Genetic identification of an autoinhibitor in CDPK, a protein kinase with a calmodulin-like domain.

CDPKs are a family of calcium (Ca2+)-dependent protein kinases which are defined by a carboxyl-terminal calmodulin-like domain. Mutational analysis indicates that the junction domain, which joins the kinase and calmodulin-like domains, contains an autoinhibitor. CDPK isoform AK1 from Arabidopsis was expressed in Escherichia coli as a fusion protein sandwiched between glutathione S-transferase and six consecutive histidines at the N- and C-terminal ends, respectively. This fusion, called AK1-6H, was purified and displayed kinase activity which was stimulated up to 127-fold by Ca2+, with a typical specific activity of 2000 nmol min-1 mg-1, using syntide-2 as peptide substrate. A truncation which deletes the calmodulin-like domain, as in mutant delta C-6H, disrupts Ca2+ activation and leaves the enzyme with a basal level of activity. Delta C-6H could be activated 87-fold by preincubation with a purified polyclonal IgG which was raised against a junction domain fusion. A further deletion of the junction domain, as in mutant delta JC, results in a constitutively active enzyme. This indicates that the junction domain in delta C-6H can function as an autoinhibitor. Its function as an autoinhibitor in a full-length enzyme was confirmed by site-specific mutagenesis, as shown by mutant KJM23-6H, which had a six-residue substitution in the junction domain between A422 and A432. Both delta JC and KJM23-6H encoded Ca(2+)-independent enzymes which had specific activities greater than 70% that of a fully active AK1-6H and displayed equivalent Km values for ATP and syntide-2. Inhibition studies on delta JC, using peptides based on the autoinhibitory domains of Ca2+/calmodulin-dependent protein kinases, are consistent with a model where the junction domain contains a similar pseudosubstrate-type autoinhibitor.

[Three dimensional finite element analysis of stress induced by orthodontic forces in the periodontal tissue].

The purpose of this study was to investigate the stress induced in the periodontal tissue of the upper central incisor by application of orthodontic forces. A three dimensional finite element model consist of tooth, periodontal ligament (PDL), cancellous and compact bone was established and the following results were obtained: (1) The highest stresses were observed on the root surface, the intermediate on the alveolar bone and the lowest in the PDL. (2) Stress condition on the surface of root and alveolar bone is very complex. The maximum, intermediate and minimum principle stresses on the same area are quite different, but the three principal stresses in the PDL are similar. (3) Regarding to tipping and torque movement, the stress value varied largely from the cervix to the apex, and there is stress concentration at the cervix and the apex. For bodily movement when M/F is -10.30, the stress induced is homogeneous and the difference of the stress values from the cervix to the apex is little, which seems to be more physiological.

[Portal vein embolization via a fine needle guided by angio-echography: experimental and clinical study].

This paper introduced a technique of portal vein embolization by injection of ethanol via fine needle under guidance of angio-echography. First introportal ethanol injection were done in 28 dogs to evaluate its embolic efficacy and safety. The result showed if a dose of injected ethanol was chosen properly, expected embolization could be obtained with slight toxicity to hepatic parenchyma and function. For clinical application, selective portal vein puncture was percutaneous transhepatic under echo guidance. Ultrasound angiography by injecting carbon dioxide (CO2) into portal vein was introduced initially. After confirmed that the injected branch was that supplied the tumor and there was no retrograde overflow of portal blood, ethanol was injected at a dose no more than 10ml. Eighteen patients with hepatocellular carcinoma and underlying liver cirrhosis underwent the procedure preoperatively. Of 14 cases who received hepatectomies, portal vein embolization developed in 12. Since the procedure was guided by angio-echography it could be used for indicated cases with satisfactory embolic effect and not harmful to liver. The procedure had advantages of simple manipulation and made selective embolization easier compared to interventional transcatheter portal vein embolization.

[Two-stage multivariant analyses for prediction of hepatic function reserve].

Eleven liver function tests were used for preoperative estimations of the hepatic function reserve in 103 patients with primary hepatocellular carcinoma (HCC) and underlying liver cirrhosis. Postoperatively, the patients' liver function could be classified as good recovering (Grade A, n = 38), functional damage (Grade B, n = 37) and liver failure (Grade C, n = 28). Single factor analyses showed 6 of those tests were significant indicators, including the ratio of blood glucose level at 120 minutes and 60 minutes by oral glucose tolerance test, total bilirubin, the ratio of albumin and globulin, prealbumin, prothrombin time and indocyanine green retention at 15 minutes. The correlations between 11 preoperative parameters (xi) and postoperative course scored (Y) were analysed by Fisher's discriminant test. The multiple regression equation Y1 was obtained by comparing from groups of Grade A with B and formula Y2 from groups of Grade B with C. The predictive accuracy of both equations were 88.0%, 83.1%, respectively. To select adequate surgical procedures with the best therapeutic effect and minimal liver damage for the patients with HCC, we proposed a method of "two-stage predications" combining use of Y1 and Y2 for evaluation of liver function reserve.

