not Vanceril.

A 24-year-old asthmatic patient reported acute wheezing immediately after using an aerosol of beclomethasone dipropionate (Beclovent). She was hospitalised for bronchial challenges. Immediate bronchoconstriction was shown after two puffs of this aerosol and after inhalation of the placebo containing all the products of the aerosol less the beclomethasone. Another brand of beclomethasone aerosol, Vanceril, did not produce any reaction. Since the contents of these two beclomethasone aerosols are similar, the authors suggest that rubber or plastic derivative(s) present in the metering valve might have been responsible for the reaction.

[Kinetics of the pulmonary clearance of inhaled cadmium and its accumulation in rat liver and kidneys].

Pulmonary clearance rate of cadmium (Cd), inhaled in the form of an aerosol of Cd acetate, is very slow (half-life 53 days). However, results demonstrate that an important part of Cd cleared from the lung is recovered in liver and kidney. Liver binds this metal rapidly but to a limited extent, whereas renal accumulation is slow but continuous throughout the three months following exposure.

[Simulation of polluted atmospheres for animal experimentation: toxic gases].

A process for the generation of standardized polluted atmospheres usable for animal exposure is described. It is based on the diffusion of compressed gases across a silicone membrane. This device has been tested with four gases of particular importance in the field of atmospheric pollution (sulfur dioxide, nitrogen oxide, carbon monoxide and ammoniac).
