Heterobimetallic oxalato-bridged Cu(II)Re(IV) complexes. Synthesis, crystal structure and magnetic properties.

Three copper(II)-rhenium(IV) bimetallic complexes of formula [ReCl(4)(mu-ox)Cu(phen)(2)] (1), [ReCl(4)(mu-ox)Cu(phen)(2)].CH(3)CN (2), and [ReCl(4)(mu-ox)Cu(terpy) (H(2)O)][ReCl(4)(mu-ox)Cu(terpy)(CH(3)CN)] (3) (ox = oxalate anion, phen = 1,10-phenanthroline, and terpy = 2,2':6,2"- terpyridine) have been synthesized and their crystal structures determined by single-crystal X-ray diffraction. Complex 1 crystallizes in the triclinic system, space group P(-1), with a = 9.776(2), b = 11.744(3), c = 14.183(3) A, alpha =102.09(2) degrees, beta = 109.42(2) degrees, gamma = 107.11(2) degrees, and Z = 2, whereas 2 and 3 crystallize in the monoclinic system, space groups P2(1)/n and P2(1)/c, respectively, with a = 12.837(3), b = 17.761(4), c = 12.914(3) A, beta = 91.32(2) degrees, and Z = 4 for 2, and a = 8.930(2), b = 18.543(4), c = 27.503(6) A, beta = 94.67(2) degrees, and Z = 4 for 3. The structures of 1 and 2 are made up of neutral [ReCl(4)(mu-ox)Cu(phen)(2)] bimetallic units. Re(IV) and Cu(II) metal ions exhibit distorted octahedral coordination geometries, being bridged by a bis(bidentate) oxalato ligand. The presence of acetonitrile molecules of crystallization in 2 causes a somewhat greater separation between the bimetallic complexes and a different packing of these units in the crystal structure with respect to 1. The copper-rhenium separation across oxalato is 5.628(2) in 1 and 5.649(3) A in 2. The structure of 3 is made up of two different and neutral bimetallic units, [ReCl(4)(mu-ox)Cu(terpy)(H(2)O)] and [ReCl(4)(mu-ox)Cu(terpy)(CH(3)CN)]. In the first one, the oxalate group behaves as a bis(bidentate) ligand occupying one equatorial and one axial position in the elongated octahedral environment of Cu(II). The water molecule is axially coordinated. In the second one, the oxalate group behaves as a bidentate/monodentate ligand occupying the axial position in the square pyramidal environment of Cu(II). The acetonitrile molecule occupies a basal coordination position around the copper atom. These units are arranged in such a way that a chlorine atom of the first unit (Cl(1)) points toward the copper atom (Cu(2))of the second one (3.077(2) A for Cl(1)(.)Cu(2)), forming a tetranuclear species. The copper-rhenium separation across bis(didentate) oxalato is 5.504(3) A, whereas that through bidentate/monodentate oxalato is 5.436(2) A. The magnetic behavior of 2 and 3 has been investigated over the temperature range 1.8-300 K. A very weak and nearly identical antiferromagnetic coupling between Re(IV) and Cu(II) through bis(bidentate) oxalato occurs in 2 (J = -0.90 cm(-1)) and 3 (J = -0.83 cm(-1)); it is ferromagnetic in 3 through both the bidentate-monodentate oxalato (J = +5.60 cm(-1)) and the chloro (J = +0.70 cm(-1)) bridges.

Determination of residues of enrofloxacin and its metabolite ciprofloxacin in biological materials by capillary electrophoresis.

A method for the analysis of enrofloxacin and ciprofloxacin in chicken muscle using marbofloxacin as internal standard is proposed. Clean-up and pre-concentration of the samples are effected by solid-phase extraction and determination is carried out by capillary electrophoresis using a photodiode array detector. The calibration graphs are linear for enrofloxacin and ciprofloxacin from 10 to 300 microg/kg. The method recoveries for enrofloxacin and ciprofloxacin are 74 and 54%, respectively. The limit of detection for the two compounds is lower than 25 microg/kg, which allows the detection of positive muscle samples at the required maximum residue limits.

