Studies on dynamics of growth and biosynthesis of artemisinin in hairy roots of Artemisia annua L.

Seven hairy root lines with the properties of fast growth and high artemisinin contents were selected from 747 hairy roots induced by transformation of Artemisia annua L. strain 025 with Agrobacterium rhizogenes ATCC15834. The differences of growth rates and artemisinin contents among the 7 selected hairy root lines were extremely significant, of which HR-9 gave the highest yield of artemisinin, reaching 33.25 mg/y.L. The differences of growth rates and artemisinin contents among hairy roots, untransformated roots and callus were also significant. There were no obvious lag phase in batch culture of hairy roots of Artemisia annua L. The exponential growth phase was 7 approximately 15 days after inculation. The growth rate reached the highest on the 11th day, and the cultures reached a stationary phase on the 20th day. The properties of artemisinin content of hairy roots were obviously "related to growth". The artemisinin content decreased slowly during the exponential phase, increased while the growth rate slowed down and remained consistent after the growth stopped. The optimum culture time for hairy roots of Artemisia annua L. was 21 days in our system.

Computer-assisted design and manufacture of posterior full crowns.

OBJECTIVE: To build an experimental system for the computer-assisted design (CAD) of full crowns and to obtain data for computer-assisted manufacturing (CAM) of full crowns. MATERIALS AND METHODS: Three steps are involved in building the system: measuring the prepared tooth, the adjacent mesial and distal teeth, and the opposing teeth by means of digital speckle correlation; reconstructing the shape of the measured teeth; and developing CAD for full crowns. The standard shape of the corresponding tooth was modified to fit the defective tooth on the basis of buccolingual and occlusal-gingival distance as well as the occlusal relationship of the defective tooth. RESULTS: CAD for full crowns was developed and was obtained for CAM of posterior full crowns. CONCLUSIONS: The CAD method for full crowns is convenient and economical and can be used for anterior and posterior crown design, copying, and veneer.

[Relationship between extracellular matrix and progressive growth of malignant tumor].

OBJECTIVE: To explore the role of extracellular matrix proteins in tumor cell invasion and metastasis. METHODS: Mouse pulmonary adenocarcinoma cell line LA795 and human nasopharyngeal carcinoma cell line CNE-2Z were implanted subcutaneously and under the renal capsule in T739 and nude mice, respectively. At different time intervals, expression of fibronectin(FN), laminin(LN) and type IV collagen(IV C) in the tumor extracellular matrix and in tumor adjacent tissues was examined by indirect immunoperoxidase and immunofluorescence techniques. In addition, various in vitro methods (random migration of tumor cells in agarose droplet, growth of tumor cell colonies in soft agar, dot blotting, etc.) were used to analyse the effect of exogenous LN, FN or IV C. RESULTS: As tumor grew, the expression of FN, LN or IV C increased and presented different distribution patterns. Exogenous FN, LN and IV C enhanced the mobility of tumor cells and the growth of the tumor cell colonies. CONCLUSION: The distribution difference and the changes in synthesis and degradation of the extracellular matrix proteins are valuable in predicting the biologic behaviour. Tumor implanted under the renal capsule of mice offers a better experimental model to study the relationship between extracellular matrix and tumor cell invasion.

[HLA-DRB alleles polymorphism in susceptibility to asthma in Beijing Chinese].

OBJECTIVES: To investigate whether susceptibility or resistance to asthma associated with HLA-DRB alleles and analyze the relationship between HLA-DRB genes and clinical phenotype of asthma (TIgE, sIgE, BHR). METHODS: Using PCR-SSP(sequence-specific primer polymerase chain reaction), we tested the frequency distribution of HLA-DRB alleles in 50 asthmatic patients and 80 healthy volunteers from Beijing China. All patients had their serum TIgE, IgE antibody specific to house dust mite measured by RAST, bronchial responsiveness assessed by methacholine bronch-provocation (if FEV1% > or = 70%), and broncho-dilation measurement by inhaling salbutamol. RESULTS: There was significantly increased gene frequency of alleles DR6(13), DR52 in asthmatics compared with normal controls (17% vs 4.3%, P < 0.01; 50% vs 17.5%, P < 0.01), and RR was 7.55 and 4.7 respectively. The frequency of DR2(15), DR51 was lower in asthmatics than in controls (7% vs 18% P < 0.01; 2% vs 33.8% P < 0.01). The percentage of HLA haplotype DR6(13)-DR52 was higher in asthmatics than in healthy volunteers (20% vs 4%, P < 0.01, RR 6.4). 70% of individuals sharing DR6(13) gene and 56% of subjects carrying DR52 gene had elevated serum d1 sIgE antibody (> or = +4). There was no relationship between HLA-DRB alleles and total IgE, BHR. CONCLUSIONS: Alleles DR6(13), DR52 are significantly implicated in their susceptibility to asthma, at least they may be closely associated with this disorder. Conversely DR2(15), DR51 alleles might confer protection against asthma. Positive associations between DR6(13), DR52 and IgE response to d1 allergen are noted. HLA-DRB genes are particularly involved in regulating human atopic immune response.

