Covalent binding of acrylonitrile to specific rat liver glutathione S-transferases in vivo.

Acrylonitrile (AN) is an industrial vinyl monomer that is acutely toxic. When administered to rats, AN covalently binds to tissue proteins in a dose-dependent but nonlinear manner [Benz, F. W., Nerland, D. E., Li, J., and Corbett, D. (1997) Fundam. Appl. Toxicol. 36, 149-156]. The nonlinearity in covalent binding stems from the fact that AN rapidly depletes liver glutathione after which the covalent binding to tissue proteins increases disproportionately. The identity of the tissue proteins to which AN covalently binds is unknown. The experiments described here were conducted to begin to answer this question. Male Sprague-Dawley rats were injected subcutaneously with 115 mg/kg (2.2 mmol/kg) [2,3-(14)C]AN. Two hours later, the livers were removed, homogenized, and fractionated into subcellular components, and the radioactively labeled proteins were separated on SDS-PAGE. One set of labeled proteins was found to be glutathione S-transferase (GST). Specific labeling of the mu over the alpha class was observed. Separation of the GST subunits by HPLC followed by scintillation counting showed that AN was selective for subunit rGSTM1. Mass spectral analysis of tryptic digests of the GST subunits indicated that the site of labeling was cysteine 86. The reason for the high reactivity of cysteine 86 in rGSTM1 was hypothesized to be due to its potential interaction with histidine 84, which is unique in this subunit.

Cytotoxic effects of antiproliferative agents on human retinal glial cells in vitro.

PURPOSE: Proliferative vitreoretinopathy (PVR) is characterized by the formation of cellular membranes on the detached retina and also in the vitreous. Glial cells can be found in epiretinal and subretinal membranes from eyes with PVR, proliferative diabetic retinopathy (PDR), idiopathic macular pucker, uveitis and other diseases affecting the retina. Proliferation and contraction of glial cells appears to play a role in the pathogenesis of PVR. This study is designed to inspect the effectiveness of harringtonine, as well as colchicine, daunomycin and fluorouracil, against cellular proliferation of cultured human retinal glial cells that might be involved in the retinal and/or vitreous proliferation. METHODS: Cultures of human retinal glial cells were prepared using the enzyme digesting method. Cells that had been in culture for 2-5 passages were used in this study. Harringtonine (0.063 microg/ml approximately 2.0 microg/ml), colchicines (0.5 microg/ml approximately 16.0 microg/ml), daunomycin (0.1 microg/ml approximately 3.2 microg/ml) and 5-fluorouracil (0.5 microg/ml approximately 16.0 microg/ml) were added to cultures of human retinal glial cells and the proliferation rates of the cells were measured by the MTT method. RESULTS: Harringtonine at the dosage of 0.063 microg/ml induced suppression of cellular growth, but the changes were not statistically significant (p > 0.05). At a dosage ranging from 0.125 microg/ml to 2.0 microg/ml, harringtonine significantly suppressed cellular growth according to the test (p < 0.01). Likewise, other antiproliferative agents inhibited cellular growth significantly at a dosage from 1.0 microg/ml to 16.0 microg/ml (colchicine), 0.2 microg/ml to 3.2 microg/ml (daunomycin) and 1.0 microg/ml to 16.0 microg/ml (5-fluorouracil), but not at 0.5 microg/ml (colchicine), 0.1 microg/ml (daunomycin) and 0.5 microg/ml (5-fluorouracil). The ID50 were 0.33 microg/ml (harringtonine), 3.11 microg/ml (colchicine), 0.79 microg/ml (daunomycin) and 5.23 microg/ml (5-fluorouracil), respectively. CONCLUSIONS: Harringtonine was extremely effective in inhibiting human retinal glial cell proliferation, like other antiproliferative drugs such as colchicine, daunomycin and 5-fluorouracil. Harringtonine, therefore, may be a candidate for further studies regarding the treatment of experimental PVR.

Evidence for cocaine and methylecgonidine stimulation of M(2) muscarinic receptors in cultured human embryonic lung cells.

