Effect of intrabronchially instilled amosite on lavagable lung and pleural cells.

Rats were instilled intrabronchially with 1 mg UICC amosite suspended in 0.2 mL of filtered saline; control animals received the saline instillation only. Five animals from each group were killed on various days after instillation, up to day 128/129. Total retrieved cell counts and differential cell analysis were performed from lung and pleural lavages. In particular, the appearance of peroxidase-positive macrophages (PPM) as indicators of newly arrived macrophages was investigated. Polymorphonuclear cells (PMN) and PPMs in lung lavages increased in number 24 hr after amosite instillation and remained at increased levels until day 62. Alveolar macrophage numbers were significantly decreased after amosite instillation. There was only a very transient increase of PPMs and PMNs in the saline group. The number of PPMs in pleural lavage fluid was already increased 24 hr after amosite instillation. The pleural PPM increase was sustained throughout the study. No pleural reaction was seen in the saline instilled group. The inflammatory reactions indicated by the composition of the lavaged cells of the lung represent the in vivo toxicity of intrabronchially instilled amosite. The stimulus for recruitment of PMNs and PPMs is different, since no PMN response was detected in the pleural space. It is suggested that the response of the pleural PPMs is caused by the early arrival of fibers at the pleural sites, which results in the recruitment of PPMs to this space by an unknown mechanism.

Particle binding, phagocytosis, and plastic substrate adherence characteristics of alveolar macrophages from rats acutely treated with chlorphentermine.

The effects of acute treatment with the phospholipidosis-inducing agent chlorphentermine (CP) on alveolar macrophage (AM) function heretofore have not been investigated. We evaluated the effects of acute CP treatment on the functional status of AM by comparing Fc receptor (FcR) and "nonspecific substance receptor"-mediated particle binding and phagocytic activities, as well as the plastic substrate adherence characteristics of AM lavaged from control (CONT-AM) and CP-treated (CP-AM) rats. Acute CP treatment caused an impairment in AM phagocytosis and binding of sheep red blood cells opsonized with immunoglobulin G (IgG). Although CONT-AM and CP-AM avidities for tanned human red blood cells were indistinguishable, phagocytosis of these particles by CP-AM was diminished in a manner similar to that found for FcR-mediated phagocytosis. Analyses of the distributions of test particles in phagocytizing CONT-AM and CP-AM indicated that the phagocytic activity of a subset of CP-AM was compromised by drug treatment. Populations of CP-AM were also found to be less adherent in a plastic substrate adherence assay verified with a poorly adherent AM population. Unlike studies involving chronic CP-treatment, our studies indicate acute CP administration results in an impairment in AM phagocytic activity and a reduction in FcR-mediated particle binding.

Pulmonary alveolar pores and alveolar macrophage-mediated particle clearance.

Pulmonary alveolar macrophages (AM) are considered to have an essential role in alveolar clearance of particles. The AM occupy only a small portion of the alveolar surface and their number is probably lower than the number of alveoli. Even at realistic experimental exposures the number of deposited particles, which are distributed all over the lung surface, is orders of magnitude higher than the number of AM. Phagocytosis of deposited particles, however, is being accomplished rapidly within hours. Based on microscopic evidence, we suggest that alveolar pores serve as important passageways for AM by shortening the distance between particles and AM. Accordingly, the pores of Kohn may, in part, lend efficiency to the in vivo particle collection by AM.

Alveolar macrophage mediated pulmonary clearance suppressed by drug-induced phospholipidosis.

Drug-induced phospholipidosis in rats treated with chlorphentermine (CP) for 4-7 days suppressed totally alveolar pulmonary clearance in the first days after a TiO2 aerosol exposure. Reversing phospholipidosis by treatment interruption led to a recovery of particle clearance. Morphological observations indicated that "foam cells" contained less TiO2 particles than alveolar macrophages (AM) of control rats. Clearance of ZnO particles which seems not to be mediated by AM was not affected by CP treatment. A grand average retention curve based on data from control groups of past experiments suggests that alveolar clearance of TiO2 particles has a phase 1 (T 1/2 = 7 days) lasting about 2 weeks and a phase 2 (T 1/2 = 69 days). The results with drug-induced phospholipidosis suggest that phase 1 is practically totally AM-mediated. Drug-induced phospholipidosis is a promising method for the study of AM involvement in defensive functions.

Soft-tissue measurements at the index finger in postmenopausal women on long-term oestrogen treatment.