Reevaluation of effect of albendazole on echinococcus multilocularis infection in mice and gerbils.

OBJECTIVE: To study the effect for albendazole therapy for alveolar echinococcus infection in gerbils and mice. METHODS: Mice and gerbils were infected of metacestode tissues by intraperitoneal (i.p.) transplantation and treated with albendazole-medicated feeds. The effects were evaluated by comparison of the treated and control groups in terms of host mortality, larval metastases to lungs and liver, final larval weight, histopathological, and ultrastructural examination of metacestode tissues. Viability of metacestode tissues at necropsy of treated animals was tested by intraperitoneal transplantation into uninfected animals. RESULTS: Albendazole-medicated feeds significantly inhibited larval growth of Echinococcus multilocularis (E. multilocularis) both in mice and gerbils with markedly reduced host mortality and pulmonary and liver metastases. Viability test showed that albendazole therapy was parasiticidal in early stage of experimental infection. Light microscopic and ultrastructural examination of metacestode tissues of the treated animals revealed severe destruction and massive necrosis with marked calcification of protoscoleces and residual tissues. CONCLUSION: Continuous long term albendazole therapy in animal models is parasiticidal against larval E. multilocularis especially in early stage of infection.

Long-term efficacy of recombinant interferon alpha 2a in the treatment of chronic hepatitis C: a randomized prospective study comparing two dose schedules in Chinese patients.

OBJECTIVE: To compare the long-term efficacy of a dose of 3 million units (MU) of r-IFN alpha 2a (IFN-alpha 2a) three times a week (t.i.w.) for 6 months with a starting dose 6 MU for 3 months and subsequent reduction to 3 MU t.i.w for further 3 months. METHODS: Sixty-eight serological and histological chronic hepatitis C patients with elevated serum alanine aminotransferase (ALT) were enrolled and randomized into two groups. Sixty-three patients were completed with full course of treatment. Five patients were withdrawn from trial (2 due to personal reasons and 3 due to adverse drug reactions during treatment). Thirty patients received 6 MU IFN-alpha 2a t.i.w., 3 months followed by 3 MU t.i.w. for another 3 months (Group A). Thirty-three patients received 3 MU IFN-alpha 2a t.i.w. for 6 months (Group B). RESULTS: The sex, age, baseline serum bilirubin, ALT and aspartate aminotransferase (AST) levels were matched in both groups. At the end of the 6th month, the complete and partial response rates in Group A were 60.0% and 16.7% respectively, and the clearance of serum HCV-RNA was 53.3%. In Group B, the complete and partial response rates were 72.7% and 6.1% respectively, and the clearance of HCV-RNA was 61.3%. The patients were followed up for 6, 12, and 18 months after stopping treatment. In Group A, the rates of complete normalization of ALT and clearance of serum HCV-RNA at 24 months were 50.0% and 60.0% respectively. In Group B, the rates of normalization of ALT and clearance of HCV-RNA at 24 months were 54.4% and 41.9% respectively. The efficacy between the two groups showed no statistically significant difference. The response rates of treatment were similar to those in the patients with HCV genotype 1b and 2a. Six patients (10.8% of the study population) developed neutralization antibodies to IFN-alpha 2a during treatment, and four of them were responded to the treatment. Adverse drug reactions (ADR), were common, but most of them were tolerable, and the incidence of ADR was in both groups, but the severity was higher in Group A. CONCLUSIONS: IFN-alpha 2a is effective in the treatment of Chinese patients with chronic hepatitis C. The sustained response rates and adverse drug reactions among two dose schedule groups are similar.

[Apoptosis induced by adenovirus-mediated wild-type p53 expression in human pancreatic cancer cells].

OBJECTIVE: To investigate the biological effects of wtp53 expression on the growth rate and apoptosis of human pancreatic carcinoma. METHODS: The Ad5CMVwtp53 a recombinant replication-deficient adenoviral vector containing a human CMV promoter, a human wild-type p53 and the SV40 polyadenylation signal was amplified in 293 packaging cells. The pancreatic carcinoma cell line (PC-2), carrying a mutation of p53 gene at codon 240, was transfected using Ad5CMV wtp53 and the control adenoviral vector Ad5pXJ. RESULTS: The cells transfected with Ad5CMV wtp53 showed that presence and expression of wtp53 gene were demonstrated using PCR and immunoprecipitation, that growth rate and 3H-TdR incorporation rate were decreased. The restoration of wtp53 encoded protein in PC-2 cells induced apoptosis assessed by in situ TUNEL apoptosis, flow cytometry, DNA agarose gel electrophoresis analyses, whereas noninfected cells or the cells infected with control virus didn't show these changes. CONCLUSION: Replication-deficient adenoviral vector is an efficient vector in transferring wtp53 gene and antitumor therapy using the p53 gene is a valuable method in inhibiting pancreatic cancer cells growth by inducing apoptosis.

[Effects of antisense cyclin D1 expressing vector on the cell growth and apoptosis of pancreatic carcinoma].

