Streptococcal erythrogenic toxin genes: detection by polymerase chain reaction and association with disease in strains isolated in Canada from 1940 to 1991.

The presence of genes encoding pyrogenic exotoxins type A (speA), B (speB), and C (speC) and streptolysin O (slo) was determined by the polymerase chain reaction (PCR) to target specific sequences in 152 strains of group A streptococci. These included reference strains, representative M and T type strains, and strains associated with scarlet fever and pharyngitis collected between 1940 to 1991 and included strains from patients with severe invasive streptococcal infections. PCR amplicons were detected by agarose gel electrophoresis, and specificity was established by restriction fragment analysis. The frequency of occurrence for each target gene among all strains tested was 33.6% for speA, 99.3% for speB, 28.9% for speC, and 100% for slo. Strains of non-group A streptococci, recognized toxigenic bacterial pathogens, and pneumolysin-producing Streptococcus pneumoniae strains were negative for all targeted gene sequences. Detection limits in the PCR were found to be 100 pg of total nucleic acids for the speB and speC genes and 1 ng for the speA and slo genes. Isolates associated with scarlet fever, pharyngitis, and severe invasive infections showed statistically significant differences in the presence of speA, with scarlet fever strains having the highest association (81.3%), severe infections the next highest association (42.9%), and pharyngitis the lowest association (18.4%). Although no significant differences were observed in speC frequencies in isolated associated with the three disease categories, a genotype of speB slo was significantly higher in isolates associated with pharyngitis (54.1%) than in strains associated with scarlet fever (18.8%) or severe invasive disease (23.8%). Streptolysin O targets were present in all the isolates tested, and only a single strain (T-11-M-11) was devoid of targeted speB sequences, thereby demonstrating that neither speB nor slo is associated with any particular clinical presentation.

Clinical evaluation of the efficacy of inoculating cattle with a vaccine containing Tritrichomonas foetus.

To test the efficacy of a polyvalent Tritrichomonas foetus vaccine, 130 nulliparous heifers were randomly assigned to either receive the test T foetus vaccine or to serve as nonvaccinated controls. The polyvalent test vaccine consisted of a Campylobacter fetus/Leptospira canicola-grippotyphosa-hardjo-icterohaemorrhagiae-pamona bacterine containing 5 x 10(7) killed T foetus/dose. The polyvalent control vaccine consisted of the aforementioned formulation without T foetus. Heifers were administered 2 doses of control or experimental vaccine at 3-week intervals. Heifers were bred to T foetus-infected bulls and their conception and pregnancy rates were determined throughout gestation. In addition, serum samples were analyzed to determine induced concentrations of antitrichomonal antibodies and vaginal secretions were sampled to determine T foetus infection rates in control and vaccinated animals. One week after each of the 15-day breeding periods, 60% (6 of 10) of tested vaccinates and 80% (8 of 10) of tested control animals were T foetus culture-positive. The mean duration of infection of vaccinates was 3.8 weeks (+/- 7.5 days), compared with 5.4 weeks (+/- 7.5 days) of infection for control heifers. All vaccinates developed increased immunofluorescence and serum neutralizing antibody titers following the first immunization, and had additional increases of at least fourfold in response to the second injection. In contrast, no consistent increase in immunofluorescence or serum neutralizing antibodies was observed in control animals. Conception rates were 89.2% for vaccinates and 85.9% for control animals 30 days after breeding and 80 to 90% of these remained pregnant 60 days after breeding.(ABSTRACT TRUNCATED AT 250 WORDS)

Removal of psoralen monoadducts and crosslinks by human cell free extracts.

Human cell free extracts are capable of carrying out damage-induced DNA synthesis in response to DNA damage by UV, psoralen, and cisplatin. We show that this damage-induced DNA synthesis is associated with removal of psoralen adducts and therefore is 'repair synthesis' and not an aberrant DNA synthesis reaction potentiated by DNA deformed by adducts. By comparing the denaturable fraction of psoralen adducted DNA which becomes labeled in the repair reaction to that of terminally labeled DNA (without repair) we have found that all DNA synthesis induced by psoralen monoadducts is the consequence of removal of these adducts. By the same approach we have obtained preliminary evidence that this in vitro system is capable of removing psoralen crosslinks as well.