[The experimental study of the canine auxiliary partial liver transplantation for the liver failure].

A model of hepatic failure was established by injection of galactosamine in CCL4-induced liver cirrhosis. Auxiliary partial liver transplantation (APLT) was carried out to treat those dogs. The survival rate, biochemical change patterns, and histologic changes were investigated in both APLT group and control group. The survival rate in APLT and control group was 59.1% and 7.1% respectively, (P < 0.01). Fair supporting graft function was demonstrated by uptake and excretion of 99mTC-HIDA and 99mTC-DIASA at cholescintigraphy, ammonia aetoxification, synthesis of clotting factors and glucohomeostasis. The results indicate that auxiliary transplantation of a partial liver proviae metabolic support and improve survival in animals with hepatic failure. The character of the model, the advantages and the disadvantages of the APLT, were also discussed.

Bite-opening mechanics as applied in the Begg Technique.

In the Begg Technique, factors controlling the anterior intrusive force provided by the archwire have yet to be fully clarified. The rationale for bite opening with very light forces, the effect of Class II elastics on intrusive force magnitude and the intrinsic quality of 'Australian' wire in bite opening are examined in this paper. A formula is presented which allows the intrusive force that archwires exert in different individuals to be predicted. The values derived from the formula were compared with case model measurements. It was found that the intrusive force of the archwire was affected by the length of the individual dental arch. The magnitude of this intrusive force was seen to increase gradually during the period of bite opening and appeared to be related to the use of Class II elastics. The study revealed factors which affected the magnitude of the intrusive force.

[An improved technique for bloodless hepatic resection on the in situ cold perfused liver].

An improved technique for bloodless hepatic resection using in vivo isolation and asanguinous hypothermic perfusion was described to deal with the huge liver tumour involving the liver hilum, the main hepatic veins and retrohepatic inferior vena cava. The original Fortner's technique was modified, including the incision; isolated cold perfusion of the healthy half of the liver through the portal vein; suprahepatic outlet for the perfusate, and shortened hepatic ischemia time by perfusing hepatic artery prior to the repair or reconstruction of the portal vein. Using this technique, two huge liver tumors were successfully resected, and the indications, intraoperative monitoring and early postoperative courses were discussed.

An improved technique for bloodless hepatic resection on in situ cold perfused liver.

An improved technique for bloodless hepatic resection using in situ isolation and asanguinous hypothermic perfusion was described to deal with huge liver tumors involved in the liver hilum, the main hepatic veins and retrohepatic inferior vena cava. The original Fortner's technique was modified, including the choice of incision; semi-isolated perfusion of the liver portion preserved through the single portal vein; suprahepatic outlet of the perfusate and the shortening of the period of hepatic ischemia by reperfusion of hepatic artery prior to the repair or reconstruction of the portal vein. The initial successful experience of the technique applied to 2 pediatric cases with giant liver tumors was reported, and the indications, intraoperative and early postoperative courses were discussed.

Effect of the vertical force component of Class II elastics on the anterior intrusive force of maxillary archwire.

Class II elastics are usually employed in the treatment of excessive overbite and overjet with the Begg technique. The effect of Class II elastics on bite opening and the extent of such an effect is uncertain. This article, based on measurements of 30 cases, illustrates that the effect of the vertical component of force from Class II elastics in reducing the intrusive force generated by the anchor bands in the upper archwire is less than previously believed. The position of the circles in the archwire for intermaxillary elastics has the greatest influence on the anterior intrusive force of the upper archwire. The direction of the Class II elastics and the length of the dental arch (a negative correlation) came second, while the force actually exerted by the elastics had the smallest influence. It is, therefore, suggested that the position of the intermaxillary elastic circles should be located according to the differing clinical objectives.

Selective total vascular occlusion for major hepatic resection.

Selective total vascular occlusion for the resection of large tumors on the right lobe of the liver or central hepatic tumors was described. The occlusion of the portal triad, infrahepatic vena cava and the total hepatic circulation was selectively used in combination to control bleeding in the process of hepatectomy. Within a year, major resection was successfully performed with this technique on 10 patients (extended right lobectomy 4 patients, right lobectomy 4, central segmentectomy 2). The mean duration of total vascular exclusion was 19.35 +/- 19.32 minutes and mean blood transfusion requirement was 7 +/- 4.5 units during surgery.

A quantum biological approach to the relations of DNA methylation with gene transcription and mutation.

DNA methylation may play a key role in some biological processes, as in DNA replication, transcription and recombination, gene expression, cell differentiation, and development. However, the experimental facts were elusive up to now. Using results calculated from quantum mechanics (at the electron level) we have solved some problems which existed concerning the relationship of DNA methylation and gene transcription and mutation. We think that the methylation of cytosine in eukaryotes does not lead directly to gene mutation, on the contrary, it can inhibit transcription.