OBJECTIVE: To observe the effects of antisense (AS) cyclin D1 expressing vector on the cell growth and apoptosis of pancreatic carcinoma. METHODS: Examination of the amplification and expression of cyclin D1 in 5 human pancreatic carcinoma cell lines. Our study found the gene amplification and overexpression of cyclin D1 in PC-7 cell line cells. We then constructed the antisense cyclin D1 vector and transfected the PC-7 cell line with lipofectin. The resultant transformant cell line, PC-7/AS-cyclin D1, showed the expression of exogenous antisense cyclin D1 mRNA and down regulation of endogenous cyclin D1 mRNA expression and inhibition of its protein synthesis detected by Northern blot and Western blot respectively. RESULTS: The transformant cells showed retardation of cell growth and partial reversion of the malignant phenotype, including decrease of the rates of cell growth, DNA synthesis, cell proliferation and metabolism, and also the ability of soft agar colony-formation. The tumorogenesis of the transformant cells in nude mice was suppressed. G1 arrest was revealed by flow cytometry. Apoptosis was identified by DNA fragmentation and in situ TUNEL detection. CONCLUSION: Down scaling of the expression of cyclin D1, which plays an important role in the regulation of the cell cycle, can effectively inhibit the proliferation of carcinoma cells and increase cell apoptosis.

Inhibiting effect of murine double minute-2 oncogene on apoptosis induced by retroviral-mediated wild-type p53.

OBJECTIVE: To investigate the effects of wild-type p53 (wtp53) on inducing apoptosis by restoring wtp53 expression in pancreatic adenocarcinoma cell line (PC-2) which contains mutant p53, and the interaction between murine double minute-2 (MDM2) and wtp53 in pancreatic adenocarcinoma. METHODS: A recombinant retroviral vector expressing wild-type p53 was constructed and packaged by packaging cell line PA317 cells using calcium phosphate coprecipitation method. The supernatant of the packaged cells PA317 was used to transfect the pancreatic carcinoma cell line (PC-2), then a transformed cell line PC-2/swtp53 was established. The recombinant vector pCMV-MDM2 was transduced into PC-2/swtp53 cell line by lipofectin-mediated method, a double transfected cell line (PC-2/swtp53/pCMV-MDM2) was formed. To determine the integration and expression of exogenous wtp53 gene in the transfected cells, polymerase chain reaction (PCR), Western blot and immunoprecipitation analyses were performed. Apoptosis was analyzed by means of flow cytometry, in situ terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) analysis and DNA agarose gel electrophoresis. RESULTS: Transduction with the retroviral vector resulted in integration and expression of wtp53 gene in host cells. The apoptotic cells in PC-2/swtp53 and PC-2/swtp53/pCMV-neo cell lines were 12.1%-12.9%, while the double transfected cell line, PC-2/swtp53/pCMV-MDM2, showed less (3.2%) apoptotic cells than its parent cell lines. CONCLUSIONS: Restoration of expression of wild-type p53 with a retroviral vector can increase programmed cell death of pancreatic adenocarcinoma cells (PC-2) containing mutant p53. The overexpression of MDM2 protein has a negative regulating role in wtp53-induced apoptosis in PC-2 cell.

[Effects of NM23/NDPK in signal transduction of metastatic mouse forestomach carcinoma cells].

OBJECTIVE: To understand the role of NM23/NDPK in the transmembrane signal transduction proccess in tumor metastasis. METHODS: The NDPK activity, IP3 contents, the invasive potentials of the cells of mouse forestomach carcinoma(MFC) substrains with different metastatic potentials and the effect of LN and EGF were observed. RESULTS: Both the NDPK activity and the contents of IP3 of the cells of the subclone with lower metastatic potential was higher than that with higher metastatic potential. Also, the cells with lower metastatic potential had lower invasive potential and higher responsibility to the challenge of EGF compared with that higher metastatic potential. However, no different effect of LN on the cells of those subclones was found. CONCLUSIONS: NDPK might regulate the metastasis of FMC cells by impacting the G protein efficacy with the selection of exotic signal ligands.

[Reoperation for recurrent carcinoma of the esophagus and cardia].

OBJECTIVE: To review the experience of reoperation for recurrent carcinoma of the esophagus and cardia. METHOD: Between 1979 and 1996, 44 patients with recurrent esophageal and cardiac cancer after the first operation underwent reoperation. Among them, 21 had esophageal cancer and 23 cardiac cancer. 36 patients were male and 8 female, with age ranged from 42 to 68 years. RESULT: The resectability rate was 77.2% (34/44) and the hospital mortality rate was 5.8%. Postoperative complications occurred in 7 patients with a morbidity of 20.5%. Pathologically, previous anastomotic relapse was seen in 26 patients, recurrent lesion of residual esophagus in 7, and recurrent cancerous residue at the esophageal bed in 1. 15 patients (44%) showed lymph node metastasis. Follow-up showed that 4 patients survived for 5 years, 4 for 3 years, and 3 for 2 years. 15 patients died within one year after reoperation. CONCLUSION: The recurrence of esophageal and cardiac cancer after first operation is not a contraindication to reoperation, but careful preoperative assessment and patients' selection should become an integral part of the evaluation of these patients.