Mucinous neoplasm in the cervix associated with a mucinous neoplasm in the ovary and concurrent bilateral sex cord tumors with annular tubules: immunohistochemical study.

The patient described synchronous mucinous tumors of the cervix and ovary and concurrent annular tubules, but without the classical stigmata of Peutz-Jeghers syndrome. The cervical tumor was an invasive mucinous adenocarcinoma with mixed components of minimal deviation and less-well-differentiated endometrioid morphology. The ovarian tumor had the benign appearance of a mucinous adenoma but histologically revealed areas of invasive carcinoma. Immunohistochemical studies of the mucinous neoplasms of the cervix and ovary are discussed. Neither the staining properties of mucin, the pattern of immunostaining for carcinoembryonic antigen, nor any other common markers were helpful in distinguishing the mucinous neoplasms. Positive immunostaining for low-molecular-weight cytokeratin in the filament profile of sex cord tumors with annular tubules was of particular interest since it has not to our knowledge been previously described.

Non-competitive NMDA-receptor antagonists and anoxic degeneration of the ERG B-wave in vitro.

Studies have been undertaken to see if the non-competitive NMDA antagonists, ketamine, MK-801 and dextromethorphan would preserve the b-wave of the electroretinogram (ERG) in vitro. The drugs had no effect on the ERG b-wave, nor prolonged its survival postmortem. The present results support previous evidence suggesting that NMDA-receptors are not involved directly in synaptic transmission between photoreceptors and ON-bipolar cells. Further, loss of the b-wave in post-mortem anoxia does not appear to be mediated via NMDA-receptors.

Electrophysiologic characteristics of human and rat retinas in vitro.

To promote studies on the human retina, we investigated the survival of function in postmortem specimens. Visual pigment has been regenerated in normal human retinas, 5 to 58 hours postmortem, by exposure to retinal isomers in the dark. Levels from 0.1 to 0.41 nmol/mg protein were reached. Photoresponses were obtained in 9 of 13 retinas: P III maximum amplitudes ranged from 20-398 microV and thresholds, taking the criterion amplitude as 3 microV, ranged from 8.8-1340 quanta/micros2. In three cases, the b-wave was also seen. The P III amplitude vs. log intensity curves gave values of n between 0.6 and 1.0, and sigma (the stimulus intensity for a half maximal response) between 132-3700 quanta/microns2. Recovery of sensitivity did not always correspond to that of maximum response.

Hepatocidal toxicity of Listeria species.

A novel rat hepatocidal test, based on morphological changes in monolayer culture and the percentage of lactate dehydrogenase (LDH) released into the medium after exposure to culture filtrates of Listeria spp. was used to determine listerial toxicity and pathogenicity. Primary cultures of rat hepatocytes exposed to brain heart infusion (BHI) culture filtrates from ATCC strains of Listeria monocytogenes and L. ivanovii, released 91-92% and 95% of LDH after 3 h and 18.5 h, respectively. Cultured monolayers changed from normal hepatocytes into nonviable round forms. Brain heart infusion broth and BHI culture filtrates of other Listeria spp. were nontoxic to hepatocytes. The rat hepatocidal test is a quantitative and rapid system for studying listerial toxicity and pathogenicity.

Distortion of magnetic evoked fields and surface potentials by conductivity differences at boundaries in brain tissue.

