Quantitative structure-property relationship studies on n-octanol/water partitioning coefficients of PCDD/Fs.

Based on some fundamental quantum chemical descriptors computed by PM3 Hamiltonian, by the use of partial least-squares (PLS) analysis, a significant quantitative structure-property relationship (QSPR) model for logKow of polychlorinated dibenzo-p-dioxins and dibenzo-p-furans (PCDD/Fs) was obtained. The QSPR can be used for prediction. The intermolecular dispersive interactions and thus the bulkness of the PCDD/Fs are the main factors affecting the logKow. The more chlorines in the PCDD/F molecule, the greater the logKow values.

Quantitative structure-property relationship study on reductive dehalogenation of selected halogenated aliphatic hydrocarbons in sediment slurries.

In this study, by the use of partial least squares (PLS) method and 26 quantum chemical descriptors computed by PM3 Hamiltonian, a quantitative structure-property relationship (QSPR) model was developed for reductive dehalogenation rate constants of 13 halogenated aliphatic compounds in sediment slurry under anaerobic conditions. The model can be used to explain the dehalogenation mechanism. Halogenated aliphatic compounds with great energy of the lowest unoccupied molecular orbital (Elumo), total energy (TE), electronic energy (EE), the smallest bond order of the carbon-halogen bonds (BO) and the most positive net atomic charges on an atom of the molecule (q+) values tend to be reductively dehalogenated slow, whereas halogenated aliphatic compounds with high values of molecular weight (Mw), average molecular polarizability (alpha) and core-core repulsion energy (CCR) values tend to be reductively dehalogenated fastest.

Quantitative structure-property relationships (QSPRs) on direct photolysis of PCDDs.

By the use of partial least squares (PLS) method and 27 quantum chemical descriptors computed by PM3 Hamiltonian, a statistically significant QSPR were obtained for direct photolysis quantum yields (Y) of selected Polychlorinated dibenzo-p-dioxins (PCDDs). The QSPR can be used for prediction. The direct photolysis quantum yields of the PCDDs are dependent on the number of chlorine atoms bonded with the parent structures, the character of the carbonoxygen bonds, and molecular polarity. Increasing bulkness and polarity of PCDDs lead to decrease of log Y values. Increasing the frontier molecular orbital energies (Elumo and Ehomo) and heat of formation (HOF) values leads to increase of log Y values.

Modified approach of hand-assisted laparoscopic nephroureterectomy for transitional cell carcinoma of the upper urinary tract.

Objectives. To report a modified approach for hand-assisted laparoscopic nephroureterectomy (HALNU).Methods. Seven patients with localized transitional cell carcinoma of the upper urinary tract underwent unilateral HALNU. Patients were placed in a 60 degrees oblique position during the entire procedure. Via a 7-cm Gibson incision on the lesion side, the distal ureterectomy and bladder cuff excision were done by an open method without opening the bladder. Then, with the surgeon's hand inserted into the peritoneal cavity by way of the same wound, HALNU was performed with two to three additional laparoscopic ports. The perioperative parameters were compared with those of 15 cases of conventional open nephroureterectomy.Results. Patients in the HALNU group had significantly less mean blood loss (140 versus 455 mL) and earlier resumption of oral intake (33 versus 61 hours), required fewer narcotics (38 versus 70 mg of morphine sulfate equivalent), and were discharged earlier (7.33 versus 9.1 days), with a faster convalescence to normal activity (3.7 versus 5.6 weeks; all P < 0.05). The total mean surgical time was 3.7 hours for the HALNU group.Conclusions. Our approach used the same incision to both excise the distal ureter and apply the hand-assist device. It also preserved the benefits of the minimal invasiveness of laparoscopic surgery compared with its open counterpart.

A preliminary in vitro study on the fabrication and tissue engineering applications of a novel chitosan bilayer material as a scaffold of human neofetal dermal fibroblasts.

