Induction of delayed-type hypersensitivity to azobenzenearsonate by a monoclonal anti-idiotype antibody.

Azobenzenearsonate (ABA)-specific sensitivity was induced in A/J mice by injecting a monoclonal anti-idiotype reagent, 14A, directed against a determinant present on a minor subpopulation of immunoglobulin molecules within the anti-ABA antibodies of A/J mice. Sensitivity was transferrable by purified T cells and this was abrogated by treating the cells with 14A, rabbit anti-mouse immunoglobulin and complement, not by treatment with only the last two reagents. The transfer was restricted by the K-end of the major histocompatibility complex.

The relation between age and mean length of utterance in morphemes.

The relationship between child age and mean length of utterance measured in morphemes (MLU) was studied in a sample of 123 middle- to upper-middle-class midwestern children, aged 17 to 59 months, conversing with mothers in free play. A significant correlation was found between age and MLU: r = .88. Age accounted for 78% of the variance when MLU was regressed on age; and MLU accounted for 77% of the variance when age was regressed on MLU. Significant nonlinear components accounted only for an additional 1% and 3% (respectively) of the variance. Ranges within one standard deviation (SD) were estimated for predicted MLUs and derived for predicted ages on the basis of linear regression. MLU increased at an average rate of 1.2 morphemes per year; predicted variability in predicted MLU increased with age. These findings can play a useful role in three activities when the children studied are from populations similar to the one sampled here, and MLUs are obtained in the same way. They can aid in (1) finding children whose development of productive syntax requires further diagnostic evaluation; (2) finding children at a particular linguistic stage quickly; and (3) predicting the age most likely to be associated with a given MLU. The substitution of local MLU norms, sampled from the population and ages of clinical interest, can further sharpen these evaluations.

Fetal loss after implantation. A prospective study.

The incidence of post-implantation pregnancy loss in 197 women was determined prospectively. The diagnosis of pregnancy was based on the detection of human chorionic gonadotropin in the urine during the luteal phase of each menstrual cycle. There were 152 conceptions in the 623 cycles studied with a pregnancy loss rate of 43%. 14 of the pregnancies ended in clinically recognised spontaneous abortions and in 50 the sole evidence of pregnancy was an increased concentration of urinary hCG. The maximum conception rate achieved was 36% in the first cycle after removal of an intra uterine device.

Suppression of T cells specific for the nonthymic parental H-2 haplotype in thymus-grafted chimeras.

The mechanism of restriction of T-cell specificity by the genotype of the thymus in allogeneic and semiallogeneic chimeras was investigated. Lack of induction of delayed-type hypersensitivity (DTH) directed against antigen in association with the nonthymic parental haplotype in naive cells adoptively transferred into chimeras suggests the existence of an in vivo suppressive mechanism. However, it was not possible to suppress the expression of DTH in sensitized cells transferred into chimeras, or to transfer this suppression to normal naive recipients.

Delayed-type hypersensitivity induced by antigen-pulsed, bone marrow-derived macrophages.

Mouse bone marrow cells, cultured in specifically conditioned medium, were found to be virtually pure macrophages after 7 days. When pulsed with antigen, these cells induced and elicited delayed-type hypersensitivity (DTH) with the same efficiency and major histocompatibility complex restrictions as antigen-pulsed peritoneal exudate cells. Both of these macrophage populations had 20% Ia+ cells, as measured by complement-mediated cytotoxicity. When most of the Ia+ cells were removed, the remaining antigen-pulsed macrophages could not elicit DTH in sensitized mice.

Language comprehension in sensorimotor stages V and VI.

A cross-sectional study of language comprehension in relation to cognitive functioning in 48 to 10-to-21 month old children, 4 at each month of age, revealed significant correlations between comprehension and five sensorimotor subscales. Age, however, was the only significant predictor in multiple regression analyses adding either age or sensorimotor subscale in second. In specific instances in which sensorimotor tasks tapped hypothetical prerequisites to specific language comprehension items, the two domains corresponded closely; but the cognitive domain was not always the first to be passed. Comprehension items were ordered from easy to hard as follows: 1) understanding single words for present people or objects; 2) for actions; 3) for absent people or objects; 4) understanding two words for the semantic relations possessor-posession, action-object, and agent-action; 5) understanding three words for agent-action-object. Implications for the Cognition Hypothesis are discussed.

Pericentric inversion (13) with two different recombinants in the same family.

A family is described in which a pericentric inversion (13) was discovered in the father after the birth of an abnormal baby. In a further pregnancy amniocentesis was carried out. The fetal karyotype showed a rec(13)dup p,inv(13)(p11q22). The fetus's abnormalities were similar to those observed in the first child. Family studies showed that a first cousin, mentally retarded, had a rec(13)dup q,inv(13)(p11q22) karyotype. In this family, the risk of occurrence of a recombinant in offspring of an inversion carrier could be as high as 40%.