Validity of a quantitative technique to study striatal dopaminergic neurodegeneration by in vivo microdialysis.

The development of a technique that allows the direct quantitative study of the damage produced by a toxin on a specific neurotransmitter system is very important. For that, we have used the microdialysis technique to validate a method to study the specific drug's toxicity on dopaminergic (DAergic) striatal terminals. We perfused different MPP(+) and 6-hydroxydopamine (6-OHDA) concentrations, with different toxicity for DAergic terminals, 24 h after the implantation of the microdialysis probe (day 1). One day later (day 2), MPP(+) was perfused through the microdialysis probe and DA extracellular output measured. We hypothesize that the amount of extracellular dopamine (DA) obtained on day 2 is directly proportional to the neurotoxic damage produced on day 1. To corroborate this hypothesis tyrosine hydroxylase (TH) immunohistochemistry was also carried out on day 2. There was a clear correlation index between the amount of DA measured after MPP(+) perfusion and the lack of TH immunoreactivity measured as the radius of the area showing decrease in TH immunoreactivity around the cannula. These results show the possibility to measure DAergic remaining terminals after a toxic drug exposure by in vivo MPP(+) perfusion. The possibility to extend this neurotoxic study to another neurotransmitter systems is suggested.

The utilization of classical spin Monte Carlo methods to simulate the magnetic behavior of extended three-dimensional cubic networks incorporating M(II) ions with an S = 5/2 ground state spin.

The numerical simulations of the magnetic properties of extended three-dimensional networks containing M(II) ions with an S = 5/2 ground-state spin have been carried out within the framework of the isotropic Heisenberg model. Analytical expressions fitting the numerical simulations for the primitive cubic, diamond, together with (10-3) cubic networks have all been derived. With these empirical formulas in hands, we can now extract the interaction between the magnetic ions from the experimental data for these networks. In the case of the primitive cubic network, these expressions are directly compared with those from the high-temperature expansions of the partition function. A fit of the experimental data for three complexes, namely [(N(CH(3))(4)][Mn(N(3))] 1, [Mn(CN(4))](n)() 2, and [Fe(II)(bipy)(3)][Mn(II)(2)(ox)(3)] 3, has been carried out. The best fits were those obtained using the following parameters, J = -3.5 cm(-)(1), g = 2.01 (1); J = -8.3 cm(-)(1), g = 1.95 (2); and J = -2.0 cm(-)(1), g = 1.95 (3).

Light- and thermal-induced spin crossover in [Fe(abpt)2(N(CN)2)2]. Synthesis, structure, magnetic properties, and high-spin<-->low spin relaxation studies.

[Fe(abpt)2(N(CN)2)2] (abpt = 4-amino-3,5-bis(pyridin-2-yl)-1,2,4-triazole) represents the first example of an iron(II) spin-crossover compound containing dicyanamide ligand, [N(CN)(2)](-), as a counterion. It shows an incomplete two-step spin transition with around 37% of HS molecules trapped in the low-temperature region when standard cooling or warming modes, i.e., 1-2 K min(-)(1), were used. The temperature, T(1/2) approximately 86 K, at which 50% of the conversion takes place, is one of the lowest temperatures observed for an iron(II) spin-crossover compound. Quenching experiments at low temperatures have shown that the incomplete character of the conversion is a consequence of slow kinetics. The quenched HS state relaxes back to the LS state displaying noticeable deviation from a single-exponential law. The rate of relaxation was evaluated in the range of temperatures 10-60 K. In the upper limit of temperatures, where thermal activation predominates, the activation energy and the pre-exponential parameter were estimated as E(a) approximately 280 cm(-)(1) and A(HL) approximately 10 s(-)(1), respectively. The lowest value of k(HL) around 1.2 x 10(-)(4) s(-)(1) (T = 10 K) was obtained in the region of temperatures where tunneling predominates. A quantitative light induced excited spin state trapping (LIESST) effect was observed, and the HS --> LS relaxation in the range of temperatures 5-52.5 K was studied. From the Arrhenius plot the two above-mentioned characteristic regimes, thermal-activated (E(a) approximately 431 cm(-)(1) and A(HL) approximately 144 s(-)(1)) and tunneling (k(HL) approximately 1.7 x 10(-)(6) s(-)(1) at 5 K), were characterized. The crystal structure was solved at room temperature. It crystallizes in the triclinic P_1 space group, and the unit cell contains a centrosymmetric mononuclear unit. Each iron atom is in a distorted octahedral environment with bond distances Fe-N(1) = 2.216(2) A, Fe-N(2) = 2.121(2) A, and Fe-N(3) = 2.160(2) A for the pyridine, triazole, and dicyanamide ligands, respectively.