The bilayer structure of chitosan film and sponge was designed as a scaffold of skin tissue engineering and a dermis substitute. It was processed successively via the formation of a dense chitosan film by casting method and a porous chitosan sponge by lyophilization. The dry thickness of the film layer was 19.6 microm and that of the sponge layer was controlled at 60-80 microm. Porogens such as sodium chloride, glucose, and sucrose were used to create large pores of the chitosan sponge layer. Human neofetal dermal fibroblasts were seeded in the chitosan sponge layer and cultured for 4 weeks. It was found that the cells could grow and proliferate well in an extended shape on the flat bottom of large pores with 15-100 microm width and in spherical form on the rough pore walls or at the edges of micropores less than 5 microm. Fibroblasts after the culture could bind tightly with the sponge layer via newly formed extracellular matrices to give a living cell-matrix-chitosan composite. The bilayer chitosan material remained stable in shape and size during the cell culture. The results suggested that the bilayer chitosan material would be an alternative of collagen materials which was obviously contracted during cell culture.

Elastic and plastic behavior of plasma-sprayed hydroxyapatite coatings on a Ti-6Al-4V substrate.

A novel approach that combines the indentation tests with nonlinear finite element modeling (FEM) is proposed to estimate the elastic/plastic constitutive relation of plasma-sprayed hydroxyapatite (HA) coatings on a Ti-6Al-4V substrate. The Ramberg-Osgood constitutive equation can well describe the deformation behavior of plasma-sprayed HA coating on Ti-6Al-4V. A reasonable estimation for the elastic modulus of the HA coatings is given, based on the fact that the coating consists of a crystalline phase, an amorphous phase and pores. The Ramberg-Osgood equations of the coatings indicate that the post-treatment increases both resistance to elastic and plastic deformation, but no effect on strain hardening behavior. The post-treatment, however, could reduce the resistance to coating/substrate separation. The influence of titanium substrate becomes more significant with decreasing coating thickness and increasing indentation load because the plasma-sprayed HA coatings exhibit much less resistance to indentation deformation than does Ti-6Al-4V.

Normal TCRbeta transcription and recombination in the absence of the Jbeta2-Cbeta2 intronic cis element.

The developmental regulation of antigen receptor gene transcription and recombination are mediated by cis regulatory elements. At the T cell receptor beta chain locus (TCRbeta), two DNase I hypersensitive sites within the Jbeta2-Cbeta2 intron contained binding sites for NF-kappaB and additional nuclear factors and were postulated to be involved in controlling TCRbeta transcription and V(D)J recombination. To test this possibility, we deleted these elements from the mouse genome by homologous recombination and assayed the effect on transcription of both the germline and rearranged TCRbeta locus, and on TCRbeta rearrangement in T and B lymphocytes. We found that TCRbeta transcription and V(D)J recombination and T cell development were normal in these mutant mice. Therefore, the Jbeta2-Cbeta2 intronic elements are dispensable for TCRbeta assembly and function.

Disruption of the Crithidia fasciculata KAP1 gene results in structural rearrangement of the kinetoplast disc.

The mitochondrial DNA (kinetoplast DNA) in trypanosomatids exists as a highly organized nucleoprotein structure with the DNA consisting of thousands of interlocked circles. Four H1 histone-like proteins (KAP1, 2, 3 and 4) are associated with the kinetoplast DNA in the trypanosomatid Crithidia fasciculata. We have disrupted both alleles of the KAP1 gene in this diploid protozoan and shown that expression of the KAP1 protein is eliminated. The mutant strain is viable but has substantial rearrangement of the kinetoplast structure. Expression of the KAP1 protein from an episome restored expression of the KAP1 protein in the mutant strain and also restored a normal kinetoplast structure. These studies provide evidence that the KAP1 protein is involved in kinetoplast DNA organization in vivo but is nonessential for cell viability.

Is it possible to develop a QSPR model for direct photolysis half-lives of PAHs under irradiation of sunlight?