1. Muscarinic cholinoceptor stimulation leads to an increase in guanylyl cyclase activity and to a decrease in adenylyl cyclase activity. This study examined the effects of cocaine and methylecgonidine (MEG) on muscarinic receptors by measurement of cyclic GMP and cyclic AMP content in cultured human embryonic lung (HEL299) cells which specifically express M(2) muscarinic receptors. 2. A concentration-dependent increase in cyclic GMP production was observed in HEL299 cells incubated with carbachol, cocaine, or MEG for 24 h. The increase in cyclic GMP content was 3.6 fold for 1 microM carbachol (P < 0.01), 3.1 fold for 1 microM cocaine (P < 0.01), and 7.8 fold for 1 microM MEG (P < 0.001), respectively. This increase in cyclic GMP content was significantly attenuated or abolished by the muscarinic receptor antagonist atropine or the M(2) blocker methoctramine. 3. In contrast, cocaine, MEG, and carbachol produced a significant inhibition of cyclic AMP production in HEL299 cells. Compared to the control, HEL299 cells treated with 1 microM cocaine decreased cyclic AMP production by 30%. MEG and carbachol at 1 microM decreased cyclic AMP production by 37 and 38%, respectively. Atropine or methoctramine at 1 or 10 microM significantly attenuated or abolished the cocaine-induced decrease in cyclic AMP production. However, the antagonists alone had neither an effect on cyclic GMP nor cyclic AMP production. Pretreatment of HEL299 cells with pertussis toxin prevented the cocaine-induced reduction of cyclic AMP production. 4. Western blot analysis showed that HEL299 cells specifically express M(2) muscarinic receptors without detectable M(1) and M(3). Incubation of HEL299 cells with cocaine, carbachol, and atropine did not alter the expression of M(2) protein levels. However, the inducible isoform of nitric oxide synthase (iNOS) was induced in the presence of cocaine or carbachol and this induction was significantly attenuated after addition of atropine or methoctramine. 5. The present data show that cocaine and MEG significantly affect cyclic GMP and cyclic AMP production in cultured HEL299 cells. Our results also show that these effects result from the drug-induced stimulation of M(2) muscarinic receptors accompanied with no alterations of receptor expression. However, the induction of iNOS by cocaine may result in the increase in cyclic GMP production.

Detoxification of endotoxin by endodontic irrigants and calcium hydroxide.

The effects of endodontic irrigants and calcium hydroxide on lipopolysaccharide (LPS; endotoxin) were analyzed using the highly selective technique of mass spectrometry/gas chromatography with selected ion monitoring. An aqueous solution of LPS was mixed with one of a variety of endodontic irrigants for 30 min. Because it is a commonly used interappointment dressing, calcium hydroxide was also applied to LPS for 1, 2, or 5 days. LPS inactivation was measured by quantitation of free fatty acid release. Water, EDTA, ethanol, 0.12% chlorhexidine, chlorhexidine + sodium hypochlorite, and sodium hypochlorite alone showed little breakdown of LPS. Long-term calcium hydroxide--as well as 30-min exposure to an alkaline mixture of chlorhexidine, ethanol, and sodium hypochlorite--did detoxify LPS molecules by hydrolysis of ester bonds in the fatty acid chains of the lipid A moiety.

Trans-(DL-isoleucinato-N,O)[tris(2-aminoethyl)amine-kappa(4)N]cobalt(III)diperchlorate..

The racemic title compound, trans-[N,N-bis(2-aminoethyl)-1,2-ethanediamine-kappa(4)N](DL-isoleucinato-N,O)cobalt(III) diperchlorate, [Co(C(6)H(18)N(4))(C(6)H(12)NO(2))](ClO(4))(2), crystallizes in the enantiomorphous space group P2(1)2(1)2(1) with Z = 12 (Z' = 3). Each of the three cations in the asymmetric unit represents a different chirality of the isoleucine ligand; two of them are R (or D) and the third is the S (or L) enantiomer. The mixture crystallizes in a so-called unbalanced crystallization, in which the cations adopt a chiral array of composition RRS or SSR, depending on the crystal selected for data collection.

Structures of three cis-beta1 and three cis-beta2 isomers of [Co(trien)(aminoacidato)]2+ complexes.

