[Interaction of polymorphonuclear leukocytes, bradykinin and carragenin. Transference reaction in the rat].

1. Injected in the paw of the rat, polymorphonuclear leucocytes do not increase the oedematogen action of bradykinin, but increase the action of lambda carrageenan. 2. This potentiation of carrageenan action is not modified when PG biosynthesis in leucocytes is inhibited by indomethacin or aspirin. It does not appear in rats previously treated by indomethacin or aspirin. 3. Our results suggest that rat polymorphonuclear leucocytes increase the inflammatory reaction, when they are stimulated by carrageenan, by the release of a phospholipase A2 activity which induces PG biosynthesis in rat paw tissues.

[Platelets, lysosomal enzymes and anaphylactic shock in the rat].

1. Rat serum levels in beta-glucuronidase and beta-galactosidase are higher than plasma levels. Rat platelets release these lysosomial enzymes during blood coagulation in vitro. 2. After anaphylactic shock, in the sensitized rat, there is no increase in beta-galactosidase and beta-glucuronidase plasma levels. The tissues of the sensitized rat do not release these enzymes during the antigen-antibody reaction. The blood platelet level is diminished after anaphylactic shock and the serum levels of the lysosomial enzymes are decreased. 3. In thrombopenic rat, anaphylactic shock is identical as in control animals. Rat platelets do not play a significant role in the anaphylactic shock.

[Inhibition of prostaglandin synthesis by (phenylthio-4 phenylamino)-2 nicotinic acids].

In the rat, (phenylthio-4 phenylamino)-2 nicotinic and [(chloro-4 phenylthio)-4 phenylamino]-2 nicotinic acids inhibit the hypotensive prostaglandin-mediated action of arachidonic acid. They inhibit also the formation from arachidonic acid, of prostaglandin-like substances by chopped rat lungs and of malonaldehyde by rat platelets. They are prostaglandin synthetase inhibitors, three to ten times less active as indomethacin.

[Inhibition of lysosomal enzyme liberation by indomethacin in vivo].

The volume of the peritoneal exudate induced in the rat by iota carrageenan is reduced by subcutaneous administration of indomethacin while the concentrations of three lysosomial enzymes in the exudate are slightly increased or not modified. Thus, the total enzymatic activities of the exudate are reduced by indomethacin. The leucocyte accumulation remains unchanged in the indomethacin treated rats. During the development of the peritoneal exudate, the circulating plasma displays a high degree of lysosomial enzymes activity which is suppressed by indomethacin at the dose of 4 mg/kg.

[Formation of human immunoreactive LMW and HMW-kininogens during coagulation. Comparison with bradykinin activity].

Variations of the levels of human HMW and LMW kininogens have been studied in serum and in plasma incubated in vitro during 30 hours, at three different temperatures: 4 degrees, 22 degrees and 37 degrees C. There is a small difference between the level of LMW kininogen in plasma and in serum, but the serum and plasma level of LMW kininogen are almost stable during the time of incubation at the three temperatures. However, HMW kininogen is reduced in serum to about fifty percent of its plasma level. It decreases in plasma and in serum during incubation, overall at 4 degrees C. A strict parallelism stays between the radioimmunoassay and the bioassay.

[Cardiovascular reactivity to arachidonic acid, prostaglandin E2, prostacyclin and bradykinin in carrageenan treated rats].

In the rat, during carrageenan-induced peritonitis, the hypotensive activity of arachidonic acid, which depends on PG biosynthesis, is increased during the first hours, and thereafter is not modified. The vasodilator action of PGE2 is reduced during the first day while the actions of PGI2 and bradykinin are not modified. The concentrations of plasma albumin and haptoglobin, two inhibitors of PG biosynthesis are reduced during the first hours. Thereafter the concentration of haptoglobin is increased by 100% while the concentration of albumin remains diminished. During this peritonitis, no plasmatic inhibitory influence on PG synthesis is seen. The anti-inflammatory action of carrageenans does not depend on PG synthesis inhibition.

[Fibrinogen and plantar edema induced by lambda carrageenan].

1. Batroxobine which induces fibrinogen consumption, accelerates the development of 48/80 and lambda carrageenan oedema: in the rat the various products released from fibrinogen and fibrin increase these inflammatory reactions. 2. Heparin and epsilon-aminocaproic acid have no influence on carrageenan oedema: the derivatives of fibrinogen do not take part in the constitution of the inflammatory reaction induced by carrageenan. 3. The anti-inflammatory action of this sulfated polygalactose does not depend on its anti-coagulant activity.

[Radioimmunologic determination of human kininogen of high molecular weight].

HHW-kininogen has been purified from human plasma. We have then studied the optimal conditions for labeling this protein with 125I. This molecule has been used for the preparation of a radioimmunoassay. We have studied the analytical performances of our dosage : sensitivity, precision, accuracy and specificity.

[Amino liberation in Brown-Norway rats].

When injected intravenously with dextran or O-hydroxyethyl derivatives of rutoside, rat Brown Norway BN/Mai Pfd f exhibit a fall in the blood pressure and an increase of vascular permeability with oedema formation. As the rat is genetically deprived of plasmatic prekallikrein an of kininogens, activation of the kinin system is not necessary neither for mast-cell degranulation nor for amine release.

[Peritoneal reactions to hydrogen peroxide in the rat].

Hydrogen peroxide, when injected in diluted isotonic solution in the peritoneal cavity of the rat, provokes an increase of the local vascular permeability. Histamine plays a minor role in the development of these lesions.