Potential role of endogenous brain-derived neurotrophic factor in long-term neuronal reorganization of the superior colliculus after bilateral visual deprivation.

The role of the brain-derived neurotrophic factor (BDNF), the BDNF receptor (TrkB), and the glutamic acid decarboxylase (GAD67) after neonatal, bilateral nerve deafferentiation during postnatal development was investigated in the rat superior colliculus (SC). BDNF and GAD67 mRNA expression were significantly increased in optic (Op) and intermediate gray (InG) layers at 5, 8, 15, and 21 days after birth, but not in adult animals. However, TrkB mRNA expression was not modified at any time tested. At 15 days, where changes in BDNF and GAD67 mRNAs were more evident, an upregulation of the NMDAR(1A) mRNA glutamate receptor in the Op and InG, a modification in the pattern of synaptic zinc in the superficial layers of SC, and an increased synaptophysin immunoreactivity in the Op was found. This indicates the existence of a synergic mechanism between BDNF and NMDA to determine refinement of connections after the loss of visual input in SC.

Aquaporins in the central nervous system.

In this review, we have tried to summarize most available data dealing with the aquaporin (AQP) family of water channels in the CNS. Two aquaporins have been identified so far in the CNS, AQP1 and AQP4. AQP1 is restricted to the choroid plexus of the lateral ventricles, which raises a role for this aquaporin in cerebrospinal fluid formation. AQP4 is the predominant water channel in the brain and it is more widely distributed than originally believed, with a marked prevalence over periventricular areas. In the first part of this review, we examine the complete distribution pattern of AQP4 in the CNS including its rostro-caudal localization to end with its subcellular location. After discussing scarce data dealing with regulation of aquaporins in the CNS, we focus in potential roles for aquaporins. Novel recent data highlights very important roles for this aquaporin in the normal and pathological brain including, among others, role in potassium buffering, body fluid homeostasis, central osmoreception and development and restoration of brain edema.

Long-lasting induction of brain-derived neurotrophic factor is restricted to resistant cell populations in an animal model of status epilepticus.

We have recently characterized an animal model of status epilepticus induced by a single intraseptal injection of kainate. Under these conditions, there is a delayed expanding apoptotic hippocampal and amygdalar cell death. In order to further characterize this animal model, we have performed a detailed time-course analysis of the appearance of cell death, brain-derived neurotrophic factor messenger RNA expression and astroglial and microglial response in different brain areas related to the limbic system. We found a long-lasting delayed apoptotic cell death in the hippocampal formation, amygdala, medial thalamus, dorsal endopiriform nucleus and multiple cortical areas from two to 21 days post-injection. There was a spatiotemporal correlation between the appearance of cell death and induction of brain-derived neurotrophic factor messenger RNA expression in the areas studied, and interestingly this induction was found in non-degenerating cells. We conclude that our animal model of status epilepticus exhibits remarkable features of recurrent seizure activity and provides evidence for a neuroprotective role of brain-derived neurotrophic factor against seizure-induced apoptotic cell death.

Evaluation of electrophoretic method versus chromatographic, potentiometric and absorptiometric methodologies for determing pK(a) values of quinolones in hydroorganic mixtures.