The dynamic focus of nursing: a challenge to nursing administration.

Nursing administrators are challenged to establish a climate that facilitates the use of appropriate frameworks to guide nursing. The author presents an extension of a self-care practice model demonstrating that the focus of nursing strategies changes as the patient's health state changes. Practical application of the proposed model is included.

Delayed-type hypersensitivity to allogeneic cells in mice. III. Sensitivity to cell-surface antigens coded by the major histocompatibility complex and by other genes.

DTH could be induced to cell-surface antigens coded by either H-2 or non-H-2 genes. Sensitivity was more readily induced across I region than across K- or D-region differences. The presence of an I-region difference during sensitization did not significantly increase the DTH response to K- or D-region-coded antigens. Macrophage processing appeared to be the major route of sensitization to background antigens. Thus, high levels of sensitivity were achieved equally well using viable or disrupted cells, the response was independent of the H-2 haplotype of the allogeneic cells, and transfer was restricted to the K end of the host H-2 complex. Although sensitization to H-2 antigens was obtained with disrupted cells, transfer of sensitivity against viable cells was unrestricted. This suggests a minor role for macrophage processing in sensitization to H-2 antigens.

Lysolecithin analogs as adjuvants in delayed-type hypersensitivity in mice. I. Characterization of the adjuvant effect.

The adjuvant activity of 5 different lysolecithin analogs (LLA) has been studied in delayed-type hypersensitivity (DTH) using fowl gamma globulin, bovine serum albumin and the terpolymer L-glutamic acid 60-L-alanine30-L-tyrosine10(GAT), as antigens. Increased DTH responses, by factors of 1.8--2.0 in CBA and BALB/c mice, showed that LLA are immunopotentiators if given intraperitoneally (i.p.) or subcutaneously (s.c.) together with the antigen. The concentration range, within which LLA are active, is limited to 10--20 micrograms/mouse s.c. and 100--300 micrograms/mouse i.p. Adjuvanticity was tested as a function of the LLA structure. The most pronounced immunopotentiation was obtained with racemic 1-octadecyl-2-methylglycero-3-phosphorylcholine (ET18-O-CH3). The LLA became less active with decreasing numbers of C atoms in the alkyl chain.

Lysolecithin analogs as adjuvants in delayed-type hypersensitivity in mice. II. Studies on the mode of action.

Lysolecithin analogs (LLA) possess adjuvant activity in delayed-type hypersensitivity (DTH). To elucidate their mode of action, we studied the influence of LLA on the recruitment of hemopoietic progenitor cells and on the induction and transfer of DTH. Whereas the number and composition of colony-forming cells (CFC) in the bone marrow remained unaltered in LLA-treated mice, the number of CFC in the spleen was augmented severalfold, and the content of macrophage progenitors was remarkably increased. DTH was induced with macrophages from peritoneal cells (PC) pulsed with keyhole limpet hemocyanin (KLH). A more marked DTH response was obtained with KLH-pulsed PC from LLA-treated mice than from mice given phosphate-buffered saline (PBS). On the other hand, there was no difference in the DTH response following adoptive transfer of sensitized cells to naive mice pretreated either with PBS or LLA. It is concluded that the adjuvant effect of LLA can be accounted for partly in terms of their effects on macrophage progenitors and partly in terms of a more effective T cell activation by antigens associated with LLA-derived macrophages.

Some estrogenic effects of two oral contraceptives consisting of norgestrel and two different doses of ethynylestradiol.

Some biological and biochemical effects (i.e. KPI, cervical mucus, SHBG and ceruloplasmin) as well as serum ethynylestradiol and serum norgestrel, following the use of two oral contraceptives containing the same amount of norgestrel (dl-norgestrel 0.5 mg) and either 30 microgram or 50 microgram of ethynylestradiol were compared. There was no difference in the clinical features in either group of patients. There was a statistically significant difference in the levels of unconjugated ethynylestradiol but not in the other biochemical parameters studied. It is concluded that the absence of any such difference might be attributable to the strong antiestrogenic effects of dl-norgestrel.

An improved method for selecting T-cell hybridomas by the fluorescence-activated cell sorter.

A two-step sorting procedure with the fluorescence-activated cell sorter (FACS) is described to select T cell hybridomas from cell populations obtained from the fusion of activated T cells and T- lymphoma cells. Cells are first sorted on the basis of differences in cell-surface antigens. After a period of growth in culture, the cells are then stained with the DNA-labeling dye dis-benzimidazole 33258 Hoechst and sorted on the basis of their DNA content. From 15 to 25% of the resulting cell clones, growing out of limiting dilutions, were hybrid lines as judged by chromosome analysis.