The crystal structures and absolute configurations of three cis-beta(1) and three cis-beta(2) isomers of [Co(trien)(L-aminoacidato)](2+), namely beta(1)-Lambda(SSS)(lambdalambdadelta)/(lambdadeltadelta)-[Co(trien)(L-isoleucinato)](ClO(4))(2), L-isoleucinato(triethylenetetramine)cobalt(III) diperchlorate (1), beta(1)-Lambda(SRS)(lambdadeltalambda)-[Co(trien)(L-valinato)](ClO(4))(2), L-valinato(triethylenetetramine)cobalt(III) diperchlorate (2), beta(1)-Delta(RSS)(deltalambdadelta)-[Co(trien)(L-pyroglutamato)]ClO(4), L-pyroglutamato(triethylenetetramine)cobalt(III) perchlorate (3), beta(2)-Delta(RRS)(deltadeltalambda)-[Co(trien)(L-isoleucinato)]I(2).H(2)O, L-isoleucinato(triethylenetetramine)cobalt(III) diiodide monohydrate (4), beta(2)-Delta(RRS)(deltadeltalambda)-[Co(trien)(L-isoleucinato)](ClO(4))(2).2H(2)O, L-isoleucinato(triethylenetetramine)cobalt(III) diperchlorate dihydrate (5), and beta(2)-Lambda(SSS)(deltalambdalambda)[Co(trien)(L-leucinato)](ClO(4))(2).2H(2)O, L-leucinato(triethylenetetramine)cobalt(III) diperchlorate dihydrate (6), have been determined. Both beta(1) and beta(2) isomers form inter-cation hydrogen-bonding interactions through double or triple hydrogen bonds which link the cations into helices. However, the motifs of the interactions are different in beta(1) and beta(2) isomers, resulting in different packing structures. A localized hydrophobic area is observed in the packing structure of (1), a unique feature amongst these six structures. This work describes the first synthesis of the beta(1) isomer by direct reaction between amino acids and cis-alpha-[Co(trien)Cl(2)]Cl in mildly basic solution.

Maximum of the weighted Kaplan-Meier tests with application to cancer prevention and screening trials.

A class of maximum weighted Kaplan-Meier test statistics is described where the weight functions are chosen from a family of smooth functions. The investigated test statistic is robust and sensitive to a variety of alternatives that are often observed in cancer prevention and screening trials. A simulation study is performed to compare the size and power properties between the proposed test statistics and some existing ones. We illustrate the procedure using data from a clinical trial of a breast cancer screening program.

Mice lacking the Nkx6.2 (Gtx) homeodomain transcription factor develop and reproduce normally.

The Nkx homeobox genes are expressed in a variety of developing tissues and have been implicated in controlling tissue patterning and cell differentiation. Expression of Nkx6.2 (Gtx) was previously observed in the embryonic neural tube, testis, and differentiating oligodendrocytes. To investigate the role of Nkx6.2 in the control of cell specification and differentiation, we generated mice with null mutations in Nkx6.2 using the standard gene targeting approach. Null mutant mice were viable and fertile without apparent histological and immunohistochemical changes in the central nervous systems and testis. The absence of detectable phenotypes suggests a redundant function of Nkx6.2 in mouse development.

Control of oligodendrocyte differentiation by the Nkx2.2 homeodomain transcription factor.

Oligodendrocytes are derived from glial precursors that arise from the ventral neural tube early in development. In the developing chicken CNS, oligodendrocyte progenitors selectively express Nkx2.2 homeodomain transcription factor, raising the possibility that Nkx2.2 may directly regulate oligogliogenesis. In this study, we have examined Nkx2.2 expression in rodent glial precursors and studied the effect of a loss of Nkx2.2 on oligodendrocyte and astrocyte differentiation. We show that Nkx2.2 is also expressed in mammalian oligodendrocyte progenitors and that the differentiation of MBP-positive and PLP-DM20-positive oligodendrocytes is dramatically retarded in Nkx2.2-null mutants along the entire rostrocaudal axis. In contrast, no effect is seen on astrocytic differentiation. Interestingly, absence of Nkx2.2 expression leads to a ventral expansion of the Olig1/Olig2 expression in neuroepithelial cells into the Nkx2.2 domain and a consequent increase in the production of Olig1/Olig2-positive and platelet-derived growth factor receptor alpha-positive oligodendrocyte progenitors. These results strongly suggest that Nkx2.2 regulates the differentiation and/or maturation, but not the initial specification, of oligodendrocyte progenitors. Consistent with this suggestion, overproduction of Nkx2.2 protein in fibroblast cells can induce gene expression from the proteolipid protein promoter.

Correlation between spontaneous apoptosis and the expression of angiogenic factors in advanced gastric adenocarcinoma.

The aim of this study was to investigate whether angiogenic factors influence the occurrence of spontaneous apoptosis in advanced gastric cancer. The apoptotic indices (AIs) of 97 tumors from 97 patients with advanced gastric cancer (pT3, pN0, pM0, Stage II) were analyzed by the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate biotin nick end labeling (TUNEL) method. Intratumoral microvessel densities (IMVDs) of tumors stained with anti-CD34 monoclonal antibody were quantified under x 200 magnification using computer-assisted image analysis. The expressions of angiogenic factors, such as vascular endothelial growth factor (VEGF), thymidine phosphorylase (dThdPase), transforming growth factor-alpha (TGF-alpha), and p53 were analyzed immunohistochemically and compared with IMVDs and AIs. The mean IMVD of the 97 tumors was 365/mm2 (range 147-990/mm2). The mean AI of tumors was 2.1% (range 0-11.3%). A significant inverse correlation between the AIs and the IMVDs was shown (p = -0.278, P = 0.0064). The mean IMVDs of tumors with high expressions of dThdPase, TGF-alpha, or p53 were significantly higher than those of tumors with low expressions of these factors. The mean AI of tumors with high expressions of dThdPase was significantly lower than that of tumors with low expressions of dThdPase (P = 0.023). However, no significant correlations were detected between AIs and the expression levels of VEGF, TGF-alpha, or p53. In gastric cancer, dThdPase may play an important role in tumor progression by increasing microvessels and by suppressing apoptosis of cancer cells.