The advantages of using capillary electrophoresis (CE) over other methodologies for determining pK(a) values of drugs in hydroorganic media are discussed. The focus of the discussion based upon the pK(a) values of a series of quinolones determined in acetonitrile (MeCN)--water mixtures by CE, liquid chromatography, potentiometric, and spectrophotometric methods.

Group II metabotropic glutamate receptor activation protects striatal dopaminergic nerve terminals against MPP+-induced neurotoxicity along with brain-derived neurotrophic factor induction.

We have studied the in vivo effect of the selective agonist for group II metabotropic glutamate receptors (2S, 2'R, 3'R)-2-(2'3'-dicarboxycyclopropyl)glycine (DCG-IV) against MPP+-induced toxicity on rat striatal dopaminergic nerve terminals by using both microdialysis and immunohistochemical techniques. Perfusion of 1 mM DCG-IV during 1 h protected dopaminergic nerve terminals against the degeneration induced by a 15-minute perfusion of 1 mM MPP+. In addition, the microglial cell population was markedly activated 24 h after DCG-IV perfusion. The astroglial cell population was only markedly activated around the microdialysis probe. This protective effect seems to be dependent on protein synthesis since 1 mM cycloheximide, an inhibitor of protein synthesis, abolished the neuroprotective effect of 1 mM DCG-IV against MPP+ toxicity. Perfusion of DCG-IV induced an upregulation of striatal brain-derived neurotrophic factor (BDNF) mRNA expressing cells which were confined precisely around the microdialysis probe. Taken together, our results suggest that the induction and release of brain-derived neurotrophic factor (BDNF) by activated glial cells induced by DCG-IV perfusion may account for its protective action against MPP+-induced dopaminergic terminal degeneration.

PCR differentiation of Saccharomyces cerevisiae from Saccharomyces bayanus/Saccharomyces pastorianus using specific primers.

The aim of the present study was to design species-specific primers capable of distinguishing between Saccharomyces cerevisiae, Saccharomyces bayanus/Saccharomyces pastorianus. The 5'-specific primers were designed from the ITS-1 region (between positions 150 and 182 from the 3'-SSU end) and the 3'-specific primers were located in the LSU gene (positions 560-590 from the 5'-end of this gene). These primers were tested with different collections and wild strains of these species and the results showed that the primers were capable of distinguishing between S. cerevisiae strains and S. bayanus/S. pastorianus. Not enough sequence differences were found between S. bayanus and S. pastorianus to design specific primers for these species using this region. This method offers an effective tool for a quick differentiation of the Saccharomyces strains of the most common species involved in industrial processes.

Low selenium diet induces tyrosine hydroxylase enzyme in nigrostriatal system of the rat.

We have studied the effect of a selenium-deficient diet on the nigrostriatal dopaminergic system for 15 and 30 days. The neurochemical analysis demonstrated significant elevations in nigral DA levels after 15 and 30 days of selenium deficiency. The most significant change in striatum was an elevation in dopamine (DA) in 30-day-deficient animals. As a further step, we measured the levels of activity and mRNA expression of tyrosine hydroxylase (TH) and dopamine transporter (DAT). There were significant elevations in all of these parameters in the nigrostriatal system following selenium deficiency at the two time-points studied except for the striatal DA uptake at day 30, which dropped to control levels. Altogether, our results suggest that the decrease in antioxidant capacity due to a selenium deficiency promotes an increase in DA synthesis and turnover, which are clearly associated to the induction of TH. This effect may trigger a positive feed-forward mechanism that could increase the oxidative stress considering the capacity of DA to generate free radicals.

Synthesis, structural characterization, and Monte Carlo simulation of the magnetic properties of two new alternating MnII azide 2-D honeycombs. Study of the ferromagnetic ordered phase below 20 K.

