Structure-activity relationship of the effect of cis-unsaturated fatty acids on heart sarcolemmal phospholipase D activity.

This study examined the role of fatty acids on the phosphatidylcholine-specific phospholipase D (PLD) function of purified sarcolemmal (SL) membranes isolated from rat hearts. The enzyme's hydrolytic activity was determined by measuring [14C] phosphatidic acid formation from exogenous [14C] phosphatidylcholine (PtdCho) in the absence or presence of the sodium salts of various saturated or unsaturated long-chain fatty acids (FA). In certain experiments the enzyme was also assayed in the transphosphatidylation mode. Cis-unsaturation and free carboxyl groups were structural prerequisites for the stimulatory effect exerted by FA on SL PLD. The most effective compounds were arachidonate and oleate, which maximally activated PLD at 4 and 5 mM concentration, respectively. To verify if a detergent-like mechanism was involved in PLD activation, anionic, zwitterionic and non-ionic detergents were used. Only anionic taurodeoxycholate had a slight effect, which was about 7% of that achieved by arachidonate or oleate. These results suggest that cis-unsaturated FA activate cardiac sarcolemmal PLD by a mechanism(s) which seems to be unrelated to non-specific perturbation of the membrane.

Changes in activity of energy dissipating mechanisms in wheat flag leaves during senescence.

Excitation energy dissipation, including the xanthophyll cycle, during senescence in wheat flag leaves grown in the field was investigated at midday and in the morning. With progress of senescence, photosynthesis (Pn) and actual PSII photochemical efficiency (PhiPSII) decreased markedly at midday. The decrease in extent of Pn was greater than that of PhiPSII. However, there was no significant decline in Pn and PhiPSII observed in the morning, except in leaves 60 days after anthesis. The kinetics of xanthophyll cycle activity, thermal dissipation (NPQ), and q (f) observed at midday during senescence exhibited two distinct phases. The first phase was characterized by an increase of xanthophyll cycle activity, NPQ, and q (f) during the first 45 days after anthesis. The second phase took place 45 days after anthesis, characterized by a dramatic decline in the above parameters. However, the qI, observed both at midday and in the morning, always increased along with senescence. A larger proportion of NPQ insensitive to DTT (an inhibitor of the de-epoxidation of V to Z) was also observed in severely senescent leaves. In the morning, only severely senescent leaves showed higher xanthophyll cycle activity, NPQ, q (f), and qI. It was demonstrated that, at the beginning of senescence or under low light, wheat leaves were able to dissipate excess light energy via NPQ, depending on the xanthophyll cycle. However, the xanthophyll cycle was insufficient to protect leaves against photodamage under high light, when leaves became severely senescent. The ratio of (Fj - Fo)/(Fp - Fo) increased gradually during the first 45 days after anthesis, but dramatically increased 45 days after anthesis. We propose that another photoprotection mechanism might exist around reaction centres, activated in severely senescent leaves to protect leaves from photodamage.

Ultrastructural changes of nucleoli in common wheat induced by actinomycin D.

Common wheat root tip meristematic cells were treated with low concentrations of actinomycin D (ActD), then stained whole by silver nitrate. We showed by transmission electron microscopy that the typical nucleolar structure did not form, but a granular and fibrillar network was exhibited in the nucleolar region. Our results support a correlation between nucleolar organization/assembly and the activation of RNA Polymerase I transcription. Furthermore, we speculate that the fibrillar network present in the nucleolar region of ActD treated cells may represent the basic skeletal structure required to support the nucleolus.

Multicenter evaluation of the performance and clinical utility in longitudinal monitoring of the Bayer Immuno 1 complexed PSA assay.

We conducted a multicenter evaluation of the analytical and clinical performance of the automated Bayer Immuno 1 complexed PSA (cPSA) assay, and compared assay performance to the Bayer Immuno 1 PSA assay. We sought to determine whether measurements of cPSA could be of clinical utility in the management of patients with prostate cancer. Results of the 10-day imprecision across three evaluation sites produced total CV < 2.50% and an analytical sensitivity of 0.02 microgram/L. There was an increased trend in clinical sensitivity for prostate cancer with increasing stage of disease (71-86%). Clinical specificity for patients with benign urogenital disease was 74.8%, and for other nonprostate diseases ranged from 91.1-100%. Retrospective serial monitoring of 155 patients with prostate cancer demonstrated concordance of cPSA measurements to clinical status for 97% of the patients analyzed. Results from the clinical studies using the Bayer Immuno 1 cPSA assay were comparable to results obtained with the Bayer Immuno 1 PSA assay. The Bayer Immuno 1 cPSA assay demonstrates analytical performance and clinical effectiveness in the management of prostate cancer patients during the course of disease and therapy.

[Cloning and characterization of a novel mouse short-chain dehydrogenase/reductases cDNA mHsdl2#, encoding a protein with a SDR domaid and a SCP2 domain].

The short-chain dehydrogenases/reductases (SDRs) play important roles in body's metabolism. We cloned a novel mouse SDR cDNA which encodes a deduced HSD-like protein with a conserved SDR domain and a SCP2 domain. The 1.8 kb cDNA consists of 11 exons and is mapped to mouse chromosome 4B3. The corresponding gene is widely expressed in normal mouse tissues and its expression level in liver increases after inducement with cholesterol food. The predicted mouse HSDL2 protein, which has a peroxisomal target signal, is localized in the cytoplasm of NIH 3T3 cells.