By the use of the partial least squares method and 11 fundamental quantum chemical descriptors computed from the PM3 Hamiltonian, a Quantitative Structure-Property Relationship model was obtained for direct photolysis half-lives of selected polycyclic aromatic hydrocarbons (PAHs) under irradiation of sunlight. Direct photolysis half-lives for some other PAHs without reported values were predicted. It was concluded from the model that the main factors affecting photolysis half-lives of PAHs under irradiation of sunlight are PAH absolute hardness and electronegativity, which are related to the energy difference between the lowest unoccupied molecular orbital and the highest occupied molecular orbital, (Elumo - Ehomo) and (Elumo + Ehomo), respectively. Increasing Ehomo and the average molecular polarizability (alpha) values of the PAHs leads to decrease of photolysis half-lives. Increasing (Elumo - Ehomo) and Elumo values of the PAHs leads to an increase of the PAH photolysis half-lives.

Intracerebral transplantation of bone marrow stromal cells in a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine mouse model of Parkinson's disease.

Adult C57BL/6 mice were injected with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). Intrastriatal transplantation of bone marrow stromal cells (MSCs) was performed 1 week after MPTP administration. MSCs were harvested from donor adult mice, and then cultured and prelabeled with bromodeoxyuridine (BrdU). MPTP-Parkinson's disease (PD) mice treated with intrastriatal injection of phosphate-buffered saline (PBS), and normal non-MPTP mice were used as controls. MPTP-PD mice with MSC intrastriatal transplantation exhibit significant improvement on the rotarod test (P<0.05) at day 35 compared with PBS controls. Immunohistochemistry shows that BrdU reactive cells survive in the transplanted areas in the MPTP-PD striatum at least 4 weeks after administration. Scattered BrdU reactive cells express tyrosine hydroxylase (TH) immunoreactivity. Our findings suggest that MSCs injected intrastriatally survive, express dopaminergic protein TH immunoreactivity, and promote functional recovery.

Characterization of a high voltage flat panel display unit using nanotube-based emitters.

Details are given of an experimental investigation carried out to study the field electron emission characteristics of a field emission flat panel display unit using a carbon nanotube-epoxy composite as electron emission material. These include: (i) dependence of direct emission current-voltage characteristic on vacuum gap spacing, (ii) the variation of the proportion of emission current passing through an aperture hole of a gate electrode with changing structural parameters of the device, and (iii) the uniformity and display characteristics of a typical display unit. Our findings indicate that it is very likely for one to produce a near-market prototype high voltage field emission flat panel display, if more sophisticated fabrication and assembly technique is adapted.

Microfabrication and characterization of gated amorphous diamond-based field emission electron sources.

Gated field emission electron sources of amorphous diamond (a-D) coated Si tips and a-D diodes on a rough Si substrate were studied, detailing the deposition and characterization of the thin film, the fabrication processes and the emission behavior of the electron sources. Mechanisms responsible for the emission process of the a-D coated devices are proposed. A comparison of the field emission performance of the two types of devices is presented. In addition, future improvements of the a-D diode on a rough Si cathode are discussed.

Study of field electron emission spectral characteristics of amorphous carbon-nitride film.

The study of field electron energy spectra from amorphous carbon-nitride (a-CN) films is essential to understand the mechanism of field emission from this promising material. In this work, the electron energy spectrum is studied by measuring the distribution of electron field emitted from individual emitting sites. The spectra are recorded at different electrical fields. It is found that the peak shift as well as the half-width increase with increasing applied fields. Furthermore, multi-peak features are observed at the low-energy side of the spectra. It is assumed that these peaks might originate from the interband states of a-CN film. We propose that although the emission mainly originates from current injection into conduction band of a-CN film, the electrons may also be emitted from interband states.

Study of the frequency response of the thin film cold cathode electron source of a lighting element.