Reaction of MnII and pyridine derivatives such as 4-methylpyridine (4-Mepy) and 4-ethylpyridine (4-Etpy) led to the new two-dimensional systems trans-[Mn(4-Mepy)2(N3)2]n (1) and trans-[Mn(4-Etpy)2(N3)2]n (2). Compound 1 crystallizes in the triclinic system, P1 group (a = 9.269(2) A, b = 9.635(3) A, c = 18.860(4) A, Z = 4), and compound 2 crystallizes in the monoclinic system, P2(1)/c group (a = 14.416(3) A, b = 8.515(2) A, c = 15.728(4) A, Z = 4). The two compounds show honeycomb structures based on dinuclear Mn-(mu-N3)2-Mn subunits linked to the four nearest-neighbor similar subunits by four end-to-end single azido bridges, but whereas the subunits of compound 1 show the end-to-end Mn-(mu 1,3-N3)2-Mn kind of bridges, compound 2 prefers the end-on Mn-(mu 1,1-N3)2-Mn fragment. Magnetically, compound 1 is an alternating 2-D system with two different antiferromagnetic interactions, whereas compound 2 corresponds to a two-dimensional ferro-antiferromagnetic system showing spin canting and permanent magnetization below 20 K. The coupling constant parameters J1 = -10.1 cm-1, J2 = -4.7 cm-1, and g = 2.019 for 1 and J1 = -5.3 cm-1, J2 = 2.9 cm-1, and g = 2.016 for 2 have been obtained from calculations using the Monte Carlo method based on the Metropolis algorithm.

Magnetic coupling through the carbon skeleton of malonate in two polymorphs of ([Cu(bpy)(H2O)][Cu(bpy)(mal)(H2O)])(ClO4)2 (H2mal = malonic acid; bpy = 2,2'-bipyridine).

Two polymorphic malonato-bridged copper(II) complexes of formula ([Cu(bpy)(H2O)][Cu(bpy)(mal)(H2O)])-(ClO4)2 (1 and 2) (bpy = 2,2'-bipyridine and mal = malonate dianion) have been prepared and their structures solved by X-ray diffraction methods. Compound 1 crystallizes in the monoclinic space group P2(1)/a, with a = 23.743(3) A, b = 9.7522(5) A, c = 27.731(2) A, beta = 114.580(10) degrees, and Z = 4. Compound 2 crystallizes in the orthorhombic space group Pbcn, with a = 23.700(5) A, b = 25.162(5) A, c = 9.693(5) A, and Z = 4. The structures of 1 and 2 are made up of uncoordinated perchlorate anions and malonate-bridged zigzag copper(II) chains grouped in an isosceles triangle running parallel to the b (1) and c (2) axes. These chains are built by a [Cu(bpy)(mal)(H2O)] unit acting as bis-monodentate ligand toward two [Cu(bpy)(H2O)] adjacent units through its OCCCO skeleton in an anti-anti conformation, whereas the OCO carboxylate bridges exhibit the anti-syn conformation. Compounds 1 and 2 contain four crystallographically independent copper(II) atoms, but the environment of all of them is distorted square pyramidal: the axial position is occupied by a water molecule, whereas the equatorial plane is formed by a chelating bpy and either a bidentate malonate or two carboxylate oxygens from two malonate groups. The equatorial Cu-O(mal) (1.911(4)-1.978(4) (1) and 1.897(5)-1.991(4) A (2)) and Cu-N(bpy) (1.983(4)-2.008(5) (1) and 1.971(6)-2.007(6) A (2)) bonds are somewhat shorter than the axial Cu-O(w) one (2.257(5)-2.524(5) (1) and 2.236(5)-2.505(6) A (2)). The angles subtended at the copper atom by the chelating bpy vary in the ranges 80.9(2)-81.8(2) degrees (1) and 80.4(2)-82.1(2) degrees (2), values which are somewhat smaller than those of the chelating malonate (80.4(2)-82.1(2) degrees (1) and 93.0(2)-93.6(2) degrees (2)). The intrachain copper-copper separations through the OCCCO fragment are 8.227(1) (1) and 8.206(2) A (2), whereas those through the OCO bridging unit are 4.579(1)-5.043(1) (1) and 4.572(2)-5.040(2) A (2). The magnetic behavior of 1 and 2 in the temperature range 2.0-290 K is very close, and it corresponds to an overall ferromagnetic coupling, the chi MT versus T curve exhibiting a maximum at 18 K. The analysis of the magnetic data through a numerical expression derived for the real topology of 1 and 2, that is, chains of isosceles triangles with two intrachain exchange pathways J1 (exchange coupling through the OCO carboxylate) and J2 (exchange coupling through the OCCCO malonate), indicates the occurrence of ferro- (J1 = +4.6 cm-1) and antiferromagnetic couplings (J2 = -4.2 cm-1). The magnetic coupling through these exchange pathways is further analyzed and substantiated by density functional theory calculations on a malonate-bridged trinuclear copper(II) model system.