Interleukin 8 is an autocrine growth factor and a surrogate marker for Kaposi's sarcoma.

Kaposi's sarcoma (KS) is the most common tumor associated with HIV-1 infection. Here, we report the expression, regulation, and biological effect of interleukin (IL)-8 in KS. AIDS-KS cell lines expressed higher levels of IL-8 than either human umbilical vein endothelial cells (HUVECs), human aortic smooth muscle (AoSM) cells or fibroblast cells (T1). The inflammatory cytokine IL-1beta up-regulated IL-8 expression in a time- and concentration-dependent manner in KS cell lines. IL-8 antisense oligonucleotides specifically reduced IL-8 mRNA and protein levels and inhibited KS cell growth in a dose-dependent manner. In addition, supernatant from a KS cell line induced the growth of HUVECs and angiogenesis in chicken chorioallantoic membrane assays, both of which were inhibited by IL-8 neutralizing antibody. Serum levels of IL-8 were also elevated in KS cases compared with matched controls. Modulation of IL-8 may thus be of therapeutic value in this disease.

[Phacoemulsification and intraocular lens implantation in patients with chronic renal failure and cataract].

OBJECTIVE: To report the experience in treatment of phacoemulsification and intraocular lens implantation and to evaluate its efficacy in patients with chronic renal failure and cataract. METHODS: Twelve patients with chronic renal failure, maintenance dialysis and cataract were treated with phacoemulsification and intraocular lens implantation in 1998-1999. The original disease of renal failure was non-diabetic. The average age was 51 years, and the average time course on dialysis was 6-12 months. All the patients were stable on maintenance dialysis, and the blood pressure was controlled in or near normal range. Before the operation, the administration of heparin was stopped, and the hemostatic drug was applied. RESULTS: All the procedures were performed on 23 eyes these patients without any complications. The naked visual acuity after operation improved significantly in comparison with that before the operation. At the postoperative one month of follow-up, the visual acuity of 52% of the surgical eyes was above 1.0. CONCLUSION: Phacoemulsification and intraocular lens implantation is effective and safe in chronic renal failure patients with cataract.

[Cerebral watershed infarcts].

OBJECTIVE: To investigate the clinical types and characteristics of cerebral watershed infarcts (CWI). METHODS: Analysis of the clinical data and imaging characteristics of 192 cases with CWI confirmed by CT scan or MRI. RESULTS: In 69 cases (35.9%), CWI appeared as wedge-shaped areas on CT scan or MRI, the infarcts located at marginal zones between the anterior and middle cerebral arteries and were usually associated clinically with hemiparesis, transcortical motor aphasia and dementia. In 74 cases (38.5%) the infarcts also appeared as wedge-shaped areas on CT scan and MRI, but located at marginal zones between the middle and posterior cerebral arteries, the patients mainly showed mild hemiparesis and apathy. In 49 cases (25.5%), the long-line or triangle-shaped infarct areas usually located at marginal zones between the superficial and deep territory of the middle cerebral arteries in the basal ganglia and posterolateral angulus frontalis. CONCLUSION: The presentation of CWI is complicated, and the diagnosis of that mainly depends on imaging studies such as CT scan and MRI.

[Significance of vascular endothelial growth factor expression in serum of patients with hepatocellular carcinoma].