We investigated the conditions under which inhomogeneity in electrical conductivity may significantly modify the magnetic evoked field (MEF) due to primary currents (i.e., neuronal currents) in the brain. In the case of an isolated turtle cerebellum immersed in a large bath of physiological saline, our theoretical analysis showed the cerebellar surface to significantly enhance the MEF due to a primary current, by a factor of as much as two, for experimentally determined values of the conductivities of the cerebellar tissue and saline. A further parametric investigation of the conductivity effect revealed that conductivity boundaries may significantly modify the MEF due to neuronal currents located within 1 mm of a conductivity boundary, as would be the case for active neurons near an edema, an anoxic fringe such as might occur during stroke, or a ventricle in the human head. For a stationary neural source, conductivity boundaries may modify the magnitude of its MEF without affecting its temporal waveform. However, this boundary effect was found to be small for a model geometry locally approximating cortical sources in a sulcus or a fissure, where the boundary effects from adjacent sulcal walls tend to cancel each other.

Survival of structure and function in postmortem rat and human retinas: rhodopsin regeneration, cGMP and the ERG.

Procedures for regenerating visual pigment and restoring phototransduction have been established with freshly isolated, bleached rat retinas. Phosphatidyl choline liposomes containing a 500 microM mixture of retinal isomers, including the 9-cis and 11-cis forms, were employed and the results compared with dark-adapted retinas, incubated similarly but without retinal. The following were recovered in a 60 min incubation, rhodopsin (plus isorhodopsin) to 91% of the original rhodopsin concentration, 87% of cGMP and 89% of PIII amplitude at saturation. PIII amplitude vs. log intensity curves gave values of n between 0.6 and 1/2.0 and sigma between 85 and 439 quanta/micron 2. Human retinas, ranging from 18 to 58 hours postmortem and treated as above, also produced photoresponses. Of the 7 retinas studied so far, rhodopsin has been regenerated to 0.1-0.35 nmol/mg protein, cGMP to 23.5-49.2 pmol/mg protein, and PIII to 20-50 microV: in some cases a b-wave was also seen. Values of n varied between 0.6 and 1.0, and sigma between 132 and 3700 quanta/micron 2. PIII responses were also seen after retinas, approximately 30 hours postmortem, were incubated for a further 24 hours in fortified medium. After incubation, retinal vacuolation was reduced.

[Induction of retarded exoerythrocytic schizonts by chloroguanide resulting in delayed parasitaemia of Plasmodium inui in Macaca mulatta].

Three Rhesus monkeys inoculated with a large number of sporozoites of Plasmodium inui dissected from the salivary glands of infected mosquitoes, Anopheles dirus. Two of the monkeys (inoculated with 8.06 x 10(5) and 1.3 x 10(7) sporozoites, respectively) were treated with chloroguanide base 6 mg/(kg.d) x 4 d starting from 24 h after the inoculations and the other one (inoculated with 5.93 x 10(6) sporozoites) was not treated with chloroguanide as control. The primary parasitaemia attacks occurred in the former 2 monkeys were 31 and 25 d respectively after inoculation, while in the control was 7.5 d. Liver biopsies were done in all of the 3 monkeys, normal schizonts were seen in the control monkey on d 8 after inoculation, while none were detected in the 2 monkeys treated with chloroguanide. However, the retarded exoerythrocytic schizonts were found by indirect fluorescent antibody test (IFAT) in liver sections of the monkey inoculated with 1.3 x 10(7) sporozoites. Therefore, it is evident that moderate doses of chloroguanide retarded the formation of schizonts and thus delayed the primary parasitaemia.

Blood selenium levels in nasopharyngeal cancer patients in south China.

Blood selenium concentrations in 111 nasopharyngeal cancer patients and 56 normal subjects were determined using a modified 2,3-diamino naphthalene fluorometric method. Cancer subjects in this study were divided into two groups and blood Se levels (mean +/- SD) were 0.105 +/- 0.024 for untreated cases, 0.113 +/- 0.034 for those who had received treatment, and 0.109 +/- 0.030 microgram/ml for all subjects. The results suggest that low blood Se levels in nasopharyngeal cancer patients are a consequence of their disease rather than the cause of the cancer.

Effect of human chorionic gonadotropin and prostaglandin F2a on progesterone production by human luteal cells.

