A gamma-ray-resistant derivative of an ataxia telangiectasia cell line obtained following DNA-mediated gene transfer.

Genomic DNA from normal human or mouse cells was transfected together with the selectable marker gpt into the simian virus 40-transformed ataxia telangiectasia fibroblast line, AT5BIVA. From a series of experiments involving over 400,000 clones selected for the gpt marker, one unambiguously radiation-resistant clone (clone 67) was recovered following selection with repeated cycles of gamma irradiation. The normal level of radiation resistance of clone 67 has been maintained for at least 11 months in the absence of further selection by radiation. The resistant clone contains one copy of the gpt gene. Its DNA synthesis following gamma-irradiation is inhibited to an extent intermediate between that of ataxia telangiectasia and normal cells. Three out of four thioguanine-resistant derivatives of clone 67 have either lost or do not express the gpt sequence and show almost the same sensitivity to gamma irradiation as the original AT5BIVA line. This suggests that the radiation resistance of clone 67 may be linked to the gpt sequence and may have arisen as a consequence of the transfection, rather than as the result of an independent mutation to radiation resistance.

Electromyographic properties of the myometrium of the pony mare during pregnancy.

Recordings of uterine electrical activity were made from 5 pregnant pony mares from Day 141 to 320 of pregnancy. Three types of activity were identified. Short, medium and long bursts were quantified as the percentage of time each occurred during the hour analysed and further categorized according to frequency, amplitude and duration. The uterus was most active during the early stages recorded and became increasingly quiescent after Day 240. Short-burst activity was greatest when the uterus was most quiescent. Long bursts showed the greatest percentage of activity until Day 220 and then decreased. Medium-burst activity was present throughout the period studied and high amplitude synchronous medium bursts peaked at Day 221-240.

Recognition of reversible and irreversible myocardial injury by technetium pyrophosphate extraction kinetics.

The need for a more accurate method of detecting episodes of myocardial ischemia during cardiac operations, particularly during the ischemic arrest interval, prompted us to investigate the usefulness of measuring the active extraction of technetium pyrophosphate in identifying and quantitating ischemic injury. Twenty-four adult mongrel dogs were subjected to cardiopulmonary bypass, and normothermic global ischemia was induced by cross-clamping the proximal aorta. Technetium pyrophosphate (1 mCi) was injected through a standard cardioplegia line with normal saline, simulating administration of cardioplegic solution, upon placement of the aortic cross-clamp (time 0), at 15, 30, 45, and 60 minutes of global ischemia, and with the onset and completion of ischemic contracture. Radioactive counts were recorded over the heart at 1 second intervals, and the extraction fraction and half-time of clearance were calculated. The extraction fraction increased from 0.22 at time 0 to 0.58 at 15 minutes, 0.82 at 30 minutes, 0.85 at 45 minutes, and 0.91 at 60 minutes. The halftime increased from a baseline of 114 seconds (time 0) to a maximum of 321 seconds at 60 minutes of ischemia. The onset and completion of ischemic contracture showed a return toward baseline of both the extraction fraction and halftime of clearance, with an extraction fraction of 0.44 and 0.46 and a halftime of 135 and 133 seconds, respectively. These data clearly show that reversible myocardial injury increased the extraction and reduced the clearance of technetium pyrophosphate and that the magnitude of change related to the extent of injury. The progression to irreversible myocardial injury decreased the active extraction of technetium pyrophosphate. This simple procedure for real-time documentation of myocardial injury promises to provide easily obtainable endpoints of injury for use during cardiac operations in humans.

Reproductive patterns in cyclic and pregnant thyroidectomized mares.

Three Quarter-horse mares were thyroidectomized at about 1.5 years of age. Three similar intact mares served as controls. The study continued through two breeding seasons. The thyroidectomized mares were lethargic, rear limbs were oedematous and hair coats were coarse. They displayed a tranquil oestrous behaviour when exposed to a stallion and were only mildly antagonistic when not in oestrus. Length of oestrous cycles varied but most often they were 19-24 days long. Duration of oestrus (mean +/- s.e.m.) for the control and thyroidectomized mares was 12.9 +/- 2.9 and 11.7 +/- 2.2 days respectively (P greater than 0.05). The peak of LH during oestrus was as high as 60 ng/ml blood serum with no difference between the two groups. Peak progestagen on Day 7 after ovulation for controls was 9.0 +/- 1.6 ng/ml and was not different (P greater than 0.05) from the peak of 6.3 +/- 1.7 ng/ml for thyroidectomized mares on Day 8. Pregnancy was achieved in both groups of mares, including the use of semen from a thyroidectomized stallion. Thyroxine was detectable in one pregnant thyroidectomized mare during the last two-thirds of pregnancy only.

Electromyographic properties of the myometrium correlated with the endocrinology of the pre-partum and post-partum periods and parturition in pony mares.