OBJECTIVE: This study was to determine the pretherapeutic serum level of vascular endothelial growth factor (VEGF) in patients with hepatocellular carcinoma (HCC) and to elucidate the relation of its level with clinical characteristics and metastasis of HCC. METHODS: 115 HCC patients, 40 patients with benign liver lesions and 30 healthy control subjects were included in this study. Serum VEGF level was measured with the quantitative sandwich enzyme linked immunosorbent assay (ELISA, R&D systems). RESULTS: Serum VEGF level in HCC [mean +/- s, (465.62 +/- 336.24) pg/ml] was significantly elevated as compared to those in patients with benign liver lesions[(159.54 +/- 120.58) pg/ml] and those of the normal controls [(123.53 +/- 51.84) pg/ml] though the mean VEGF level of benign liver lesions and normal controls were not significantly different. Serum VEGF concentration showed a positive rate of 77.4%, 25%, 3.3% respectively in HCC, benign liver lesions and normal controls. Of 115 HCC patients, serum VEGF level in patients with portal vein(PV) emboli [n = 26, mean, (582.76 +/- 441.89) pg/ml], with metastasis [n = 43, (548.29 +/- 438.57) pg/ml] or with large HCC lesions (> 5 cm in diameter) [n = 69, (554.43 +/- 369.99) pg/ml] were significantly higher than those without PV-emboli [n = 89, (431.39 +/- 292.84) pg/ml], without metastasis [n = 72, (416.24 +/- 247.27) pg/ml] or with small HCC lesions [n = 42, (328.67 +/- 227.47) pg/ml]. Serum VEGF levels in stage I, II, III, IV a and IV b HCC patients were 340.6 pg/ml, (451.55 +/- 307.84) pg/ml, (397.44 +/- 257.18) pg/ml, (486.10 +/- 397.73) pg/ml, (647.93 +/- 344.56) pg/ml, respectively. CONCLUSIONS: The pretherapeutic serum VEGF level of HCC appears to reflect its potential activity of vascular invasion and metastasis.

[Nested case-control study on the risk factors of colorectal cancer].

OBJECTIVE: To investigate the risk factors of colorectal cancer. METHODS: A nested case-control study was conducted based on a cohort of 64,693 subjects in Jiashan county, Zhejiang, China. One hundred and ninety-six cases of colorectal cancer were detected during a follow-up program for 10 years. Nine hundred and eighty non-colorectal cancer subjects were randomly selected from the cohort as controls. Univariate analysis and mutivariate non-conditional logistic regression were used to analyse associations between the exposure factors and colorectal cancer. RESULTS: The age of case group was significantly older than that of the control group. Moreover, the age at diagnose of colon cancer was significantly older than that of the rectal cancer. Other than age, the final regression model showed that mucous blood stool history and colon polyp history were significantly associated with colon cancer (OR = 2.961, 95% CI:1.202-7.298 and 8.941, 95% CI:1.820-43.926, respectively) while mixed sources of drinking water was found to be associated with rectal cancer with a OR value of 1.823 (95% CI:1.024-3.247). CONCLUSION: The risk factors of colon cancer were different from that of the rectal cancer. Age was an important factor influencing the incidence of both colon cancer and rectal cancer. Mucous blood stool history and colon polyp history were associated with colon cancer, while drinking mixed water was closely associated with rectal cancer.

[Reconstruction of heel: long-term follow up of 12 cases].

OBJECTIVE: To study long-term follow up result of fibula transplantation to reconstruct partial or complete defect of the heel. METHODS: Heel defects were reconstructed by means of reverse transferring of combined vascular pedicled fibulaflap. The calcaneus was repaired by two segments of the fibula. Skin defects were covered by skin flap, and sensation was restored by anastomosing the sural nerve and medial pedal nerve. The residual cavity was filled by muscles surrounding the fibula to reconstruct the appearance of foot. RESULTS: Four patients with complete heel defect and 8 with partial defect were operated on successfully. Two to 13 years(average 5.2 years) follow-up showed that the function and appearance of foot were normal and adaptive changes happened in the tissue of foot. The fibula was getting into a real cal caneous gradually. Besides this, the skin was thickened and the sensation was restored with the two-point discrimination of 0.8 to 3.0 cm. All patients resumed physical work. Comprehensive evaluation revealed that 11 patients were excellent and 1 was good in their functional recovery. CONCLUSION: The results suggested that this operation is ideal for reconstructing heel defect. All kinds of tissues in heel can be reconstructed and more satisfactory function and appearance can be obtained through adaptive heel reshaping along with long-term walk.

[Cloning and sequencing of Chinese squash leaf curl virus].

Sequence analysis of virus isolation DNA of Squash leaf curl disease shows that Squash Leaf Curl Virus (SLCH-CHI) contains 2741nt, which encode six Open Reading Frames (ORFs). Two of them are on virus sense strand: AV1 (256aa) and AV2 (140aa). AV1 encode coat protein. The other of ORFs are on complementary sense strand: AC1 (replicase gene, 243aa), AC2 (transactivtor, 134aa), AC3 (136aa) and AC4 (172aa). The virus belongs to one number of begomoviruses from old world.