Details of the recent experimental and theoretical studies of the frequency characteristic of the field emission electron source of a lighting element are given. The response times at different frequencies (0.02-200 kHz) and applied gap fields, acting on cathode surface, have been studied. A correlation has been found to exist between the response time and the frequency of applied voltage pulse. The response time remains almost constant within a range of frequencies between 1 and 30 kHz, and it remains nearly constant with increasing applied gap field higher than the threshold field. Finally, the cutoff frequency of the electron source is found, and with the current design of the electron source, it can be as high as 40 kHz. An equivalent circuit model is proposed, and theoretical results based on this model agree well with experimental findings.

Comparative demography of isogenic populations of Caenorhabditis elegans.

Demographic characteristics of the bacterial-feeding nematode Caenorhabditis elegans were determined in two long-lived mutant strains, TJ1052 (age-1), CB4876 (clk-1), and a wild-type strain, N2. Within each strain, there was little correlation between longevity and reproduction for individuals that lived longer than 10days. Long-lived mutant strains produced fewer eggs than the wild type. Mean total life spans were 13.2days for the wild type, 21.9days for age-1, and 15.8days for clk-1; maximum life spans were 24days for the wild type, 47days for age-1, and 32days for clk-1. Differences in total life span resulted primarily from longer post-reproductive survival. The mean post-reproductive life spans were longer than the wild type by 183% in age-1 and 60% in clk-1. We conclude that (i) post-reproductive survival is not correlated with egg production within isogenic populations of C. elegans, and (ii) the relationship between reproduction and longevity differs among isogenic populations with specific longevity genes.

Predicting one-year mortality among elderly survivors of hospitalization for an acute myocardial infarction: results from the Cooperative Cardiovascular Project.

OBJECTIVES: We sought to develop a model based on information available from the medical record that would accurately stratify elderly patients who survive hospitalization with an acute myocardial infarction (AMI) according to their risk of one-year mortality. BACKGROUND: Prediction of the risk of mortality among older survivors of an AMI has many uses, yet few studies have determined the prognostic importance of demographic, clinical and functional data that are available on discharge in a population-based sample. METHODS: In a cohort of patients aged > or = 65 years who survived hospitalization for a confirmed AMI from 1994 to 1995 at acute care, nongovernmental hospitals in the U.S., we developed a parsimonious model to stratify patients by their risk of one-year mortality. RESULTS: The study sample of 103,164 patients, with a mean age of 76.8 years, had a one-year mortality of 22%. The factors with the strongest association with mortality were older age, urinary incontinence, assisted mobility, presence of heart failure or cardiomegaly any time before discharge, presence of peripheral vascular disease, body mass index <20 kg/m2, renal dysfunction (defined as creatinine >2.5 mg/dl or blood urea nitrogen >40 mg/dl) and left ventricular dysfunction (left ventricular ejection fraction <40%). On the basis of the coefficients in the model, patients were stratified into risk groups ranging from 7% to 49%. CONCLUSIONS: We demonstrate that a simple risk model can stratify older patients well by their risk of death one year after discharge for AMI.

Multimodal standardization of voice among four multicultural populations formant structures.

A stratified random sample of 20 males and 20 females matched for physiologic factors and cultural-linguistic markers was examined to determine differences in formant frequencies during prolongation of three vowels: [a], [i], and [u]. The ethnic and gender breakdown included four sets of 5 male and 5 female subjects comprised of Caucasian and African American speakers of Standard American English, native Hindi Indian speakers, and native Mandarin Chinese speakers. Acoustic measures were analyzed using the Computerized Speech Lab (4300B) from which formant histories were extracted from a 200-ms sample of each vowel token to obtain first formant (F1), second formant (F2), and third formant (F3) frequencies. Significant group differences for the main effect of culture and race were found. For the main effect gender, sexual dimorphism in vowel formants was evidenced for all cultures and races across all three vowels. The acoustic differences found are attributed to cultural-linguistic factors.