A complete set of electromyographic recordings, plasma samples and behavioural observations were collected from 2 mares beginning 7 days pre partum, through parturition and into the early post-partum period. During the week pre partum, EMG activity was elevated, occurring 26-73% of the time. Activity was least during the day and greatest at night with no significant difference for the hours of the day or between days pre partum. During the 24 h before delivery, EMG activity was increased for 7-13 h (55-80%) during the daylight hours. EMG activity decreased 2-4 h immediately preceding delivery of the foal, with an abrupt increase at rupture of the chorioallantois. At delivery, EMG activity consisted of events containing a series of 10-13 discrete bursts of increasing amplitude occurring in rapid succession. After fetal delivery there was a reduction in activity until placental delivery followed by very long (2-22 min) trains of potentials.

Left ventricular dysfunction and dilatation resulting from chronic supraventricular tachycardia.

It has been suggested that patients with chronic supraventricular tachycardia may have impaired ventricular function, which returns to normal after surgical procedures that eliminate the tachycardia. The purpose of this study was to determine the functional consequences of prolonged supraventricular tachycardia in 12 awake dogs in which permanent asynchronous atrial pacemakers were implanted and programmed to a rate of 190 +/- 5 beats/min. Serial radionuclide angiograms were obtained immediately after pacemaker activation and at regular intervals over a 3 month period. Chronic tachycardia resulted in a significantly depressed ejection fraction (49% +/- 1% to 29% +/- 3%; p less than 0.0005) compensated for by a dramatic increase in left ventricular end-diastolic volume (69 +/- 4 to 105 +/- 9 ml, p less than 0.005). Stroke volume and cardiac output were not significantly changed. Five dogs were allowed to recover, and serial radionuclide angiograms were obtained for 12 weeks. Although ejection fraction returned to control values (50% versus 47%, p = no significant difference), end-diastolic volume remained persistently elevated after a 12 week recovery period in all animals (67 +/- 5 versus 91 +/- 6 ml, p less than 0.05). Thus prolonged tachycardia resulted in significant functional changes associated with cardiac enlargement, which were not immediately reversible.

Computer-assisted intraoperative mapping of the entire ventricular epicardium in the Wolff-Parkinson-White syndrome.

Intraoperative mapping with a hand-held, roving electrode requires a sustained rhythm lasting 5 to 10 minutes. To overcome this limitation, a computerized mapping system that records from 60 epicardial electrodes simultaneously was used to study 16 patients with Wolff-Parkinson-White syndrome. A sock containing 6 rows of electrodes arranged concentrically from base to apex was place over the ventricles. The total time from placing the sock to analyzing the most basal row of electrode recordings was 5 minutes. A 39 X 44-mm plaque containing 56 electrodes was than placed across the atrioventricular (AV) groove for detailed simultaneous mapping of the ventricle and atrium in the preexcited region identified from the most basal row of sock electrodes. During plaque placement and recording, the remaining sock recordings were analyzed and a complete isochronal epicardial map was drawn. The plaque recordings were then analyzed. This technique rapidly detects early activation at the AV groove as do other computer systems using only a band of electrodes around the AV groove. Also, complete epicardial mapping supplied important additional information. One patient with a posterior paraseptal accessory pathway had ventricular epicardial breakthrough below the strip recorded by the AV band. When more than 1 early activation site was present along the AV groove, complete maps allowed multiple pathways to be differentiated from normal activation fronts ascending from the bundle branches. Complete epicardial maps allowed the study of rapidly changing or short-lived electrical events including isolated premature impulses, initiation and termination of reciprocating tachycardia by pacing, entrainment and changing degrees of fusion created by pacing during reciprocating tachycardia, and ventricular responses during atrial fibrillation.(ABSTRACT TRUNCATED AT 250 WORDS)

Incidence of major injuries, severe colic, and acute laminitis at American Horse Shows Association A- and B-rated shows.

A questionnaire was designed to determine the incidence of major injuries, severe colic, and acute laminitis in horses at American Horse Shows Association (AHSA) A- and B-rated shows, ie, those shows requiring a veterinarian in attendance. The questionnaire was sent to the show stewards and filled out by the attending veterinarians of the AHSA A- and B-rated shows held from January 1982 to October 1984. Of 2,952 questionnaires sent out, 799 (27%) were completed and returned by veterinarians; 99 of them were discarded because of incomplete or misleading information. The study revealed a low incidence of major injuries, severe colic, and acute laminitis in horses at AHSA A- and B-rated shows. In 700 horse shows surveyed, there was a total of 231 major injuries, severe colic, or acute laminitis. Considering that the shows surveyed had a median number of 193 horses and a median duration of 3 days, the 231 injuries represent a relatively small number, compared with the general population at risk.

Simple step gradient elution of the major high-energy compounds and their catabolites in cardiac muscle using high-performance liquid chromatography.

