Experimental infection with Scedosporium inflatum.

The experimental pathogenicity of Scedosporium inflatum (three isolates) and Scedosporium apiospermum (one strain) was determined following intraperitoneal and intravenous injection into immunosuppressed and normal mice. Immunosuppressed animals were treated with cortisone acetate. The results show that the strains studied displayed positive tropism to kidney and brain. In the histopathological study of the first organ, we initially observed abscesses at the glomerular level and later in the renal pelvis. In the brain, we detected large areas with necrosis, which produced important neurological disorders resulting in convulsive movements and a stiff neck. The isolates of S. inflatum were more virulent (45% mortality) than S. apiospermum (16% mortality). The inoculation method did not significantly affect the mortality rate of the mice by either fungus. In infections caused by S. inflatum the percentage of immunosuppressed mice which died was higher in comparison to that of the untreated group. On the other hand no differences were observed in the mortality of either group animals inoculated with S. apiospermum.

Re-evaluation of the synonymy between Keratinomyces ceretanicus and Trichophyton ajelloi.

In 1987, Keratinomyces ceretanicus was reduced to synonymy with Trichophyton ajelloi based on mating between the putative ex-type culture, ATCC 58594, and minus mating strains of Arthroderma uncinatum. Although we confirm that ATCC 58594 produces fertile ascomata when mated with A. uncinatum, we demonstrate that this strain is not representative of the type of K. ceretanicus and conclude that K. ceretanicus should be maintained as a separate taxon.

Age effects on monoamine turnover of the rat substantia nigra.

Measurement of turnover of dopamine (DA), noradrenaline (NA) and serotonin (5-hydroxytryptamine (5-HT), and their metabolites has been performed in 6- and 24-month-old rats. Dopamine synthesis in 24-month-old rats did not show any change with respect to 6-month-old rats. However, our results seem to indicate decreased DA release in 24-month-old rats. This hypothesis could be supported by the changes found in the 3-methoxytyramine (3-MT) accumulation rate after monoamine oxidase inhibition with pargyline. The turnover of NA and its main metabolite, 3-methoxy-4-hydroxyphenylglycol (MHPG) decreased in 24-month-old rats compared with 6-month-old rats. Serotonin synthesis did not change in 24-month-old rats with respect to 6-month-old rats. However, the metabolism of 5-HT quantified as turnover of 5-hydroxy-3-indoleacetic acid (5-HIAA) increased in 24-month-old rats with respect to 6-month-old rats. The monoamine oxidase B:monoamine oxidase-A ratio increased in 24-month-old rats. The significance of these changes is discussed.

Age-dependent changes in the activity and isoenzymatic pattern of the phosphofructokinase in different areas of the central nervous systems.

Regional activity of phosphofructokinase enzyme and the amount of the isoenzyme C in 7 areas of young, adult and aged rats brain have been studied. The phosphofructokinase activity in substantia nigra decreased in adult and aged rats, but the maximum decrease was found in the septum of aged rats. There is a regional distribution of the isoenzyme C in the different areas studied. There was a general decrease in the amount of this isoenzyme in the areas studied with the exception of the hippocampus and the amygdala. The highest decrease was found in the septal area and in the diagonal band of Broca. These results are discussed in relation to the aging in different regions.

Effects of neonatal bilateral eye enucleation on postnatal development of the monoamines in posterior thalamus of the rat.

Levels of dopamine (DA), noradrenaline (NA) and 5-hydroxytryptamine (serotonin, 5-HT) and their metabolites, and the activities of tyrosine hydroxylase (TH), tryptophan hydroxylase (TPH) and monoamine oxidase A and B (MAO-A and MAO-B) have been determined in the rat posterior thalamus after enucleation during postnatal development. DA and 5-HT turnover rate have been measured as 3,4-dihydroxyphenylalanine (DOPA) and 5-hydroxytryptophan (5-HTP) accumulation rates after central decarboxylase inhibition by 3-hydroxybenzylhydrazine (NSD-1015). The major changes were an increase in noradrenergic and serotoninergic metabolism in enucleated animals compared with control animals. A decrease of the MAO-A to MAO-B ratio during postnatal development was found.

Turnover of dopamine and serotonin and their metabolites in the striatum of aged rats.

