Fresh and predegenerate nerve allografts and isografts in trembler mice.

In order to investigate whether Schwann cell or myelin was the principal antigen responsible for nerve graft rejection, fresh nerve grafts and those in which myelin had been previously allowed to degenerate (predegenerate grafts) from both isogeneic BALB/c and allogeneic C57/B1 mice were inserted into trembler BALB/c mice. Schwann cells within nerve allografts from C57/B1 mice were rejected, whether or not the grafts contained myelin. Nerve isografts from normal BALB/c animals produced normally myelinated trembler axons within the grafted segments, and across these segments conduction velocity was restored towards the normal value. It is concluded that Schwann cells, not myelin, constitute the principal antigen within nerve allografts and it is Schwann-cell rejection that limits the successful use of nerve allografts.

A critical review of therapies in acute and chronic inflammatory demyelinating polyneuropathies.

Acute and chronic inflammatory demyelinating neuropathies are among the most common treatable neuropathies seen by neurologists. Evidence for effective therapy has only recently been provided by randomized or controlled trials. In the Guillain-Barré syndrome such evidence does not support the use of corticosteroids or immunosuppressive agents. However, when used early in the course, plasma exchange (PE) has been shown to lessen the severity and shorten the duration of the disease; it is indicated only in severely paralyzed patients or those whose rapid deterioration suggests the imminent need for ventilatory support. Some patients with chronic inflammatory demyelinating polyneuropathy (CIDP) respond to corticosteroid therapy or other immunosuppressive agents. PE is also effective in certain patients, but there is no sound evidence to date concerning combined immunosuppression and PE. The rationale of PE in these conditions and whether it is the removal of a toxic factor or the replacement fluid used that is beneficial remains to be clarified.

The effect of cell numbers on Schwann cell radioimmunoassay for serum cytotoxicity.

Immune cytotoxic effects on cultured Schwann cells can be monitored by a 51Cr release cytotoxicity assay. This paper presents the results of experiments using different cell numbers in the assay and a revision of the method for calculating the % specific release of 51Cr. The results give a more reliable and quantitative method for distinguishing between different sera.