The majority of high-energy nucleotides and their catabolites are separated in a single 13-min run using reversed-phase high-performance liquid chromatography. Economical step gradient elution of these compounds with a Nova Pak-A C18, 5-microns, 10 cm X 8 mm column accomplishes this separation with recoveries of 94-100% and sensitivities of 1-5 pmol. Furthermore, the total adenine nucleotide pool can now be quantitated precisely in a simple and easily automated procedure.

Diagnosis and recommended management of esophageal perforation and rupture.

Over the past 47 years (1937 to 1984), a total of 127 patients with esophageal perforation or rupture were evaluated at Duke Medical Center or the Durham Veterans Administration Medical Center. In 13 patients, the diagnosis was established at the time of autopsy and in the remaining 114, the diagnosis was established clinically. The etiology, radiological findings, underlying esophageal disease, time interval between onset of symptoms and therapy, and eventual outcome were evaluated. Patients with anastomotic leaks and those in whom carcinoma resulted in perforation or fistula were excluded. Iatrogenic causes were responsible for 55% of perforations, followed by spontaneous rupture in 15%, foreign body perforation in 14%, and traumatic perforation in 10%. Of the 127 patients, 114 underwent treatment involving primary closure (43%), drainage alone (28%), resection (9%), or nonoperative therapy (20%). The overall mortality among these 114 patients was 21%. Fourteen patients sustained a major complication requiring additional operative intervention. The overall mortality among patients requiring reoperation was 57%. Survival was significantly influenced by a delay in treatment of greater than 24 hours. With the exception of nonoperative therapy, survival was improved for all forms of treatment instituted within 24 hours. Primary closure within 24 hours resulted in the most favorable outcome (92% survival). In addition to early treatment, other factors associated with a favorable outcome included traumatic perforation (100% survival), foreign-body perforations (94% survival), and iatrogenic causes (80% survival). Spontaneous rupture resulted in the lowest survival (37%). The incidence of esophageal perforation has increased dramatically since 1967.(ABSTRACT TRUNCATED AT 250 WORDS)

A derivative of an ataxia-telangiectasia (A-T) cell line with normal radiosensitivity but A-T-like inhibition of DNA synthesis.

Ataxia-telangiectasia (A-T) cells are hypersensitive to the lethal effects of ionizing radiation and fail to inhibit DNA synthesis following radiation exposure. A cell line derived from an A-T line following DNA-mediated gene transfer has normal radiation sensitivity, but the kinetics of DNA synthesis after gamma-irradiation are similar to those of A-T cells.

Review of large intestinal motility and mechanisms of impaction in the horse.

The large intestine has distinct motility patterns which include non-rhythmic haustral kneading of ingesta and stronger rhythmic retropulsive and propulsive contractions which move ingesta along the tract. A variable site electrical pacemaker exists at the pelvic flexure where the strong rhythmic contractions begin. The large intestine can contract adequately with only the intrinsic nerve supply intact. Neurotransmitters have profound effects on large intestinal activity.

Time dependence of airways and lung parenchymal recoil hysteresis.

Hysteresis of airways and lung parenchymal recoil was examined in normal subjects by measuring specific conductance (sGaw) and lung elastic recoil (Pst,L) before and 5, 10, 15, and 30 s after deep inspiration (DI). Routine lung function tests were normal before and after inhaled metaproterenol. sGaw increased significantly for 10 s after DI. Also, sGaw(DI) was greater than sGaw in 11 of 12, 8 of 12, 7 of 12, and 6 of 12 subjects at 5, 10, 15, and 30 s, respectively, after DI. The response of sGaw to DI and metaproterenol correlated significantly with each other (r = 0.82, P less than 0.001). However, after metaproterenol, sGaw(DI) did not exceed sGaw. Pst,L decreased significantly for 15 s after DI, with the lowest measured Pst,L(DI) values occurring 5 s after DI (P less than 0.01-0.001). Both sGaw(DI) and Pst,L(DI) values returned to base line (preinspiration) in a time-dependent exponential manner, with time constants of 9.2 +/- 4.9 and 11.3 +/- 6.1 s, respectively; these time constants were not significantly different from each other. We conclude that airways hysteresis is the predominant finding in normal subjects (even without prior pharmacological bronchoconstriction) before but not after metaproterenol; Pst,L decreases after DI and, in normal individuals, returns to base line in a time-dependent manner; and the time-dependent behavior of airways and lung parenchymal hysteresis have opposite (and unequal) effects on airway caliber.

Comparison of once-a-day and twice-a-day theophylline in asthma.

Sustained-release theophylline compounds given once (Uniphyl) or twice (Theodur) daily were compared in adult asthmatics. Following a single dose of oral medication, large and peripheral airways bronchodilation occurred; response to theophylline correlated significantly with the log plasma theophylline concentration. Cardiac output and stroke volume, measured noninvasively using the acetylene technique, also increased significantly. During maintenance therapy, both preparations caused similar improvements in pulmonary function and symptoms; however, side effects were less with once-daily therapy.