To determine and compare the direct effects of prostaglandin F2a (PGF2a) and human chorionic gonadotropin (hCG) on luteal cell progesterone production in vitro, 9 human corpora lutea obtained at tubal ligation were minced and treated with collagenase to disaggregate luteal cells. Dispersed luteal cells (80% viable) were incubated in air at 37 degrees C in a shaking water bath for 3 h and total progesterone in the media and cells was determined by radioimmunoassay. Optimum progesterone production was obtained using 25,000 or more cells per incubate and an incubation time of 2-4 h. hCG-stimulated progesterone production increased significantly with 0.01 IU to as high as 100 IU. In the early luteal phase (days 1-5 post ovulation or days 15-20 of the luteal phase), PGF2a (10-1000 ng) significantly inhibited progesterone production but significantly stimulated progesterone production in the mid-luteal phase (days 21-25). PGF2a had no effect on luteal cell progesterone production in the late luteal phase (days 26-30). This age-dependent direct effect of PGF2a on human luteal cell progesterone production in vitro indicates a role for PGF2a in the total intragonadal regulation of progesterone output, possibly through a paracrine or autocrine manner directed towards synchronizing luteal progesterone secretion and endometrial preparation for nidation.

Investigations of the incidence of bovine trichomoniasis in nevada and of the efficacy of immunizing cattle with vaccines containing Tritrichomonas foetus.

Trichomonas cultures taken from 2389 bulls showed that approximately 4.7% of them were infected. Correlation of these data with the ranches from which diagnostic samples were obtained indicated that in the period of 1984 through 1987 26.7 to 44.1% of ranches had at least one infected bull. Thirty-four 18-month-old Holstein heifers were assigned to one of three groups, controls n = 12 animals, soluble vaccine n = 11 animals, and whole vaccine n = 11 animals to determine the effect of Tritrichomonas foetus vaccines on the reproductive performance of T . foetus infected animals. Heifers were bred with T . foetus infected bulls beginning two weeks after the second T . foetus vaccination. All immunized animals developed antibody titers of at least 1:1000 following vaccination. In addition, all control and immunized animals became infected with T . foetus . However, the duration of infection was approximately two weeks shorter in immunized animals. Approximately 42% (5 of 12) of control heifers remained infected with T . foetus for the duration of the experiment, while only 18% (2 of 11) of each of the vaccine groups remained infected for the duration of the experiment. Finally, 27% (3 of 11) of heifers in each of the vaccine groups were pregnant at slaughter, while none of the control heifers were pregnant at slaughter. Therefore, both vaccine formulations appeared to protect heifers (P<0.05) from fetal loss due to trichomoniasis.

Antigenic relationship among field isolates of Tritrichomonas foetus from cattle.

Analysis of protein and antigen profiles of Tritrichomonas foetus isolates from cattle from 5 western states was accomplished by sodium dodecyl sulfate polyacrylamide-gel electrophoresis, immunoblot, immunoprecipitation, and fluorography techniques. Total protein profiles of all isolates were compared by Coomassie brilliant blue staining of T foetus protein samples prepared by 4 protein-extraction methods. Antigenic tritrichomonas proteins were identified by immunoblot assay with polyclonal bovine or rabbit anti-T foetus serum. Additionally, [14C]glucosamine-labeled T foetus was used for total and antigenic glycoprotein analyses. Detectable differences in the composition of total proteins or antigenic tritrichomonal proteins were not observed among all isolates. However, intensity differences in some antigenic protein bands were apparent. Bovine and rabbit sera from immunized animals possessed antibodies to the same antigenic tritrichomonal proteins. Each T foetus isolate contained 4 to 7 molecular weight size classes of glycoprotein, which were labeled by [14C]glucosamine; however, only 3 to 4 glycoproteins were identified as antigens by bovine or rabbit antiserum.

Baboon corpus luteum oxytocin: an intragonadal peptide modulator of luteal function.