Turnover of dopamine (DA), serotonin [5-hydroxytryptamine (5-HT)], and their metabolites has been measured in adult and aged rats. Turnover rates of 3,4-dihydroxyphenylacetic acid (DOPAC), homovanillic acid (HVA), and 5-hydroxy-3-indoleacetic acid (5-HIAA) have been assayed from the disappearance rates after blocking by pargyline inhibition of monoamine oxidase (MAO) and from the accumulation rates by probenecid inhibition of the probenecid-sensitive transport system. DA and 5-HT turnover rates have been measured as accumulation rates of 3,4-dihydroxyphenylalanine and 5-hydroxytryptophan, respectively, after central decarboxylase inhibition by 3-hydroxybenzylhydrazine (NSD-1015) and as accumulation rates of DA and 5-HT after pargyline inhibition of MAO. The DA turnover rate after NSD-1015 was 23.9% lower in aged rats than in adults, whereas after pargyline there was no significant difference between the two age groups. The HVA fractional rate constant and turnover after pargyline were lower in aged rats than in adults, and HVA turnover after probenecid was higher in aged rats than in adults. The DOPAC-HVA pathway seems to be reinforced at the expense of DOPAC conjugation. In aged and adult rats whose 5-HT steady-state levels were not statistically different, the 5-HT turnover rate after pargyline and NSD-1015 treatment was lower in aged rats than in adults. An increase of 5-HIAA levels after pargyline and probenecid treatment in aged rats could be due to the handling stress.

Pharmacologic characterization of GABAA/benzodiazepine receptor in rat hippocampus during aging.

We have characterized the pharmacologic properties of the gamma-aminobutyric acid (GABAA)/benzodiazepine receptor complex in hippocampal membranes from 3-month- and 24-month-old Wistar rats. No major changes were found in [3H]flunitrazepam or [3H]muscimol binding parameters. Neither the dissociation constant(s) nor the Bmax for either ligand seemed to be modified during aging in hippocampus. Furthermore, the allosteric interaction between the barbiturates binding site and the GABA binding site, determined by pentobarbital stimulation of [3H] muscimol binding, remained unaltered. However, there was a significant increase with aging in the efficacy of the GABA-enhancement of [3H]flunitrazepam binding. On the other hand, we have also detected a significant increase in the proportion of type I benzodiazepine receptor in 24-month-old hippocampal membranes. We propose that the age-related increase in the efficacy of GABA-enhancement of [3H]flunitrazepam binding could be correlated with the increase in the proportion of type I benzodiazepine receptor. Based on these results it is tempting to speculate that the age-dependent modifications on the GABAA/benzodiazepine receptor might reflect an age-dependent neuronal degeneration of the hippocampus or the hippocampal formation.

Effects of enucleation on postnatal development of catecholamines and serotonin metabolism in the superior colliculus of the rat.

We have measured the level of catecholamines and serotonin and their principal metabolites, and the activities of tyrosine hydroxylase, tryptophan hydroxylase and monoamine oxidase-A and -B in the rat superior colliculus during postnatal development after bilateral removal of the eyes. The visual deprivation has different effects on the catecholamines and serotonin metabolism. The major changes in both amines were at day 15. Tyrosine hydroxylase and tryptophan hydroxylase activities increased during postnatal development but this increase was higher in enucleated compared with controls. An increase of the MAO-B to MAO-A ratio during postnatal development was found. The significance of these changes has been discussed.

Determination of levels of biogenic amines and their metabolites and both forms of monoamine oxidase in prefrontal cortex of aged rats.

Levels of dopamine, noradrenaline and serotonin and their metabolites and the activities of monoamine oxidase A and B, have been determined in the prefrontal cortex of the rat during ageing. Serotonin turnover rate has been measured as 5-hydroxytryptophan accumulation rate after central decarboxylase inhibition. The major changes were an increase of dopamine and noradrenaline levels and a decrease of serotonin in aged animals compared with control animals. A decrease of the MAO-A to MAO-B ratio was found in aged rats.

Experimental pathogenicity of Aphanoascus spp.

Immunodepressed mice were inoculated intraperitonealy with a suspension of conidia and micelia macerate of four species of Aphanoascus (A. keratinophilus, A. fulvescens, A. reticulisporus and A. verrucosus). All produced nodular lesions in the peritoneal cavity, especially in the liver, spleen, diaphragm and peritoneum. The recovery rate of the fungi on phytone-yeast extract agar after one month varied approximately from 20 to 80% depending on the organs. Sections of these revealed the presence of ascospores and hyphal fragments though only in the centre of the nodules. Fungal elements were not detected in organs which were apparently not affected, nor intravascularly.

[A drug prescription study in chronic pathology at a primary care center].

To evaluate the characteristics of the drug used for the therapy of chronic diseases, 1588 prescriptions from a sample of 549 continued drug prescription forms were evaluated. Every patient had a mean number of 2.9 drugs, 2.14 diagnoses and 1.41 drugs per diagnosis. The patients over 65 years had a higher number of drugs and diagnoses. A smaller number of diagnoses but a higher number of drugs per diagnoses was found for males. The drug subgroups with higher prescription rates were cardiac drugs (12.3%), diuretics (11.2%) and analgesics (9.9%). The most common diagnoses were hypertension and osteoarthritis, with 17.6% and 13.6% of prescriptions, respectively. A high rate of patients receiving vasodilators (12%) and benzodiazepines (14.7%) was found. The usefulness of the continued prescription forms for the control of chronic treatments is emphasized.

Early postnatal development of the monkey globus pallidus: a Golgi and electron microscopic study.