Nitrogen and methyl jasmonate induction of soybean vegetative storage protein genes.

Vegetative storage protein (VSP) and VSP mRNA levels in soybean (Glycine max) leaves correlated with the amount of NH(4)NO(3) provided to nonnodulated plants. The mRNA level declined as leaves matured, but high levels of N delayed the decline. This is consistent with the proposed role for VSP in the temporary storage of N. Wounding, petiole girdling, and treatment with methyljasmonate (MeJA) increased VSP mRNA in leaves 24 hours after treatment. The magnitude of the response depended on leaf age and N availability. N deficiency essentially eliminated the response to wounding and petiole girdling. MeJA was almost as effective in N-deficient plants as in those receiving abundant N. Inhibitors of lipoxygenase, the first enzyme in the jasmonic acid biosynthetic pathway, blocked induction by wounding and petiole girdling but not by MeJA. This supports a role for endogenous leaf jasmonic acid (or MeJA) in the regulation of VSP gene expression.

Methyl jasmonate treatment eliminates cell-specific expression of vegetative storage protein genes in soybean leaves.

Soybean (Glycine max) plants accumulate a vacuolar glycoprotein in the parenchymal cells of leaves, petioles, stems, seed pods, and germinating cotyledons that acts in temporary nitrogen storage during vegetative growth. In situ immunolocalization of this vegetative storage protein (VSP) revealed that it accumulates in those parenchymal cells in close proximity to existing and developing vasculature, as well as in epidermal and cortical cells. The protein was more prevalent in younger, nitrogen-importing tissues before pod and seed development. Removal of actively growing seed pods greatly enhanced VSP accumulation, primarily in bundle sheath and paraveinal mesophyll cells. In situ hybridization of a VSP RNA probe to mRNA in leaf sections demonstrated that cell-specific mRNA accumulation corresponded with the pattern of protein localization. Treatment of leaf explants with 50 micromolar methyl jasmonate resulted in accumulation of VSP mRNA and protein in all cell types.

[Modified normothermic avascular hepatic resection].

The conventional Heaney's avascular hepatic resection was modified based on an experimental study on normothermic total hepatic vascular exclusion in dog. The modification was cancellation of simultaneous exclusion of the abdominal aorta and proper prolongation of total hepatic vascular exclusion time. The method was successfully used in 3 patients, 2 with primary hepatocellular carcinoma involving the second porta hepatis and 1 with a huge hepatic cavernous hemangioma in segments VII and VIII. The clinical application of this method is presented and discussed. The modified Heaney's technique, being simpler and safer than the conventional one, may increase the resection rate for central hepatic tumors (segments I, IV, V and VIII according to Couinaud's classification).

[A modified technique for normothermic bloodless hepatic resection. Experimental and clinical studies].

On the basis of experimental study of canine normothermic total hepatic vascular blockade (NTHVB) through portal decompression by means of jugulo-splenic shunting. We proposed a critical period of the hepatic ischemic time with NTHVB be 45 minutes. The conventional technique of Heaney's bloodless hepatic resection was modified, including the cancellation of the synchronous blockade of the abdominal aorta and effective application of limited hepatic ischemic time to the critical manipulation during operation through the combination of various techniques for hepatectomy. The technique was successfully employed in 4 patients with central hepatic tumors. The modified Heaney's technique is simpler and safer than the conventional one. It may increase the resectability of central hepatic tumors (segments I, IV, V, VIII of Couinaud's classification) and is of clinical value.

Effects of inaccurate reference lifetimes on interpreting frequency-domain fluorescence data.

The effects of assigning inaccurate reference lifetimes in lifetime determinations are predicted theoretically by using standard equations. This theory leads to a method to remove reference error effects using common least-squares software. This method cannot, however, be used to deconvolute data collected with isochronal references. Uncorrected data can always be exactly solved with models containing one more degree of freedom than the true model. Monoexponential decays are fit by double-exponential decays or excited-state processes. Unimodal distributed decays often appear as discrete, double-exponential decays.

[Intrahepatic biliary-jejunal anastomosis in perihilar bile duct carcinoma].

A series of 15 patients with perihilar biliary carcinoma treated by intrahepatic biliary-jejunal anastomosis at our department was studied to determine its adequacy for bilirubin clearance, the decrease of alkaline phosphatase (AKP) value, the duration of relief from jaundice, the length of hospital stay, life quality and survival time of the patients. Intrahepatic bypasses were shown to allow bilirubin to return to normal or nearly normal value. Free from jaundice persisted until death for most cases. The postoperative decrease of AKP value was in an irregular pattern. Postoperative reflux cholangitis was not found in any patients. There was only one death during hospital stay. Most patients survived more than 6 months. A satisfactory palliation was achieved by the intrahepatic biliary-jejunal anastomosis. A new method for palliative diversion of the bile by means of creating an internal fistula between a bile duct in segment v and gallbladder and an anastomosis of the gallbladder to the jejunum was introduced.