Oxytocin concentrations were determined in baboon (Papio anubis) corpora lutea, and the effect of oxytocin on dispersed luteal cell progesterone production was evaluated. Oxytocin concentrations increased significantly from an early luteal phase value of 2.1 +/- 1.1 ng/gm to a peak concentration of 18.1 +/- 4.3 ng/gm wet weight in midluteal phase corpora lutea. Corpora albicantia and ovarian stroma had comparatively low oxytocin concentrations. Reverse-phase high-pressure liquid chromatography of corpora lutea extracts gave a peptide peak (retention time, 17.25 min) similar to a standard oxytocin peak. Plasma oxytocin levels, which were significantly higher in the ovarian vein draining a corpus luteum than in the contralateral side or the femoral vein, declined significantly after luteectomy. Oxytocin was localized by immunocytochemical methods in luteal cells. In the early luteal phase oxytocin (4 to 800 mU; 1 mU is equivalent to 2 ng) inhibited basal and human chorionic gonadotropin-stimulated progesterone production by dispersed luteal cells, but in the late luteal phase 200 to 800 mU oxytocin inhibited only human chorionic gonadotropin-stimulated progesterone output. Oxytocin did not affect luteal cell progesterone production in the midluteal phase. Thus oxytocin is present in corpora lutea, can be localized in the luteal cells, is probably produced locally, and may modulate luteal cell progesterone production.

Serum thyroid hormone levels of heifers grazing leucaena leucocephala in southern Taiwan.

This study investigated the effect of prolonged ingestion of Leucaena leucocephala, a leguminous shrub with a potential as a source of animal feed in Southern Taiwan, by heifers on serum thyroid hormone levels. Ten crossbred beef heifers were divided into treated (grazing Hawaiian wildtype leucaena) and control (grazing Pangola grass only) groups of 5 animals each. The heifers grazing leucaena exhibited significantly (P less than 0.001) lower serum total triiodothyronine levels and also showed a tendency (P less than 0.10) for a lower thyroxine level. Growth rates to 90 days were 0.26 +/- 0.02 and 0.17 +/- 0.02 kg/day for heifers on Pangola grass and leucaena pastures, respectively. The difference was significant (P less than 0.05). Three of five heifers grazing leucaena showed severe hair loss from the tail and other parts of the body. Our results suggest that leucaena toxicity to animals may be present in Southern Taiwan.

Observations on early and late post-sporozoite tissue stages in primate malaria. V. The effect of pyrimethamine and proguanil upon tissue hypnozoites and schizonts of Plasmodium cynomolgi bastianellii.

Two rhesus monkeys were each infected with 2.1 x 10(6) sporozoites of Plasmodium cynomolgi bastianellii; one was treated with 1.0 mg of pyrimethamine base per kg body weight for 5 d after sporozoite inoculation. A further 2 monkeys were each infected with 9.75 x 10(6) sporozoites of the same parasite; one was treated with 10 mg of proguanil per kg body weight for 4 out of 5 d after inoculation. The treated monkeys showed a delayed primary parasitaemia and relapses. In sections of liver biopsies taken 7.5 d after sporozoite inoculation, all monkeys showed numerous hypnozoites. However, there were no full grown schizonts and only rare retarded schizonts in the treated monkeys, in contrast to the untreated monkeys which had many mature or nearly mature schizonts. Later biopsies confirmed the continued presence of hypnozoites in all monkeys.

Identification of 5-hydroxytryptamine binding site subtypes in rat spinal cord.

High-affinity, specific [3H]5-hydroxytryptamine (5-HT) binding was analyzed in rat spinal cord homogenates. Drug competition studies were performed using 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT) to compete selectively for 5-HT1A sites, RU 24969 to compete for 5-HT1B sites and mesulergine to compete for 5-HT1C sites. Competition data were analyzed by computer-assisted iterative curve-fitting analysis. The results demonstrate that 5-HT1A, 5-HT1B and 5-HT1C binding sites are present in rat spinal cord. In addition, approximately 33% of total 5-HT1 sites do not appear to represent either 5-HT1A, 5-HT1B or 5-HT1C binding sites. Therefore, 5-HT1D and/or some other 5-HT1 binding site subtype may also be present in rat spinal cord.
