Microheterogeneity of alpha 1-acid glycoprotein in the sera of patients with cancer or inflammatory states of the ovaries.

alpha 1-Acid glycoprotein (AGP) and its concanavalin A-dependent four microheterogeneous fractions were measured in sera from women with ovarian carcinoma and those with an inflammatory ovarian disease. Total concentrations of AGP in inflammatory and cancerous sera were significantly higher than in control group, but did not differ from each other. In the fourth stage cancer the distribution of Con A-dependent fractions 1, 3 and 4 was significantly different as compared with control, second stage cancer and inflammatory groups.

T-lymphocyte colony formation of murine thymocytes in agar contained in glass capillaries.

Optimal growth conditions are presented for a new colony test with mouse thymocytes in agar contained in glass capillaries. The kinetics of colony growth and the dependence from the PHA-, I1-2-, agar- and 2-mercaptoethanol concentration are shown. The colony forming cells are identified as T-lymphocytes by usual morphology and by an indirect immunoperoxidase method using mouse anti-Thy 1.2 antibodies.

Affino-immunoelectrophoresis of haptoglobin with wheat germ agglutinin. Diagnostic significance in ovarian carcinoma.

Affino-immunoelectrophoresis with wheat germ agglutinin (WGA) was applied to the analysis of microheterogenous fractions of human haptoglobin (Hp), present in cancerous ascitic fluids and in sera from patients either with ovarian carcinoma or with inflammatory ovarian disease. Normal and inflammatory sera contained two WGA-dependent Hp fractions: strongly- and weakly-retarded while cancer sera and ascitic fluids: non-retarded and weakly-retarded fractions. The content of WGA-dependent Hp fractions following curative surgery and chemotherapy of the patients was found to resemble the pattern of the control group.

Synthetic factor for evaluation of the effectiveness of the therapy in ovarian carcinoma.

The synthetic factor based on determinations of 10 parameters in sera of patients with ovarian carcinoma, was constructed. The factor was found to be useful in evaluation of the effectiveness of the cytotoxic treatment and in indication of the recurrence of the malignancy. Reduction of the number of biochemical markers to five (haptoglobin, seromucoid, lactate dehydrogenase, alkaline phosphatase, aspartate aminotransferase), according to significance of particular markers in the laboratory diagnostics of ovarian cancer did not affect the diagnostic sensitivity of the synthetic factor.

Haptoglobin and proteinase inhibitors in the blood serum of women with inflammatory, benign and neoplastic lesions of the ovary.

Blood sera of women with myomas of the uterus, benign or malignant neoplasms of the ovary and with inflammatory conditions were examined by determining the haptoglobin concentration and functional activities of: alpha 2-macroglobulin, alpha 1-proteinase inhibitor and alpha-cysteine proteinase inhibitor. In the women with ovarian cancer, as compared with the control group, a significant increase of haptoglobin, alpha 1-proteinase inhibitor and alpha-cysteine proteinase inhibitor was found (p less than 0.001). On the contrary bioactivity of alpha 2-macroglobulin was depressed (p = 0.01). The observed increase of alpha-cysteine proteinase inhibitor activity in the groups with ovarian tumors and ovarian cysts was not related to the inflammatory state as estimated by the haptoglobin concentration.

Clinical usefulness of serum acute-phase reactants in patients with ovarian tumors.

Concentrations of the following acute-phase reactants (APR) were determined in the sera of patients with benign and malignant ovarian tumors: haptoglobin, ceruloplasmin, sialic acid, seromucoid (in sulphosalicylic acid supernatant), and trypsin inhibitory capacity. In patients with ovarian carcinomas significant increase (p less than 0.001) in all measured APR parameters as compared to healthy women and women with benign tumors was found. Moreover, the levels of haptoglobin, ceruloplasmin and trypsin inhibitory capacity were statistically lower (p less than 0.001) in nonmalignant than in malignant ovarian disease. No distinct statistical difference was confirmed between healthy controls and the benign tumor group as well as between ovarian carcinomas of Stages I--III and Stage IV.

Serum enzymes in ovarian carcinoma.

The following enzymatic activities were measured in serum of patients with benign and malignant ovarian tumors before treatment: alkaline and acid phosphatases, aspartyl (AspAT) and alanyl (AlAT) aminotransferases, leucyl (LAP) and alanyl (AAP) aminopeptidases, lactate dehydrogenase (LDH), gamma-glutamyl transpeptidase, cathepsin, alkaline ribonuclease (RNase) and beta-glucuronidase. It was shown that at least three determinations (phosphatases and LAP) are practically useless in a discrimination between the examined groups. RNase in combination with AspAT (AlAT) or RNase with AAP and LDH were found to give the best results as marker enzymes.

The acoustic reflex and temporary threshold shift: temporal characteristics.

One ear of each of seven normal-hearing subjects was exposed to a continuous 1000-Hz tone at 110 dB SPL for three minutes. During exposure, a broad-band noise at 100 dB SPL was presented to the contralateral ear. The noise was either continuous or pulsed. Four pulsed conditions employed repetition periods of 360, 180, 90, or 9 msec with a 50% duty cycle. A control condition in which no noise was presented was also included. Temporary threshold shift was measured at selected postexposure times at the frequency one-half octave above the exposure frequency. TTS2 was greatest for the control condition and least for the 360- and 180-msec conditions. Results are discussed in relation to the dynamics of the acoustic reflex, particularly reflex relaxation, reflex adaptation, and reflex temporal summation.