Measurements of the soft-tissue at the index finger on X-rays were made in 33 women after a natural menopause and who were on a long-term oestrogen replacement therapy for at least 5 yr. The data were compared with values obtained in an age-sex matched control group. The oestrogen treated women had a soft-tissue thickness (total finger width minus phalanx width) significantly smaller than the control group. The soft-tissue index (the ratio width of the phalanx to the total finger width) was significantly larger than expected. The soft-tissue values observed in the oestrogen treated subjects correspond to those observed in premenopausal women.

Relaxin in non-pregnant women during ovarian stimulation.

Around pharmacologically induced ovulation, inconstant immunoreactive plasma relaxin concentrations were observed. Increasing levels appear during HCG or LH-RH induced lutinization. These values remain high as long as the corpus luteum is sustained. These observations may suggest that relaxin has a role during early luteinization and during the non-gestational lifespan of the corpus luteum in women.

Lung clearance of particles in two strains of rats.

Two strains of commonly used experimental rats were compared with regard to lung clearance of TiO2 particles. The Fisher 344 inbred rats retained predictably fewer particles after a 7-hr exposure than the larger outbred Long-Evans rats. This fact can be expected in view of the different lung size of the two strains. In addition, clearance of the retained particles was significantly slower in Fisher 344 rats. Differences in lung clearance capacity of different animal strains should be considered when experiments in inhalation toxicology are performed.

[Microscopic study of human fallopian tubes after laparoscopic sterilization (author's transl)].

Histological study was carried out on human fallopian tubes one hour and one month after various methods of laparoscopic sterilization (unipolar electrocoagulation, bipolar electrocoagulation, silicone ring and Hulka-Clemens clip). Microscopic lesions were observed. Ciliated cells were counted. Deciliation is proved at a distance from the site of sterilization. It has been possible to display the substance loss of the tubal tissues which have to be taken into consideration in case of surgical reanastomosis.

The effect of long-term oestrogen treatment on the development of osteoarthrosis at the small hand joints.

The effect of long-term oestrogen treatment of at least 5 yr on the development of osteoarthrosis was studied in 48 postmenopausal women. Osteoarthrosis at the hand joints was graded according to Kellgren's criteria, using hand X-rays. Data obtained in a normal population of 155 women served as controls as well as data obtained in 24 postmenopausal women who didnot receive oestrogen replacement therapy. No significant difference in osteoarthrosis between the treated and non-treated groups was found. The development of osteoarthrosis at the hand joints seems to be independent of the hormonal status in postmenopausal women.

[Radioimmunoassay of HCG and HCG beta in blood during pregnancy and trophoblastomas].

This work was carried out on 151 estimations of the radioimmune levels of hCG-hCG beta in the first half of normal pregnancy and 390 levels estimated in 10 molar pregnancies, in 4 choriocarcinomata following moles and 1 primary choriocarcinoma of the ovary using homologous hCG beta. In normal pregnancy the level of hCG-hCG beta rises rapidly. The levels are highest between the 8th and 9th weeks of pregnancy, and then gradually decrease. In active molar pregnancies the levels of hCG-hCG beta in the plasma are significantly higher than those found in normal pregnancies of the same gestational age. When the outcome is going to be favourable the levels drop progressively. Complete absence in peripheral blood is usually found about 14 weeks after a mole has been expelled or evacuated. In choriocarcinomata, if the level of hCG-hCG beta rises it is a certain sign of reactivation of the tumour.

Pre-ovulatory plasma levels of 17beta-oestradiol (E2) and conception in patients treated with gonadotrophins.

Twenty-six patients seeking advice for sterility were given courses of treatment with HMG-HCG. Ovarian maturation was followed by daily evaluation of plasma 17beta-oestradiol concentrations. HCG was administered as soon as 17beta-oestradiol levels reached 250 pg/ml. In a first group of 7 amenorrheic patients with lack of oestrogen activity, a pregnancy rate of 71.4% and an ovulation rate of 91.7% were achieved. In a second group of 5 amenorrheic patients showing evidence of oestrogen activity, a pregnancy rate of 80.0% and an ovulation rate of 100% were obtained. In a third group of 14 oligomenorrheic patients, the pregnancy rate attained 71.4% and the ovulation rate 96.9%. The overall pregnancy rate was 73.0%. With this procedure of monitoring HMG-HCG treatment by means of plasma 17beta-oestradiol levels, the multiple pregnancy rate reached only 11.7% and only one case of mild ovarian hyperstimulation was observed. Pre-ovulatory and pre-conceptional 17beta-oestradiol concentration were identical with those observed in spontaneous ovulatory cycles with or without conception.