Magnetic coupling in end-to-end azido-bridged copper and nickel binuclear complexes: a theoretical study.

The influence of structural parameters on the exchange coupling J between metal atoms in end-to-end azido-bridged binuclear complexes of Cu(II) and Ni(II) has been studied by means of density functional calculations. For the case of double-bridged Cu(II) compounds, four ideal pentacoordinate models have been employed in which the coordination spheres of the two metal atoms are either a trigonal bipyramid or a square pyramid, connected through equatorial or axial bridges. The distortion from those ideal geometries along a Berry pathway has also been analyzed. For the hexacoordinate Ni(II) compounds, models with two or one bridging ligands have been studied. The effect of the bridging M-N-N bond angles on the exchange coupling has been analyzed for both the Cu(II) and Ni(II) complexes.

Neurotoxic relationship between dopamine and iron in the striatal dopaminergic nerve terminals.

The neurotoxic effect of dopamine (DA) and iron(III) on DAergic terminals in striatum has been studied by intracerebral microdialysis technique. Twenty-four hours after surgery (day 1), DA and/or iron(III) with and without DA reuptake inhibitor, nomifensine, were perfused for 1 h. Forty-eight hours after surgery (day 2), MPP(+) 1 mM was perfused for 15 min and the output of DA was measured, its amount being directly proportional to the remaining striatal DAergic terminals, supported by tyrosine hydroxylase immunohistochemistry technique. Perfusion of exogenous DA, as well as iron(III) 10 and 100 microM, did not produce any neurotoxic effect. However, perfusion of iron(III) (333 and 1000 microM) produced a concentration-dependent toxic effect. Co-perfusion of iron(III) at non-toxic concentration (100 microM) with DA (15 microM) produced a toxic effect. Elevation of the endogenous extracellular levels of DA by inhibiting its uptake with nomifensine increased the neurotoxic effect of iron(III) in a dose-dependent manner. The use of tetrodotoxin after elevation of DA with nomifensine partially prevented the neurotoxic effect of its co-perfusion with iron(III) (100 microM). These results suggest that DAergic system could be synergistically damaged by DA and iron(III). Thus, alterations in the clearance of DA from extracellular space along with an increase of iron may have significant consequences for DAergic system toxicity.

Synthesis, structural characterisation, and Monte Carlo simulation of the magnetic properties of the 3D-stacked honeycomb Cs(n)

Two new polymeric manganese-azido systems with formula Cs(n)-[[Mn(N3)3]n] (1) and [[N(C2H5)4]n][[Mn2-(N3)5(H2O)]n] (2) were synthesised and structurally characterised. Compound 1 crystallises in the P2(1)/n group and consists of a three-dimensional system with end-to-end and end-on azido bridges with the caesium atoms in the holes of the net. Magnetically, compound 1 is a rare case of a three-dimensional network with alternate ferro-antiferromagnetic interactions. Compound 2 crystallises in the P1 group and consists of double chains of manganese atoms bridged by end-on and, the exceptional, (mu-1,1,1)-azido bridges. Magnetically, compound 2 shows net ferromagnetic behaviour. Exact fit of the magnetic data was performed for the two compounds by means of Monte Carlo simulations based on the Metropolis algorithm on sets of 10 x 10 x 10 (1) and 1 x 1 x 320 (2) S = 5/2 classical spin centres.