The globus pallidus of 20 monkeys ranging in age from newborn to 4 months was examined in Golgi-impregnated material and ultrastructurally. There was no discernible difference between the lateral and medial segments of the structure. At the light microscope level, all neuronal types described in the adult are found at birth. The most common, the large fusifom cell, shows initial signs of immaturity such as blunt protrusions and dendritic dilations at bifurcation points, as well as growth cones, filopodia, and filiform processes. These features become more rare with age, and by 4 months, the neurons appear fully mature save for the terminal dendritic arborizations which are still underdeveloped. From the earliest ages examined, the large globular cells and the interneurons are more mature than the previous type. The afferent radial fibers of striatal origin are observed from birth, but they are grouped in bundles only after 8 weeks. The density of their climbing branches increases over time, reaching a mature appearance by 16 weeks. Afferents entering from the ventral surface do not yet show clusters of varicosities at 2 weeks. At the latter age, plexuses of fine beaded fibers are already seen covering large extensions of the nucleus. The fine structure correlates well with the Golgi material. The basic features of the neuropil are present at birth, albeit with immature characteristics such as the incomplete covering of the dendrites with axonal boutons and the low level of myelination of the radial fibers. Growth cones and profiles with signs of degeneration are observed during the first month. In the early ages examined, most dendrites show large varicosities and protrusions, some of which are spinelike and can be postsynaptic to multiple terminals. The other dendritic type, with only an occasional axodendritic synapse, is also seen from birth and increases in size as a function of time. The type I axonal boutons, of probable striatal origin, are quite immature at birth, and their characteristic interdigitations are seen only after the first week. The type II, III, IV, and V boutons appear mature at all ages examined but crest synapses formed by the type III terminals are observed in the later stages of the study. Finally, postsynaptic vesicle-containing profiles are present at 4 weeks, but triadic synaptic arrangements are formed only by 16 weeks.(ABSTRACT TRUNCATED AT 400 WORDS)

Determination of monoamines and both forms of monoamine oxidase in the rat's substantia nigra during postnatal development.

Changes in biogenic amine content in the substantia nigra and in both forms of monoamine oxidase in substantia nigra and striatum of the rat during postnatal development (15-180 days) have been studied. Dopamine and serotonin had the same levels at day 15, however, each monoamine showed a different developmental profile. Dopamine levels and their metabolites (except 3-methoxytyramine) decreased during postnatal development. Serotonin levels and their main metabolite, 5-hydroxyindolacetic acid, underwent an increase during all stages studied. There were no statistically significant changes in noradrenaline levels until day 180 when they increased with respect to day 15. The highest activity of the monoamine oxidase-A in substantia nigra coincided with the highest 5-hydroxyindolacetic acid:serotonin ratio. Monoamine oxidase-A in the striatum did not change contrary to that which happened in substantia nigra. The monoamine oxidase-B:monoamine oxidase-A ratio increased during development both in the substantia nigra and the striatum. The significance of these changes is discussed.

Age-related changes of catecholamines and their metabolites content in the visual system of the rat.

The changes in the content of the catecholamines in each structure of the geniculate and extrageniculate visual system of the rat during the aging period (6-30 months) have been studied. Dopamine was found at lower levels than noradrenaline in all the structures. The dopamine and noradrenaline showed different developmental profiles. Dopamine and its metabolite levels decreased in the lateral geniculate and visual cortex and increased in superior colliculus and posterior thalamus. Noradrenaline and its metabolites increased in all structures during the aging period. However, 3-methoxy-4-hydroxyphenylglycol/noradrenaline and normetanephrine/noradrenaline ratios decreased in all structures except in superior colliculus. These results suggest age-related changes in the catecholamines in the visual system of the rat.

Alterations produced by sertaconazole on the morphology and ultrastructure of Candida albicans.

We studied the changes produced in Candida albicans yeast cells after treatment in vitro with sertaconazole, with light microscopy, transmission and scanning electron microscopy. Four different concentrations were evaluated (10(-6) M, 10(-5) M, 10(-4) M, 10(-3) M) and two times of action (12 and 17 h) for each concentration. This new antimycotic agent shows a considerable destructive effect in C. albicans cells. The alterations produced by sertaconazole were already manifested after 12 h at the concentration of 10(-6) M and a strong fungicidal activity recorded at 10(-4) M.

Cytoplasmic organelles in neurons of the dorsal lateral geniculate nucleus of aged rats.

The ultrastructural characteristics of the neurons containing complex convolutions have been studied in the dorsal lateral geniculate nucleus of the 31-month-old rat. Neurons were seen to contain oval or round dense bodies which were surrounded by a nuclear membrane and granular endoplasmic reticulum. Their perikarya showed rarely clusters of pleomorphic and small clear vesicles intermingled with a few larger vesicles of dense material. Dendrites occasionally exhibited intermediate forms between laminated bodies and complex convolutions. The significance of these features has been discussed.
